Objective To explore the relation between endothelial function and Alzheimer′s disease(AD).Methods The concentration of asymmetric dimethylarginine(ADMA)and L-arginine were measured by high-performance liquid chromatography(HPLC),the concentration of nitric oxide(NO) detected by the method of nitrate reductase,and the activity of nitric oxide synthase(NOS)examined using NOS assay kits,in the plasma of 45 mild/moderate AD patients and 48 controls.Results Compared with controls,the concentration of ADMA(0.95?0.09?mol/L,P=0.00)was significantly increased,however the activity of NOS((13.9?3.9)U/L,P=0.00),inducible NOS(iNOS)((5.1?1.8)U/L,P= 0.00)and concentration of NO((41.7?5.0)?mol/L,P=0.00)were significantly decreased.In addition,Logistic regression analysis found significant positive correlations between the plasma concentration of NO and the score of the CDR(y=36.64?0.398_x;r=0.398,P
3.Effects of Tanshinone ⅡA on Arrhythmia and Levels of Ischemia Modified Albumin and Heart Type Fatty Acid Binding Protein in Rats with Myocardial Ischemia-reperfusion Injury

Objective To observe the effect of tanshinoneⅡA (TⅡA) on arrhythmia and levels of ischemia modified albumin (IMA) and heart type-fatty acid binding protein (H-FABP) in rats with myocardial ischemia-reperfusion injury.Methods Forty Wistar rats were equally randomized into 5 groups:pseudo-operation group (normal saline),model group (normal saline),and high-,middle-and low-dose TⅡA groups (in the dose of 20,10 and 5 mg?kg-1?d-1 respectively).The rats were treated for 7 days,and received operation 2 hours after the last medication.Rats in pseudo-operation group received threading without ligation,while animal model of myocardial ischemia-reperfusion injury was established by ligating the left-anterior coronary artery for 15 minutes and then loosening for 30 minutes.During the operation,electrocardiogram was traced continuously to observe the arrhythmia.HE staining was used to observe the pathological changes of myocardium.Levels of IMA and H-FABP in myocardium were determined by enzyme-linked immunosorbent assay (ELISA).Results Ischemic changes of the myocardium and arrhythmia occurred,myocardial IMA level increased and H-FABP level decreased in the model rats.TanshinoneⅡA obviously attenuated arrhythmia caused by ischemia-reperfusion injury (P
4.Efficient transient expression to analyze miRNA targets in rice protoplasts.

Compared with the transgenic approach, transient assays provide a convenient alternative to analyze gene expression. To analyze the relationship between miRNAs and their target genes, a rice protoplast system to detect target gene activity was established. The MIRNA and GFP-fused target sequence (or GFP-fused mutated sequence as a non-target control) were constructed into the same plasmid, and then delivered into rice protoplasts. The GFP expression level decreased significantly when the protoplasts were transfected with the plasmid containing GFP-fused target compared to that of the plasmid with non-target sequence either by fluorescence microscopy or qRT-PCR method. Two microRNA genes, osaMIR156 and osaMIR397, and their target sequences were used to prove the feasibility of the rice protoplast transient assay system. This method will facilitate large-scale screening of rice miRNA target in vivo, and may be suitable for functional analysis of miRNAs of other monocot plants that might share the evolutionarily conserved small RNA processing system with rice.

5.Value of MRS in the differential diagnosis of corpus callosum lesions

Objective To explore the value of multivoxel 1H-MRS in the differential diagnosis of corpus callosum lesions.Methods 17 patients with corpus callosum lesions confirmed by clinical or pathological method underwent MRS examination.These patients included 2 cases of lymphoma,4 cases of Marchiafava-Bignami disease,3 cases of glioma, 7 cases of infarction,and 1 case of multiple sclerosis.MRS features of lesions were analyzed.Results 2 cases with corpus callosum lymphoma revealed significantly increased Cho peak, decreased Cr and NAA peak,towering Lip peak.Among 4 cases with Marchiafava-Bignami diseases, 2 cases revealed increased Cho/Cr and decreased NAA/Cr,1 case showed normal,1 case revealed inverted Lac peak.For 3 cases with corpus callosum glioma,NAA peak decreased or disappeared,and Cho peak increased in varying degrees.7 cases with corpus callosum infarction revealed significant inverted Lac peak, and NAA peak decreased in varying degrees.1 case of multiple sclerosis revealed increased Cho peak,decreased NAA peak,and inverted Lac peak during acute period with enhancement,while these features returned normal during inactive period.Conclusion Multivoxel 1H-MRS plays an important role in the differential diagnosis of corpus callosum lesions.
6.Liposomes-mediated chemosynthesis siRNA transfection to primary cardiomyocytes: Selection of an ideal concentration

BACKGROUND: siRNA transfection is a key step in RNA interference. The methods of cardiomyocytes transfection were most use of plasmids as vector to transfect long-chain shRNA. However, the processes were complex. It was a simple efficient and low-toxic method that liposomes-mediatad chernosynthesis siRNA transfection. It was useful for expanding RNA interference application.OBJECTIVE: To choice the optimal concentration of Iiposomes-mediated chemosynthesis siRNA transfection, and to discover a simple efficient RNA interference application.METHODS: CY3-Negative siRNA was mediated by lipid-besed agent siPORT~(TM) NeoFX~(TM) to transfect cardiomyocytes. A blank control group was set. After 24 hours, the transfection efficiency and apoptotic rate were evaluated by fluorescent microscope and flow cytometer to select an optimal concentration. Based the best concentration, siRNA PHB was transfected to cerdiomyocytes. 48 hours later, the expression of PHB was tested.RESULTS AND CONCLUSION: With the increased concentration of CY3-Negative siRNA, the number of cells emitted red fluorescence grew under fluorescence microscope, and the transfection efficiency was also increased (P<0.05). The best concentration was 30 nmol/L (P<0.05). There was no significant difference in apoptotic rate between transfected groups and the control group (P > 0.05). The PHB expression of cardiomyocytes transfected siRNA PHB was dropped by 74.11% on average (P<0.05). The results indicated that lipid based agent siPORT~(TM) NeoFX~(TM) was suitable to transfect chemosynthesis siRNA to cardiomyocytes, and the best transfection concentration of siRNA was 30 nmol/L.

BACKGROUND:Artificial teeth computer aided design and computer aided manufacture (CAD/CAM) is a high technology involving various subjects.Many CAD/CAM systems have designed.However,they have some disavantages and a high cost.Therefore,only the system with proprietary intellectual property can solve this problem.OBJECTIVE:To optimize the connector of the CAD/CAM fixed bridge.METHODS:Three-dimensional finite element models of the connector were established which was used to design different styles of connectors.Stress under different vertical loading conditions was compared.RESULTS AND CONCLUSION:The connectors with different appearances displayed varying stress.Stress on the connector with arc transition was less than connector without arc transition.Connector was supposed to design to style with arc transition.

9.Analysis of cone beam CT for the skeletal structure of temporomandibular joint in patients with sleep bruxism

Objective To analyze the skeletal structure features of temporomandibular joint (TMJ) by using cone beam computed tomography (CBCT) in patients with sleep bruxism. Methods CBCT scanning was performed in the intercuspal position in 30 patients with sleep bruxism (sleep bruxism group) and 30 health adults (normal group). The reconstruction of TMJ images were analysed by Invivo5 software. Measurements of horizontal section include antero-posterior diameter, medial-lateral diameter and axial angel. Measurements of sagittal section include acetabulum depth, aricular eminence inclination, anterior space, superior space and posterior space. Measurements of coronal section include force angle. Results There were no significant differences in both sides of TMJ between sleep bruxism group and normal group(P>0.05). Compared with control group, there were significant differences in antero-posterior diameter, medial-lateral diameter, acetabulum depth, anterior space and force angle in sleep bruxism group (P<0.05). There were decreased antero posterior diameter, me?dial-lateral diameter and increased acetabulum depth, anterior space and force angle in sleep bruxism group than those of control group. Conclusion The bilateral bone stuctures of TMJ are basically symmetry in patients with sleep bruxism. There is no significant change in condylar position in acetabulum , but there is a trend of absorbtion of anterior space and de?crease of condyle in patients with sleep bruxism.
10.Anti-oxidation of Tanshinone ⅡA and Prohibitin on Cardiomyocytes

Objective To investigate the anti-apoptotic mechanism of tanshinone ⅡA and the function of prohibitin(PHB)onmyocardial cells apoptosis induced by hydrogen peroxide(H2O2).Methods Myocardial cells were primary culturedneonate rat were cultured in medium with 200 μmol/L H2O2,and the medium was supplemented with tanshinone ⅡA(1 × 10-4 mol/L)in advance for 24 h.PHB in myocardial cells was knocked down by RNA interference,and theexpression level of PHB was determined by Western blotting analysis.Flow cytometric analysis was used to detectapoptosis rate,intracellular calcium concentration([Ca2+]i),and mitochondrial membrane potential(MMP).ResultsH2O2-mediated cell apoptosis resulted in activation of PHB,increasing of[Ca2+]i,and decreasing of MMP.TanshinoneⅡA profoundly inhibited myocardial cell apoptosis induced by H2O2,decreased[Ca2+]i,and increased MMP.Specificsilence of PHB by siRNA down-regulated the expression level of PHB,increased apoptosis rate and[Ca2+]i,and decreasedMMP.Conclusion The results demonstrate that tanshinone ⅡA could attenuate apoptosis induced by H2O2,and theactivation of PHB induced by H2O2 is the major regulatory pathway of cyto-protective gene expression against oxidativestress.
