1.Histopathological study of renal biopsies and its correlation with clinical features in primary Sjgren's syndrome

Objective To explore the histopathological features of renal lesions in patients with prima- ry Sjgren's syndrome(pSS)and the correlation between the findings with clinical manifestations.Methods Kidney and labial salivary gland biopsies from 16 patients with pSS were studied by a semiquantitative assess- ment approach.Results Among these pSS patients,light microscopy showed acute or chronic interstitial nephritis(IN,8/16),diffuse mesangial proliferative glomerulonephritis(MePGN,3/16),mild mesangial prolifer- ative glomerulonephritis(3/16),focal segmental glomerulosclerosis(FSGS,2/16),membranoproliferative glomeru- lonephritis(MPGN,1/16)and membranous nephropathy(MN,2/16).There were 11/16(68.8%)cases had renal tubular acidosis(RTA),8 cases of them presented with IN.In two chronic renal failure cases,one showed MN and another showed IN accompanied with FSGS.Immunofluorescent examination revealed deposition of IgG, IgA,IgM,C3,Clq on the glomerular basement membrane or tubular basement membrane,mesangium focally.It was noted that the interstitial inflammation was focal in 85% of the patients and was always combined with dif- fusely distributed cellular infiltrates.The percentage of obsolescent glomeruli was found correlated with the level of vessel wall thickening(r=0.77,P=0.001)but not with the grade of tubular atrophy and interstitial fi- brosis.Creatinine clearance was correlated with the adjusted chronic tubulointerstitial nephritis(CTIN)score (r=-0.52,P=0.04)and the percentage of obsolescent glomeruli(r=-0.53,P=0.04).No association could be found between the labial salivary gland and the renal focus scores(r=0.28,P=0.30).Conclusions The renal tubular acidosis and IN are the most common findings in pSS patients with renal involvement.Glomerular dam- age is not uncommon,althought mild and is not merely secondary.Timely renal biopsy can help to estimate prognosis and guide therapy.
2.Comparison of 3 Methods for Determination of Chemical Oxygen Demand of Water

ve To compare the advantage and disadvantages of spectrophotometry assay, microwave di-gestion and standard circumfluence method for determination of chemical oxygen demand (COD) in various water samples. Methods The levels of COD of the water samples were determined by spectro-photometry, microwave di-gestion and standard circumfluence method respectively. Results The relative standard variations were 4.3% ~ 9.59% for spectrophotometry, 3.58% ~5.61% for microwave digestion method and 4.03% ~ 4.27% for standard circumfluence method. The recovery rates of adding standard material were 90% -95% , 90% ~96% and 90% ~98% respectively. Conclusion The microwave digestion method showed easy, rapid and accurate. It presented good practical prospect in monitoring environmental samples.
3.Clinicopathologic characteristics of eight patients with small cell carcinoma of the cervix

Objective To investigate the clinicopathologic characteristics,therapy and prognostic factors of small cell carcinoma of the cervix.Methods Clinical and pathological data of eight patients were analyzed retrospectively.Results Eight patients all presented with symptoms of abnormal vaginal bleeding or postcoital spotting.One patient had stage Ⅰ b1 disease,2 had stage Ⅰ b2 disease.2 had stage Ⅱ b disease and 3 had stage Ⅲ b disease.Histopathologic findings showed the small tumor cells had scant cytoplasm,round nuclei,absence of nucleoli,and finely dispersed chromatin.Immunohistochemical findings were positive in 7 cases for neuron-specific enolase.Three patients with stage Ⅰ b disease and 1patient with stage Ⅲb disease underwent radical hysterectomy and postoperative chemotherapy.with or without radiotherapy,and the survival period was 64,22,14 and 6 months respectively.Two patients with stage Ⅱ b disease and 2 with stage Ⅲ b disease underwent chemotherapy and radiotherapy,and the survival period was 25,9,10 and 5 months respectively.Conclusions Immunohistochemical analysis using several kinds of neuroendocrine markers is helpful in establishing the correct diagnosis in addition to focusing on characteristic histopathological features.It is necessary to use comprehensive treatment including surgery,chemotherapy and radiotherapy for patients with small cell carcinoma of the cervix.Chemotherapy may play an important role in the treatment.
4.Screening and Identification of Egg Mimotope of Schistosoma japonicum

Objective To explore egg mimotope of Schistosoma japonicum that can be used in the development of diagnostic reagents for schistosomiasis.\ Methods\ By performing three rounds of biopanning in the affinity selection before picking out single clones for identification, target specific phages were effectively enriched from a random 15\|peptides phage library with an immobilized mAb 6B12 which is specific to egg antigen of S\^japonicum as a bait. By using ELISA, competitive ELISA with natural egg antigen as competitor, and Western blotting, 13 phage clones with high affinity and specificity to 6B12 were obtained and amplified from 400 single clones. \ Results \ DNA sequencing revealed that all the 13 selected single clones were identical in displaying gene sequence. Serum samples were tested by ELISA for the presence of IgG antibodies to the phages with mimic epitope, and showed that there was a significant difference between healthy volunteers and schistosomiasis patients.\ Conclusion \ It is possible that the phage display peptide antigen, egg mimotope of S.japonicum may be a replacement of natural egg antigen for the diagnosis of schistosomiasis.
5.Acupuncture clinical trials published in high impact factor journals.

Acupuncture clinical trials are designed to provide reliable evidence of clinical efficacy, and SCI papers is one of the high-quality clinical efficacy of acupuncture research. To analyze these papers published in high impact factor journals on acupuncture clinical trials, we can study clinical trials from design to implementation, the efficacy of prevention and cure, combined with international standard practices to evaluate the effectiveness and safety of acupuncture. That is the core of acupuncture clinical trials, as well as a prerequisite for outstanding academic output. A scientific and complete acupuncture clinical trial should be topically novel, designed innovative, logically clear, linguistically refining, and the most important point lies in a great discovery and solving the pragmatic problem. All of these are critical points of papers to be published in high impact factor journal, and directly affect international evaluation and promotion of acupuncture.

7.A clinical analysis of intensive gastrointestinal lavage method for treatment of 100 severe cases with stress ulcer bleeding in neurosurgery department

Objective To discuss the intensive gastric cavitary lavage method in treatment of stress ulcer bleeding(SUB).Methods The therapeutic effects of 100 patients with severe SUB in intensive care unit (ICU)from July 2005 to February 2008 treated with intensive gastric cavity lavage method were observed and analyzed.Results In these 100 cases,the bleeding was stopped successfully in 97 cases,the therapeutic effective rate being 97%;there were 17 cases died of complicated pulmonary infection(17%)and 3 cases of bleeding(3%),the total fatality rate being 20%.All the other cases were cured.Conclusion Intensive gastrointestinal lavage therapy is an effective method as acid inhibitor for treatment of SUB,but it can decrease the inhabitant flora in the stomach to pass through gastric-pulmonary route,thus the incidence of pulmonary infection was reduced.
8.Effects of Rhizoma Acori Tatarinowii on Amino Acids Neurotransmitter in Mice Brain

Objective To study the effects of Rhizoma Acori Tatarinowii (RAT) on amino acids neurotransmitter in mice brain and to explore its mechanisms. Methods After treated by different parts of RAT,the cerebral contents of the amino acids neurotransmitter in rats were detected by high performance liquid chromatography (HPLC) Results Volatile oil, fluid extract and defatted decoction of RAT could significantly lower cerebral glutamic acid level. Volatile oil and fluid extract decreased the content of aspartic acid, and volatile oil could also reduce the taurine content in brain tissue. Conclusion RAT can lower the contents of excitatory amino acids(EAA ) and has protective effect on the brain.
9.Cleavage activity of anti-transforming growth factor ?_1 RNA by U1 snRNA chimeric ribozyme in vitro

AIM: To determine the cleavage activity of anti-transforming growth factor ?1 hammerhead ribozymes which was inserted into U1 small nuclear RNA in cell-free system.METHODS: The hammerhead ribozyme targeting against transforming growth factor ?1 was designed through the analysis of computer software.The ribozyme fragments were synthesized and cloned into the U1 snRNA ribozyme vector pZeoU1EcoSpe,which contained U1 snRNA promoter/enhancer and terminator.TGF ?1 cDNA partial fragment was generated by RT-PCR,and then cloned into the T-vector at the downstream of T7 promoter.The transcripts of ribozyme and target RNA incorporated into isotope were transcribed in vitro and purified by denaturing polyacrylamide gel electrophoresis.-labeled U1 snRNA chimeric ribozyme transcripts were incubated with target-RNAs at different conditions and autoradiographed after running denaturing PAGE.RESULTS: U1snRNA chimeric ribozyme(U1Rz803) cleaved TGF?1 mRNA efficiently and specifically at 37 ℃,while the disable ribozyme(U1Rz803m) showed no cleavage activity,so these indicated the design of U1Rz803 was correct.CONCLUSION: U1Rz803 prepared in this study possesses the perfect specific catalytic cleavage activity in cell-free system.These results indicate that U1 snRNA chimeric ribozyme U1Rz803 may suppress the expression of TGF?1 in vivo,therefore it may provide a new means for exploring the role of TGF?1 in hematopoietic regulation in the future.
