1.Experimental Study on Anti-inflammatory, Diuretic Effect and Acute Toxicity of Herb of Lanceolate Sedge

This study was aimed to investigate the anti-inflammatory, diuretic effect and acute toxicity of Herb of Lanceolate Sedge. The mice were randomly divided into 5 groups, which were the physiological saline group, aspirin group (0.05 g·kg-1), high dose (HD), middle dose (MD) and low dose (LD) of water extract of Herb of Lanceolate Sedge (10, 5, 2.5 g crude drug per kg) group. Intragastric administration of medication was given to mice once a day for 3 days. Auricular swelling induced by dimethyl benzene was used in the model establishment to calculate the swollen ear degree and the swollen inhibiting rate. And rats were randomly divided into 5 groups, which were the physiological saline group, aspirin group (0.05 g·kg-1), high dose (HD), middle dose (MD) and low dose (LD) of wa-ter extract of Herb of Lanceolate Sedge (10, 5, 2.5 g crude drug per kg) group. Intragastric administration of medi-cation was given to rats once a day for 5 days. Carrageenan induced paw edema was used in the model establishment to calculate the paw swollen rate. And rats were randomly divided into 5 groups, which were the physiological saline group, Furosemide tablets group (0.01 g·kg-1), high dose (HD), middle dose (MD) and low dose (LD) of water extract of Herb of Lanceolate Sedge (10, 5, 2.5 g crude drug per kg) group. Intragastric administration of medication was giv-en to rats once a day for 5 days. Water-loaded was used in the model establishment to calculate the 5-hour urine amount. The maximum tolerance dose (MTD) was used in the determination of the acute toxicity. The results showed that the three groups of HD, MD and LD of water extract of Herb of Lanceolate Sedge had obvious inhibiting effects on the swollen ear degree and the swollen inhibiting rate among mice, and the paw swollen rate of rats. It had signif-icant increasing effect on the 5-hour urine amount. The MTD of intragastric administration of water extract of Herb of Lanceolate Sedge was 640 g crude drug per kg for mice. It was concluded that Herb of Lanceolate Sedge dis-played obvious effects on anti-inflammatory and dieresis with low acute toxicity.
2.Simultaneous Content Determination of Hesperidin and Phillyrin in Bao-He-Wan by HPCE

This study was aimed to establish a HPCE method for the content determination of hesperidin and phillyrin in Bao-He-W an (BHW). Fused silica capillary (75 cm í 50 μm) was employed and 30 mmol·L-1 borax so-lution (8% acetonitrile, pH9.64) was served as the running buffer. Other conditions were as follows: electrokinetic injection was 50 kPa í 20 s; analytical voltage was 20 kV; temperature was 20℃; and detection wavelength was 254 nm. The silica capillary was flushed with 0.1 mol·L-1 sodium hydrate and the running buffer for 10 min before each injection, respectively. The results showed that the linearity of hesperidin was in the range of 0.10~2.40 mg·mL-1 (r=0.999 4), the average recovery was 99.85% and RSD=2.34%. The phillyrin was in the range of 0.07~0.84 mg·mL-1 (r=0.999 2), the average recovery was 99.16% and RSD=2.78%. It was concluded that the method was rapid and sensitive. It can be used for the quality control of content determination of hesperidin and phillyrin in BHW.
3.Optimization on Microwave Extraction Crafts for Polysaccharide from Radix Platycodonis Based on Orthogonal Test

This study was aimed to select the best microwave extraction craft conditions of polysaccharide from Radix platycodonis. The composite scores of polysaccharide content and yield rate were used as evaluation indexes. By ex-amining the impact of microwave power, microwave time, extraction times and solid-liquid ratio on crafts, the best craft conditions were optimized by orthogonal test. The orthogonal test showed that when the microwave power was 1800 W, microwave time was 8 min, extraction times were 3, solid-liquid ratio was 1:15, the composite scores were the highest and the extraction craft conditions were the best. It was concluded that the crafts are efficient and stable. It had laid a foundation for the further study on polysaccharide from R adix p latyc odonis .

BPPV is the most common cause of dizziness in adults, and the symptoms, causes, behavior of adult patients are clearly defined. Pregnant woman may also suffer from BPPV. To our knowledge, there are few reports on BPPV in pregnant women. In this paper, the clinical characteristics and results of 3 pregnant women with BPPV are reported.

5.Isolation of algicolous fungi and screening of their antitumor activity

Objective To isolate algicolous fungi from algae and obtain antitumor strains.Methods Selective isolation of algicolous fungi from algae was done,and antitumor activity corresponds to their lethiferous effect on brine shrimp and cytotoxicity against K562 cell was determined.Results 221 strains of algicolous fungi were isolated from 28 algae samples collected from intertidal zones of the Yellow Sea.Eight strains showed lethiferous activity and eight showed cytotoxicity.Conclusion The results indicated that algicolous fungi would be one of the important resources to search for new antitumor compounds.
6.Clinical Experience of Minimally Invasive Direct Cardiac Surgery With Right Anterolateral Thoracotomy Incision by Closed Cardiopulmonary Bypass

Objective: To summarize the short-term clinical experience of minimally invasive direct cardiac surgery (MIDCS) with right anterolateral thoracotomy incision by closed cardiopulmonary bypass. Methods: A total of 42 patients received MIDCS in our hospital from 2013-09 to 2015-05 were summarized. There were 18 male and 24 female patients including 16 with atrial septal defect (ASD) repair, 4 with ventricular septal defect (VSD) repair, 16 with mitral valve replacement (MVR), 1 with mitral valve plasty (MVP) and 5 with aortic valve replacement (AVR). Direct cardiac surgery was performed by cardiopulmonary bypass through femoral artery-vein and right jugular vein annulations. A right anterolateral thoracotomy incision (length 3-5 cm) was made to enter the chest and complete the operation. Results: All 42 patients received successful operation, no peri-operative or early post-operative death, no incision infection occurred. Cardiopulmonary bypass time was [98-142 (122.4 ± 23.7) min], aortic cross-clamp time [0-118 (48.3 ± 26.2) min]. Post-operative mechanical ventilation time was [8-76 (17.4±13.1) h], intensive care unit stay time [45-124, (54.6 ± 32.6) h], hospital stay time [6-12, (8.2 ± 1.3) d]. Incision length was [3-7, (4.8 ± 1.5) cm], the draining volume at the 1st post-operative day was (356.9 ± 283.8) ml and there were 27 (64.3%) patients without transfusion. Conclusion: The short-term outcomes for MIDCS were good, it with superior safety, broad application range with minimal invasion and less complication.
7.Establishment and application of HPLC-QAMS for quality evaluation of Chuanxiong Rhizoma.

A quantitative analysis method of multi-components with a single marker (QAMS) for simultaneous determination of six marker compounds (one from phenolic acids and five from phthalides) in Chuanxiong Rhizoma was established by applying HPLC and using butylidenephthalide as the internal reference substance. And also the feasibility and accuracy of the established method for quality evaluation and application of Chuanxiong Rhizoma were investigated and validated. The analysis was performed with the mobile phase consisting of acetonitrile - 0.2% aqueous formic acid. The flow rate was 1.0 mL . min-1 and the column temperature was maintained at 30 °C. The detection wavelengths were set at 252 nm (for ferulic acid, Z-ligustilide, and butylidenephthalide) and 266 nm (for senkyunolide I, senkyunolide A, and coniferyl ferulate), separately, and 20 µL was injected for analysis with gradient elution. The results showed that there were no significant differences observed between the HPLC-QAMS method and the external standard method (RSD <5%). The relative correction factors were credible (RSD < 5%) in changed chromatographic conditions. The established HPLC-QAMS method can be accurately used for simultaneously evaluating and controlling the quality of Chuanxiong Rhizoma with multi-components.

9.Efficacy of quick repositioning maneuver for posterior semicircular canal benign paroxysmal positional vertigo in different age groups.

OBJECTIVE:
To observe the short and long-term efficacy of quick repositioning maneuver for posterior semicircular canal benign paroxysmal positional vertigo (PC-BPPV) in different age groups.
METHOD:
The clinical data of 113 adult patients with single PC-BPPV who underwent quick repositioning maneuver from July 2009 to February 2015 were retrospectively analyzed. The quick repositioning maneuver was to roll the patient from involved side to healthy side in the coronal plane for 180° as quickly as possible. The patients were divided into 3 groups according to different ages: young group (< 45 years), middle-age group (45 ≤ age < 60 years) and old group (≥ 60 years). The short and long term outcomes of the three groups were observed.
RESULT:
The left ear was involved in 58 cases (51.3%) and the right ear in 55 cases (48.7%). The short term improvement rates of the young, middle-age and the old groups were 92.5%, 93.6% and 92.3% respectively, and the long term improvement rate was 90.0%, 85.1% and 73.1% respectively. There was no significant difference among the three groups in short and long term outcomes (P > 0.05). The recurrence rate of the three groups was 5.0%, 6.4% and 15.4% respectively, also no significant difference (P > 0.05).
CONCLUSION
The quick repositioning maneuver along the coronal plane for PC-BPPV has a definite effect for every age groups. The method is simple, rapid and easy to master, and the patients are tolerated the maneuver well without evident side effect.

10.The sustained releasing ability of CEL-PLGA-MS in vitro and its inhibitory effects on experimental choroidal neovascularization after intravitreal injection

Background Choroidal neovascularization (CNV) is a common pathological basis of many ocular fundus diseases.Some treating methods are proved to be effective on CNV but there exist their own shortages.Celecoxib can inhibit experimental neovescularization.Sustained release drug of celecoxib and application approach can offer a basis for the therapy of CNV.Objective This study was to evaluate the sustained release ability of celecoxib-poly lactide-co-glycolide microsphere (CEL-PLGA-MS) in vitro and its inhibitory ability on experimental CNV in vivo.Methods CEL-PLGA-MS was prepared by Hebei Medical University and examined under the scanning electron microscope.The size of CEL-PLGA-MS was measured by Laser Particle Size Analyzer.The drugloading in vitro releasing was monitored by high performance liquid chromatograph (HPLC).Experimental CNV was induced by laser photocoagulation of retina in the right eyes of 72 male brown Norway (BN) rats and then were randomized into the CEL-PLGA-MS group,celecoxib group,blank PLGA group and PBS group.CEL-PLGA-MS with 320 μmol/L celecoxib,80 μmol/L celecoxib,blank PLGA microspheres solution and 0.01 mol/L PBS was intravitreally injected separately according to the grouping.CNV was assessed by fundus fluorescein angiography (FFA) on the 14th day after injection.The fibrovascular proliferation (FVP) thickness at photocoagulation spots was measured by OCT.The retinal pigment epithelium (RPE)-choroid-sclera sections were prepared for the histopathologieal examination of FVP.On the 7th and 28th day after intravitreal injection,the relative expression levels of VEGF mRNA and COX-2 mRNA in the photocoagulation area were detected by reverse transcription PCR (RTPCR).The use and feeding of the experimental animals were followed by the ARVO statement.Results CELPLGA-MS showed the spherical shape with the mean size of 2 467.9 nm and the drug-loading of 7.77% and the drugrelease rate of 80.91% in vitro for 45 days.It presented the controllable release characteristics.CEL-PLGA-MS agglomerated in vitreous body after injection.On the 14th day after intravitreal injection,the mean FVP thicknesses were (94.67±4.64),(98.56±4.72),(71.00±4.77),(50.44±3.01) μm in the blank PLGA microspheres group,PBS group,celecoxib group and CEL-PLGA-MS group,respectively,showing significant increases in mean FVP thickness in the blank PLGA microspheres group and PBS group compared with the celecoxib group and CEL-PLGAMS group (all at P<0.01),and the CEL-PLGA-MS group appeared a lower mean FVP thickness value than the celecoxib group (P<0.01).FFA revealed a large number of strong hyperfluorescences at the photocoagulation area in the rat eyes of the blank PLGA microspheres group and PBS group;while only weak hyperfluorescences were seen in the eelecoxib group and CEL-PLGA-MS group.Histopathological examinations verified the same results in the FVP thickness to OCT image.The relative expression levels of COX-2 mRNA and VEGF mRNA in the RPE-choroid-sclera were all significantly elevated in the blank PLGA microspheres group compared with the celecoxib group and CELPLGA-MS group both on the 7th and 28th day after intravitreal injection (all at P<0.01).On the 7th day after injection,the relative expression levels of COX-2 mRNA were lower on the 7th day and the relative expression levels of COX-2 mRNA and VEGF mRNA were higher on the 28th day in the celecoxib group in comparison with the CEL-PLGA-MS group (all at P<0.01).Conclusions CEL-PLGA-MSs are even in size with the spherical shape and controllable release characteristics in vitro.CEL-PLGA-MS can inhibit experimental CNV and was more durable effective than celecoxib after intravitrea] injection.
