Objective:To investigate the inhibitory effects of cytokine induced-killer(CIK) cells combined with docetaxel(DTX) on the expansion of lung adenocarcinoma cell line SPC-A1/DTX in vitro and in vivo.Methods:Peripheral blood mononuclear cells(PBMC) from healthy donors were incubated in vitro to induce CIK cells in the presence of interferon-gamma(IFN-?),IL-2 and anti-CD3 monoclonal antibody.MTT assay was employed to evaluate the cytotoxic activity of DTX,CIK cells and their combination against SPC-A1/DTX cells in vitro.SPC-A1/DTX lung adenocarcinoma cells were injected subcutaneously into nude mice.On the 14 th day,normal saline(control group),docetaxel(DTX 1 mg/kg in 0.2 ml,DTX group),CIK cells(1?107,CIK group),and CIK cells combined with docetaxel(CIK+DTX group) were administered intraperitoneally respectively.All the nude mice were sacrificed at day 15 after treatment and the tumor were weighted out.Results:MTT assay showed that the IC50 to docetaxel in SPC-A1/DTX cells was 110.5 ?g/ml and 8.5 ?g/ml in wild SPC-A1 cells.CIK cells possessed a higher anti-tumor cytotoxic activity in SPC-A1/DTX cells in vitro than in wild SPC-A1 cells(P

Cytokine-induced killer(CIK) cells are heterogeneous lymphocytes generated by incubation of human peripheral blood mononuclear cells with various cytokines in vitro,characterized by both the T lymphocyte's powerful cytotoxic activity and the natural killer(NK) cell's non-major histocompatibility-restricted antitumor activity in vitro and in vivo.CIK cells have their peculiar superiority over standard lymphokine activated killer(LAK) cells,tumor-infiltration lymphocytes(TIL) and anti-CD3 antibody induced activated killer(CD3AK) cells in application to the cancer adoptive immunotherapy and have hence received wide attention from researchers.This article updates recent researches on CIK cells,their interaction with dendritic cells,bispecific antibodies and oncolytic viruses,as well as their use in gene transfer.
3.Clinical Analysis of Diabetes Mellitus Complicating with Pneumonia in Old People.

Objective To investigate the clinical features of diabetes mellitus complicating with pneumonia in old people,and to improve the diagnose and treatment tactics.Methods In this retrospective analysis,the symptom of respiratory tract,body temperature,hemogram,radiologic characteristics,etiologic characteristics,the use of antibiotic and the course of disease of 30 old patients T2DM complicated by pneumonia and 34 the aged with pneumonia were compared.Results There were statistically significant differences in the use of drug and the course of disease among the two intervention groups.By contrast,the incidence of the symptom of respiratory tract,body temperature,hemogram,main etiologic agents,and the radiologic characteristics,such as empyema did not show significant differences in this investigation.Conclusions The keys to treating diabetes mellitus complicated by pneumonia are the severe control of blood glucose,application of two or more than two effective antibiotics.And the former,s course is longer.

ObjiectiveTo investigate the clinical efficacy of Chinese medicineShuxin-Huoxue decoction on coronary heart disease and angina pectoris.MethodsThis study was a randomized controlled trial. 86 patients with coronary heart disease and angina pectoris and conformed to the indusive criteria were randomly divided into a control group and a treatment group, with 43 cases in each group. The control group was treated with conventional western medicine, the treatment group was treated with traditional Chinese medicineShuxin-Huoxue decoction. Serum homocysteine (Hcy) was measured by enzymatic cycling assay, high sensitive C reactive protein (hs-CRP) was measured by immunotubidimetric assay and left ventricular ejection fraction (LVEF), stroke volume (SV), cardiac output (CO), cardiac index (CI) were measured by color doppler echocardiography before and after treatment. The clinical effect of the two groups was evaluated.ResultsThe total effective rate of the treatment group and the control group was 90.7% (39/43) and 74.4% (32/43) respectively,there was significant difference (χ2=3.957,P=0.047). Followed up for 4 weeks, LVEF(65.7% ± 8.7% vs. 61.5% ± 8.4%,t=2.277), SV(86.9 ± 16.8 ml vs.79.2 ± 16.5 ml,t=2.144), CO(49.3 ± 9.1 ml/m2 vs. 44.7 ± 8.8 ml/m2,t=2.383), CI (3.7 ± 0.9 L/min?m2 vs. 3.3 ± 0.7 L/min?m2,t=2.301) of the treatment group were significantly improved after treatment than those of the control group (P<0.05), and the decrease of serum Hcy (8.7± 3.7mmol/L vs. 11.4± 3.5mmol/L,t=3.476), hs-CRP (2.7± 1.1 mg/L vs. 3.5± 1.3 mg/L,t=3.081) level of the treatment group were better than those of the control group(P<0.01).ConclusionTraditional Chinese medicineShuxin-Huoxue decoction has good therapeutic effect on coronary heart disease and angina pectoris.
6.The cross-sectional study and health management evaluation on chronic diseases in the city zone of Shaoyang

Objectives To analysis the prevalence and major risk factors of common chronic diseases among adult residents in the city zone of Shaoyang. To formulate and evaluate the measures of health intervention. Methods Using multistage stratified random sampling, 5267 residents aged 18 or above from 26 communities of Shaoyang were invited to participate in the survey, which was 1.24% of Shaoyang municipal residents. Questionnaire, physical examination, biochemistry test and B-ultrasonography on the liver and cholecystis were undertaken. Health intervention and evaluation was undertaken too. Results The prevalence of hypertension, fatty liver, cholelithiasis, high blood sugar, dystipidemia and overweight obesity was respectively 22.0% , 24.2%, 7.4%, 8.2%, 43.7%, 47.6%. The prevalence of hypertension, high blood sugar, dystipidemia and overweight obesity was down respectively significatively to 19.6%, 5.0%,35.6% and41.1% after intervention(P<0.05). Conclusions There is a high prevalence of common chronic diseases in the city zone of Shaoyang, which can be effectively reduced through health management programme.
7.The application of preoperative ultrasonography in small incision appendectomy

Objective To investigate the value of ultrasound locates the appendectomy and guide appendec -tomy incision in small incision appendectomy .Methods According to the digital table ,80 patients with acute appen-dicitis diagnosed by B ultrasound examination , were randomly divided into the observation group , and the control group.Each of the group has 40 cases and use small incision appendectomy .In the observation group , mark corre-sponding appendix root in the abdominal wall surface when use B ultrasound examination ,and mark again in anesthe-sia before surgery.Design of a 2~2.5cm minimal incision according to appendix after anesthesia marker ,distance measurement before and after .The control group did not use B ultrasound localization ,minimal incision by McBurney point or near tenderness point .Between the two groups in operation time ,bleeding volume ,postoperative exhaust time , hospitalization time,complications.Results Ultrasound before and after positioning distance was (1.9 ±0.6)cm,and the preoperative immediate ultrasound localization of appendix and intraoperative exploration results ,and preoperative ultrasound localization in observation group ,the average operation time was better than no ultrasound localization of group(P<0.05).The amount of bleeding volume,postoperative exhaust time,hospitalization time,complications had no obvious difference .Conclusion Preoperative immediate ultrasound can more accurately reflect the position of ap-pendix ,favorable operation in small incision appendectomy is quickly ,shorten the operation time .

Objective: To investigate the effect of rat hyperplasia suppressor gene-1(rHSG-1) on the proliferation of aortic vascular smooth muscle cells(VSMCs) from spontaneously hypertensive rats(SHR). Methods:VSMCs were transfected with an adenoviral vector expressing rHSG-1(Ad5rHSG-1). The effect of rHSG-1 on the proliferation of VSMCs was investigated by cell counting, MTT assay and 3H-thymidine incorporation. We also analyzed the cell-cycle using flow cytometry and detected the expression of p27 Kip1 and p21 Cip1 by Western Blot. Objective To evaluate the cardioprotective effect of trimetazidine in elderly patients with myocardial ischemia and chronic heart failure of coronary artery disease. Methods Eighty nine patients with angiographically proven coronary artery disease were randomized to therapy group (n=45) and controlled group (n=44). The patients in therapy group were given both the routine treatment and trimetazidine 20 mg, 3 times daily, for 6 months. The patients in controlled group were given only the routine treatment. Results After the therapy, the episode of angina pectoris every week was fewer and weekly consumption of nitroglycerin tablets was less in patients of therapy group than those in controlled group: (3.0?1.7 )episodes/w vs (3.9?2.1 )episodes/w,P0.05). 10.Hypertriglyceridemia and insulin resistance in the obese elderly

Objective To study the changes of circulating triglyceride (TG) in the obese elderly, and to investigate the effect of hypertriglyceridemia on the development of insulin resistance. Methods A total of 82 subjects were divided into simple obesity group, obesity with IGT group, obesity with T2DM group and normal control group. The body height, body weight, blood pressure, fasting plasma glucose (FPG) and insulin(FPI), circulating TG and total cholesterol (TC) were measured. The homeostasis model assessment-insulin resistance (HOMA-IR) was calculated. Results The levels of serum TG were significantly higher in the simple obesity group ( 1.3 ± 0. 6) mmol/L, obesity with IGT group (1.9 ± 0.9)mmol/L and obesity with T2DM group (2.1±0.7) mmol/L than in the normal controls [(1.0±0.2)mmol/L, all P<0. 05], Furthermore, the levels of TG, FPG, FPI, HOMA-IR and systolic pressure were significantly higher(all P<0. 05) in obesity with IGT group and obesity with T2DM group than in simple obesity group. There were significantly positive correlations between TG and body mass index (BMI), FPG, HOMA-IR, TC, systolic pressure respectively (all P < 0.05). Multiple linear regression analysis indicated that FPG and HOMA-IR were the independent factors affecting TG (both P < 0. 05 ). Conclusions Hypertriglyceridemia may play an important role in the development of insulin resistance.
