Objective To study the combined effect of chlorotoluron and atrazine on the testis of mice. Methods Kunming mice were divided into groups and treated with chlorotoluron and atrazine alone or in combination by gavage at different doses for 25 consecutive days. Microscope and electron microscope were used to observe the morphological changes. Results The herbicides used alone or in combination, at all the test doses, caused the morphological changes in degrees in the testis. Under the light microscope, seminiferous epithelium arrayed loosely and disorderedly, spermatogenic cell shed, and layers lessened compared with the control group. Under the electron microscope, mitochondria in the seminiferous epithelium appeared vacuolated, karyotheca swelled, bent and the function of sustentacular cell declined. Compared with alone use groups, the pathological changes were more serious in combination use groups. Conclusion Chlorotoluron and atrazine can produce a combined toxic effect on the testis of mice.

Objective Analyzing the distribution of 1476 strains of clinical common pathogens and their antimicrobial resistance, provide reference the reasonable use of antimicrobial drugs for clinical.Methods Using BD company PHOEIX-100 Automatic Pathogen Identification to Pathogen identify 1476 strains of pathogens which were got from the various isolated and cultivated samples which were examined from 2009 January to 2010 August and its antibacterial Drug Susceptibility Test, then with WHONET5. 3 system to analyze monitoring data of the pathogen distribution and resistance. Results In 1476 strains of pathogens isolated, gram-negative bacteria had 1206 strains occupying 81.7% ; Gram-positive cocci had 270 stains occupying 18. 3%. The most common detected pathogen were Escherichia coli, Klebsiella pneumoniae,Staphylococcus aureus and Pseudomonas aeruginosa, which accounted for 20. 2% ,17. 5% ,13. 2% ,11.4%.The ESBLs producing E. coli and K. pneumoniae accounted for 65. 9% ,48. 1% ; Also methicillin-resistant S. aureus (MRSA) accounted for 79. 2% and methicillin-resistant coagulase-negative staphylococci (MRCNS) accounted for 68.4%. It isnt found yet that grape genus to vancomycin, nitrofurantoin can resist drug.Gram-negative bacilli to antimicrobial vinyl hydrocarbon mold degree of sensitivity is high. Conclusion The isolated pathogen's resistance of drug is widespread. It is important to guide clinical reasonable use of antibiotics and control infectious pathogens that carry out continuous monitoring of drug resistance.

Objective To study the expression and clinical significance of CD34 and smooth muscle actin (SMA) in stroma of breast tissues and lesions. Methods Seventy cases of breast tissues and lesions, including 20 fibroadenomas, 10 sclerosing adnoses, 30 invasive ductal carcinomas and 10 invasive lobular carcinomas were investigated, and 10 normal breast tissues were served as controls. Immunohistochemical staining was applied to compare the distribution of CD34+ fibrocytes and SMA-reactive myofibroblasts. Results The stroma of normal breast tissue contained CD34+ fibrocytes, whereas SMA-reactive myofibroblasts were absent (100% for both). All benign breast lesions exhibited astromal CD34+ fibrobytes, and fibroadenomas showed SMA-reactive myofibroblasts as well. In invasive ductal and lobular carcinoma the stroma was devoid of CD34+ fibrocytes, but a varying number of stromal SMA-reactive myofibroblasts were detectable (100%). Conclusion In breast carcer, immunohistochemical staining used in detecting expressions of CD34 and SMA is helpful in distinguishing benign lesions from malignancies.

OBJECTIVE： To develop a HPLC method for the determination of the contents of lidocaine and prilocaine in compound lidocaine cream as a quality control means.METHODS： Lidocaine and prilocaine in compound lidocaine cream were determined by high-performance liquid chromatography on C18 column with the detection wavelength at 254nm.The mobile phase was 0.5％ ammonium dihydrogen phosphate(pH=7)-methanol(20∶ 80).RESULTS： The calibration curves of both lidocaine and prilocaine were linear within the concentration range of 130～250μ g/ml(r=0.9 996).The recovery rates of lidocaine and prilocaine were 99.05％ and 99.27％ respectively， RSDs were 0.67％ and 1.15％ ， intra-day RSDs 0.81％ and 1.45％ ， inter-day RSDs 0.55％ and 0.63％ respectively.CONCLUSION： The method was sensitive， stable and accurate.It can be used to determine and control the quality of compound lidocaine cream.

Objective Comparatively studying the method,efficiency and anti-hypoxia ability of cardiomyocytes which are directionally induced from human embryonic stem cells and mouse embryonic stem cells in vitro so as to provide an experimental basis for further study of inducing the differentiation of embryonic stem cells into cardiomyocytes in vitro.Methods Human embryonic stem cells are induced into cardiomyocytes by suspension method which dosen't using inducersand adherence method which using inducers respectively.Mouse embryonic stem cells are induced into cardiomyocytes by hanging drop method which dosent using inducers and using inducer respectively.Staining the specific marker cTnT of cardiomyocytes by immunofluorescence.Comparing the time,percentage and beating frequency of cardiomyocytes by counting under the microscope.Using apoptosis-hoechst staining kit to detect the apoptosis ratio of beating cardiomyocytes which have been treated by hypoxia with 24 h.Results The group of hESC without inducers results in that the mean time days of appearing beating cardiomyocytes is (13.9 ± 0.9) days,the percentage is 20.8 ％ and the average frequency of beating is (63.8 ± 5.6) times/min.The group of hESC with inducers results in that the mean time days of appearing beating cardiomyocytes is (13.0 ± 1.1) days,the percentage is 66.7％ and the average frequency of beating is (63.0 ± 7.0) times/min.The group of mESC without inducers results in that the mean time days of appearing beating cardiomyocytes is (14.3 ± 1.0) days,the percentage is 12.5％ and the average frequency of beating is (80.2 ± 3.9) times/min.The group of mESC with inducers results in that the mean time days of appearing beating cardiomyocytes is (12.2 ± 1.2) days,the percentage is 81.3％ and the average frequency of beating is (79.9 ±7.7) times/min.Beating cardiomyocytes of each group are positive to cTnT staining.Different apoptosis ratio are detected of beating cardiomyocytes of each group.Conclusion The four methods can all successfully induce the differentiation of embryonic stem cells into myocardiocytes,and the adherent method of hESC induced with activin A + BMP4 is the first successful induction in China.The groups adding inducers improve the differentiation efficiency more significantly than the groups without adding inducers.Inducing mouse embryonic stem cells into cardiomyocytes is more simple and efficient compared with human embryonic stem cells.Without the presence of other protective factor,anti-hypoxia ability of cardiomyocytes induced from human embryonic stem cells is stronger and the beating time are longer in vitro compared with mouse embryonic stem cells.

Objective To explore the effect of protease-activated receptor 1(PAR-1) activation on the expression of CXCR4 mR-NA of mouse endothelial progenitor cells (EPCs) and proliferation ,migration of EPCs .Methods Mouse EPCs were activated by different concentration of SFLLRN (one agonist of PAR-1) ,or transfected by small interfering RNA of PAR-1 .The expressions of PAR-1 and CXCR4 mRNA of EPCs were detected by fluorescent quantitative real-time PCR .Proliferation ,migration of EPCs were detected by MTT ,Transwell chambers respectively .Results The expressions of PAR-1 and CXCR4 mRNA in SFLLRN group were higher than that in other groups(P< 0 .05) .The expression of CXCR4 mRNA was highly positive correlation with PAR-1 mRNA(r=0 .991) .The proliferation ,migration of mouse EPCs were induced by activation of PAR-1 .Conclusion Activation of PAR-1 promotes cell proliferation and migration of mouse EPCs ,this effect may be depended on CXCR4 .
