1. Clinicopathologic and prognosis features of Claudin-low breast cancers

Objective:
To investigate the clinicopathologic and prognostic features of Claudin-low breast cancers (CLBC).
Methods:
Tissue microarray sections were scored semiquantitatively for the immunohistochemical expression of claudin-1, -3, -4, -7 and -8 in 233 cases of invasive breast cancers collected from Qingdao Central Hospital from January 2010 to December 2011.
Results:
The expression rate of Claudin-3 (72/212, 33.9%) and -4 (56/212, 45.2%) was most similar, and Claudin-4 showed the highest expression. Twenty one cases (21/212, 9.0%) were diagnosed as CLBC, with triple-negative breast cancer (TNBC) accounted for the highest proportion (11/21, 52.4%). Among the CLBC cases, the invasive carcinoma no special type (66.7%, 14/21) and metaplastic carcinoma (14.3%, 3/21) were mostly seen, while metaplastic squamous carcinoma did not show Claudin-low pattern. Compared to the non CLBC in this cohort, CLBC had higher proportion of histologic grade 3 and tumors larger than 2 cm, and the proportions were slightly lower than TNBC. 2.Relationship Between Serum Pepsinogen and OLGA/OLGIM Stages and its Value in Early Screening of Gastric Cancer

Background:Gastric cancer is a malignant tumor originating from the gastric mucosal epithelium.OLGA/OLGIM staging systems are scoring systems for gastric mucosal atrophy and intestinal metaplasia based on gastroscopic pathology.Serum pepsinogen(PG)and PG Ⅰ/PG Ⅱ ratio(PGR)can reflect the gastric mucosal function and status.Studying the relationship between PG and OLGA/OLGIM stages and the changing trends is of great value for early screening of gastric cancer.Aims:To explore the relationship between serum PG and OLGA/OLGIM stages,and its application value in screening of early gastric cancer.Methods:A total of 188 healthy individuals underwent gastroscopy from June 2021 to June 2023 at Shanghai Health and Medical Center were selected.The serum PG level was measured,and the status of Helicobacter pylori(Hp)infection was evaluated.OLGA/OLGIM stages were assessed according to the findings of gastroscopic pathology,and correlated with the serum PG level.After two years of follow-up,changes of PG and PGR in subjects with different baseline OLGA/OLGIM stages were analyzed.Results:There were significant differences in PG Ⅰand PGR among subjects with various baseline OLGA/OLGIM stages(P<0.05).OLGA/OLGIM stages were correlated with Hp infection,family history of malignant gastrointestinal tumors and spicy diet(P<0.05).The incidences of gastric mucosal atrophy in PG Ⅰ ≤70 μg/L group and PGR≤3.0 group were significantly higher than those in PG Ⅰ>70 μg/L group and PGR>3.0 group,respectively(P<0.05).The follow-up results showed that for baseline OLGA/OLGIM stage 0,PG Ⅰ showed a decreasing trend over time,while no significant change of PG Ⅰ was found in other OLGA/OLGIM stages;PG Ⅱ showed a decreasing trend while PGR showed an increasing trend over time in all stages.But all these trends were not statistically significant(P>0.05).Conclusions:The combined detection of PG and Hp infection has important value in evaluating the severity of chronic atrophic gastritis and the risk of gastric cancer.It can be applied as an early screening project for gastric cancer in individuals undergoing physical examination.
3.Effects of blood pressure variability on the progression and prognosis of carotid artery stenosis in elderly patients with cerebral infarction

Objective To explore the effect of blood pressure variability (BPV) on the progression and prognosis of carotid artery stenosis in elderly patients with cerebral infarction. Methods A total of 98 elderly patients with cerebral infarction admitted from May 2019 to May 2022 were selected as the observation group, and 100 healthy people with physical examination during the same period were enrolled as the control group. According to the degree of carotid artery stenosis, the elderly patients with cerebral infarction were divided into four groups. The blood pressure variability parameters in each group were compared, and the relationship between BPV and the degree of carotid artery stenosis was analyzed. The risk factors of vascular events were analyzed and the predictive efficiency of BPV on vascular events was evaluated. Results The BPV parameter values in the observation group were higher than those in the control group (P<0.05), while the nocturnal decline rate of blood pressure was lower than that in the control group (P<0.05). The higher the degree of carotid artery stenosis, the higher the BPV parameter value and the lower the nocturnal decline rate of blood pressure (P<0.05). The proportion of total non-dipper type in the severe stenosis group was higher than that in the moderate stenosis group, mild stenosis group and non-stenosis group (P<0.05). Spearman correlation analysis showed that the BPV parameter values were positively correlated with the degree of carotid artery stenosis, and the nocturnal decline rate of blood pressure was negatively correlated with the degree of carotid artery stenosis (all P<0.05). Multivariate logistic regression analysis showed that the increases of BPV parameter values were all risk factors for vascular events in elderly patients with cerebral infarction (P<0.05). Receiver operating characteristic (ROC) curve analysis revealed that among the BPV parameters, dSSD had the highest efficiency in predicting the occurrence of vascular events in elderly patients with cerebral infarction. Conclusion BPV significantly affects the progression and prognosis of carotid artery stenosis in elderly patients with cerebral infarction.
4.Biological effects of acute high-dose radon exposure on mice

Objective:To investigate the biological effects of acute high-dose radon exposure on mice.Methods:BALB/c male mice aged 6 to 8 weeks were exposed once in an HD-3 ecological radon chamber with an average radon concentration of 7 × 10 5 Bq/m 3 for 10 h. Mice were weighed, their lung tissues and blood samples were collected at 1, 2 and 3 months after exposure. Control groups were set up at the three time points with four mice in each group. For these mice, the lung tissue pathology was observed using hematoxylin-eosin (HE) staining method, routine blood tests were conducted using a hematology analyzer and the levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in the serum and lung tissues were measured using corresponding assay kits. Results:The HE staining result revealed that compared to the control groups, the experimental groups exhibited thickening of alveolar walls and increased infiltration of granulocyte, whose degrees, however, reduced over time and displayed no significant difference at 3 months after exposure. There was no significant difference in body weight or blood routine between the experimental and control groups. The detection result revealed decreased SOD levels in the lung tissues at 2 months after exposure, which were (11.34 ± 1.03) U/mgprot and (9.75 ± 0.71) U/mgprot, respectively for the control and experimental groups ( t = 2.54, P < 0.05). The MDA levels in lung tissue increased at 1 month after exposure, which were(2.30 ± 0.24) and (2.77 ± 0.29) nmol/mgprot, respectively for the control and experimental groups ( t = 2.49, P < 0.05). At 3 months after exposure, the SOD and MDA levels differed insignificantly between the control and experimental groups ( P > 0.05). Conclusions:After acute high-dose radon exposure, the mice suffered damage to the lung tissue, with changes in their oxidative stress indicators being detected. However, these effects gradually diminished at 3 months after exposure. Additionally, acute high-dose radon exposure did not give rise to significant changes in the body weight or routine blood result of the mice.
