Objective To get a new approach to conserve the germplasm of Platycodon grandiflorum. Methods A method of vitrification was studied to cryopreserve in vitro shoot-tips of P. grandiflorum, and the regenerated plantlets were observed subsequently. Results Shoot-tips, 2—3 mm length, gotten from in vitro shoots of P. grandiflorum precultured on MS medium supplemented with 5% DMSO and 103 g/L sucrose for 3 d, were loaded with the 60% PVS_2 for 20 min at 20 ℃, and incubated in PVS_2 for 90 min at 0 ℃ prior to a direct plunging into liquid nitrogen (LN) and keeping for 1 d. After rapid thawing in water at 40 ℃, the shoot-tips were rinsed in the MS medium supplemented with 410 g/L sucrose for 20 min, and plated on the filter paper sustained by the MS regeneration medium supplemented with 0.6 mg/L KT, 0.2 mg/L BA, and 0.05 mg/L NAA for 1 d in dark and subcultured on the above regeneration medium for one week in dark prior to exposure to the light. The survival of shoot-tips was up to 80%, and they grew normally. Conclusion The method of vitrification to cryopreserve the germplasm of P. grandiflorum is simple in handling with high in survival and normal in regeneration and can be applied in practice.

Objective To investigate the effectiveness of TACE in combination with percutaneous microwave coagulation therapy(PMCT) in treatment of hepatocellular carcinoma.Methods 82 cases of hepatocellular carcinoma confirmed by pathology were randomly divided into 2 groups by odd or even hospitalization number.A total of 40 patients were enrolled in the TACE only group(group A) and 42 patients were enrolled in the synthetic therapy group(group B,underwent treatment by TACE in combination with PMCT).Results The survival rates in 1,2 and 3 years were 70%,54% and 20%,respectively,with a median survival of 1.72 years in group A,and 88%,76% and 51%,respectively,with a median survival of 2.1 years in group B.The survival rates in group B were significantly higher than that in group A(P

Objective To study the appropriate neck irradiation volume for neck lymph nodenegative nasopharyngeal carcinoma patients by Meta-analysis.Methods The related references published from Jan 1990 to Jun 2013 were searched in Pubmed,Embase,Cochrane and Wangfang Databases.The qualified references for enrollment criteria were screened and the relative data were collected.The OR (Odds ratio) value by fixed effect model was used to evaluate the data for patients who received radiotherapy between the upper neck and whole neck irradiation.The endpoints including 5-year neck local control rate (LCR),neck-in-irradiation LCR,and neck-outsides-irradiation LCR.Results Five qualified references were screened and belonged to respective research.There enrolled 1 333 patients,including 970 patients who received the upper neck irradiation and 363 patients who received the whole neck irradiation,respectively.The forest plots revealed that there were no significant differences for 5-year neck LCR,neckin-irradiation LCR,and neck-outsides-irradiation LCR for patients received radiotherapy between the upper neck and whole neck irradiation.These OR values were 0.89 (95％ CI:0.41-1.94),1.29 (95％ CI:0.58-2.88) and 0.42 (95％ CI:0.07-2.36),respectively.Conclusions The Meta-analysis results suggest that irradiation to the upper neck for neck lymph node-negative nasopharyngeal carcinoma can be appropriate.

BACKGROUND:Studies have confirmed that the surface treatment methods such as silane coupling agent and sand blasting could improve bonding strength of zirconia ceramics and resin binder. In addition, the increase of micro cracks on the surface of zirconia ceramics can also improve the bonding strength of zirconia ceramics and resin binder. But there is a lack of related studies addressing whether repeatedly sintering would have an impact on shear bond strength of zirconia ceramics.
OBJECTIVE:To determine the effect of decorative porcelain temperature firing on the shear bond strength between dental zirconia ceramics and resin binder.
METHODS:Twenty pieces of zirconia ceramics specimens were selected from 40 tablets, and then randomly divided into five groups according to the number of sintered times:control group (without sintering), sintering groups for 2, 4, 6, 8 times. Heat starting temperature was 500 ℃, and final temperature was 1 000 ℃ with a heating rate of 55 ℃/min. The vacuum time was 7 minutes. During the sintering, the final temperature was constant. Unsintered ceramic pieces were bonded using resin binder. Universal testing machine was used to test the shear strength of the interface between the two pieces of zirconia ceramics. The interface after shearing was characterized by scanning electron microscope.
RESULTS AND CONCLUSION:The shear strength of sintering groups for 4, 6, 8 times was significantly higher than that of the control group (P<0.05). Shear strength of specimens sintered twice was slightly higher than that of specimens without sintering, but the difference was not significant (P>0.05). Shear strength of specimens sintered 8 times was slightly higher than that sintered 4 and 6 times (P<0.05). Under the scanning electron microscope, there was no crack on the surface of non-sintered zirconia ceramics. After 2 times sintering, the surface appeared to have fine cracks. After sintering 4 times, visible cracks were increased. After six rounds of sintering, the surface began to have significant changes;cracks rose along with slight gaps and a smal amount of adhesive residues were found. After sintering for 8 times, cracks and voids were obviously increased on the sintered surface and adhesive residues were found. It indicated that after sintering for 4, 6 and 8 times, zirconia ceramics could have a good shear strength binding to resin adhesives;with the increasing of sintering times, the shear bond strength could be increased.

ObjectiveTo investigate the effect of basic fibroblast growth factor-polyactide release nanospheres on proliferation and adipogenic induction of adipose-derived stem cells in vitro.MethodsAdipose-derived stem cells were isolated and induced for three-line differentiation in vitro.The culture medium and inductive medium of stem cells were prepared containing 0,1,2,3,4 and 5 mg/ml basic fibroblast growth factor-polyactide release nanospheres,respectively.Adipose-derived stem cells were cultured ina 96-well plate and replaced the culture medium containing release nanospheres the second day.The cells proliferation was detected by the method of MTT every day and quanti fication of oil red O every other day.The data obtained were analyzed with SPSS13.0 statistical software.ResultsThe basic fibroblast growth factor polyactide release nanospheres had the ability promoting proliferation and adipogenic induction of adipose-derived stem cells.The best concentration of nanospheres was 3 mg/ml and 4 mg/ml,respectively.ConclusionsThe basic fibroblast growth factor-polyactide release nanospheres could promote proliferation and adipogenic induetionof adipose-derived stem cells significantly.It could be used as an ideal cytokine release nanospheres in adipose tissue engineering.

Objective To investigate the interventional therapy for the complication in the treatment of advanced primary hepatocellular carcinoma.Methods 62 cases of primary hepatocellular carcinoma confirmed by pathology,imagings and AFP,were randomly divided into 2 groups according to odd or even hospitalization number.32 patients were enrolled in the TACE group(control group)and 30 patients were enrolled in the combination therapy group(TACE + other kinds of therapy).Patients in the combination therapy group underwent the TACE procedure adding with another kind of therapy for treating the complication such as arterial,venous,portal vein cancer embolism,or the inferior vena cava tumor embolisms and may further include ablation of residual cancer,jaundice,hepatic abscess or biloma.Results 1,2,3-years survival rates and median survival rates of the control group and combination therapy group were 68%,50%,19% and 87%,75%,48%;1.5 years and 2 years,respectively.Survival rate and duration in the combination therapy group was significantly greater than those of control group(P

Objective To assess the differences in delineation of organs at risk (OAR) and dosimetry between junior and senior physicians during intensity-modulated radiation therapy (IMRT) for nasopharyngeal carcinoma (NPC) and to evaluate the role of specific training in reducing the differences.Methods Sixteen patients newly diagnosed with NPC were selected in the study.The OAR was delineated separately by three junior physicians and three senior physicians,and the geometric and dosimetric differences were assessed relative to the reference OAR.Delineation was performed again for the two OARs with the biggest difference after specific training in the two groups of physicians,and the differences were evaluated again.The difference was determined by paired t test.Results The maximum dose differences (Dmax) of OAR in the junior and senior physicians were (2.33 ± 12.06) ％ (-48.06％-137.82％) and (0.09 ± 4.72) ％ (-49.54％-42.96％),respectively (P =0.039),and the difference in the optic chiasm was the greatest ((5.85 ± 19.63) ％ ∶ (1.36 ± 4.64) ％,P =0.042).The mean dose differences (Dmean) of OAR in the junior and senior physicians were (3.10 ± 8.07)％ (-46.76％-59.76％) and (-0.93 ± 2.03) ％ (-45.54％-35.69％),respectively (P =0.021),and the difference in the parotid gland was the greatest ((13.23 ± 13.39) ％ ∶ (3.20 ± 6.71) ％,P =0.002).In the secondary delineation after training,the Dmax of the optic chiasm in the junior and senior physicians was (1.68 ± 3.34)％ and (1.50 ± 1.87) ％,respectively (P =0.841),and the difference in junior physicians was reduced significantly compared with before training ((1.68 ± 3.34) ％ ∶ (5.85 ± 19.63) ％,P =0.048) ; the Dmean of the parotid gland in the junior and senior physicians was (2.46 ± 3.06) ％ and (1.35 ± 3.00) ％,respectively (P =0.2 7 4),significantly reduced compared with before training ((2.46 ± 3.0 6) ％ ∶ (13.23 ± 13.39)％,P=0.002; (1.35 ± 3.00)％ ∶ (3.20 ± 6.71) ％,P =0.033).Conclusions The differences in delineation of OAR lead to dose uncertainties during IMRT for NPC,and specific training can improve the accuracy of delineation.

Objective To investigate the distribution and antimicrobial resistance profile of clinical gram-negative bacterial isolates in the First Afifliated Hospital of Nanjing Medical University during 2014.Methods Bacteria identiifcation was performed by API system or the VITEK-2 Compact automatic identiifcation system. Disk diffusion susceptibility testing or VITEK-2 Compact automatic identification system was used to determine the susceptibility to antimicrobial agents. All data were analyzed using WHONET 5.6 software.Results Among the total 7 931 clinical isolates in 2014, gram-negative bacteria accounted for 64.2% (5 088/7 931). The top three pathogens wereE. coli,A. baumannii andK. pneumoniae. Notably, during the year 2014, 195 strains of carbapenem-resistantEnterobacteriaceaewere isolated, about 6.9% of all theEnterobacteriaceae isolates. Meanwhile, 613 (66.5%) strains of multiple drug resistantA. baumannii and 197 (28.7%) strains of multiple drug resistantP. aeruginosa were isolated.Conclusion During the year 2014, the resistance of the gram-negative bacteria in this hospital is mainly characterized by carbapenem-resistantEnterobacteriaceae, multiple drug resistant A. baumanniiand multiple drug resistantP. aeruginosa. Surveillance of antimicrobial resistance is beneifcial for rational use of antibiotics.
