1.Therapeutic strategy for HER-2-positive advanced breast cancer

Patients with HER-2-positive advanced breast cancer were associated with poor prognosis. Meanwhile, HER-2-targeted ther-apy has dramatically improved survival and prognosis among breast cancer patients. Over the years, multiple HER-2-targeting drugs stepped into clinical practice, and the targeted agents are now considered as the standard of care in the first-line setting and beyond. This review basically summarizes the importance of HER-2-targeted therapy, the significance of the clinical trial results, and the clinical practice guidelines for the management of patients with HER-2-positive advanced breast cancer.
2.Differential expressions of mitochondrial genome and apoptosis related genes in A549 cell line induced by cisplatin

Objective To investigate the differential expression of mitochondrial genome and apoptosis-related genes in human lung adenocarcinoma A549 cell line induced by cisplatin. Methods A549 cells were cultured in RPMI 1640 medium supplemented with 10% fetal bovine serum (FBS; Hyclone) in 150-mm dishes, and the test cells were exposed to 0.5?g/ml cisplatin continuously for 20h. Same volume PBS solution was added to culture of control cells instead of cisplatin. Human mitochondrial genome oligonucletide micoarray was used to detect the differential expression of 26 target genes. The apoptosis status was analyzed by the flow cytometry technique. Results Compared with the control cells, the expression levels of tRNA-Cys, tRNA-Asn, 12S rRNA and cytochrome oxidase subunit I were remarkably elevated in test cells. The expression levels of bax, NADH dehydrogenase subunit I, NADH dehydrogenase subunit Ⅱ and tRNA-Ser, tRNA-Asp, tRNA-Arg and tRNA-Ala also were elevated to some extent. OBJECTIVETo study the inhibitory effect of Xiaotan Sanjie Recipe (XSR) on the microsatellite instability of orthotopic transplantation tumor in MKN-45 human gastric cancer nude mice.
METHODSThe 3rd passage subcutaneous transplantation tumor was taken as the origin of the model by using MKN-45 human gastric cancer cell lines. MKN-45 human gastric cancer nude mouse model was established using OB glue adhesive method. Then 30 nude mice were divided into the model group, the XSR group, and the chemotherapy group. Mice in the XSR group were intragastrically given XSR at the daily dose of 0.4 mL. Mice in the chemotherapy group were intragastrically given Fluorouracil at the daily dose of 0.4 mL. No intervention was given to mice in the model group. After 6 weeks of medication, the tumor weight was measured, and the tumor inhibition rate calculated. The size, the peak height, and the peak area of 5 microsatellite instability sites were detected.
RESULTSThe tumor inhibition rate was 40. 84% in the XSR group. The tumor weight was significantly lower in the XSR group than in the model group (P < 0.01), showing no statistical difference when compared with the chemotherapy group (P >0.05). The incidence of high microsatellite instability (MSI-H) in the model group was 70%, and the incidence of low microsatellite instability (MSI-L) was 30%. Microsatellite stable site tended be stable after 6 weeks of XSR treatment.
CONCLUSIONXSR showed inhibition on microsatellite instable orthotopic transplantation tumor in MKN-45 human gastric cancer nude mice.

7.Study for cardiac systolic synchronization in patients with ischemic cardiomyopathy

Objective:To explore value of pulsed wave (PW) and tissue synchronization imaging (TSI) evaluate cardi-ac systolic synchronization in patients with ischemic cardiomyopathy (ICM) .Methods:A total of 50 ICM patients were enrolled as ICM group and 35 healthy volunteers without organic heart disease were regarded as healthy control group ,PW was used to measure mitral diastolic blood flow duration/RR interval (LVFT/RR) to evaluate synchroni-zation of left atrial and left ventricular synchronization ;interventricular mechanical delay (IVMD) was measured to evaluate left and right ventricular synchronization ;TSI software was used to measure mitral annular mean systolic peak velocity (LV-Sm) ,systolic time to peak (Ts) and Ts standard deviation (Ts-SD) of all segments in order to e-valuate left ventricular systolic synchronization .Results:Compared with healthy control group ,there were signifi-cant reductions in LVFT/RR [ (44.74 ± 1.58)% vs .(41.08 ± 4.65)% ] and LV-Sm [ (9.72 ± 0.53) ms vs .(4.09 ± 1.06) ms] ,and significant rise in IVMD [ (15.51 ± 5.52) ms vs .(41.96 ± 4.20) ms] and Ts-SD [ (16.47 ± 4.16) ms vs .(34.13 ± 11.68) ms] in ICM group , P<0.01 all;Ts of anterior wall ,inferior wall et .6 positions were significantly longer ,and its synchronization significantly reduced , P<0. 05 all ,percentages of the most de-layed systolic part of left ventricle were no significant differences between two groups (P> 0.05) .Conclusion:Compared with healthy control group ,left ventricular total systolic function ,left atrial&ventricular ,left&right ventricular synchronization and systolic synchronization within left ventricular significantly reduce in ICM patients .

OBJECTIVE:
To study the effect of MDR1 (P-gp) and ABCG2 on the drug resistance in Hep 2 cells.
METHOD:
Flow cytometry was used to detect the variations of the antitumor drugs accumulation and discharging, and activity variations when MDR1 and ABCG2 inhibitors were used in Hep-2.
RESULT:
The accumulation and discharging of mitoxantrone was significantly higher than the control group when ABCG2 inhibitor FTC was used in Hep-2 (P<0. 05). In contrast, P-gp did not appear similar case; To the mitoxantrone and cisplatin, there was no statistical correlation about activity of Hep-2 between P-gp or ABCG2 antagonist and the control; To the doxorubicin, combining FTC and P-gp, the activity of Hep-2 was higher than the control and difference was significant (P<. 05), In contrast, FTC and P-gp did not appear similar case when used alone; To the 5-FU, when PGP used, the activity of Hep-2 was higher than that in the control and difference was significant (P<0. 05), In con- trast, FTC and FTC+P-gp did not appear similar case; To the paclitaxel, when P-gp or FTC+P-gp used, the activity of Hep-2 was higher than that in the control and difference was significant(P<0. 05).
CONCLUSION
ABCG2 may lead to drug resistance mainly by changing the ability of cell in accumulating and discharging chemotherapy drugs. P-gp has other way. P-gp and ABCG2 play different roles in different drug resistance.
