2. Total intravenous anesthesia with propofol and remifentanil in cervical vertebra surgery: An assessment of efficacy

Objective: To assess the efficacy of total intravenous anesthesia (TIVA) with propofol and remifentanil in cervical vertebra surgery. Methods: O ne hundred patients receiving elective cervical vertebra surgery were randomly allocated to the following 4 groups: group DR, desflurane 1-2 MAC and remifentanil 0.15-0.40 μg · kg-1 · min-1; group DF, desflurane 1-2 MAC and repetitive boluses of fentanyl 0.05 mg; group PR, propofol 50-100 μg · kg-1 · min-1 and remifentanil 0.15-0.40 μg · kg-1 · min-1; and group PF, propofol 50-100 μg · kg-1 · min-1 and repetitive boluses of fentanyl 0.05 mg. During the anesthesia, the adverse cardiovascular response, the use of vascular active drug, time of extubation and time of directive movement resuming were recorded. The mental state and degree of pain were assessed by a blinded observer using simplified State-Trait-Anxiety Inventory (STAI) and Visual Analogue Scale (VAS) 90 min and 24 h after anesthesia. Moreover, the incidence of nausea and vomiting was also recorded. Results: The adverse hemodynamic response, time of extubation and directive movement resuming in group DR and PR were earlier than those in group DF and PF (P<0.05). The STAI scores of TIVA were significantly lower than those of group DR and DF(P<0.05) 90 min after anesthesia, but the difference disappeard after 24 h. The incidence of nausea and vomiting in group DR and DF were significantly higher than those in other groups(P<0.05) 90 min and 24 h after anesthesia. There was no difference in VAS scores among all groups. Conclusion: The application of TIVA with propofol and remifentanil in cervical vertebra surgery has stable hemodynamics, quicker and complete awakening, good mental state, and reduced incidence of nausea and vomiting. It is worth recommending in clinical practice.
3.Microsatellite analysis of BAT-26 and BAT-25 in nodal non-Hodgkin′s lymphomas

Purpose:To investigate the existence of micro sa tellite instability in nodal non-Hodgkin's lymphomas by microsatellite analysis of BAT-26 and BAT-25.Methods:Frozen tissues of 51 nodal non-Hodgkin's lymphomas and 9 lymphoid reactive hyperplasia were collected by surgical biopsy. Genomic DNA was extracted. Microsatellite alterations of BAT-26 and BAT-25 were detected b y polymerase chain reaction(PCR)followed by fragment analysis using automatic DNA sequencer and GeneScan 3.1 software. 10 cases of hereditary nonpolyposis col orectal cancers with known high frequency microsatellite instability (MSI-H) we re retrieved as positive controls. Results:No aberration of mon onucleotide duplication in the microsatellite markers BAT-26 and BAT-25 was ob served in all successfully amplified specimens.Conclusions:The microsatellite analysis of BAT-26 and BAT-25, two highly sensitive markers to microsatellite status in colorectal tumors, demonstrated no evidence of MSI-H i n nodal non-Hodgkin's lymphomas. The existence of low frequency microsatellite instability and participation of mismatch repair genes in lymphomagenesis remain to be determined by further studies.
4.Study on Pathogenesis of Complex Syndrome of Functional Gastrointestinal Disorders in 72 cases

Objective:To probe distribution law and pathogenesis of TCM complex symptoms of functional dyspepsia and irritable bowel syndrome.Methods:Generally recognized criteria was to establish database of comprehensive analysis of the data gained by the four methods of diagnosis,and analyze with SPSS8.0.Results:Syndrome distribution was most closely related with spleen deficiency,accounting for 66.7%;and the next,with stagnation of liver-Qi,accounting for 40.3%;the last,with the kidney,only accounting for 5.6%.Conclusion:Cause of disease and inducing factors of this disease are closely correlated with pathogenic cold, diet of lipids,mental depression,prolonged disease,wound due to abdominal operation.The disease is located at the stomach, intestine with most coexistence of deficiency and excess,and spleen deficiency is the key of pathogenesis.
5.Study on PHBHHx co-cultured with mouse induced pluripotent stem cells for use as myocardial patches in vitro

Objective To study the effect of polymer material on adhesion , proliferation and differentiation of mouse in-duced pluripotent stem cells via co-culturing in vitro, to find a suitable polymeric biomaterials for miPSCs attachment , prolifera-tion and differentiation , forming myocardial patches as a new repair method for myocardial infarction .Methods miPSCs were recovered, passaged and cultured with PHBHHx two-dimension films and PHBHHx three-dimension films together.The mor-phology of miPSCs attached on the films was visualized under scanning electron microscope ( SEM) .The mouse induced pluri-potent stem cells were induced by differentiation medium that containing vitamin C .Control group did not add any inducer.The survival and differentiation of miPSCs were observed through immunofluorescence and flow cytometry .Results MiPSCs can grow, proliferate and differentiate on PHBHHx films both two-dimension and three-dimension.Vitamin C, as a favourable in-ducer, can markedly improve the efficiency of miPSCs differentiation of cardiomyocytes on PHBHHx films .Immunofluorescence results demonstrated positive cTnT expression.Flow cytometry measured cTnT expression: Vitamin C inducer group(47.54 ± 1.46)%>without any inducer group(7.02 ±0.95)%(P<0.05).In the process of miPSCs differentiating into myocardial cell, after adding Vitamin C, the expression quantity of cTnT:PHBHHx three-dimensional films(60.32 ±1.76)%>PHBHHx two-dimensional films(53.31 ±1.41)%>traditional cell culture(47.54 ±1.46)%(P<0.05).Conclusion iPSCs can ad-here, survive and differentiate on the PHBHHx film.Vitamin C, as a favourable inducer, can markedly improve the efficiency of miPSCs differentiation of cardiomyocytes .Relative to PHBHHx two-dimensional film culture and traditional culture , the three-dimensional PHBHHx film culture has a great advantage in the process of miPSCs differentiating into myocardial cells .
6.Diagnostic Value of Fine Needle Aspiration Biopsy for Children with Thyroid Nodules

Objective To investigate the diagnostic value of fine needle aspiration biopsy for children with thyroid nodules.Methods Eleven children with thyroid nodule were examined by fine needle aspiration biopsy.The cytologic features of thyoid nodules were observed by the light microscope.The pathological sections of thyroid nodules were acquired and compared with fine needle aspiration biopsy.Results Eleven cases were diagnosed by fine needle aspiration biopsy including nodule goiter 4 cases,papillary thyroid carcinoma 2 cases,follicular adenoma 2 cases,medullary carcinima 1 cases,Hashimoto′s thyroiditis 1 case and cyst 1 case.Compared with pathological section,10 cases were diagnosed correctly and one was misdiagnosis.Conclusion Fine needle aspiration biopsy is a valuable methods to the diagnosis of children with thyroid nodules.
7.Expression of antibacterial peptide gloverin in vitro

AIM: To construct a recombinant vector containing gloverin and express it in vitro by Rapid Translation System(RTS500). METHODS: The gloverin cDNA was amplified by PCR and inserted into the prokaryotic expression vector pIVEX2.3. The recombinant product was identified by PCR and enzyme digestion. The positive reconstructed expression plasmid pIVEX2.3-G was expressed by RTS500 in vitro. The expressed protein was identified by SDS-PAGE and western blotting. RESULTS: Positive recombinant plasmid pIVEX2.3-G was successfully constructed. Target protein of 13.8 Kda with 6-his tag was detected by SDS-PAGE and western blotting. CONCLUSION: Gloverin polypeptide is successfully expressed in inactive E.coli in vitro.

The results revealed the status quo of career planning and the demand for guidance in this aspect. Recommendations for career planning education of pharmacy postgraduate students were made thereby.

Objective To study the mechanisms of Curcumin-induced apoptosis on human gastric cancer cell line SGC-7901.Methods SC,C-7901 cells were treated with various concentrations of Curcumin and the growth inhibition rates of it were accessed by MTT method.Apoptosis of gastric cancer cells were in- spected by flow cytometry.The expression of Fas and survivin in gastric cancer cells were evaluated by west- ern blot.Results Curcumin could effectively inhibit the growth of gastric cancer cells in dose-dependent and time-dependent manners,the sub-peak appeared and the apoptotic rate was increased.The expressions of Fas was higher in Western blot,meanwhile,the expressions of survivin was decreased.Conclusion Curcumin could significantly inhibit the growth and induce apoptosis of gastric cancer cells(SGC-7901),Curcumin could probably through up-regulating Fas and down-regulating surviving to induce apoptosis.

Objective To study whether methicillin-resistant Staphylococcus aureus (MRSA)will increase the burden of patients with hospital-acquired pneumonia (HAP).Methods Patients with Staphylococcus aureus HAP in a hospital between January 1 ,2013 and November 31 ,2014 were selected,patients with MRSA HAP were as case group,patients with methicillin-sensitive Staphylococcus aureus (MSSA)HAP were as control group,propen-sity score matching (PSM)analysis were conducted to compare the prognosis of MRSA HAP and MSSA HAP (length of hospital stay, duration from infection to discharge, mortality, total therapeutic cost ). Results APACHE II score in case group was higher than control group before PSM was conducted,length of hos-pital stay and duration from infection to discharge were both longer than control group (40[20,94]d vs 28[21 ,53] d;19[10,46]d vs 17[8,29]d,respectively,both P <0.05).APACHE II score in case group and control group af-ter conducting PSM was not significantly different(P >0.05 ),data were balanced and comparable;there were no significant difference in length of hospital stay between two groups (28[21 ,52]d vs 28[21 ,53]d),duration from in-fection to discharge (15[9,25]d vs17[8,29]d),mortality(10.87% vs 15.22%),and total therapeutic cost (121 013.5[80 747.21 ,176 200]yuan vs 119 911 .2[66 994.08,241 184.7]yuan)(all P >0.05).Conclusion APACHE II score is an important factor affecting prognosis;after balancing this factor,there is no difference in the prognosis of patients with MRSA and MSSA HAP,MRSA HAP can not increase the burden of disease.
