1.Expression and clinical significance of PTEN in bladde r transitional carcinoma

Objective Investigates expression of PTEN(Phosphatase and tensin homology deleted on chromosome ten , PTEN) in bladder transitional cell carcinoma (BTCC) and its clinical significance.Methods Fifty-one cases of bladder transitional carcino -ma, forty-two cases of Cystitis Glandularis(CG) and ten cases of normal bladder tissue 10 cases of normal bladder tissue were collect -ed.All samples were derived from cystoscopic biopsy and were subjected to histopathological examination to confirm the diagnosis .Im-munohistochemistry S-P methods is used to demonstrate the expression of PTEN .Results The positive rate of PTEN expression in BTCC, CG and normal bladder tissue are 58.8%, 81% 100%,and there is statistic significance ( P <0.01) among three groups.51 cases of BTCC are divided into group G Ⅰ ,GⅡ ,GⅢ and Tis -T1 ,T2-T4 ,and there is statistic significance among group G Ⅰ and GⅡ, GⅢ ( P <0.01),between group Tis -T1 and T2-T4 ( P <0.05).Conclusions The reduced expression or loss of PETN might play an important role in carcinogenesis and progression of bladder cancer .The positive rate of PTEN expression was negatively correlated to the clinical and grade staging of tumor .
2.Relationship between seedling grade and plant growth, yield and quality of medicinal Chrysanthemum morifolium.

OBJECTIVETo provide the basis for standardization cultivation of medicinal Chrysanthemum morifolium, the relationship between the seedling grading and plant growth, yield and quality were studied.
METHODThe morphological index of the seedlings was measured and the method of principal component and correlation analysis were used to determine the grading index, and step-wise cluster analysis was applied for clustering analysis. Pot experiments were used to measure the indicators of plant growth and development, yield and quality.
RESULTThe height and ground diameter were determined for the quality indicators of the seedlings grading, and the standard quality grading of seedlings of Ch. morifolium was initially set up. The ground diameter of the class I and II were larger than that of the class III, and the number of branches of class I and II was more than that of the class III, on the contrary, the plant height of the class III was higher than that of the class I and II. The shape and appearance of the plant had no effect on the intrinsic quality. Flower center diameter and tubular floret number of the class I and II were significantly larger than those of the class III, so as the yield. The seedling grading had no obvious effect on the internal quality of medicinal material.
CONCLUSIONSeedlings of the class I and class II were suitable for transplanting.

3.Investigation of the compliance and its impact factors of amblyopic children treating with visual perceptual learning

Objective To investigate the compliance and its impact factors of amblyopic children treating with visual perceptual learning.Methods 116 amblyopic children treating with visual perceptual learning were investigated using self-made compliance questionnaire for amblyopic children treating with visual perceptual learning.Results The average compliance score of amblyopia in treatment of visual perceptual learning was(71.45±10.10) points,ranging from 36 to 92 points.The univariate linear regression analysis showed thatawareness of parents on the amblyopia training,communication with medical staff,parents supervise children training and eyesight improvement speed of children were the statistical significant impact factors.The further multivariate linear regression analysis showed that awareness of parents on the amblyopia training and parents supervise children training were also the statistically significant impact factors.Conclusions The compliance of amblyopic children treating with visual perceptual learning was satisfactory and its impact factors were associated with parents,the supervision and awareness of parents were the key impact factors for amblyopic children treating with visual perceptual learning.
4.Preliminary research of 1 H-MRS for moderate and severe OSAHS patients before and after CPAP treatment

Objective To investigate the variation of multivoxel 1 H magnetic resonance spectroscopy(1 H-MRS)before and after continuous positive airway pressure(CPAP)treatment in obstructive sleep apnea hypopnea syndrome(OSAHS).Methods Brain multivoxel 1 H-MRS examinations were performed in 25 cases of moderate or severe OSAHS patients before and after CPAP treat-ment,and 25 cases of healthy.The ratios of brain metabolites of the frontal lobe were recorded and analyzed respectively.To observe whether the lactate(Lac)peak appeared or not.Results In the frontal lobe,the NAA/Cr,NAA/Cho of the patients before treatment (2.021 2±0.231 2 and 1.608 8±0.257 1,respectively)was decreased compared with the healthy (2.726 8±0.607 1 and 2.445 6± 0.437 5).The NAA/Cr,NAA/Cho of the patients after treatment (2.314 0±0.312 8 and 2.01 6 4±0.424 0,respectively)was in-creased compared with the patients before treatment (2.021 2±0.231 2 and 1.608 8±0.257 1).The NAA/Cr,NAA/Cho of the pa-tients after treatment (2.314 0±0.312 8 and 2.01 6 4±0.424 0,respectively)was decreased compared with the healthy (2.726 8± 0.607 1 and 2.445 6±0.437 5),and the difierences were statistically significant (P <0.01).The Cho/Cr of the patients before treatment (1.293 2±0.261 5)was increased compared with the healthy (1.129 2±0.157 7),the difference was statistically significant(P <0.05).Lac peak was not detected in all.Conclusion Multivoxel 1 H-MRS can demonstrate sensitively the changes of brain metabolism in pa-tients with OSAHS before and after CPAP treatment,and may provide imaging evidence for clinical therapeutic effect and prognostic evaluation.
5.Epidemiological analysis of 5 577 cases of primary liver cancer in Xinjiang

Objective To analyze the clinical epidemiology of primary hepatocellular carcinoma (PHC) in the Xinjiang region. Methods Clinical data of the patients with PHC were collected at First Affiliated Hospital of Xinjiang Medical University from 5 577 cases from January 2002 to December 2014, their gender, race, age, household distribution, hepatitis virus-positive rate were analyzed retrospectively. Results Among the 5 577 eases, the men/women gender ratio was 3.45∶1;the proportion of Han, Uighur, Kazakh, and other ethnic groups (Hui, Mongolian, Manchu, Xibo) was 79.67%, 9.86%, 4.55%, 3.31%and 2.61%, respectively. The Constituent ratio difference between Uighur and Han was significant (P<0.05);4 232 patients had hepatitis B surface antigen (HBsAg) detection, and 3 833 patients had HCV antibody (HCV-Ab) detection. HBsAg was positive in 2 560 cases (60.49%), HCV-Ab was positive in 490 cases (12.78%). Hepatitis B virus detection positive rate in Uygur was 35.52%, in Kazak was 40.00%, which was lower than the Han's (65.68%, P<0.05). Urban and rural population had 3589 cases (64.35%) and 1988 cases (35.65%). Conclusion An increased risk for PHC was found in hepatitis virus-positive patients, the Xinjiang Uygur and Kazak people had significantly lower prevalence of HBV infection than the Han's. Appropriate measures should be taken for clinical diagnosis, treatment and prevention of PHC.

7.Rapid identification system for seedlings of medicinal Chrysanthemum morifolium.

OBJECTIVETo achieve the rapid identification for seedlings of medicinal Chrysanthemum morifolium, the discriminant equation was established and the software for rapid identification was designed.
METHODLeaf structure of medicinal Chrysanthemum of 12 cultivars was analyzed to establish the discriminant equation based on variance analysis and discriminant analysis. On this basis, the identification program and software (based on the python language) were designed.
RESULTThrough the analysis of variance and multiple comparisons for the 11 leaf parameter index data of 12 different cultivars, it was found that that the leaf parameters were significant different from each other and reached significant levels. The discriminant equation and the rapid identification software were set up based on the analysis of various indicators.
CONCLUSIONThe rapid identification system of seedlings of medicinal Chrysanthemum could be achieved through the establishment of discriminant equation combined with computer technology.

8.Numerical analysis of morphological variation of germplasm resources of Chrysanthemum morifolium for medicine.

OBJECTIVEBotanical characters of germplasm resources of Chrysanthemum morifolium for medicine were observed and compared, which could offer reference for its genetic improvement and germplasm resources protection.
METHODBased on the random blocks field experiments design, twenty-six morphological traits were observed. The morphological differences among germplasm resources were compared by principal component analysis and cluster analysis.
RESULTThe coefficient of variation values for 17 of 26 traits indicated a high level of variation (above 20%). Six principal components which accounted for 77.14% of total variance were extracted from the principal component analysis. The 29 germplasm resources could be divided into two clusters.
CONCLUSIONThere were large morphological variation among germplasm resources on Ch. morifolium for medicine.

9.Targeted nano-vector for gene delivery into human bone marrow mesenchymal stem cells and cellular MR imaging in vitro

Objective To explore the feasibility and efficacy of an MRI-visible,targeted,nano-vector which is synthesized by attaching a targeting ligand,the GD2 single chain antibody (scAb GD2),to the distal ends of PEG-g-PEI-SPION as a carrier for gene delivery into human bone marrow mesenchymal stem cells (hBMSCs) and in vitro cellular MR imaging.Methods scAbGD2-PEG-g-PEI-SPION was synthesized as previously reported.Gel electrophoresis was performed to assess the pDNA condensation ability of scAbGD2-PEG-g-PEI-SPION.The particle size and Zeta potential of scAbGD2-PEG-g-PEI-SPION/pDNA nanocomplexes were observed by dynamic light scattering.Cytotoxicity of scAbGD2-PEG-g-PEI-SPI-ON was evaluated by CCK-8 assay using hBMSCs.Gene transfection efficiency of scAbGD2-PEG-g-PEI-SPION in hBMSCs was quantified by flow cytometry,PEG-g-PEI-SPION,scAbGD2-PEG-g-PEI-SPION,scAbGD2-PEG-g-PEI-SPION+ free AbGD2 and scAbIgG2a-PEG-g-PEI-SPION group was established.The cellular internalization of scAbGD2-PEG-g-PEI-SPION/pDNA nanocomplexes was observed by confocal laser scanning microscopy and Prussian blue staining.MRI of scAbGD2-PEG-g-PEI-SPION was performed by cellular MRI scanning in vitro.Results scAbGD2-PEG-g-PEI-SPION condensed pDNA to form stable nanocomplexes of 80-100 nm in diameter and showed low cytotoxicity to hBMSCs.At the same N/P ratio,the transfection efficiency of scAbGD2-PEG-g-PEI-SPION group was significantly higher than those of other groups (P<0.001).At the optimal N/P ratio of 20,scAbGD2-PEG-g-PEI-SPION/pDNA obtained the highest transfection efficiency of (59.60 ± 4.50) % in hBMSCs.Furthermore,hBMSCs labeled with scAbGD2-PEG-g-PEI-SPION showed sensitive low signal intensity on MRI T2/T2 *-weighted images in vitro.Conclusion scAbGD2-PEG-g-PEI-SPION is an efficient MRL visible targeted nano vector for gene delivery into hBMSCs.
10.Effects of radiation on echinococcus in vitro culture and its mRNA expression of Gadd45α

Objective:To investigate the killing effect of radiation on echinococcus in vitro culture and its effect on the mRNA expression of growth arrest and DNA damage 45 alpha (Gadd45α) gene. Methods:Echinococcus from naturally infected sheep liver was cultured in vitro and divided into 7 groups. The echinococcus was irradiated with 6 MeV at doses of 0 (control), 20, 40, 60, 80, 100, and 120 Gy, respectively. The growth of echinococcus was observed under light microscope at 1, 3, 5 and 7 d after the radiation. The expression of Gadd45α mRNA in control, 20, 40 and 60 Gy groups of echinococcus was detected by RT-PCR technique at 7 d after the radiation. Results:The disintegration and exfoliation of echinococcus were observed under light microscope at 1, 3, 5 and 7 d after the radiation, and the death rate of echinococcus was positively correlated with the radiation dosages ( r = 0.81, P < 0.05). After the radiation at 7 d, compared with the control group (100.00 ± 0.00), the mRNA expression levels of Gadd45α in echinococcus of 20, 40, and 60 Gy groups were significantly increased (279.74 ± 80.08, 759.38 ± 160.98, 1 666.68 ± 316.36, P < 0.01), and it was positively correlated with the radiation dosages ( r = 0.93, P < 0.01). Conclusion:Radiation has a certain killing effect on echinococcus cultured in vitro, and there is a certain dose-effect relationship with the radiation dosages, and Gadd45α gene may be involved in the molecular mechanism of radiation-induced killing of echinococcus in vitro.
