4.The roles of occludin in changes of Wood-brain barrier permeability

As an important transmembrane protein in brain microvascular endothelial cells tight junction, occludin plays important roles in regulating and maintaining blood-brain barrier permeability. Its abnormal expression can increase blood-brain barrier permeability. In order to provide some theoretical supports for the roles of studying occludin intensively in the mechanisms of blood-brain barrier permeability-related diseases and targets for pharmacological intervention, this article reviews the roles of occludin in the molecular composition and functional maintenance of blood-brain barrier tight junction, as well as the changes of cerebral microvascular permeability during hypoxia.
5.Determination of the Content of Gentamycin Sulfate C Component in Medicinal Albumin Gel by HPLC

OBJECTIVE:To establish a HPLC method for the determination of gentamycin sulfate C component in medicinal albumin gel. METHODS: Samples was separated on Agilent ZORBAX Eclipse XDB-C8 at a column temperature of 26℃. The mobile phase was methanol-acetic acid-water (70∶5∶25, added with 0.02mol?L-1 sodium heptanesulfonate solution) for gradient elution at a flow rate of 1.0mL?min-1. The detection wavelength was set at 330nm. RESULTS: Baseline separation of gentamycin sulfate C component in medicinal albumin gel was achieved. The linear range of gentamycin sulfate C component was 0.26?g~6.50?g and its average recovery rate was 90.0%, RSD=0.43%, the lowest limit of detection was 0.368ng. CONCLUSION: The method was proved to be simple, accurate and reliable, and suitable for the content determination of gentamycin sulfate C component in related preparations.

7.the Application of Homemade Stapler in Anus-retained Operation and it′s Relative Aspects.

Objective To investigate the application of homemade stapler in anus-retained operation on low and super-low colon or rectal cancer.Methods From August 1991 to August 2006,this technique was used in 333 patients with low colon or rectal cancer,among which 96 patients were right hemicolon excision,ileum colon anastomosis,colon sigmoideum excision,colon descendens rectum anastomosis and 183 patients were rectum pro-excision,low pro-excision,54 patients were low pro-excision,super low anastomosis.The other traditional operations(59 patients manipulation anastomosis) were used in 166 patients.Results Fourteen patients had anastomotic fistula(one rectovaginal fistula) in group1 and two in group2.Two patients had pro-sacral bone bleeding,and one dead.Conclusion The stapling device was a safe and feasible choice in anus-saving operation of low rectal cancer.
8.Effect of paeoniflorin on protein expression of HFCL of human bone marrow stromal cells

Objective To investigate the effects of paeoniflorin on proliferation and the protein expression of HFCL of human bone marrow stromal cells and to discuss molecular mechanism of blood enriching functions of paeoniflorin. Methods Flow cytometry and proteomics were used respectively to measure the effect of paeoniflorin on cell cycle and protein expression of HFCL. Results Paeoniflorin could promote HFCL from G_0/G_1 phase to S phase, increase proliferative index, up-regulate nine kinds of proteins, and down-regulate five kinds of proteins of HFCL. The proteins up-regulated include Ras-related nuclear protein, lamin A/C, isocitrate dehydrogenase 3 (NAD+), triosephosphate isomerase, ATP synthase, ribosomal protein P2, and chaperonin containing t-complex polypeptide 1. Conclusion Through promoting HFCL proliferation, acting on multiple protein targets, enhancing the synthesis of cytoarchitecture proteins and the expression of protein chaperonin, increasing the energy metabolism of HFCL, and suppressing the apoptosis of HFCL, paeoniflorin plays indirectly blood enriching function.
9.The Effect of Pseudolaric Acid on the Expression of VEGF in Hepatocellular Carcinoma Cells

Objective To investigate the effect of pseudolaric acid B(PAB) on the expression of vascular endothelial growth factor (VEGF) in human hepatoma BEL-7402 cells. Methods BEL-7402 cells were cultured with PAB (0, 0.5, 1.0, 2.0, 4.0 and 8.0μmol · L-1) for 6 h and stained with 0.4%trypan blue. The cell viability was observed and calculated under the microscope. The cell proliferation was detected by WST-8 analysis. The expression of VEGF mRNA in BEL-7402 cells was detected using RT-PCR method. The expression of VEGF protein in BEL-7402 cells was detected using Western blot assay. Results There was no significant difference in the cell proliferation of BEL-7402 cells between different PAB treat-ment groups and control group. With the increased concentration of PAB, the expressions of VEGF mRNA were significantly decreased in different PAB treatment groups (1.90±0.02,1.31±0.03,0.86±0.04,0.60±0.03 and 0.34±0.02) compared with that of control group (2.53±0.07, P<0.01). The expressions of VEGF protein were significantly decreased in different PAB treat-ment groups (2.60±0.09, 2.42±0.07, 1.85±0.05, 0.92±0.05 and 0.60±0.04) compared with that of control group (3.06±0.15, P<0.01). Conclusion PAB can inhibit the expressions of VEGF mRNA and protein in BEL-7402 cells.
10.The characteristics of metabolic syndrome among staffs of Guangdong

Objective To investigate and analyze the prevalence of metabolic syndrome and its components among staffs of Guangdong. Methods One thousand six hundred and fifteen persons were enrolled in this survey. The medical check up data from the physical examination were analyzed. Results The prevalence of metabolic syndrome, obesity, hyper-triglyceridemia, hypo-HDL-C, hypertension, glucose metabolic disorder were 16.3% ,27.6% ,22.7%, 15.0% ,51.8%, 10.7% respectively. The MS prevalence was increased with aging. Prevalences of 0-5 MS components were 31.5% ,31.2% , 22.2% , 10.0% ,4.7%,0.3% respectively. It was significant difference between 2 groups that UA, ALT, AST, CHOL, LDL-ch (t value is 6. 473,4. 238,7. 406,6.810,8. 350, seperatly, P<0.01). Conclusions The prevalence of MS is high in the staffs of Guangdong. The blood pressure increasing prevalence was maximal. The other components prevalences were high too. MS tends to be a many data abnormality disease, early detection and intervention are important.
