2.Progress in relationship between long non-coding RNAs and gastric cancer

Long non-coding RNAs (lncRNAs) are a group of regulatory and non-coding RNA molecules which are longer than 200 nucleotides. They are involved in a variety of biological processes such as cell proliferation, differentiation and apoptosis mainly by regulation of gene expression at various levels, including epigenetics, transcription and post-transcription. The evidence has demonstrated that many lncRNAs are dysregulated and closely related to tumorigenesis, progression and prognosis in gastric cancer. Based on an introduction to lncRNAs and the domestic and foreign research status, this review summarizes the function, regulatory mechanisms, potential diagnostic and therapeutic values of lncRNAs in gastric cancer.

The hypotensive effect of PAMC and its mechanism were studied.In conscious renal hypertensive rats, intraperitonal injection of PAMC once a day for 7d at the dosage of 11 mg/kg ( 1 time/day ) and oral administration of the drug 3 times a day for 14 d at the dosage of 12 mg/kg/d, the arterial pressure decreased 29.0?2.8% and 20.3 ? 1.4%, respectively.In normal anesthetized dogs and rats, intravenous injection of PAMC at the dosage of 0.4 mg/kg, the mean arterial pressure deceased 43.7 ? 7.9 and 39.7 ? 2.3%. In anesthetized rabbits and rats, intraduodenal injection of PAMC at the dosage of 4 mg/kg, the mean arterial pressure decreased 24.5 ? 3.8% and 38.2 ? 11.4%, respectively.The experiments suggested that inhibition of the (?) -adrenal receptors may be the principal mechanism of the hypotensive action of PAMC. Ganglion blockade, vasodilation as Well as excitation of ?2-adrenal receptors may also be involved.The acuts iv PAMC LD50 in mice were 25.5?5.0 mg/kg.
5.Colorimetric evaluation of dentin is significant for accurate color selection in cast ceramic prostheses

BACKGROUND:VITA 3D Master shade guide is a newly launched colorimetric system in recent years, with a wide clinical prospect.
OBJECTIVE:To evaluate the significance of dentin colorimetric evaluation for accurate color section of cast ceramic prostheses.
METHODS:Using conventional colorimetric assay and conventional colorimetric assay combined with dentin colorimetric assay (combined colorimetric assay), 30 patients were subject to colorimetric evaluation 1/3 to the neck, central part, and cut end of the tooth. In CIE1976L*a*b*color system, a digital SLR camera (Canon D50) was used for colorimetric measurement and analysis of cast ceramic prostheses prepared with two colorimetric methods and teeth with the same name. And a variety of new colorimetric methods were analyzed based on examples.
RESULTS AND CONCLUSION:In the conventional colorimetric group,△L*was 1.22±0.16,△C*was 1.19±0.20,△H*was 0.31±0.05,△E*was 1.32±0.13. in the combined colorimetric group,△L*was 1.03±0.11,△C*was 1.12±0.19,△H*was 0.29±0.03,△E*was 1.23±0.11. Cast ceramic prostheses prepared by conventional colorimetric method were satisfactory in 22 cases, while cast ceramic prostheses prepared by the combined colorimetric method were satisfactory in 23 cases. There was no difference in patient satisfaction for color rendition (P>0.05), but chromatic difference analysis was significantly different between the two groups (P<0.05).
6.Effect of low-frequency transcutaneous electrical nerve stimulation on the action potential of spinal cord posterior horn cells in rats after peripheral nerve injury

BACKGROUND: Up to now, few studies related to the mechanism of low-frequency transcutaneous electrical nerve stimulation (TENS) in relieving pain, and the effect of low-frequency TENS on the activity potential of dorsal horn cells in rats after peripheral nerve injury. OBJECTIVE: To observe the effects of low-frequency TENS on the activity potential of dorsal horn cells induced by mechanical allodynia and thermal allodynia by using animal models of peripheral nerve injury, and observe the efficacy after interfering of naloxone. DESIGN: A randomized control animal study. SETTING: Department of Neurology, Affiliated Hospital of Medical College, Yanbian University. MATERIALS: The experiment was carried out in the central laboratory of Medical College, Yanbian University between February and October 2004. Eighty male Sprague-Dawley rats were used, and 60 random selected ones were operated to separate sciatic nerve, two branch tibial nerves and sural nerves of sciatic nerve were amputated after ligation, and peroneal nerve was left as the experimental group; the other 20 rats were placed at the origin after sciatic nerve was separated, and then the skin was sutured as the control group. METHODS: ① Pain detection (Behavioral test): At 1 week postoperatively, the rats were given mechanical allodynia and thermal allodynia once every 5 seconds for 10 times, and then the frequency of foot withdrawal was detected (0%-40% for mild pain, 40%-70% for moderate pain; 70% and above for severe pain). ② The spontaneous activity potential of dorsal horn cells and that induced by mechanical allodynia and thermal allodynia were detected among the rats with moderate and severe pain in the control group and study group. ③ Low-frequency TENS of 3 mA and 10 Hz was applied to the legs of rats in the experimental group with annular electrode for 10 minute, and the membrane potential of dorsal horn cell was detected before and after stimulation. ④ At the same time of low-frequency TENS was given, rats in the experimental group were injected with naloxone via audal vein, and the membrane potential of dorsal horn cell was detected before and at 10 minutes after naloxone injection.RESULTS: Finally 80 rats were involved in the analysis of results after compensation. ① The foot withdrawal frequencies induced by mechanical allodynia and thermal allodynia in the experimental group were obviously higher than those in the control group (P < 0.01). ② The membrane potential of dorsal horn cell by mechanical allodynia and thermal allodynia in the experimental group were obviously higher than those in the control group (P < 0.01). ③ The membrane potential of dorsal horn cell by mechanical allodynia and thermal allodynia at 10 minutes after low-frequency TENS in the experimental group were obviously higher than those in the control group [(102.6±0.9), (136.9±1.46) impulses per 10 seconds; (175.2±1.28), (240.8±1.51) impulses per 10 seconds, P < 0.01]. ④ In the experimental group, the membrane potential of dorsal horn cell by mechanical allodynia and thermal allodynia at 10 minutes after naloxone injection were obviously higher than those before injection [(174.5±0.4), (235.4±1.4) impulses per 10 seconds, P < 0.01].CONCLUSION: Low-frequency TENS can effectively inhibit the activity potential of spinal dorsal horn cells induced by non-noxious stimulation,and the intravenous injection of naloxone (8 mg/kg) can recover it to the pretreatment level, indicating that low-frequency TENS may alleviate pain by stimulating central nervous system to make it secrete endogenous opium system, and acting on spinal dorsal horn cells to reduce the activity.

8.Effect of ginsenoside(Rg3)combined with delaying tamoxifen resistance in breast cancer

Objective To evaluate the effect and the best concentration of Rg 3 combined with delaying tamoxifen resistance in breast cancer .Methods The tamoxifen-resistant(TAM-R)cell line with TAM was es-tablished.TAM-R cell line and WT -MCF-7 cell line were cultured in TAM at different concentrations and then detected the growth of cells .WT-MCF-7 cell line was cultured medium in TAM which contained Rg 3 at different levels .The growing situation of cells was detected with MTT method .Results Compared with wild -type WT-MCF-7 cells,TAM-R showed higher curves when they were both at TAM treatment .Rg3 could be associated with delaying the emergency of tamoxifen resistance and the best concentration was 1 ×10 -8 mol/L and 5 ×10 -8 mol/L.Conclusion It might be useful to reduce the tamoxifen resistance incidence by clinically com-bined Rg3 with TAM treatment .
9.Novel compounds in Momordica charantia

Objective To study new active constituents in the mature fruits of Momordica charantia.Methods M.charantia was extracted by alcohol and purified by D-101 macroporous adsorptive resin.The isolation and purification were carried out by silica gel and semiprepared HPLC,the compounds were identified and elucidated by spectral and chemical methods.Results Eight compounds were obtained and identified as charantadiol A(5?,19-epoxycucurbita-6,23(E),25(26)-triene-3?,19(R)-diol,Ⅰ),(+)-eduesmin(Ⅱ),bluemenol A(Ⅲ),karatavilagenin D(Ⅳ),5?,19-epoxycucurbita-6,23-diene-3?-19,25-triol(Ⅴ),5,19-epoxy-5?-cucurbita-6,23(E)-diene3?,25-diol(Ⅵ),3?,7?-trihydroxy-cucurbita-5,23(E)-diene-19-al(Ⅶ) and ?-sitosterol(Ⅷ).Conclusion Compound Ⅰ is a novel one,named charantadiol A.Compounds Ⅱ and Ⅲ are obtained from M.charantia for the first time.
10.Survival of engineered myocardium constructed by bone marrow mesenchymal stem cells in vitro

BACKGROUND:So far, engineered myocardium is stil facing many problems. Research has demonstrated that bone marrow mesenchymal stem cells can be differentiated into myocardial cells. Polyglycolide and polycaprolactone are commonly used artificial polymers, which have good biocompatibility.
OBJECTIVE:To observe the growth of the poly(glycolic acid)/poly(e-caprolactone) copolymer patch in vitro in normal myocardium and infarcted myocardium.
METHODS:Bone marrow mesenchymal stem cells in Sprague-Dawley rats were separated using adherent separation and selection method, cultured in vitro. The third passage was labeled with DAPI. The bone marrow mesenchymal stem cellsuspensions (2×106/cm2) were produced and planted on poly(glycolic acid)/poly(e-caprolactone) copolymer scaffolds to form poly(glycolic acid)/poly(e-caprolactone) copolymer patch. After culturing for 48 hours, the specimens were observed under electron microscope, stained with hematoxylin and eosin, and then observed under light microscope. Rat models of myocardial infarction were established by ligating left anterior descending coronary artery. Poly(glycolic acid)/poly(e-caprolactone) copolymer patch was implanted into the normal and infarcted myocardium for 5 weeks. The survival of bone marrow mesenchymal stem cells was determined by the detection of pathology.
RESULTS AND CONCLUSION:Results of light microscope and electron microscope demonstrated that bone marrow mesenchymal stem cells grew three-dimensional y on poly(glycolic acid)/poly(e-caprolactone) copolymer patch. cells and patch were adhesive wel . Under laser confocal microscopy, compared with the first week, bone marrow mesenchymal stem cells were marked by DAPI in the myocardium at the fifth week. There were bone marrow mesenchymal stem cells marked by DAPI in the infracted area. Results of hematoxylin-eosin staining exhibited that bone marrow mesenchymal stem cells were detected in the infarct area. These results suggested that bone marrow mesenchymal stem cells adhered to the poly(glycolic acid)/poly(e-caprolactone) copolymer stent wel . The complexes of poly(glycolic acid)/poly(e-caprolactone) copolymer and bone marrow mesenchymal stem cells can be used for reparation of myocardium.
