DNMT1,the main member of the DNA methyltransferase (DNMT) family,plays a core role in maintaining and regulating tumor cell genome-wide and the local area methylation.DNMT1 is highly expressed in gynecological malignancies,which leads to abnormal DNA methylation of genes,especially high methylation of tumor suppressor genes,gene expression silencing,and malignant cell growth.
2.Characteristics of Outdoor Air Microbes Pollution in Summer in Huangshan Scenic Spot

The concentration of airborne microbes in summer tourism season in Huangshan Scenic Spot was measured by fallen plate method at l4 collection sites. The study on the community structure and distribution characteristics of outdoor airborne microbes was carried out. The results showed that the average concentra- tion of the bacteria, fungi and total microbes in the outdoor environment were 0.41?104 CFU/m3, 0.34? 104 CFU/m3 and 0.75?104 CFU/m3 respectively, and the average percentage of fungi of the total was 45.9%. These indicated that the outdoor air had been polluted by airborne microbes. The preliminary identification was made on the bacteria and fungi at different sampling sites and the results showed that the dominant bac- teria mainly belonged to Bacillus, Staphylococcus, Micrococcus and Pseudomonas, and the average per- centage of the total amount of the bacteria was 37.7%, 17.2%, 10.1% and 9.8% respectively; the dominant fungi belonged to Cladosporium, Mycelia sterilia, Penicillium and Alternaria, and the average percentage of the total amount of the fungi were 40.4%, 35.0%, 5.6% and 4.3% respectively. The results indicated that thedistribution characteristics of air microbes are affected not only by topographical and climatic characteristics, but also by factors such as tourist activities, animals and plants.

6.lnnercapsular phacoemulsification with primary intraocular lens implantation in the treatment of lens dislocation

To observe the clinical results of innercapsular phacoemulsification with primary intraocular lens ( lOL ) implantation in the treatment of lens dislocation.METHODS: A total of 23 cases ( 23 eyes ) of lens dislocation ( lla and llb ) were underwent innercapsular phacoemulsification combined with primary lOL implantation. lOL implantation were underwent during operation, the complications of intraoperative and postoperative, postoperative vision, intraocular pressure ( lOP ) , corneal endothelial cell, lOL location were analyzed.RESULTS: The operations were successfully completed for all patients in accordance with the pre - surgery program; lens nucleus or its fragments did not crash into the vitreous cavity; 20 cases of corneal edema and 17 cases of lOP presented at the first day after surgery, the deviation or displacement of lOL and serious complications such as retinal detachment were not appeared. At the first week postoperation, the average lOP was 15. 81 ± 2. 10mmHg, with statistically significant differences when compared with the preoperative ( P<0. 01 ) , the visual acuity in all eases increased, with statistically significant differences when compared with the preoperative ( P < 0. 01 ). Corneal endothelial cell density and percentage of hexagonal cells decreased, the variation coefficient increased in first week of postoperative, with no statistically significant differences when compared with the preoperative (P>0. 05) CONCLUSlON: lnnercapsular phacoemulsification combined with primary lOL implantation in the treatment of the whole lens dislocation (‖a and ‖b ) can restore function in patients with diplopia and control lOP effectively, reduce corneal endothelial cell damage, which is an effective method to treat the whole traumatic lens dislocation.
7.Role of silent information regulator 1 in ischemic preconditioning-induced alleviation of myocardial ischemia-reperfusion injury

Objective To evaluate the role of silent information regulator 1 (SIRT1) in ischemic preconditioning (IPC)-induced alleviation of myocardial ischemia-reperfusion (I/R) injury in rats.Methods Forty-eight male Sprague-Dawley rats,aged 12 weeks,weighing 200-250 g,were randomly divided into 4 groups (n =12 each):sham operation group (S group),I/R group,IPC group and IPC + SIRT1 inhibitor ex527 group (IPC + ex527 group).Myocardial I/R was induced by 30 min occlusion of left anterior descending branch of coronary artery followed by 120 min reperfusion.IPC was induced by 3 episodes of 5 min occlusion of left anterior descending branch at 5 min intervals before myocardial ischemia.SIRT1 inhibitor ex527 1 μg/kg was injected intravenously 15 min before myocardial ischemia and at 1 min before reperfusion.Blood samples were collected before myocardial ischemia and at 120 min of reperfusion for measurement of serum TNF-α and IL-6 concentrations by ELISA.The rats were then sacrificed and myocardial specimens were obtained for determination of myocardial lactic dehydrogenase (LDH) and creatine kinase MB (CK-MB) activities,SIRT1 expression and acetylation of NF-κB p65.Results Compared with S group,the serum TNF-α and IL-6 concentrations at 120 min of reperfusion,and myocardial LDH and CK-MB activities and acetylation of NF-κB p65 were significantly increased,and SIRT1 expression was down-regulated in I/R and IPC + ex527 groups (P < 0.05).Compared with I/R group,the serum TNF-α and IL6 concentrations at 120 min of reperfusion,and myocardial LDH and CK-MB activities and acetylation of NF-κB p65 were significantly decreased,SIRT1 expression was up-upregulated in group IPC (P < 0.05),and no significant change was found in the parameters mentioned above in group IPC + ex527 (P > 0.05).Compared with IPC group,the serum TNF-α and IL-6 concentrations at 120 min of reperfusion,and myocardial LDH and CK-MB activities and acetylation of NF-κB p65 were significantly increased,and SIRT1 expression was down-regulated in group IPC + ex527 (P < 0.05).Conclusion SIRT1 is involved in IPC-induced alleviation of myocardial I/R injury in rats.

9.Evaluation of the application of regional citrate anticoagulation in sustained low efficiency hemodialysis

Objective To compare the anticoagulation effect of regional citrate and heparin in patients with sustained low-efficiency hemodialysis (SLED).Method This study was conducted in the teaching hospital of Sichuan University between November 2011 and January 2013.Sixty-three patients suffering from acute kidney injury or end-stage renal diseases (ESRD) were enrolled and further randomized to 2 groups:citrate and heparin anticoagulation treatment groups in SLED.SLED was conducted by Fresenius 4008sARrTplus dialyzer for 8 hours each session,and blood flow was set at 150 ml/min.Prothrombin time (PT),activated partial thromboplastin time (APTT) and platelet (PLT) count were analyzed.Result Sixty-three patients underwent 118 sessions of SLED.Among them,59 patients (93.7%) was discharged after treatment or converted to outpatient intermittent hemodialysis,and 4 patients died of multiple organ failure during hospitalization.Compared with that in the citrate group,both PT and APTT in heparin group was significantly higher [PT:(15.5 ± 2.0) s vs (12.3 ± 2.7) s,P < 0.001 ; APTF:(56.0 ± 10.9) s vs (32.8 ± 6.1) s,P < 0.001 ; respectively] at 2 h during SLED.However,the PT and APTT levels in heparin group decreased afterwards and were similar with those in the citrate groups at 5 h during treatment.There is no difference on PLT counts between these two groups after treatment.Conclusion The anticoagulation effect of regional citrate and heparin was similar in patients when receiving SLED.Regional citrate may be an alternative anticoagulant approach for the patients at high risk of bleeding who require the treatment of SLED.
10.Development and clinical application effect of venipuncture auxiliary devices under special condi-tions

Objective To design and develop a kind of venipuncture auxiliary device to use in the absence of light condition or patients with poor vein condition, and validate its clinical effect. Methods A total of 70 patients with shock were divided into experimental group (venipuncture auxiliary device group with 37 cases) and control group (non-device group with 42 cases) by random digits table method. The one-time puncture success rate and operating time were compared between two groups. Results The one-time puncture success rate was 94.59%(35/37) in experimental group and 78.57%(33/42) in control group, and there was significant difference, χ2=4.214, P<0.05. The time of establishing a single vein passage was (73.63±29.35) s in experimental group and (107.61±45.94) s in control group, and there was significant difference, t=3.656, P<0.05. Conclusions The device is small in size, low cost, easy to carry, the screen is clear, is a rapid infusion with explore vascular equipment. It can guide nurses accurately on vascular puncture, fast, smooth and effective transfusion puncture process.
