Objective To investigate the expression of P21ras and PIK3CA proteins in hepatitis B virus-related hepatocellular carcinoma(HBV-HCC), post-hepatitis B hepatic cirrhosis (HBV-hepatic cirrhosis)and normal hepatic tissues specimen, and their correlation between HBV-HCC and HBV-hepatic cirrhosis tissues. Methods Using immunohistochemistry, the expression of P21ras and PIK3CA proteins in 34 cases of HBV-HCC, 37 cases of HBV-hepatic cirrhosis and 30 cases of normal liver tissues specimen were detected and compared. Results The mean gray scales of P21ras protein in HBV-HCC, HBV-hepatic cirrhosis and normal hepatic tissue specimen were 138.86 ± 2.9, 145. 34 ± 2.06 and 152.07 ± 1.17 (P < 0. 0l), respectively, and were related to the progression of hepatopathy (P <0.01). The mean gray scales of PIK3CA protein in HBV-HCC, HBV-hepatic cirrhosis and normal hepatic tissue specimen were 138.20 ± 3. 14, 149.49 ±0. 78 and 154.71 ± 1.29 (P < 0.01), respectively, and were related to the progression of hepatopathy (P < 0. 01).There were apparent correlation between P21ras and PIK3CA in HBV-HCC and HBV-hepatic cirrhosis respectively (r =0. 64, P <0. 05; r =0. 42, P <0. 05). Conclusion The overexpression of P21ras and PIK3CA in HCC and hepatic cirrhosis tissue suggests that they participate in the tumorigenesis and development of hepatocellular carcinoma and hepatic cirrhosis, and there may be a signal transduction pathway of P21ras-PI3K in HBV-HCC and HBV-hepatic cirrhosis.

Objective To investigate the effects of aerobic exercise on tau phosphorylation and PI3K/ Akt pathway in the hippocampus of obese rats, and provide some theoretical basis for physical activity improving obesity-related neurological disorders. Methods Male Sprague-Dawley rats aged 3 weeks were randomly assigned to either a high-fat or a normal diet protocol for 12 weeks.Animals submitted to the high-fat diet were further divided into two groups:a sedentary group (HF-Sed) and an exercise group (HF-Ex).The rats fed the normal diet were also divided into a sedentary group (ND-Sed)and an exercise group (ND-Ex).The rats in the HF-Ex and ND-Ex groups underwent a treadmill training for 8 weeks. Then the hippocampus was isolated at 48h after the last exercise. The protein and phosphorylation levels of tau, glycogen synthase kinase 3β (GSK3β),phosphoinositide 3-kinase (PI3K) and Akt were assayed using Western blotting. Results After 12 weeks of feeding,55% of rats in the high-fat diet group reached the conditions for the obesity model. After 8 weeks of treadmill exercises,in the HF-Sed group the phosphorylation level of tau was significanlty higher than that in the ND-Sed group, while in the HF-Ex group that was significantly lower than the HF-Sed group. Moreover,in the HF-Sed group the phosphorylation level of GSK3β Ser9 was significantly lower, and the phosphorylation level of GSK3β Tyr216 was significantly higher than the ND-Sed group, indicating the activity of GSK3β was significantly higher than the ND-Sed group. However, after 8 weeks of treadmill exercise,in the HF-Ex group the phosphorylation level of GSK3β Ser9 was signfiicantly higher, and the phosphorylation level of GSK3β Tyr216 was signficanlty lower than the HF-Sed group, indicating significantly lower activity of GSK3β than the HF-Sed group. Then,in the HF-Sed group the protein levels of PI3K p110 and p85 subunits, and the phosphorylation levels of Akt Thr308 and Ser473 were significantly lower than those in the ND-Sed group, indicating inhibited activity of the PI3K/Akt pathway. However,in the HF-Ex group the protein levels of PI3K p110 and p85 subunits and the phosphorylation levels of Akt Thr308 and Ser473 were significanlty higher than those in the HF-Sed group, showing the activity of PI3K/Akt pathway was enhanced. Conclusion Obesity induces tau hyperphosphorylation in the rats hippocampus and long-term aerobic exercises can reduce tau hyperphosphorylation by increasing PI3K/Akt pathway activity and inhibiting GSK3β activity. It has a positive effect on delaying the formation of neurofibrillary tangles and improving obesity-related neurological disorders.

Objective To observe the effect of 8-week climbing ladder exercise with load on the expression of proliferator-activated receptor gamma coactivator 1-alpha (PGC1α)/ fibronectin type Ⅲ domain containing 5 (FNDC5)/ myostatin (MSTN)in soleus and extensor digitorum longus muscle and the serum concentration of irisin in Sprague-Dawley (SD)rats. Methods Twenty SD rats were randomly divided into a control (Con,n=10)group and a climbing ladder (Lad,n=10)group.The Lad group took an 8-week and once-every-3-day program of resistance exercise using an improved model of climbing ladder with load. Then the soleus and extensor digitorum longus muscles were isolated at 48 h after exercise. The PGC1α/FNDC5/MSTN mRNA and protein expression in the skeleton muscle were detected using quantitative PCR and Western blotting respectively. The serum concentration of irisin was measured using enzyme-linked immunosorbent assay. Results Compared with Con group, the expression of PGC1α in the soleus muscle reduced significantly (P<0.05),that of FNDC5 reduced but without significance and that of MSTN increased significantly (P<0.05)in Lad group. However, the expression of PGC1α in the extensor digitorum longus muscle increased significantly (P<0.05),that of FNDC5 increased but without significance and that of MSTN decreased significantly (P<0.05)in Lad group compared with Con group. There were no significant differences in the serum concentration of irisin between the two groups. Conclusion Eight-week climbing ladder exercise with load on rats induces differential expression of PGC1α/FNDC5/MSTN in soleus and extensor digitorum longus muscle, but doesn't influence the serum concentration of irisin. It is suggested that there may be a complex mechanism of balancing and antagonism in the body after long-term resistance exercises.

OBJECTIVETo provide the basis for standardization cultivation of medicinal Chrysanthemum morifolium, the relationship between the seedling grading and plant growth, yield and quality were studied.
METHODThe morphological index of the seedlings was measured and the method of principal component and correlation analysis were used to determine the grading index, and step-wise cluster analysis was applied for clustering analysis. Pot experiments were used to measure the indicators of plant growth and development, yield and quality.
RESULTThe height and ground diameter were determined for the quality indicators of the seedlings grading, and the standard quality grading of seedlings of Ch. morifolium was initially set up. The ground diameter of the class I and II were larger than that of the class III, and the number of branches of class I and II was more than that of the class III, on the contrary, the plant height of the class III was higher than that of the class I and II. The shape and appearance of the plant had no effect on the intrinsic quality. Flower center diameter and tubular floret number of the class I and II were significantly larger than those of the class III, so as the yield. The seedling grading had no obvious effect on the internal quality of medicinal material.
CONCLUSIONSeedlings of the class I and class II were suitable for transplanting.

Objective To explore the correlation between the glucose variability and the severity of acute isolated traumatic brain injury(TBI). Method According to the inclusion/exclusion criteria,a total of 125 cases of acute isolated TBI admitted in Department of Neurosurgery of China Medical University from July 2012 to June 2015 were included. According to Glasgow coma scale(GSC),the patients were divided into five groups including control(GCS 15),mild(GCS 13?14),moderate(GCS 9?12),severe(GCS 6?8),and extra?severe(GCS 3?5)groups. Blood glucose control(including relief of the stress and the application of insulin)were carried out immediately. The average, standard deviation, and variation co?efficient of blood glucose of all groups were recorded at admission,48 hours and 3?7 days after hospitalization. The clinical records and glycemic in?dex were compared among different groups and during different periods,so as to analyze the relationship of the variability of glucose and the duration of hyperglycemia with the severity of TBI and the effects of glycemic intensive care management. Results The results of Kruskal Wallis test and Mann?Whitney Utest showed that the average, standard deviation, and variation coefficient of glucose in the extra?severe group and the severe group were statistically higher than those in the control group(P<0.05)during the same period. Meanwhile, the average, standard deviation, and variation coefficient of glucose at admission,48 hours and 3?7 days after hospitalization were also different among each group(P<0.05). The duration of hy?perglycemia and conscious disturbance in both the extra?severe group and the severe group were longer than those in the control(P<0.05). The analyses using rank correlation indicated that glucose variability, the level and duration of hyperglycemia were positively correlated with the severity of TBI(r>1). Conclusion The glucose variability in acute isolated TBI patients could be considered as the index of the severity of TBI.
