Objective To explore the expression of TLR4/MyD88/NF-κB signaling and its correlation with the progression of acute intestinal graft-versus-host disease (iGVHD) after allogeneic bone marrow transplantation in mice.Method Recipient BALB/c female mice were lethally irradiated and were reconstituted within 4-6 h with a transplant of bone marrow cells (1 × 107) and different amounts of splenocytes (1 × 107,n =12 or 2 × 107,n =12) from MHC-mismatched C57BL/6 donors to induce iGVHD,and 6 healthy BALB/c mice served as controls.A globe survey observation of GVHD by survival, clinical manifestation, and histological detection was performed.RT-PCR,immunohistochemistry,and Western blotting technology were used to detect the mRNA and protein levels of TLR4,MyD88 and NF-κB p65 in the small intestine tissue.Result The exacerbation of iGVHD was associated with the increasing dose of allogeneic spleen lymphocytes. The mRNA expression of TLR4,MyD88 and NF-κBp65 was increased as the iGVHD progressed. All of them in severe iGVHD model were significantly increased as compared with the healthy controls (P<0.05).The expression of corresponding proteins had the same tendency as mRNA.All of the three genes expression was not only positively correlated with each other, but also with the clinical GVHD score:TLR4 (R =0.814,P<0.001),MyD88 (R=0.828,P<0.001),and NF-κB p65 (R=0.568,P =0.034).Conclusion Excessive activation of TLR4/MyD88/NF-κB signaling pathway does exist in iGVHD,and the enhanced levels of gene transcription and translation are positively correlated with the deterioration of iGVHD.

OBJECTIVETo investigate the clinical application of pedicled superficial circumflex iliac artery (SCIA) flaps in Burns and Plastic Surgery.
METHODS19 cases with skin and soft tissue defects were repaired with the Superficial circumflex iliac artery flaps. The defects located in the hand and forearm, the lower abdomen and perineal area, the radicle area of thigh, etc. Before the SCIA flaps were transfered to the recipient areas, 15 cases with different kinds of wounds were debrided thoroughly, 3 cases with scar were removed directly. The size of the flaps ranged from 3.0 cm x 4.5 cm to 26.0 cm x 22.0 cm, and the pedical was 5 cm to 7 cm in length.
RESULTSThe flaps in the 18 cases survived completely. Skin necrosis in the distal end of the flap appeared in 1 case, and the wound healed after the second repair. The follow-up period rang from 3 to 18 months. The apperance and function of the hand or foot was satisfactory.
CONCLUSIONSBecause the pedicled SCIA flaps can be obtain conveniently and contain sufficient blood-supply, so the flap is easy to survive and the flap can be designed in a large size. We believe it is an ideal method to use this flap to repair skin and soft tissue defects located in hands, forearms, the lower abdomen and perineal areas, the radicle area of thigh and so on.

Objective To approach a way to induce MSCs to dopaminergic neuron by mesencephalic conditional media and cytokines in vitro, and supply an ideal cells source for the treatment of Parkinson's disease. Methods The rat MSCs were isolated primarily from the femurs and tibias of the Wistar rats.MSCs were cultured, proliferated and purified by passage culture.Cultuered MSCs were divided into the control and the experimental group. In control group,MSCs were cultured without any induction medium.MSCs of experimental group were first cultured at medium containing bFGF for 24 hours. Then media were replaced with induction media which contained the agents as follows,respectively:GM_1,GDNF, GDNF+GM_1,GDNF+GM_1+mesencephalic conditional media. The surface markers of the differentiated neuron, such as NSE and TH were detected by immunocytoehemistry after MSCs were cultured in induction media for 3 and 7 days. Results In control groups, the NSE expression of MSCs was very lower than experimental groups. The percentage of NSE-positive cells of GDNF+GM_1+mesencephalic conditional media group in 7 day((45.257?5.999)/HP)was significantly more than other groups(control group is 2.214?0.779,GM_1 group is 22.014?3.624,GDNF group is 31.345?2.850,GDNF?GM_1 group is 40.314?4.203,P
7. Effects of red ginseng saponin components on neurochemicals in rat plasma
Chinese Traditional and Herbal Drugs 2017;48(14):2918-2924
Objective To investigate the effects of red ginseng saponin components A and B on 15 neurochemicals in rat plasma. Methods Saponin components A and B were prepared from red ginseng total saponin by macroporous adsorption resin. The composition and contents of saponin components A and B were detected using UPLC-Q-TOF-MS/MS method. A total of 15 kinds of neurochemical substances such as dopamine (DA), adrenaline (E), norepinephrine (NE), serotonin (5-HT), 5-hydroxytryptamine (5-hydroxytryptamine), tyrosine (Tyr), glycine (Gly), glutamic acid (Glu), glutamine (Gln), aspartic acid (Asp), taurine (Tau), serine (Ser), tryptophan (Try), acetylcholine (Ach), and histamine (His) were measured by high performance liquid chromatography- mass spectrometry (UPLC-QQQ-MS/MS). Results By analyzing the intrinsic relationship between saponin composition and amount of neurochemicals, it was found that components A and B had significant differences in saponin composition, and the larger polar saponins were mainly concentrated in component A. The polar of saponin contained in B is relatively small. The levels of Ach, Tau, Glu, Gln, Asp, Tyr, E, NE, and DA in rat plasma of each dose group were increased, and the plasma levels of Gly, His, 5-HT, Ser, HIAA, and Try were significantly increased and positively correlated with the dose, suggesting that the larger polar saponins could increase the excitatory neurochemical content and the smaller saponin fraction inhibitory neurochemicals. Conclusion Red ginseng has a moderating effect on the content of central nervous system. The content of smaller polar saponins is related to the maintenance of brain activity, improvement of memory function, prevention of brain fatigue, maintenance of awakening and nervous system of anti-aging, while the larger polar saponin fraction is associated with the regulation of psychiatric rhythm, reduction of stress response and improvement of sleeping.

Journal of Biomedical Engineering 2015;32(5):1131-1134
Magnetic susceptibility is an intrinsic physical quantity which describes the relationship between material magnetization and applied external magnetic field. Quantitative susceptibility mapping (QSM) is an MRI technology which can quantify the buck magnetic susceptibility of tissue in vivo. It is particularly effective at elucidating anatomy with paramagnetic or diamagnetic components. QSM technology is a method for solving the ill-pose problem of unconventional de-convolution of the measured tissue magnetic field with the unit magnetic dipole field to obtain the susceptibility source map. Many multi orientation scan based QSM and clinically acceptable single orientation QSM methods have been proposed to solve this ill-posed problem. In this paper, the QSM concept is introduced and the various QSM methods are systematically categorized and discussed. The aim of this paper is to summarize the current research progress of QSM, popularize the knowledge of QSM and promote the improvements and the rational application of QSM in clinical field.

Chinese Journal of Clinical Oncology 2015;(6):323-328
Objective:To investigate the inhibitory effects of halofuginone on radiation-induced pulmonary injury and to explore the therapeutic mechanism of this drug. Methods:A total of 72 healthy female C57BL/6 mice were randomized into 4 groups, namely, control, irradiation, halofuginone, and irradiation plus halofuginone groups, with 18 mice in each group. No treatment was performed in the control group. In the halofuginone group, the halofuginone lavage was conducted once a day, with a continuous course treatment for a month or until sacrifice of the mice. In the therapeutic alliance group, the treatment mode was the same as that in the halofuginone group. Then, a 6MV-X ray single fraction irradiation was performed after the completion of a 15-day intragastric administration. At 24 h, 1 week, 2 weeks, 4 weeks, 12 weeks, and 20 weeks after the irradiation, 3 mice from each group were randomly sacrificed, and total lung tissues were harvested. The lung was dissected to prepare pathological sections. The sections were stained with hematoxylin and eosin staining (H&E) to explore morphologic changes. The protein and mRNA expression levels of TGF-β1 were analyzed by a combi-nation of immunohistochemistry and polymerase chain reaction. The level of hydroxyproline was also measured. Results: The out-comes of H&E staining showed that halofuginone markedly ameliorated the acute pulmonary inflammation and fibrosis induced by irra-diation. The combination group had a lower level of hydroxyproline than the irradiation group, with statistically significant differences at 20 weeks after irradiation (P=0.037). The protein and mRNA expression levels of TGF-β1 were higher in the irradiation and combi-nation groups than in the control group and (or) halofuginone group at different time points (P<0.05). The combination group had lower TGF-β1 protein expression than the irradiation group at different time points, with statistically significant differences at 2, 4, 12, and 20 weeks after the irradiation (P<0.05). Meanwhile, TGF-β1 mRNA level was lower in the combination group than in the irradiation group only at 4 and 12 weeks after the irradiation (P<0.05). Conclusion:Halofuginone can ameliorate the irradiation-induced lung inflamma-tion and fibrosis probably by inhibiting the radiation-induced TGF-β1 expression. Therefore, halofugione is expected to be a therapeu-tic drug for preventing irradiation injury of the lung.
