2.Study on a Pre-enrichment Medium for the Simultaneous Recovery of Salmonella spp., Escherichia coli and Staphylococcus aureus

This text involved with a pre-enrichment medium for the simultaneous recovery of Salmonella spp.,Escherichia coli and Staphylococcus aureus.This medium is named buffered saline broth(BSB),which contains peptone 10g,beef exact 3g,disodium hydrogen phosphate 9g,potassium dihydrogen phosphate 1.5g,additive 50g,deionized water 1 000mL,pH7.2.1cfu per one milliliter of Salmonella spp.,Escherichia coli and Staphylococcus aureus in saline were stimutaneously added to 97 mL of BSB,buffered peptone water,lactose broth,nutrient broth,Escherichia coli broth,rappaport-vassiliadis enrichment broth,7.5% sodium chloride enrichment medium,respectively,and incubated for 18h at 37℃.The results showed BSB was the best enrichment medium,in which Salmonella spp.,Escherichia coli and Staphylococcus aureus multiplied at nearly same speed,and reached at 10~6 、10~6 、10~7 cfu/mL,respectively.Multiplex PCR produced specific amplicons of expected sizes,284bp for Salmonella spp.invA gene,622bp for Escherichia coli phoA gene,484bp for Staphylococcus aureus nuc gene.In contrast,the three bacteria couldn't multiply harmoniously in the other six media.So BSB might be considered as the medium,which could enrich above mentioned three bacteria.
3.Study on Hydrolysis of chitosan by Lysozyme

The effects of reaction temperature, pH, substrate and enzyme concentration, deacetylation degree of chitosan on the hydrolytic velocity of chitosan by Lysozyme were investigated. The results showed that the specificity of catalyst obey the dynamics of michae-lis-Menten. The optimum temperature and pH were 50℃ and 6. 0 respectively, the reaction velocity was reduced while the deacetylation degree of chitosan was raised. The mean molecular weight of low molecular chitosan has been determined by high performance liquid chro-matography method, the mean molecular weight was 8328.

7.Effect of Simotang oral liquid on anal exhaust in patients after abdominal gynecological operation.

OBJECTIVETo study the effect of Simotang oral liquid and glycerin enema on the patients' bowel sound (BS) restoration and anal exhaust after abdominal gynecological operation.
METHODNinety patients with benign tumor who had undergone gynecological operation were randomly divided into the Simotang group, treated after operation with Simotang oral liquid; the enema group, treated with glycerin enema, and the control group, non-treated. The restoration time of BS and anal exhaust were observed.
RESULTSCompared with the control group, the restoration time of BS and anus exhaust were both significantly shorter in the Simotang group and the enema group, showing statistical significance (P < 0.05); but the difference between the two treated groups was insignificant (P > 0.05).
CONCLUSIONSimotang oral liquid and glycerine enema both could benefit the restoration of anal exhaust and BS after abdominal operation.

8.Therapeutic effects of artesunate on cytomegalovirus pneumonia in mice

To investigate the therapeutic effect of artesunate on mouse cytomegalovirus pneumonia, the BALB/c-nu mice were infected with murine cytomegalovirus-green fluorescent protein (MCMV-GFP) by nose dropping method. The experimental protocol was approved by the Medical Laboratory Animal Ethics Committee of Guangzhou Medical University. The BALB/c-nu mice were randomly divided into five groups: control group, MCMV pneumonia group, and artesunate (60, 120, and 240 mg·kg-1) groups. The survival rate, weights, and virus loads in lungs among the groups were observed. The degree of histopathologic changes in lungs was assessed directly by hematoxylin-eosin (HE) assay. 9.A genetic diversity comparison between captive individuals and wild individuals of Elliot's Pheasant (Syrmaticus ellioti) using mitochondrial DNA

Maintaining genetic diversity is a major issue in conservation biology. In this study, we demonstrate the differences of genetic diversity levels between wild and captive individuals of Elliot's Pheasant Syrmaticus ellioti. Wild individuals showed a higher genetic diversity level than that of the captive individuals. Nucleotide diversity and haplotype diversity of wild individuals were 0.00628 and 0.993, while those of captive individuals were 0.00150 and 0.584 respectively. Only 3 haplotypes of mtDNA control region sequence were identified among 36 captive individuals, while 16 unique haplotypes were identified among the 17wild individuals in this study. One captive haplotype was shared by a wild individual from Anhui Province. It is concluded that a low number of founders was the likely reason for the lower level genetic diversity of the captive group. Careful genetic management is suggested for captive populations, particularly of such an endangered species, to maintain genetic variability levels.
10.Protective effects of methylene blue on ischemia/reperfusion-induced mitochondrial injury in isolated rat hearts

AIM:To study the effects of methylene blue (MB) on myocardial ischemia/reperfusion (I/R)-induced mitochondrial injury in isolated rat hearts.METHODS:Spragure-Dawley (SD) rats were divided into 3 groups randomly (n=6): control group, I/R model group and MB treatment group (IR+MB group).The isolated rat hearts were prepared and set up to Langendorff perfusion.The rats in I/R+MB group received MB (2 mg/kg) by intraperitoneal injection 2 h before operation.The hearts in control group were perfused with K-H solution for 110 min consecutively.The hearts in I/R group and I/R+MB group were in equilibrium for 20 min, following by 45 min of global ischemia, and then reperfused for 60 min.The heart rate (HR), left ventricular developed pressure (LVDP), left ventricular pressure maximum change rate (±dp/dtmax) and left ventricular end-diastolic pressure (LVEDP) were recorded.The perfusate was collected to determine the activity of creatine kinase-MB (CK-MB) and lactate dehydrogenase (LDH).The contents of reactive oxygen species (ROS), malondialdehyde (MDA) and adenosine triphosphate (ATP), and the activity of superoxide dismutase (SOD) in the myocardial tissues were all determined.Histopathological examination of left ventricle was performed.The mitochondria from the heart tissues was isolated and the mitochondrial swelling and mitochondrial membrane potential (MMP) were analyzed.RESULTS:Compared with control group, the hearts in I/R group showed poorer function, higher CK-MB and LDH levels in the perfusate, increased ROS and MDA contents, higher SOD activity and less ATP content in the heart tissues (P<0.05).Furthermore, the mitochondrial swelling level increased and MMP reduced in I/R group (P<0.05).Compared to I/R group, MB improved heart function and reduced the release of CK-MB and LDH (P<0.05).MB also decreased ROS and MDA contents, and increased the activity of SOD and the content of ATP (P<0.05).In addition, MB alleviated mitochondrial swelling and restored the reduced MMP (P<0.05).CONCLUSION: MB protects the isolated rat hearts from I/R-induced injury by attenuating the damage of mitochondria.
