1.Clinical analysis of myocardial infarction in young adults with and without diabetes

Objective To compare the clinical features of myocardial infarction (MI) in young adults with and without diabetes. Methods A total of 94 young adults with MI were enrolled in this study and divided into two groups according to whether they had diabetes:diabetic group (DM) and Non diabetic group (NDM).The clinical features,angiographic results,risk factors and prognosis were compared between the two groups. Results The incidence of atypical chest pain (25.0% vs 7.6%),non-ST segment elevation myocardial infarction (50.0% vs 9.1%),ventricular wall motion abnormalities (78.6% vs 42.4%),double vessels (35.7% vs 21.2%),triple vessels (39.3% vs 3.1%) and the incidence of mortality(10.7% vs 1.5%)were all significantly higher in DM group than in NDM group (P<0.05 or P<0.01).The number of affected vessels and Gensini scores were (2.110±0.875) and (52.61±10.47) in DM group,and(1.140±0.677),(34.02±10.24) in NDM group.The between group difference was statistically significant (P<0.05).Multiple linear regression showed that HbA1c and smoking were the independent risk factors for Gensini score. Conclusion The clinical features of young adult patients with MI and diabetes are more complex,their coronary stenosis are more serious,and the prognosis is poor.Smoking and HbA1c are positively associated with Gensini scores.
2.Application of PHB in transplantation of adrenocortical cells among allorats

Objective To investigate the effect of PHB on the proliferation and secretion function of adrenocortical cells and the efficiency of PHB as a cell carrier in transplantation of adrneocortical cells. Methods Adrenocortical cells from rat adrenal gland were separated and cultured in vitro. The effect of PHB on the proliferation and secrete function of adrenocortical cells were evaluated by MTT and RIA method. Adrenocortical cells were seeded into PHB. After cells were cultured in vitro for about 7 days, they were implanted into rats subject to bilateral adrenalectomy. The changes of blood corticosterone and aldosterone and local histological changes were observed. Results Adrenocortical cells could grow and passage on PHB. PHB had no effect on the proliferation and secretion function of adrenocortical cells. 3.Transplantation of adrenocortical cells using poly-?-hybroxybutyric acid as a cell carrier

Objective To investigate the morphology change of transplants of adrenocortical cells using poly-? -hybroxybutyric acid (PHB) as a cell carrier. Methods Adrenocortical cells were seeded on PHB and then implanted into rats bearing bilateral adrenalectomy. Histological changes and CD3 expression by using immunohistochemical staining were observed in local area of transplants on the postoperative week 2, 4, 8. Results Histological study showed inflaming cells were decreased as time passed, fibrous tissue disappeared step by step, PHB degraded gradually and adrenocortical cells tended to be organized; Ultrastratural observation revealed these cells maintained identical secreting function; Immunohistochemical assay indicated the expression of CD3 in the transplanted was similar to that in the normal adrenal cells. Conclusion The transplanted cells seeded in PHB can not only be survival, but also organized to the same tissue structure and ultrastructure as normal adrenal tissue.
4.Three-dimensional reconstruction and visualization of internal microstructure of median nerve

[Objective]To explore the application of computer in dealing with two-dimensional image information of human median nerve series freezing tissue sections,in order to develop nerve three-dimensional visualization system(3D Nerve),and finally to reconstruct 3D internal microstructure of human median nerve and realize 3D visualization.[Method]One fresh cadaver median nerve was taken,located with human hair and embedded in OCT.Series freezing tissue sections were made and stained with ACHE histochemistry method,and 2D image information was obtained through high resolution scanner.Microstructure of median nerve was finally reconstructed with 3D Nerve.[Result]Different cross sections of median nerve had quite different number and positions of fasciculi.In addition,characters of fasciculus' s internal nerve fiber were also quite different.Scross sections observation showed that all fasciculi were mixed fasciculi.With the 3D Nerve,the microstructure of median nerve was able to be observed in magnifying visual field at any cross section,and the tracking of stereo course of fasciculi in median nerve became possible.[Conclusion]Reconstructed 3D visualization can reveal the whole microstructure of median nerve and the three dimensional stereo-structure of fasciculi and fasciculus groups exactly and truly.It can provide exact topographic atlas and facilitate precise clixical repair of median nerve injury.
5.In vitro experience with tissue-engineered heart valve leaflets

Objective: The experiment was designed to evaluate the in vitro creation of tissue engineered heart valve leaflets. Methods: Aortic artery was harvested from 6 hybrid young pigs. The endothelial and myofibroblast cells were isolated and cultured to get enough cells for seeding. The myofibroblast and endothelial cells were seeded on the scaffolds sequentially. The cellular scaffolds were cultured 28 days and sent for scanning electron microscopy (SEM), histological and hydroxyproline examination. Results: SEM showed that a large amount of cells attached on the scaffolds. The cell had covered a large area of scaffold with a lot of matrix synthesized. The HE stain showed large amount of cells growing on the scaffolds and some cells were found in the central part of scaffolds. Gomori stain showed that there was elastin synthesized on the scaffolds. Hydroxyproline content is (10.93?2.89) ?g/g (scaffolds). Conclusion: With vascular cell, it is possible to seed them onto biodegradable scaffolds and create a viable tissue-engineered heart valve leaflets in vitro.
6.Direct Determination of Impurity Elements in High Purity Lithium Hexafluorophosphate Dissolved with Ethyl Alcohol by Inductively Coupled Plasma Mass Spectrometry

An inductively coupled plasma mass spectrometry ( ICP-MS) was developed for the direct determination of Be, Na, Mg, Al, K, Ca, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ga, Zr, Mo, Sr, Cd, Ba, Pd, Ag, Sn, Au and Pb in lithium hexafluorophosphate. The sample was dissolved with absolute ethyl alcohol to analyze the above 26 impurity elements in the solution by ICP-MS. The condensation and deposition of high concentrations of carbon in mass cone interface and ion lens, which will decrease the sensitivity of element analysis, can be avoided effectively by introducing O2 to plasma. The mass spectral interferences of polyatomic ions to analytes were corrected by collision reaction cell (CRC). The matrix matching method was used to corrected matrix effect. Satisfactory linearity of each working curve of 26 impurity elements was obtained. The correlation coefficients being over 0. 9995, the detection limits for the investigated elements were in the range of 0. 6-32 ng / L, the relative standard deviation of each element within 2. 5% -7. 3% , and the recovery of each element at 90. 6% -108. 6% . The method had been applied to the determination of trace elements in lithium hexafluorophosphate with satisfactory results.

9.Injuring effect of DMSO-soluble particles from cigarette smoke on human umbilical vein endothelial cell line EA. hy 926 in vitro

AIM: To investigate the injuring effect of DMSO-soluble particles from cigarette smoke(DSP) on human umbilical vein endothelial cells. METHODS: Human umbilical vein endothelial cell line EA. hy 926 was used as target cells in the study. The growth and viability of the cells treated with various dosages (1, 2, 4 or 4 mL/L) of DSP and low dose (2 mL/L) of DSP at different time points were evaluated by MTT colorimetric assay and celllular protein assay in 96-well plates. Transmission electron microscopy study was carried out to observe the ultrastructure of human umbilical vein endothelial cells under DSP treatment.RESULTS: DSP inhibited the proliferation of human umbilical vein endothelial cell line EA. hy 926. Under DSP treatment, the reducing cellular protein and increasing cell death(mainly necrosis) were observed in time-dependent and dosage-dependent manners.CONCLUSIONS: These results indicated that the toxic effect of DSP caused functional disturbance and structural damage of human endothelial cells.
