2.Analysis on Expression of NK Cells in Peripheral Blood in the Patients of Lung-Qi Deficiency and Lung-Yin Deficiency

Objective:To observe expression of NK cells in peripheral blood in the patients of lung-Qi deficiency and lung-Yin deficiency. Methods:CD_(16)/CD_(56) were determined by flow cytometry in 58 cases of lung-Qi deficiency,12 cases of lung-Yin deficiency and 20 cases of healthy persons.Results:Activities of NK cells in both the patient of lung-Qi deficiency and the patient of lung-Yin deficiency decreased;There were significant differences in activities of NK cells among the various groups of lung-Qi deficiency syndrome.Conclusion:The immunological function in the patient of lung-Qi deficiency syndrome developed gradually to immunodepression of different degrees from active state at early stage,and then developed to lung-Yin deficiency syndrome,and finally leading to maladjustment of hemeostasis of immunological system.
3.Effect of hypoxia combined with exercise training on mitochondrial con-tent in skeletal muscle of rats

[ABSTRACT]AIM:Toinvestigatetheeffectofhypoxiacombinedwithexercisetrainingonmitochondrialcon-tent, and the role of mitochondrial biogenesis and mitophagy in this process .METHODS:Male Sprague-Dawley rats were randomly divided into 4 groups:normoxia control (NC) group, normoxia+training (NT) group, hypoxia+control (HC) group, and hypoxia+training (HT) group.The hypoxic animals were housed in normobaric hypoxic tent (11.3 % oxy-gen) for consecutive 4 weeks.The exercise training animals were exercised on a motor-driven rodent treadmill (5°) at a speed of 15 m/min, 60 min/d, 5 d/week for 4 weeks.Mitochondrial membrane potential was determined using JC-1 fluo-rescent probe .ATP synthesis capacity was determined using a bioluminescence technique .The protein expression of cyto-chrome C oxidase IV (COXIV), voltage-dependent anion channel-1 (VDAC-1), peroxisome proliferator-activated receptor gamma cofactor 1 alpha (PGC-1α), mitochondrial transcription factor A (Tfam),Bcl-2/adenovirus E1B 19 kD-interacting protein 3 (Bnip3) and beclin-1 in the muscles was detected by Western blotting .RESULTS:Compared with NC group, hypoxia attenuated mitochondrial membrane potential , ATP synthesis capacity , and the expression of COXIV , VDAC-1, PGC-1αand Tfam.Furthermore, hypoxia increased the expression of Bnip 3 and beclin-1.Compared with HC group , the exercise training elevated mitochondrial membrane potential , ATP synthesis capacity , and the expression of COXIV , VDAC-1, PGC-1α, Tfam, Bnip3 and beclin-1.CONCLUSION:A combination of reduced mitochondrial biogenesis and increased mitophagy seems to be responsible for the decrease in mitochondrial content after hypoxia .Exercise training with hypoxia elevates mitochondrial content and function in hypoxia , which may be mediated by appropriate increase and co-reg-ulation of mitochondrial biogenesis and mitophagy .
4.Expression, Purification of Recombinant Flounder MRF4 Protein in Escherichia coli and Analysis of Its Polyclonal Antibodies

MRF4 is one of muscle regulatory factors and plays critical roles during skeletal muscle development. The muscle development is important for the fish growth which is an important economic factor for the fish culture. To analyze the function of MRF4 in fish, the founder MRF4 antibody was prepared. The flounder MRF4 was cloned, ligated into prokaryotic expression vector pET-30b and expressed in strain E. coli BL21 (DE3). The recombinant flounder MRF4 fusion protein was soluble and purified with cobalt IMAC resins. To prepare MRF4 polyclonal antibodies, rabbits were immunized with the soluble protein and the increasing level of antibodies was determined by Western blot. Also, the endogenous flounder MRF4 was recognized by the anti-serum. The result further proved the existence of the anti-MRF4 antibody in the anti-serum, which will be useful for studies on the function of flounder MRF4.
5.Protective effect of ischemia preconditioning and the role of nitric oxide on the small intestines injury following hind limbs ischemia reperfusion in rat *

Objective To observe the protection effect of ischemic preconditioning to small intestines injury after limbs ischemia reperfusion ,and investigate the role of nitrogen monoxidum in the process .Methods 52 male Wistar rats were randomly divided in-to Sham group ,limbs ischemia-reperfusion (IR) group ,IPC + IR group and Nω-nitro-L-arginine methylester (L-NAME) group . The value of diamine oxidase (DAO) and nitrogen monoxidum (NO) in plasma and gut tissue ,the content of fluorescein isothiocya-nate lipopolysaccharide (FITC-LPS) in plasma ,and the value of cyclic guanosine monophosphate (cGMP) and malondialehyde (MDA) in gut tissue were measured ,respectively .The expression of P-selection and Caspase-3 were measured by immunohisto-chemistry .The apoptosis of intestinal cells was detected by terminal deoxynucleotidyl transferase -mediated dUTP nick end labe-ling (TUNEL) and the results were analysed quantitatively by automatic image analytical system .Results Compared with the LIR group ,the values of DAO and FITC-LPS in plasma decreased obviously in the IPC group ,and the level of NO in plasma increased obviously (P<0 .01) .The values of DAO ,NO and cGMP in gut tissue increased obviously ,the level of MDA and the expression of Caspase-3 and the apoptosis index (AI) were significantly decreased in IPC group (P<0 .01) .Compared with the IPC group ,the value of DAO and FITC-LPS were higher and the level of NO was lower in plasma of the L-NAME group (P<0 .01) .The values of DAO ,NO and cGM P in gut tissue obviously decreased ,the level of MDA significantly increased ,and the expression of Caspase-3 and the AI significantly upregulated in L-NAME group than those in IPC group (P<0 .01) .Conclusion IPC can improve the small intestine injury and the apoptosis following hind limbs ischemia reperfusion which may correlated with the increase in the level of NO .
6.Effects of Phentolamine and Dopamine Combined with Cardiopulmonary Resuscitation on Sudden Cardiac Arrest

Objective:To study the effects of phentolamine and dopamine combined with cardiopulmonary resuscitation on the patients with cardiac arrest.Methods:60 patients with cardiac arrest who were treated from February 2015 to April 2016 in our hospital were selected and divided into the control group and the observation group according to different treatment methods.The control group was treated with routine treatment.while the observation group was treated with phentolamine and dopamine based on the control group.The changes ofcTnI,CK,CK-MB levels,MDA,SOD concentration and hemodynamics were observed before and after treatment.Results:After treatment,the total mortality was 20% in the observation group,which was significantly lower than that of the control group (33.33%,P<0.05).The levels of cTnI in the observation group were significantly lower than those of the control group at 2 h and 24 h after the cardiopulmonary resuscitation(P<0.05).After the cardiopulmonary resuscitation,the serum levels of CK and CK-MB in the observation group were significantly lower than those in the control group (P<0.05).The concentration of MDA and SOD in the observation group were significantly higher compared with the control group [(1.86± 1.65) μg/L vs(3.81± 1.24) μg/L、(6.58± 0.95)μg/L vs(3.74± 0.56) μg/L](P<0.05).After the cardiopulmonary resuscitation,the MAP value of observation group was significantly higher than that of the control group (P<0.05).The CO value and CI value of observation group were significantly increased from the 6th day to the control group (P<0.05),and the PCWP values of observation group were significantly lower than those of the control group (P<0.05).Conclusion:Phentolamine combined with dopamine could effectively improve the cardiac hemodynamics and prognosis of patients with cardiac arrest,reduce the myocardial injury and enhance the survival with high safety.
7.Growth,Siderophore Production and Outer Membrane Protein Expression of Vibrio alginolyticus by Iron Regulation

Iron uptake mechanism of Vibrio alginolyticus was primarily investigated. V.alginolyticus could survive in the medium with high-concentration iron chelator. The strain of V. alginolyticus isolated from diseased fish produced more siderophore than that from marine environment. The extract of siderophore from V. alginolyticus could stimulate the growth of Escherichia coli mutant AN93. Under iron limitation,the growth rate was decreased and several outer membrane proteins were induced. Adding iron into the iron-limited medium the normal growth could be recovered.
8.Content Comparison of Polysaccharide and Protein in Male and Female Trichosanthis Radix

Objective:To compare the contents of polysaccharide and protein in male and female Trichosanthis Radix to provide reasonable explanation for that male plant was better. Methods:Polysaccharides in Trichosanthis Radix were obtained by water-extrac-tion and ethanol-precipitation method, and the content was determined by anthrone-sulfuric acid method. Tichosanthin ( TCS) was ob-tained by hydrochloric acid extraction and acetone precipitation. Based on the light absorption of Coomassie brilliant blue G-250 com-bined with proteins, the content of TCS was detected. Results: The polysaccharides content in male and female Trichosanthis Radix was 2. 628% and 3. 237%, TCS content was 0. 193% and 0. 280%, respectively. Conclusion: The contents of polysaccharides and TCS in male plant are higher than those in female plant. The male plant is a better source for Trichosanthis Radix.
9.Content Comparison of Amino Acid in Female and Male Plants of Trichosanthes Kirilowii

Objective:To compare the content of amino acid in female and male plants of Trichosanthes kirilowii. Methods:Total protein was hydrolyzed by hydrochloric acid, and then determined by HPLC. Results:The percentage of amino acid in female and male plants of Trichosanthes kirilowii was 7. 35% and 7. 04%, respectively. Conclusion:The method provides a new idea for the determi-nation of amino acid in the root of Trichosanthes kirilowii. The total amino acid content in the root of male plants is lower than that of fe-male plants, while the content of alkaline amino acid is opposite. The content of histidine in male plants is about twice as much as that in female plants. The essential amino acid content has no significant difference.
10.Pharmacokinetics of topically applied econazole nitrate nanoparticles in rabbit eye

Background Econazole nitrate is not effective as an antifungal eyedrop because of its poor intraocular permeability,therefore changing the formulation of econazole nitrate to improve its intraocular permeability become a critical point in the treatment of intraocular fungal infection. Objective The present study was to observe the penetration of 0.5% econazole nitrate nanoparticles in the corneas and aqueous humors following its topicaladministration. Methods Econazole nitrate nanoparticles were prepared by quasi-emulsion solvent diffusion.Characteristics and size of nanoparticles were examined with transmission electron microscope and laser scatteringmethod,respectively.Econazole nitrate nanoparticles drops (0.5% )was topically administered in 27 New Zealandwhite rabbits bilaterally,and aqueous humor and corneas were obtained after the application of the eye drops for 5,15,30,45,60,90,120,180,240 minutes respectively to detect the concentration of econazole nitrate with highperformance liquid chromatography (HPLC). The pharmacokinetic parameters were calculated with 3 p97pharmacokinetic computer software.The use of the animals followed the Regulation for the Administration of AffairsConcerning Experimental Animals by State Science and Technology Commission. Results The diameter of thenanoparticles was 50 nm with the round shape and encapsulation efficiency was 96.0%.Econazole nitrate nanoparticlesat the concentration of 0.5% could be rapidly separated with other elements by HPLC with a lowest quantitativeconcentration of 0.1 mg/L.The mean recovery rates of econazole nitrate nanoparticles were 98.09% in cornea and 99.66% in aqueous humor,respectively after topical administration.The peak levels of econazole nitrate nanoparticles in cornea and aqueous humor were achieved at 5 minutes after application ( cornea:40.620 μg/g± 7.756 μg/g;aqueous humor:0.504 mg/L±0.153 mg/L),and its half-life( t1/2 )in cornea and aqueous humor was 23.5 minutes and 18.6 minutes,respectively. Conclusions Econazole nitrate nanoparticles at 0.5% concentration can remain a feasible bioavailability in ocular tissue and therapeutic level in cornea and aqueous humor.
