2.Effects of acupuncture on mood and glucose metabolism in the patient of type 2 diabetes.

OBJECTIVETo observe effects of acupuncture on depression and anxiety, and glucose metabolism in the patient of type 2 diabetes.
METHODSOne hundred cases of type 2 diabetes mellitus patients were randomly divided into the control group treated with oral administration of Diaformin and the observation group treated with oral administration of Diaformin combined with acupuncture at Baihui (GV 20), Fengfu (GV 16), 50 cases in each group. Before and after treatment, depression scale (HAMD, SDS) and anxiety scale (HAMA, SAS) scores, and fasting blood glucose (FPG), blood glucose 2 h after meal (PG 2 h), glycosylated hemoglobin (GHb) levels were detected.
RESULTSAfter treatment, the mood disorder improved and FPG, PG 2 h, GHb levels decreased in the two groups, with the observation group being better than the control group (P < 0.05).
CONCLUSIONAcupuncture can improve negative mental state, glucose metabolism and blood glucose in type 2 diabetes patients.

3.Study on clinical therapeutic effect of activating brain and regaining consciousness needling method on poststroke depression and the mechanism.

OBJECTIVETo probe into an effective method for treatment of poststroke depression.
METHODSTwo hundred and fifty-six cases of poststroke depression were randomly divided into a control group (n = 76) treated by oral administration of amitilin and an acupuncture group (n = 180) treated by activating brain and regaining consciousness needling method. Their clinical therapeutic effects and changes of relative factors of poststroke depression and plasma contents of monoamine neurotransmitters were observed.
RESULTSIn the acupuncture group, the clinical symptoms and the relative indexes in the depression scales improved and plasma contents of the monoamine neurotransmitters increased significanty as compared with the control group (P < 0.05).
CONCLUSIONThe needling method for activating brain and regaining consciousness can improve relative factors to cure poststroke depression.

4.Surgical treatment for abdominal aortic aneurysm in 46 cases

Objective To study how to improve the safety of the operation for abdominal aortic aneurysm(AAA).Methods Forty-six cases of AAA received resection of AAA plus artificial blood vessel transplantation in recent three and a half years in our 2 hospitals.The lesions involved only the abdominal aorta in 20 cases,and extended to unilateral common iliac and internal and external iliac arteries in 8 cases,to bilateral common iliac and external and internal iliac arteries in 16 cases,and involved the renal artery in 2 cases.Emergency operation was done in 3 cases.Results In these 46 cases,45 were cured and 1 died,and no operative complications occurred.Conclusions Surgical operation is the best way to treatment AAA.
5.Studies on the chemical constituents of Cirsium japonicum DC.

AIMIn order to look for new bioactive compounds, investigation on the chemical constituents, especially on the typical polyacetylenes from the rhizomes of Cirsium japonicum DC. was carried out.
METHODSChromatographic techniques including silica column chromatography and preparative silica thin-layer chromatography were used to separate and purify the constituents. Their structures were elucidated by physicochemical properties and spectral analyses including UV, IR, 1HNMR, 13CNMR, HMQC, HMBC and HREIMS.
RESULTSTwelve compounds were isolated from the rhizomes of Cirsium japonicum DC., and their structures were identified as cis-8, 9-epoxy-heptadeca-1-ene-11, 13-diyne-10-ol (1), ciryneol A (2), 8,9,10-triacetoxyheptadeca-1-ene-11,13-diyne (3), ciryneone F (4), cireneol G (5), ciryneol H (6), ciryneol C (7), p-coumaric acid (8), syringin (9), linarin (10), beta-sitosterol (11) and daucosterol (12).
CONCLUSIONCompounds 4, 5 and 6 are new compounds, compound 3 is a new natural product and compound 8 was isolated from this plant for the first time.

6.Chemical and pharmacological advances of study on Lysimachia.

The progress in the research of the chemical components and pharmacological activities of Lysimachia was reviewed. Lysimachia mainly contain flavonoids, saponins and phenolic acids, and some of the plants have been used in the treatment of fever, ulcers, diarrhea and as analgesic or anti-inflammatory agents. The plants of genus Lysimachia are widespread in China, so it is a singificants work to study their chemical ingredients and pharmacological activities.

7.HPLC-MS3 analysis of chemical constituents in Epimedium brevicornum.

OBJECTIVETo investigate the chemical constituents of effective part of Epimedium brevicornum.
METHODThe sample was extracted with ethanol and purified by macroporous resin. The structures were identified by HPLC-MS3 experiments.
RESULTNine compounds were identified from the effective part of E. brevicornum.
CONCLUSIONThe method is simple and rapid for the identification of the flavonoids from E. brevicornum.

8.Effect of a microRNA-132 antagonist on pilocarpine-induced status epilepticus in young rats.

OBJECTIVETo study the effect of a microRNA-132 antagonist on lithium-pilocarpine-induced status epilepticus (SE) in young Sprague-Dawley (SD) rats.
METHODSForty-five 3-week-old SD rats were randomly and equally divided into epilepticus model group, microRNA-132 antagonist group, and microRNA-132 antagonist negative control group. The young SD rat model of SE was established using lithium-pilocarpine. For the microRNA-132 antagonist group and the negative control group, pretreatment was performed 24 hours before the model establishment. Behavioral observation was performed to assess the latency of SE and success rate of induction of SE. The scale of Lado was used to evaluate the seizure severity. Electroencephalography (EEG) was used to assess the frequency and amplitude of epileptiform discharges. The mortality rate was calculated in each group.
RESULTSThere was no significant difference in the success rate of induction of SE between the three groups (P>0.05). Compared with the microRNA-132 negative control group and the epilepticus model group, the microRNA-132 antagonist group had significantly prolonged SE latency after model establishment (P<0.05), a significantly lower Lado score of seizure (P<0.05), significantly lower frequency and amplitude of epileptiform discharges on EEG (P<0.05), and a slightly reduced mortality rate.
CONCLUSIONSThe treatment with the microRNA-132 antagonist shows an inhibitory effect on the development and progression of lithium-pilocarpine-induced SE in young SD rats. The inhibition of microRNA-132 is likely to be a potential target or direction for drug treatment of SE.

9.Transmission electron microscopy investigation of the denudation of the envelope of influenza virus treated with Nonidet P-40 and the binding influence of antibody.

OBJECTIVEThrough observing the morphological changes of the prepared influenza viruses (H1N1) treated with the different concentration of Nonidet P-40 solutions and added with antibody against the influenza virus using transmission electron microscopy (TEM), to explore the principle of application of the internal antibody for immunoassay of influenza virus.
METHODSThrough treating the virus samples with serial diluted Nonidet P-40 solutions from 0.01% to 0.2% and/or not adding antibody against the influenza virus, then investigating the samples by TEM to obtain the morphological changes of the virions.
RESULTSThe serial images show that the denudation degree of the virions is proportional with the rise of NP-40 concentration, and partly denuded virion image appeared at 0.1% NP-40 treatment, also under this condition no influence was observed on antibody binding to the virus.
CONCLUSIONThis work demonstrated the interaction between influenza virion and its antibody under nonionic surfactants existing, which supports the advantages of sample preparation for immunoassay enveloped virus using internal antibody theoretically.

OBJECTIVEThe purpose of this study was to assess weather the immortalized mouse brain endothelial cell line Bend.3 displays the comparative barrier characteristics as the primary brain microvascular endothelial cells (BEMC).
METHODSImmortalized mouse brain endothelial cell line, Bend.3 cells were cultured in transwell inserts and their restrictive characteristics were assessed by transendothelial electrical resistance (TEER) and horseradish peroxidase (HRP) permeability assays. Western blot and direct fluorescent staining methods were used to detect the tight junction protein expression and F-actin distribution.
RESULTSThe TEER in Bend.3 cells increased with the prolonged culture time and increased to 82.3+/-6.0 Omega cm2 10 days after culture, which was significantly higher than that 3 days after culture (37.3+/-3.1 Omega cm2; P<0.05). There were significant differences in the permeability rates for HRP 3 and 10 days after culture (4.3+/-0.20)% vs (2.2+/-0.05)% (P<0.05). Western blot indicated high level expression of tight junction proteins occludin and ZO-1 in Bend.3 cells 10 days after culture. F-actin was visualized around the cell membrane and presented scrobiculate linear fluorescence 10 days after culture.
CONCLUSIONSBend.3 cells have similar barrier characteristics to BEMC, and their barrier function may reach to the best effect 10 days after culture.
