1.Treatment and prognosis of 135 nasal non-Hodgkin′s lymphoma patients

Objective To evaluate the effects of nasal non-Hodgkin′s lymphoma(N-NHL) treated with chemotherapy alone, radiotherapy alone, chemotherapy plus radiotherapy and autologous peripheral blood stem cell transplantation(APBSCT) combined with total body irradiation(TBI);and to analyze the impact factors of prognosis. Methods 135 patients were treated between 1980 and 2000. All were confirmed by histopathology as N-NHL, including 122 T cell in origin, 12 B cell and 1 NK cell in origin. The main radiotherapy portal was set in front of the nose with a spade-like protrusion, supplement with a portal next to the ear on one side or both sides. Combined portal in facial cervical area was first used when there was invasion of the oropharynx. The median dose to the nasal cavity was 56.0Gy with a range of 35.2 to 75.5Gy, with added 30Gy to the primary lesion in two patients. Six patients received TBI combined with APBSCT, with 8Gy in the TBI group. Chemotherapy, given before or during after radiotherapy or alone, consisted of 2-6 cycles of COP, COPP, COMP, CHOP or COBDP. Prognostic factors were analyzed with Cox model. Results The local control rate was 12%,69%,76% and 83% in chemotherapy alone, radiotherapy alone, chemotherapy plus radiotherapy and APBSCT combined with TBI, respectively(P=0.057).The 5-year survival rate was 9%,52%,63% and 83%,respectively(P=0.032). Multi-factor analysis showed that tumor extension and treatment methods were the most important prognostic factors besides Ann-Arbor stage, but gender, pathology, age and symptoms had little effect on prognosis .Conclusions Chemotherapy plus radiotherapy group achieves a better survival rate than radiotherapy alone. It is helpful to evaluate prognosis to make more detail subareas on basis of local extensions in Ann Arbor staging system.For some N-NHL patients with good financial condition, APBSCT combined with TBI is a good choice.
2.Compare the curative effect of Non-Hodgkin lymphoma by four types of therapy

Objective To evaluate the curative effect of nasal non-Hodgkin lymphoma(N-NHL) by four types of therapy such as pure chemotherapy, pure radiotherapy, chemotherapy combined with radiotherapy, and APBSCT combined with TBI. Methods One hundred and thirty five patients with nasal NHL were treated between 1980 and 2000. All patients received radiotherapy alone or chemotherapy alone or radiotherapy combined with radiotherapy or TBI combined with APBSCT. The median radiation dose to the nasal cavity was 56.0 Gy with a range of 35.2 to 75.5 Gy. Six patients received TBI combined APBSCT. The TBI dose was 8 Gy. Two patients received 30 Gy in nasal of the six patients. The chemotherapy regimen consisted of 2-6 cycles of COP, COPP, COMP, CHOP, COBDP. Results The local control rate and 5-year survival rate of the four groups of pure chemotherapy, pure radiotherapy, combined chemotherapy and radiotherapy and APBSCT combined with TBI were 12 %, 69 %, 76 %, 83 % and 9 %, 52 %, 63 %, 83 %. For the four groups, the best is APBSCT, then combined chemotherapy and radiotherapy group ,then pure radiotherapy, the last is pure chemotherapy. 3.Baicalein induced Nrf2 activation and its protection against hepatotoxicity

Aim To study the activation of Nrf2 in-duced by baicalein ( BAI ) , and its protection against carbon tetrachloride ( CCl4 ) , ethanol and acetamino-phen ( APAP )-induced hepatotoxicity. Methods A reporter gene assay was conducted in human normal liver L-02 cells to detect the activation of transcription factor Nrf2 induced by baicalein. APAP ( 10 mmol · L-1 ) , CCl4 (10 mmol·L-1 ) and Ethanol (100 mmol · L-1 ) were used to induce hepatotoxicity in L-02 cells. After the pre-incubation with Baicalein (0, 1, 10, 25, 50, 100 μmol·L-1 ) for 15 min, cells were administrated with or without those above hepatotoxins. 48 h later, cell viability was detected by 3-(4, 5-dim-ethylthiazol-2-yl ) 2 , 5-diphenyltetrazolium bromide (MTT) method. Results Baicalein (25, 50 μmol· L-1 ) induced the activation of Nrf2 ( P <0. 01 , P <0. 05) in the reporter gene assay. As compared with control, three hepatotoxins ( APAP, CCl4 , Ethanol ) all decreased cell viability ( P<0. 01 ) , and baicalein significantly reversed such decreases in a concentra-tion-dependent manner ( P<0. 01 ) . Conclusion Ba-icalein can induce the activation of transcription factor Nrf2 , which is probably one of the mechanisms con-tributing to the protection of baicalein against hepato-toxins (APAP, CCl4, Ethanol)-induced hepatotoxici-ty.
4."Medical records data mining research of Zhuang Guokang’s""Nourishing Yin method""in the treatment of eczema"

Objective To explore the diagnosis and treatment rule of eczema by Zhuang Guokang. Methods Data of 58 eczema patients treated by Zhuang Guokang were collected as research objects to set up the database. Support vector machine (SVM) method was adopted to take data mining research of the key points of syndrome differentiation and the rule of medication. Results The nourishing Yin method was extracted as the category property to get 3 classification models of the syndrome differentiation key points, the related treatment methods and medication rule and 63 relevant quantitative data. The accuracy rate of model classification of the main differentiation points of nourishing Yin method corresponding to Yin reached 77.586%, and Kappa value was 0.536. Among these points, the weight of chapping(1.558), papules(1.456), thready pulse(1.036)and desquamation(0.858) got higher significance. The accuracy rate of model classification of medication feature reached 86.2069%, and Kappa value was 0.719. Among these medicines, Pseudostellaria heterophylla(0.776), Semen plantaginis (0.550), Asparagus root(0.525) and Solomonseal (0.388) got higher significance. Conclusion The research revealed the syndrome differentiation key points and the medication rule of Zhuang Guokang’snourishing Yin methodin the treatment of eczema, and explored the path of experience transforming to knowledge of the famous old TCM doctors.
5. Infusion of mesenchymal stem cells culture supernatant ameliorates hyperglycemia disorders in STZ-induced diabetes mellitus rats

Objective To evaluate the therapeutic effect of systemic infusion of MSC culture supernatant in STZ-induced diabetic rats, and explore the mechanism of effect of MSC secretion on promoting regeneration of the islet tissues. Methods The diabetic animal model was reproduced in 35 SD rats by intraperitoneal injection of a single large dose of streptozotocin (STZ, 65mg/ kg). The 30 successfully induced diabetic rats were randomly divided into MSC culture supernatant infusion group (CM, n=15) and medium infusion group (M, n=15), and in addition, 15 normal rats were used as control. Animals were intravenously transfused with MSC supernatant (CM group) or raw medium (M group), then the contents of blood glucose were determined 3 days after infusion. The serum insulin and C-peptide levels were monitored and the intraperitoneal glucose tolerance test (IPGTT) was performed on the 7th day of infusion to evaluate the therapeutic effect of MSCs supernatant infusion in diabetic rats. Finally, all the experimental animals were sacrificed at indicated time points and the pancreatic tissues were collected for multiple immunofluorescence staining (MIFS), in order to observe the β-cell regeneration after MSCs supernatant infusion, and to further explore the possible mechanism involved in the experiment. Results At the early stage after infusion (<7 days), the blood glucose level declined and the contents of serum insulin and C-peptide increased obviously in CM group as compared with that of M group (P<0.05). IPGTT showed that the islet function was significantly enhanced in CM group compared with M group. MIFS showed that the number of β cells in the destroyed islets in CM group rats was significantly increased as compared with that of M group rats. In addition, the proliferation rate of β cells was obviously higher in CM group (4%) than in control group (0.5%, P<0.01). Conclusion Treatment with MSCs culture supernatant obviously prompted β cell proliferation in the destroyed islets, resulting in regeneration of the islets with islet function recovery, thus it effectively controls the advance of diabetes.

Objective: To examine the distribution of IL-1B+3953 genotypes of Han nationality people with different periodontal health status and to evaluate if there is an association between the genotype of IL-1B+ 3953 and the severity of periodontitis. Methods: 271 subjects of Han nationality were selected, among them there were 85 cases with severe chronic perioodontitis(group①), 97 cases with light and mild chronic periodontitis (group ②) and 89 subjects of periodontal healthy control(group ③). Full periodontal examinations were taken including clinical periodontal tissue attachment loss measurements of full-mouth, periodontal pocket depths and bleeding on probing. DNA samples were obtained with buccal swabbing technique and were further analyzed for IL-1B+3953 genotype polymorphisms using PCR-RFLP-based method. 8.Application and projects approved for fundamental research on integrated traditional Chinese and Western medicine in National Natural Science Foundation of China from 2000 to 2004

From 2000 to 2004, the National Natural Science Foundation of China (NSFC) accepted 1 171 applications and funded 160 projects for fundamental research on integrated traditional Chinese and Western medicine. The success rate is 13.64%. Being supported by NSFC, a number of achievements well known in China and abroad have been made, such as acupuncture complementary anaesthesia, treatment of leukemia, viral hepatitis, cardio-cerebrovascular diseases, acute abdomen, burns and fracture with integrated traditional Chinese and Western medicine and their therapeutic mechanisms, and some new concepts and theories have been put forward, such as the theories of activating blood to resolve stagnation, simultaneous treatment of bacteria and toxin, etc. But there still exit some problems in the research of the funded projects. The research ways are mainly combination of different methods instead of integration of both traditional Chinese and Western medicine. The research results are mainly confirmation of traditional Chinese medical theories instead of exploration of their innate regularities. The relationship among disease, syndrome and symptom is not clearly explored. The principal-subordinate relationship between macro- and micro-differentiation of syndromes is not clear. Academic research is short of new ideas. Improper or biased explanation of the theories of traditional Chinese medicine still exists. On analysis of above problems, some preferable aspects of projects for fundamental research on integrated traditional Chinese and Western medicine funded by NSFC in future are suggested in this article.
9.Expression of peroxisome proliferator-activated receptors in the epidermis of skin from patients with psoriasis vulgaris

Objective To investigate the expression of peroxisome proliferator-activated receptors (PPARs) in epidermal keratinocytes of patients with psoriasis vulgaris and its significance.Methods Immunohistochemical method was used to examine the expression of PPARα,β/δand.γ in tissue specimens from the normal skin of 5 human controls,lesional and normal skin adjacent to the lesions of 17 patients with psoriasis vulgaris.A semi-quantitative analysis was carried out by image analyzer.Results Different levels of expression of the three PPAR isotypes were observed in the nuclei of epidermal keratinocytes of normal human controls.The expression intensity of PPAR α,β/δand γ was statistically higher in the epidermis of adjacent normal skin than that in psoriatic lesions(all P<0.01)and normal control skin(all P<0.01).Conclusions The decreased expression of PPARα may be associated with the overproliferation and parakeratosis of epidermal keratinocytes in psoriatic lesions,and PPAR β/δ and γ may display a synergistic effect on the maintenance of homeostatic proliferation and difierentiafion of epidermal cells.
10.Effect of glycoprotein (transmembrane) nonmetastatic melanoma protein B on the proliferation and migration of as well as melanogenesis in melanoma cells

Objective To estimate the effect of glycoprotein (transmembrane) nonmetastatic melanoma protein B (GPNMB) on the proliferation and migration of as well as melanogenesis in melanoma cells.Methods The expression of GPNMB was detected by immunofluorescence assay in two melanoma cell lines M14 and G-361,as well as in primary human melanocytes.Then,the three kinds of cells each were classified into three groups:experimental group treated with small interfering RNA targeting GPNMB (GPNMB-siRNA),negative control group treated with the negative control siRNA,blank control group remaining untreated.Methyl thiazolyl tetrazolium (MTT) assay,transwell invasion assay and spectrophotometry were performed to evaluate cell proliferation activity,invasion potential and melanin levels,respectively.Statistical analysis was done using Student's t test.Results GPNMB was expressed in both melanoma cells and melanocytes.The transfection with GPNMB-siRNA down-regulated the mRNA and protein expressions of GPNMB in,and markedly suppressed the proliferation and migration of,melanoma cells.In detail,the proliferative activity (expressed as the absorbence value at 570 nm) of M14 and G361 cells was reduced by 35% and 40% respectively,the migration activity of M14 and G361 cells by 49% and 51% respectively,and the melanin levels in melanocytes,M14 cells and G361 cells by 73%,82% and 69% respectively,in the experiment group compared with those in the blank control group.Conclusions The siRNA-mediated silencing of GPNMB could effectively inhibit the proliferation of,invasion of and melanogenesis in melanoma cells,which suggests that GPNMB plays critical roles in the initiation and progression of melanoma.
