1.Hair Lead Level of Children Living in Lead Polluted Area

Objective To understand the environmental lead pollution status in Tongzhou and the biological burden of lead in children living in the lead polluted area.Methods The 353 pupils aged 7-14 years were randomly selected in Zhangzhishan town central elementary school that was near to steel wire factories,283 pupils were selected as the control by using the same methods in an elementary school around where there is no any steel wire factory.Every students were investigated by questionnaires,meanwhile their hair were collected to determine lead with ASS method.Results In the polluted area group,hair lead of 77 pupils exceeded the normal level,the median was 4.437 ?g/g,the rate of exceeding the normal level(≤10 ?g/g)was 21.81%,and 65 pupils with lead poisoning related symptoms(18.41%).In the control group,the median was 3.965 ?g/g,the rate of exceeding the normal level was 11.31%,38 pupil with lead poisoning related symptoms(13.43%).After controlled the passive smoking confounder,the difference of hair lead level between these two groups was still significant(P
2.Establishment of allele-specific real-time polymerase chain reaction for detecting neonatal hyperbilirubinemia related gene SLCO1B1 A388G polymorphism

Objective To establish the allele-specific real-time polymerase chain reaction (ASPCR) for detection of neonatal hyperbilirubinemia related gene SLCO1B1 A388G polymorphism and apply this assay to identify the clinical samples.Methods According to SLCO1B1 A388G polymorphism loci,specific primers were designed and the assay was established.Wide type plasmid and mutant plasmid were constructed.Fifty clinical samples were selected,including 30 samples of neonatal hyperbilirubinemia that had been diagnosed with SLCO1B1 A388G mutant and 20 samples of healthy newborns without SLCO1B1 A388G mutant were selected as the controls.Wide type plasmid,mutant plasmid and clinical samples were tested by specific and non-specific primers.A388G polymorphism was determined by difference in Ct (cycle threshold) between specific and non-specific primers.Then,the accuracy,sensitivity and specificity of assay were evaluated.Results The difference in Ct (cycle threshold) between specific and non-specific primers that amplified equivalent wide type template was 13.97 ±0.75.The assay could correctly distinguish the wide type and mutant plasmid.Probit regression analysis showed the sensitivity of the assay could reach to 5.28 copies/μL.For clinical samples,the Ct values of the samples with A388G mutation was less than 37.75 and showed positive results,while the samples without A388G mutation did not show any amplification nor Ct values were larger than 37.75,which showed negative results.Conclusions ASPCR is a fast,simple and effective method for SLCO1B1 A388G polymorphism detection of the clinical simples.It can be used for large sample screening for neonatal hyperbilirubinemia gene loci.
3.Study on the Serum Levels of Placenta Growth Factor During Normal Pregnancy and in Patients with Pregnancy Induced Hypertension

Objective:To detect serum levels of placenta growth factor(PIGF)throughout normal pregnancy and in patients with pregnancy-induced hypertension(PIH)as well as to explore the relationship between P1GF and pathogenesis of PIH. Methods: Serum specimens were collected from 51 healthy pregnant women as a control group and 33 women suffered from PIH as third trimester of normal pregnancy(P<0.001).There was a trend that serum levels of P1GF in PIH group decreased with the severity of PIH(P<0.001).Conclusion:PlGF play important roles in placental angiogenesis throughout pregnancy, and decreased serum levels of PlGF is associated with PIH.
4.Analysis on vision changes and related factors of transnasal transsphenoidal pituitary adenoma resection

Objective To investigate the factors affecting the vision recovery after transnasal transsphenoidal pituitary adenoma resection.Methods The vision of the left eyes and the right eyes were compared respectively.The vision after and before the operation was also compared by using paired t-test,related factors were analyzed by multiple variable logistic regression model.ResultsThe paired t-test indicated that both eyes' visions were improved after transnasal transsphenoidal pituitary adenoma resection( P <0.01 ).The tumor diameter and the vision before operation were two factors affecting the post-operative visions by cumulative logistic regression model.The larger the diameters of the tumors were,the better the vision recoveries after operation( Wald x2 =0.047,OR =2.240,P =0.047).Worse vision lose before operation came up with a better recovery after operation ( Wald x2 =9.684,OR =0.010,P =0.001 ) ; however,age,gender,pathological category,the extent of tumor removal and the time of the vision loss have no significant affects on the vision recovery after operation.ConclusionThe transnasal transsphenoidal pituitary adenoma resection can improve the vision of the patients,and the degree which would to be improved is correlated with the tumor diameter and the vision lose before operation.

6.Effects of surgery-induced menopause on neuropsychological symptoms in women with perimenopause

Objective To investigate the effects of surgery-induced menopause on neuropsychological symptoms in women with perimenopause. Methods Sixty-eight women with perimenopause who underwent bilateral oophorectomy for benign indications were evaluated by the modified Kupperman index on the perimenopause syndrome pre-and post-surgery,including the score in neuropsychological symptoms.Patients were further requested to remake a decision on the acceptance of surgery,and were divided into "accept the surgery" group(n=30) and "postpone the surgery" group(n=38).The neuropsychological symptoms were reevaluated and comparison was made between these two groups. Results The prevalence of perimenopause syndrome pre-surgery was 67.65%(22/68),and that of moderate and severe symptoms was 30.88%.The prevalence of perimenopause syndrome post-surery(97.06%)(66/68)was significantly increased,as well as that of moderate and severe symptoms(70.59%)(P
7.Informed consent right of the appraised individuals in forensic clinical examination.

Informed consent right is not just for basic ethical consideration, but is important for protecting patient's right by law, which is expressed through informed consent contract. The appraised individuals of forensic clinical examination have the similar legal status as the patients in medical system. However, the law does not require informed consent right for the appraised individuals. I recommend giving certain informed consent right to the appraised individuals in the forensic clinical examination. Under the contracted relationship with the institution, the appraised individuals could participate in the examination process, know the necessary information, and make a selected consent on the examination results, which can assure the justice and fairness of judicial examination procedure.

8.Experimental investigation on iodine-induced autoimmune thyroiditis in NOD.H-2~(h4) mice

Objective To investigate the kinetic changes of inflammatory cell infiltration and thyroid autoantibodies in the development of iodine-induced autoimmune thyroiditis in NOD.H-2~(h4)mice.Methods Either 128 five-week-old NOD.H-2~(h4)mice or 128 Kunming mice were randomly divided into two groups,and received plain water or water containing 0.05% sodium iodide.At the time points of 1,2,4,8,12,16,and 24 week after receiving iodinated water,mice were anesthetized by diethyl ether and bled from eye socket vein,and their thyroid glands were collected.Indirect ELISA method was used to measure the levels of serum thyroglobulin autoantibodies (TgAb)and thyroid hormone.After being fixed with paraformaldehyde and embedded in paraffin,thyroid sections were stained with HE and used for morphometrieal analysis.Results In the iodine treated group of NOD.H-2~(h4) mice,autoimmune thyroiditis was observed as early as 1 week after they began receiving indinated water.The prevalence as well as the degree of autoimmune thyroiditis reached the maximum at 12 weeks and remained until 24 weeks.Serum TgAb level increased after 8 weeks of iodine ingestion in NOD.H-2~(h4) mice,then increased steadily throughout the 24 weeks of experiment.On the contrary,serum TgAb was not increased in the control group of Kunming mice.Conclusion Iodine may induce and exacerbate lymphocytic infiltration of the thyroid in genetically susceptible NOD.H-2~(h4) mice,and serum TgAb is just a marker of autoimmune thyroiditis.

Arnebia euchroma is the main source for medicinal herb Zicao. and its most important component shikonin compounds have high medicinal and industrial value. This research is aimed to build overexpression vectors and RNAi vectors for key secondary metabolism genes of A. euchroma, and bulid platform for constructions of related transgenic lines using GATEWAY technology. To build genetic material based genetic research platform is to provide a great convenience for digging and functional verification of the genes on secondary metabolic pathway, and also to fill the gaps in transgenic research of A. euchroma. This study is also important for the cultivation of shikonin high-yielding strains of A. euchroma.

10.Transcriptome-based bioinformatics analysis of Arnebia euchroma ERF transcription factor family.

Twenty-seven ERF transcription factor family genes were isolated from Arnebia euchroma, with an average size of 1,010 bp, each gene encoded a 212 amino acids on average. The gene structure and expression of physicochemical properties, subcellular localization, signal peptides, senior structural domains and conservative forecasting, and analysis of A. euchroma were studied comparing with ERF gene gi261363612 of Lithospermum erythrorhizon, and phylogenetic analysis of A. euchroma ERF family was carried out. The results showed the existence of three conserved domains in this family, the senior structure based on random coil and it clustered into CBF/DREB and ERF subfamilies.
