1.Association between SERPINH1 gene expression level in gastric cancer tissue with pathology and prognosis

Objective To investigate the association between SERPINH1 gene expression in gastric cancer tissue with the pathology and prognosis and its action mechanism in gastric cancer.Methods The gastric cancer data in the TCGA and GEO public databases were collected,and the retrospective analysis and survival analysis were conducted on the SERPINH1 expression data of the tumor tissue samples and related clinical information;the gene set enrichment analysis(GSEA) was used to analyze the SERPINH1 expression related functional gene sets and to investigate the SERPINH1 action mechanism.Results In 421 cases of gastric cancer sample,the SERPINH1 expression level had no significant differences with the tumor grade,N stage and metastasis.However,its high expression was significantly correlated with the tumor invasion depth(T stage)(P=0.049).The higher the expression level,the preater the invasion depth.In the survival analysis,the patients with high SERPINH1 expression showed a poorer prognosis than those with low SERPINH1 expression[P<0.001,HR(95%CI):1.97 (1.61-2.41)].The gene set enrichment analaysis found that the related gene sets such as tumor signal pathway and tumor microenvironment were enriched in the SERPINH1 high expression samples.The gene expression chip data in 69 cases of gastric cancer and paracancerous tissue also suggested that SERPINH1 was highly expressed in gastric cancer (P<0.000 1) with a greater diagnostic value (AUC=0.988 1).Conclusion High expression of SERPINH1 is correlated with the occurrence and development of gastric cancer,and has a greater clinical significance.
2.Alterations of the proteasome in the CA1 neurons after transient ischemia

Objective To investigate the alteration of chaperonine hsp40 and its influence on delayed death of neurons in the CA1 region of hippocampus after transient cerebral ischemia. Method After transient global ischemia for 20 minutes, rat model was made. Following different lengths of reperfusion, all the 28 wistar rats were divided into sham-operation group,4 hour recovery group, 24 hour recovery group and 72 hour recovery group ( n = 7 rats in each group). Immunochemistry and laser scanning confocal microscopy were used to observe the distributional alteration of hsp40 in the neurons. Differential centrifuge and western blot assay were used to analyze the quantitative alteration of hsp40 and its redistribution in the neurons. Results Immunochemistry and laser scanning confocal microscopy showed the progressive reduction of hsp40 occurred at first in the cytosol, then in the nucleus until the death of all the neurons in the CA1 region died. Differential centrifuge and western blot assay showed the level of hsp40 decreased from 1.00 ± 0.21 to 0.23 ± 0.13 ( P < 0.01 ) 24 hours after reperfusion; the quantity of hsp40 in the protein aggregates increased from 1.00±0.18 to 8.61 ± 1.89 (P <0.01 =24 h after reperfusion.Conclusions The reduction of hsp40 in the neurons of hippocampus CA1 region is an important role in protein aggregates formation.
3. Imaging study of second sacral alar-iliac screw placement in adult degenerative scoliosis

Objective:
To discuss the imagingcharacteristics of S2AI screw trajectory in ADS patients.
Methods:
Forty patients with degenerative scoliosis were scanned with Simens Sliding 40-slice spiral CT scanner. Three-dimensional scanning and reconstruction were performed in these patients with the scanning range including thoracolumbar spine, lumbar spine, lumbosacral region, pelvis and bilateral hip joints. The base of the lateral sacral crest on the midline between the lower edge of S1 dorsal foramina and the upper edge of S2 dorsal foramina was the starting point. The placement plane of S2AI screw trajectory was determined from the starting point to the lower margin of anterior inferior iliac spine. A 10mm diameter screw was the design template. A circle with a diameter of 5 mm as the center of the lowest point of the ilium inner cortex was made, and a tangent line from the starting point to the outer diameter of the circle (the inner part of the ilium) was selected as the axis of the screw trajectory. The lateral angle and the length of the axis of the screw trajectory and iliac width were measured in transverse plane. The caudal angle, the distance from the axis of the screw trajectory to iliosciatic notch, and the caudal angle, the distance from the axis of the screw trajectory to the upper edge of the acetabulum were measured in sagittal plane. These parameters were recorded and analyzed.
Results:
The trajectory length of S2AI screw in ADS patients was 12.00±0.99 cm, the lateral angle was 41.24°±3.92°, the caudal angle was 27.73°±6.45°, and the distance from the axis of the screw trajectory to iliosciatic notch was 1.05±0.81 cm, the distance from the axis of the screw trajectory to the upper edge of the acetabulum was 1.85 ± 0.33 cm, and the iliac width was 2.12±1.65 cm. The trajectory length, lateral angle, caudal angle, distance from the axis of the screw trajectory to iliosciatic notch, distance from the axis of the screw trajectory to the upper edge of the acetabulum and iliac width of S2AI screw was respectively 12.40±0.83 cm, 39.47°±1.76°, 28.00°±6.39°, 1.08±0.32 cm, 1.76±0.34 cm, 2.26±0.25 cm in male patients, and was respectively 11.75±1.01 cm, 42.30°±4.48°, 27.56°±6.61°, 1.21±1.00 cm, 1.90±0.32 cm, 2.04±0.18 cm in female patients. Breast cancer is the worldwide most common malignant tumor in women,and early diagnosis and treatment is the key to improve prognosis.Conventional screening imaging techniques,such as mammography and ultrasonography,have only limited diagnostic efficacy for micro breast cancer.Combining diffuse optical tomography(DOT)and ultrasonography for assessing blood vessel distribution and metabolic level inside the breast lesions,optical tomography image ultrasonography system(OPTIMUS)has unique advantages for diagnosing micro breast cancer with diameter≤1.0 cm.The research progresses of OPTIMUS for diagnosing micro breast cancer were reviewed in this article.
5.Safety analysis of microscope-assisted lumbar interbody decompression,fusion and internal fixation in the treatment of recurrent lumbar disc herniation

Objectives:To analyze the safety of the assistance of microscope in lumbar interbody decompres-sion,fusion and internal fixation in the treatment of recurrent lumbar disc herniation.Methods:Retrospective analysis was made on 73 patients recurred lumbar disc herniation after percutaneous endoscopic lumbar dis-cectomy(PELD)and underwent a revision surgery of single-segment lumbar decompression and fusion in our hospital from January 1,2020 to April 1,2022.Among the patients,42 received microscope-assisted opera-tion(microscope group),including 31 males and 11 females,aged 53.4±16.4 years;the other 31 received an operation without microscope assistance(conventional group),including 18 males and 13 females,aged 49.6± 11.4 years.There were no significant differences between the two groups in general data such as gender,age and BMI(P>0.05).Data such as the surgical approach and surgical segment of initial PELD,and operative time,intraoperative bleeding volume,total postoperative drainage volume,postoperative length of stay,dural tears,nerve injury and superficial infection of the revision surgery were collected and compared between the two groups.Results:All the operations were successful.In the microscope group,the operative time was 168.0±33.1min,intraoperative bleeding volume was 125.0±40.2mL,total drainage volume was 379.6±220.6mL and postoperative length of stay was 5.2±1.3d,and dural tears occurred in 2 patients,none nerve injury or superficial wound infection was found.In the conventional group,the operative time was 155.8±29.4min,in-traoperative bleeding volume was 133.9±59.7mL,total drainage volume was 452.2±464.5mL,and postoperative length of stay was 5.0±1.7d,and dural tears occurred in 4 patients,nerve injury was noticed in 1 patient,and none superficial wound infection was found.There was no statistical difference in perioperative and com-plication data between the two groups(P>0.05).Conclusions:Microscope does not increase the operative time,intraoperative bleeding volume,postoperative length of stay,and the rate of infection,dural tears and nerve injury in the revision surgery after PELD.
6.Establishing Multi-Modal Ultrasound-Based Prediction Model for Placental Insufficiency of Second Trimester

Purpose Using conventional ultrasound,3D energy Doppler and shear wave elastography to develop one multi-modal ultrasound-based prediction model for placental insufficiency of second trimester.Materials and Methods A prospective analysis was conducted by reviewing 209 pregnant women with anterior wall placenta who underwent multi-modal ultrasound examination in the Affiliated Renhe Hospital of China Three Gorges University from April 2021 to April 2022,and all participants were divided into 118 cases in the normal group and 91 cases in the placental insufficiency group based on clinical diagnosis.Their placental function was evaluated through hemodynamics[blood flow parameters of umbilical artery(UA),uterine artery(Ut A)and cerebral placental rate(CPR)],blood stream perfusion[vascular index(VI),blood flow index(FI)and vaso-blood flow index(VFI)]and mechanical tissue properties[mean of elasticity modulus(Emean)].Logistic regression analysis was performed on the indicators with statistically significant differences between groups,establishing a prediction model of placental insufficiency and evaluating its efficacy.Results There were statistically significant differences in Ut A blood flow parameters,CPR,VI,VFI and Emean between the two groups(t=-12.74,-10.28,-11.01,7.02,7.00,-11.97,all with P<0.05).In regression analysis,Ut A-RI(≥0.65),CPR(≤1.53)and Emean(≥4.14 KPa)were found to be the independent risk factors,of which Emean had the greatest diagnostic efficacy(area under the curve,AUC=0.904).The AUC of the prediction model established by combining the three parameters was 0.945,with a sensitivity of 87.91%and a specificity of 94.92%,which has significantly higher diagnostic efficacy than that of a single parameter.Conclusion The prediction model based on the multi-modal ultrasound parameters has certain predictive value and high specificity for placental insufficiency of second trimester and can guide early clinical intervention to improve perinatal outcomes.

Objective To explore the feasibility and efficacy of an MRI-visible,targeted,nano-vector which is synthesized by attaching a targeting ligand,the GD2 single chain antibody (scAb GD2),to the distal ends of PEG-g-PEI-SPION as a carrier for gene delivery into human bone marrow mesenchymal stem cells (hBMSCs) and in vitro cellular MR imaging.Methods scAbGD2-PEG-g-PEI-SPION was synthesized as previously reported.Gel electrophoresis was performed to assess the pDNA condensation ability of scAbGD2-PEG-g-PEI-SPION.The particle size and Zeta potential of scAbGD2-PEG-g-PEI-SPION/pDNA nanocomplexes were observed by dynamic light scattering.Cytotoxicity of scAbGD2-PEG-g-PEI-SPI-ON was evaluated by CCK-8 assay using hBMSCs.Gene transfection efficiency of scAbGD2-PEG-g-PEI-SPION in hBMSCs was quantified by flow cytometry,PEG-g-PEI-SPION,scAbGD2-PEG-g-PEI-SPION,scAbGD2-PEG-g-PEI-SPION+ free AbGD2 and scAbIgG2a-PEG-g-PEI-SPION group was established.The cellular internalization of scAbGD2-PEG-g-PEI-SPION/pDNA nanocomplexes was observed by confocal laser scanning microscopy and Prussian blue staining.MRI of scAbGD2-PEG-g-PEI-SPION was performed by cellular MRI scanning in vitro.Results scAbGD2-PEG-g-PEI-SPION condensed pDNA to form stable nanocomplexes of 80-100 nm in diameter and showed low cytotoxicity to hBMSCs.At the same N/P ratio,the transfection efficiency of scAbGD2-PEG-g-PEI-SPION group was significantly higher than those of other groups (P<0.001).At the optimal N/P ratio of 20,scAbGD2-PEG-g-PEI-SPION/pDNA obtained the highest transfection efficiency of (59.60 ± 4.50) % in hBMSCs.Furthermore,hBMSCs labeled with scAbGD2-PEG-g-PEI-SPION showed sensitive low signal intensity on MRI T2/T2 *-weighted images in vitro.Conclusion scAbGD2-PEG-g-PEI-SPION is an efficient MRL visible targeted nano vector for gene delivery into hBMSCs.

Objective:To systematically analyze the basic characteristics and contents of the current health standards for infectious disease, environmental health, school health and disinfection in the context of COVID-19 prevention and control, and provide support for the further optimization of epidemic prevention and control guidelines and reference for the revision and improvement of related health standards.Methods:Public health standards used in COVID-19 prevention and control were selected for a systematic comparison with "The Plan of COVID19 Prevention and Control (the 6 th Edition)" and other epidemic prevention and control guidelines from the perspectives of application scope and technical elements. Results:The current standards of public health are with scientificity, timeliness and feasibility. The application scope and technical elements of the current public health standards basically meet the needs of the prevention and control of COVID-19 epidemic, but the public health standardization system still needs improvement, and some public health standards need to be revised.Conclusions:The implementation of current public health standards can provide strong technical support for the prevention and control of COVID-19 epidemic. The experience obtained from COVID-19 epidemic prevention and control might promote the further improvement of the health standardization system.
