Objective:To observe the effect of liver-soothing and mind-regulating acupuncture method on the resting-state electroencephalography (EEG) in rats with post-traumatic stress disorder (PTSD),and to provide evidence for the effect mechanism study and clinical application of acupuncture intervention for PTSD.Methods:Sixty Sprague-Dawley (SD) rats were randomly divided into a blank control group,a model group,a grasping group,a paroxetine group and an acupuncture group, with 12 rats in each group.Except for rats in the blank control group, rats in the other groups were subjected to preparing the PTSD models using 'incarceration plus electric shock' method. After interventions, changes in rat behavior of each group were observed;changes in resting-state EEG were collected and analyzed with multichannel EEG acquisition and analysis system, and image analysis and statistical processing were performed. Results:Compared with the blank control group, the average escape latency in the model group was significantly longer (P<0.05),and the times of crossing the platform and the effective areas were all significantly reduced (P<0.01).Compared with the grasping group, the average escape latencies in the paroxetine group and acupuncture group were significantly shortened (P<0.05),and the times of crossing the platform and the effective areas were all significantly increased (P<0.05).There were no significant differences in the average escape latency, the times of crossing the platform and the effective areas between the acupuncture group and paroxetine group (all P>0.05).Compared with the blank control group, the α-wave power spectrum value in the model group was significantly decreased, and the power spectrum values of β-wave,δ-wave and a-wave were significantly increased (all P<0.01);compared with the grasping group,α-wave power spectrum values in the paroxetine group and acupuncture group were significantly increased (both P<0.01),and the power spectrum values of β-wave,δ-wave and a-wave were decreased significantly (all P<0.01).The power spectrum values of α-wave,β-wave,δ-wave and (e)-wave of rats in the acupuncture group were not significantly different from those in the paroxetine group (all P>0.05).Conclusion:Liver-soothing and mind-regulating acupuncture method can significantly improve the abnormal EEG activity in PTSD rats, which may be one mechanism of liver-soothing and mind-regulating acupuncture method in effectively affecting the brain function in PTSD rats.

A chiral high-performance liquid chromatography method was developed for the simultaneous determination of ibuprofen enantiomers in dog plasma. It was used to study the pharmacokinetics in the Beagle dog after intravenous administration of racemic-ibuprofen, S-ibuprofen and R-ibuprofen. Ketoprofen was chosen as the internal standard. After a simple precipitation using methanol as the precipitating solvent, both analytes and IS were separated on a Kromasil 100-5CHI-TBB chiral column (250 mm x4.6 mm, 5 μm) with isocratic elution using acetonitrile - 20 mmol x L(-1) phosphate buffer (pH 3.0, containing 5% methanol) (6 : 4) as the mobile phase. The detection wavelength was 220 nm. Liner calibration curves for both of the ibuprofen enantiomers were over the concentration range from 0.5 to 50 μg x mL(-1) with a lower limit of quantification of 0.5 μg x mL(-1), the accuracies were all in standard ranges. The intra- and inter- assay precisions were all below 7%. The recovery rate was 93.1% to 100.4%. The experiments proved that the method was simple, rapid and sensitive. It can be used in the quantitative determination of ibuprofen enantiomers in dog plasma. The method was used to determine the concentration of ibuprofen enantiomers in Beagle dog plasma after a single intravenous administration of racemic-ibuprofen, S-ibuprofen and R-ibuprofen (9 mg x kg(-1)) and the pharmacokinetics parameters were calculated based on the concentration-time curves. The C(max) of S-ibuprofen in Beagle dog plasma after a single intravenous administration of racemic-ibuprofen, S-ibuprofen and R-ibuprofen were 30.8 ± 4.7, 46.1 ± 5.9 and 20.0 ± 2.6 μg x mL(-1), respectively. In terms of the exposure of active ingredient, it revealed a significant difference between the administration of S-ibuprofen and the other two groups. The systematical R- to S- chiral inversion was discussed. Comparing the pharmacokinetic parameters at different doses, chiral inversion were 70.1% ± 36.6% and 76.4% ± 36.2%, respectively, after intravenous administration of racemic- and R-ibuprofen. This study provides a theoretical basis for the safety of ibuprofen formula of injection drug.

4.Studies of heat shock protein 60 and heat shock protein 70 in oral lichen planus.
Chinese Journal of Stomatology 2003;38(4):275-278
OBJECTIVETo study the role of heat shock protein (HSP) 60 and HSP70 in the pathogenesis of oral lichen planus (OLP).
METHODSThe immunohistochemical SP methods were used to detected the expression of HSP60 and HSP70 in 62 cases of oral lichen planus, 10 cases of normal oral mucosa, 21 cases of chronic discoid lupus erythematosus (DLE), 10 cases of benign lymphoadenosis of mucosa, 46 cases of leukoplakia. RT-PCR was used to evaluate the expression of HSP60 mRNA and HSP70 mRNA in 12 cases of OLP and 5 cases of normal oral mucosa.
RESULTSHSP60 was expressed at much higher levels in OLP than other groups. The expression of HSP70 decreased in epithelial cells in erosive lichen planus group. HSP60 mRNA and HSP70 mRNA were expressed at high level in OLP.
CONCLUSIONHSP60 and HSP70 play an important role in pathogenesis of OLP.

Chinese Journal of Dermatology 2003;0(07):-
Objectives To report a generalized atrophic benign epidermolysis bullosa family, identify the deficient protein and related pathogenic gene mutation. Methods The diagnosis was confirmed based on clinical manifestations and necessary examinations. Electron microscopy and immunofluorescent staining were used to detect the deficient protein. The pathogenic gene mutation was identified by PCR amplification of ge-nomic DNA with primers targeting the flanking introns, followed by direct automated sequencing. Results In the family, the affected individuals were homozygous for a novel 4-bp deletion in COL17A1, 3897delATCT, which resulted in a downstream premature termination codon. Conclusions 3897delATCT of COL17A1 is the pathogenic gene mutation in the patients and probably results in nonsense-mediated mRNA decay and abrogation of type XⅦ collagen synthesis, as documented in the literature.

Objective To evaluate the application of Mammotome minimally invasive biopsy(MMIBS) in the diagnosis of non-palpable small lesions in breast. Methods A total of 42 patients with non-palpable small breast lesions and abnormal imaging findings were performed MMIBS.After MMIBS,21 patients received open operations, and the pathological diagnosis was compared and analysed. Results All these 42 patients were successfully performed MMIBS,with no severe complications, and the pathological diagnosis of MMIBS were breast cancer(8 patients) and benign pathological changes(34 patients).Among the 21 patients with open operations, the post-operative pathological findings of 20 patients were same as the results of MMIBS,with the sensitivity of 88.9% and specificity of 100%.The other 21 patients without operations were followed up for 6 to 12 months, and no abnormal mammographic and ultrasonographic findings were observed. Conclusion MMIBS boasts the characteristics of minimal invasion, safety, high sensitivity and high specificity, which can serve as the first choice in the diagnosis of non-palpable small lesions with abnormal imaging findings.

Microbiology 1992;0(04):-
200 citrinin mutants were screened with the inhibition zone method from the transformants library of Monascus ruber M-7 by Agrobacterium-mediate DNA transfer, which contains more than 5,000 transformants. Then 53 mutants, whose citrinin contents ranged from 0.04?g/g to 154.57?g/g in the red fermented rice (RFR), were achieved by high performance liquid chromatography (HPLC).Color values of RFR prepared by these mutants were also detected. The results showed that there was a positive correlation between the citrinin content and the color value among the mutants. These results provide materials and research bases for ferrther studying the relationship between the production of citrinin and pigment of Monascus ruber at molecular level.
