2.Microsatellite analysis of BAT-26 and BAT-25 in nodal non-Hodgkin′s lymphomas

Purpose:To investigate the existence of micro sa tellite instability in nodal non-Hodgkin's lymphomas by microsatellite analysis of BAT-26 and BAT-25.Methods:Frozen tissues of 51 nodal non-Hodgkin's lymphomas and 9 lymphoid reactive hyperplasia were collected by surgical biopsy. Genomic DNA was extracted. Microsatellite alterations of BAT-26 and BAT-25 were detected b y polymerase chain reaction(PCR)followed by fragment analysis using automatic DNA sequencer and GeneScan 3.1 software. 10 cases of hereditary nonpolyposis col orectal cancers with known high frequency microsatellite instability (MSI-H) we re retrieved as positive controls. Results:No aberration of mon onucleotide duplication in the microsatellite markers BAT-26 and BAT-25 was ob served in all successfully amplified specimens.Conclusions:The microsatellite analysis of BAT-26 and BAT-25, two highly sensitive markers to microsatellite status in colorectal tumors, demonstrated no evidence of MSI-H i n nodal non-Hodgkin's lymphomas. The existence of low frequency microsatellite instability and participation of mismatch repair genes in lymphomagenesis remain to be determined by further studies.
3.Total saponins of Aralia elata(Miq)Seem protect tumor necrosis factor-α-induced human umbilical vein endothelial cells via PI3K/Akt and NF-κB signal pathways

It is now thought that atherosclerosis,although due to enhanced lipid deposition,is mainly the result of a series inflammatory process.Total saponins of Aralia elata (Miq)Seem(TASAES)from the Chinese traditional herb Longya Araliachinensis L.,a folk medicine used for treating various diseases, increasing energy and improving the body′s ability to prevent hypoxia in Asian countries has attracted widespread attention. However, the ability of TASAES on inflammation-triggered vascular endothelial cell injury, a key early event in the pathogenesis of atherosclerosis, and its potential mechanisms of this protection have never been demonstrated. The present study determined the anti-inflammatory and anti-apoptoticactivities and protective mechanisms of the total aralosides of Araliaelata(Miq)Seem (TASAES) ameliorate tumor necrosis factor-α (TNF-α)-induced human umbilical vein endothelial cells (HUVECs) injury. Our results indicate that TASAES pretreatment provided cytoprotective effects by suppressing TNF-α-induced HUVECs apoptosis, mitochondrial membrane depolarization, caspase-3 activation, and modulation of inflammatory factors (IL-6, MCP-1 and VCAM-1), meanwhile inhibiting NF- κB transcription. Furthermore, the effect was correlated with the activation of the PI3K/Akt signal pathway. Blocking Akt activation with the PI3K inhibitor LY294002 effectively reversed the protective effect of TASAES against TNF-α-induced cell apoptosis.Moreover,the PI3K inhibitor partially blocked the effects of TASAES on the increasing of Bcl-2 and Bcl-xl protein expression,and inactivation of Bax protein expression. In conclusion, the results showed that TASAES decreased the inflammation and apoptosis of HUVECs caused by TNF-α treatment,and PI3K played a crucial role in enhancing cell sur-vival during this process.
5.Mechanism of T follicular cells in experimental autoimmune encephalomyelitis

Objective: To evaluate the mechanism of T follicular helper cells ( Tfh ) in experimental autoimmune encephalomyelitis (EAE) via in vivo experiments. Methods:C57BL/6 mice were randomly divided into four groups,CFA group,EAE group,anti-ICOSL group and control group. Lymphocytes of different time points isolated from draining lymph nodes and spleen were stained for T follicular helper cells surface marker and T cells activation surface marker and analyzed by FACS. Observed parameters include inflammatory infiltration,demyelination in spinal cord and germinal center in spleen. ELISA was used to measure the level of antigen specific antibodies. Results: Mice in anti-ICOSL treated group developed mild disease was with lower clinical scores when compared with the EAE group. HE staining results turned out with alleviated inflammation and Luxol Fast Blue staining( LFB) showed no demyelization in anti-ICOSL treated mice compared with non-treated EAE models. Flow cytometry results revealed that percentages of T follicular helper cells decreased though the whole activated degree T cells was not influenced in anti-ICOSL treated group. Fewer ger minal center was found in both anti-ICOSL group and CFA group with reduced secretion of MOG-specific Ab. Conclusion:T follicular helper cells supported the development of cognate B cells,promoted the formation of germinal center,facilitate pathogenic MOG-specific Ab secretion,thus enhance EAE.
6.Analysis of postoperative complications after ventriculo-peritoneal shunt in children

Objective To analyze the curative effects and complications of ventriculo-peritoneal shunt in treating hydrocephalus caused by different diseases in children. Methods The data of 289 children with hydrocephalus who were performed ventriculo-peritoneal shunt were examined and the incidence rate of complications was analyzed. Results Thirty-seven incidences of complications occurred in 24 patients with the total incidence rate of 12.46%.There was obstruction of proximate tube in five cases,distal end in two cases,with the obstruction rate of 2.42%.Postoperative infection happened in nine cases(3.11%).Bacterial culture was positive in three cases,two of which were Staphylococcus epidermidis and Bacillus coli in the other one.Negative results were found in six patients for the bacterial culture.Subdural hematoma occured in one case,and tube break after injury in two cases.Slit ventricle syndrome broke out seven times in three cases,leakage of cerebrospinal fluid in two,exposure of pumps in three,and subcutaneous hydrops in two.Four cases(1.38%)were dead from infection. Conclusion Ventriculo-peritoneal shunt is preferred for hydrocephalus in children if the complications are well prevented and controlled.
7.Comparison of iNOS mRNA expression in colorectal carcinoma cell strains with different proliferation protentials

AIM: To evaluate the expression of iNOS mRNA in different invasion ability colon carcinoma cell strains. METHODS: MTT was used to detect the growth and reproduction of colon cancer cell strain CW-2 and LS174T. RT-PCR and Northern blot were used to detect expression of iNOS mRNA in colon cancer. RESULTS: MTT growth curve displayed that colon cancer cell strain LS174T grew and reproduced faster than cell strain CW-2. RT-PCR showed that iNOS mRNA expressed strongly in CW-2 cell strain, while iNOS mRNA expressed weakly in LS174T cell strain. Northern blot detected that iNOS mRNA expressed obviously in CW-2 cell strain, but cell strain LS174T have no obvious iNOS mRNA expression. All-trans retinoic acid (ATRA) had no obvious effect on iNOS mRNA expression in CW-2 cell strain of colon cancer. CONCLUSIONS: ATRA has no obvious effect on iNOS mRNA expression. iNOS has a dual effect on tumor growth. In low-metastatic colon carcinoma CW-2, iNOS may exert a anti-tumor influence by cytotoxicity or inducing cell apoptosis. In high-metastatic colon cancer LS174T, iNOS produced low concentration of NO, which may be an important signal-transduction molecule for increasing blood supply and angiogenesis, which improve the growth, invasion and metastasis of tumor.

10.Study on the correlation between Chinese medical syndrome types and serum levels of PLTP and CETP in coronary heart disease patients.

OBJECTIVETo study the correlation between the serum levels of phospholipid transfer protein (PLTP), cholesteryl ester transfer protein (CETP), and Chinese medical syndrome types of coronary heart disease (CHD) patients, thus probing a new pathway for the objectivity of CHD syndrome typing and developing therapeutic drugs.
METHODS201 patients with CHD confirmed by coronary angiography were selected. The comprehensive analysis database by the four examination methods was established using generally accepted standard for Chinese medical syndrome typing. Twenty healthy subjects were randomly recruited as the control group. Serum samples were separated from venous blood. The serum activities of PLTP and CETP were assayed by ELISA. The triglyceride (TG) content was determined using acetic acetone coloring method. Contents of total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were determined using precipitation floating enzyme couplet method.
RESULTSSerum levels of PLTP, CETP, and TC of CHD patients of all Chinese medical syndrome types were significantly higher than those of the control group, showing statistical difference (P<0.05, P<0.01). Statistical difference existed in serum levels of TG, HDL-C, and LDL-C of all Chinese medical syndrome types when compared with the control group (P<0.05, P<0.01). Statistical difference existed in serum levels of PLTP between the sthenia in superficiality groups of all Chinese medical syndrome types and the asthenia in origin groups of all Chinese medical syndrome types (P< 0.05). Statistical difference existed in serum levels of TG, HDL-C, and LDL-C between the sthenia in superficiality groups of all Chinese medical syndrome types and the asthenia in origin groups of all Chinese medical syndrome types (P<0.05, P<0.01).
CONCLUSIONSThe serum PLTP levels of CHD patients of sthenia in superficiality significantly increased. Changes in serum lipids was more significant in CHD patients of the sthenia in superficiality syndrome than in those of the asthenia in origin syndrome.
