1.Effects of large dose of ambroxol hydrochloride on lung ischemia reperfusion injury in rats

Objective To evaluate the protective effect of large dose of ambroxol hydrochloride on lung ischemia reperfusion injury in rats and analyze the relationship between ambroxol hydrochloride and pulmonary vascular permeability mediators including EphrinAl, HIF-1α and VEGF at mRNA and protein levels.Methods Rats were operated by the thoracic posterolateral approach.The left lung is-chemia was induced by clamping the hilum of the left inflated lung for 60 minutes, and then followed by a 6-hour reperfusion.Forty rats were divided into ischemia-reperfusion group, ambroxol hydrochloride inter-vention group, sham operation group and blank control group.At the beginning of reperfusion, rats in am-broxol hydrochloride group were infused with ambroxol hydrochloride solution (3.75 mg ·kg~(-1) ·h~(-1)) via femoral vein for six hours.Then, PaO_2, wet-to-dry lung weight ratio, concentrations of IL-1β and TNF-a, activity of myeloperoxidase (MPO), mRNA and protein expressions of EphrinAl, HIF-1a and VECF were detected.Histopathological changes were observed under light microscope.Results (1) Histo-logical observation with light microscope showed significant decrease of diffuse alveolar damage consisting of septal edema, intraalveolar hemorrhage and exsudation in ambroxol hydrochloride group compared with ischemia-reperfusion group.The wet-to-dry lung weight ratio in ambroxol hydrochloride group was de-creased and PaO_2 was elevated significantly.(2) Compared with ischemia-reperfusion group, the concen-trations of IL-1β and TNF-α and activity of MPO in ambroxol hydrochloride group were decreased signifi-cantly.(3) The mRNA and protein expressions of EphrinAl, HIF-la and VEGF in ambroxol hydrochl-oride group did not recover to normal but were down-regulated significantly compared with ischemia-reper-fusion group.Conclusions (1) Large dose of ambroxol hydrochloride can alleviate the LJRI damage by down-regulating the expressions of IL-1β and TNF-α and the activity of MPO in the lung parenchyma.(2) The mRNA and protein expressions of EphrinA1, HIF-1α and VECF can be down-regulated by large dose of ambroxol hydrochloride, which contributes to alleviation of the LJRI damage.The results indicate possible role of gene regulation and fresh pharmacological effect of ambroxol hydrochloride during forma-tion of LIRI.
2.Status quo of stroke rehabilitation care development in our country:a bibliometrics analysis

Objective To analyze the present research situation of stroke rehabilitation nursing in China, find out the defects and shortcomings existing in the development, and lay a good foundation for the development of stroke rehabilitation nursing. Methods The documentation published in Chinese core journals was searched in Chinese National Knowledge Infrastructure (CNKI), VIP database (VIP), Wanfang database by computer retrieval. The publishing year, journal name, scientific research funds, the method of literature research, research objects and research contents were analyzed by bibliometrics. Results A total of 253 articles were included with an increasing trend. These literature distributed in 34 kinds of Chinese core journals, fund rate was relatively good, but very few national fund, the experimental research was given priority to, and the research content was wide. Conclusions In order to better promote the rehabilitation of brain stroke patients, community rehabilitation care needs to be vigorously promoted, emphasis should be paid to the importance of patients family members, at the same time, great importance should also be attached to the combination between disciplines, in order to find better, more effective and easy methods to promote the rehabilitation nursing.
3.Reconstruction and prokaryotic expression of human high mobility group box 1 protein

Objective To design and prepare recombinant mutant human high mobility group box 1 protein (HMGB1s) that can combine with HMGB1 receptors but cannot produce inflammatory effect, and accordingly lead to the creation of a new potential agent for anti-inflammatory therapy. Methods This experiment was based on successful clone and expression of human HMGB1.Six mutant HMGB1 cDNA were designed and constructed by one step inverse PCR. They were cloned into prokaryotic expressive vector pQE80L and followed with production of mutant HMGB1s and identification by Western blotting. Results Six mutant proteins were designed and constructed into prokaryotic expressive vector pQE80L. The recombinant HMGB1 proteins were obtained and identified by Western blotting. Conclusion Human HMGB1 mutants have been successfully constructed and the expression and characterization of intent proteins are identified. It will lay a foundation for further study on biological functions of HMGB1.
4.The effect of nursing intervention on postnral hypotension of aged with heart disease

Objective To study the effect of nursing intervention on postural hypotension of aged with heart disease.Methods Nursing intervention were gived to the 68 aged patients with heart disease and postural hypotension.The effect of nursing intervention on postural hyporension and hypotension associated symptoms were observed.Results The postural hypotension was improved and the incidence of hypotension associated symptoms were reduced after nursing intervention.Differences were statistically significant(P<0.01).Conclusion Nursing intervention was effective on the improvement of postural hypotension.
5.The value of otolith function test in the prognosis of sudden sensorineural hearing.

OBJECTIVE:
To evaluate the characteristics and prognosis of sudden sensorineural hearing loss through vestibular evoked myogenic potentials.
METHOD:
Fifty patients with unilateral sudden hearing loss underwent an ear test battery, including audiometry, cervical vestibular evoked myogenic potential (cVEMP) and ocular vestibular evoked myogenic potential(oVEMP). The cVEMP and oVEMP in patients with sudden sensorineural hearing loss were investigated. Their associations with initial hearing threshold, the type of audiogram, the results of coloric test and hearing recovery were also accessed.
RESULT:
After one-month treatment, the average threshold declined significantly in affected ears, with normal VEMP rates improved significantly (P < 0.05). Initial hearing threshold and hearing recovery were significantly associated with the results of coloric test and the results of VEMP test (P < 0.05).
CONCLUSION
Our study indicated that vestibular evoked myogenic potential examination was not only a useful additional diagnostic tool in the neurotological evaluation of patients suffering sudden hearing loss, but also very valuable in the prognosis of sudden sensorineural hearing loss.

6.Study on inhibitory effect of baicalein on mycoplasma pneumonia and protection mechanism of pulmonary epithelial cells of mice

Objective To investigate the inhibitory effect of baicalein on mycoplasma pneumonia and its the protective mecha -nism in body , and to provide scientific experimental basis for prevention and treatment of mycoplasma pneumoniae infection .Methods The mycoplasma pneumoniae and baicalein treated BALB /c mice lung tissues were stained with hematoxylin eosin (HE), and his-topathological grading .Minimum inhibitory concentration ( MIC) of baicalein on mycoplasma pneumonia was determined by real-time quantitative polymerase chain reaction ( PCR) .The expression of P 1 adhesion molecules mRNA and protein in lung tissue of BALB /c mice was determined with reverse transcription-PCR and Western blot .The expression of epidermal growth factor ( EGF) mRNA and protein in lung tissue was detected by quantitative RT-PCR and immunofluorescence .Results Baicalein significantly reduced the my-coplasma treated mice's lung tissue pathological score .The minimal inhibitory concentration of baicalein was 32 μg/ml.Baicalein sig-nificantly downregulated P 1 gene transcription and protein translation , and upregulated EGF gene transcription and protein expression . Conclusions Baicalein shows significant resistance to mycoplasma pneumoniae , and can protect the body against mycoplasma damage by inhibiting the expression of P 1 protein and promoting the expression of EGF protein .
7.Clinical analysis of intractable epistaxis under nasal endoscope in the treatment of elderly people

Objective To summarize the clinical experience of intractable epistaxis under nasal endoscope in the treatment of elderly people .Methods 312 cases of endoscope of intractable epistaxis patients were retrospectively analyzed.Results Nasal septum and olfactory corresponding head of middle turbinate bleeding site was mainly the upper part of the nasal cavity and the split-plot,back-end middle turbinate at the back of the nose and nasal were fol-lowed,as well as inferior turbinate and the rear end of inferior nasal meatus ,the Littell nasal septum of the front nose were bleeding less .After nasal endoscopic radio hemostatic therapy or local packing treatment with expansive sponge , 310 cases were cured,2 cases were invalid.The total efficiency was up to 99.4%.During the following 3 months,there was no recurrence .Conclusion For the elderly intractable epistaxis hemostatic therapy ,under nasal endoscope oper-ation method,radio hemostatic therapy or local packing treatment with expansive sponge is simple and effective ,which is worthy of popularization and application .
8.Correlation study between the reason of heterotopic ossification after cervical artificial disc replacement and the degeneration of cervical facet joints

Objective To investigate the correlation between the reason of heterotopic ossification after cervical artificial disc replacement and the degeneration of cervical facet joints.Methods From May 2009 to May 2012,133 patients who had undergone cervical artificial disc replacement were included into this study.There were 74 males and 59 females with an average age of 42.63±4.15 years old (range,23-56 years old).There were 109 patients who had undergone single level implant,and 24patients double level implant.Cervical spine A-P and the flexion-extension X-rays were taken to assess the range of motion (ROM)of the surgical level.Degeneration degree of patients' cervical facet joints were evaluated by Park grading standards with the preoperative cervical CT scan images on GE-PACS system.Heterotopic ossification (HO) situation were evaluated by McAfee classification through cervical lateral X-ray film.Patients were divided into two groups (HO group and non-HO group) according to whether heterotopic ossification appeared during the follow-up period.The data were collected before surgery to the latest follow-up.Results The average follow-up time was 2.9 years (range,2.0-4.8 years).HO was detected in 25 patients (18.80%,25/133) at latest follow-up.The ROM of surgical level of patients in HO group was significantly lower than non-HO group (6.8°±3.9° vs 9.1 °±2.4°).In addition,the degeneration degree of cervical facet joints of patients in HO group was significantly higher than that in non-HO group.There was a significant correlation between preoperative cervical facet joint degeneration and the occurrence of HO after CDR (r=0.683,P=0.033).Conclusion There was a significant correlation between preoperative cervical facet joint degeneration and the occurrence of HO after CDR.Patients present HO after surgery have significantly higher degeneration degree of cervical facet joints than patients without HO.Patients with higher degree of HO may have a higher level of cervical facet joints degeneration.
9.Toufeng Capsule's Influence on Concentration of Neurotransmitter and Blood Active Substance in Brain and Blood of Magraine Mice

Objective:To research the influence and mechanism of Toufeng capsule (TFC) on the concentratioin of blood active substance and single amine neurotransmitter in the brain and blood of magraine mice.Methods: We chose animal disease model of migraine made by hypodermic nitroglycerin and measured the concentration of 5 HT. NE and DA in the rat's brain and blood by means of enzymeimmunoassay experimental methods.Results: TFC could obviously increase the concentration of 5 HT and NE in the bloold and of 5 HT, DA and NE in the brain.Conclusions: TFC might improve neurosystematic regulation functions and enhance the neurotransmitters' secretion and metabolism.
10.Stromal cell derived factor-1 promotes the proliferation of bone marrow stem cells:the optimal concentration is 200 μg/L

BACKGROUND:Stromal cel derived factor-1 is a smal molecular protein with a wide range of biological activity that can cause immune cel chemotaxis, and it also has a chemotactic effect on bone marrow stem cels and periodontal ligament cels.
OBJECTIVE:To investigate the effect of stromal cel derived factor-1 with different concentrations on the proliferation of bone marrow stem cels and to probe the best concentration.
METHODS:Bone marrow stem cels from beagle dogs were culturedin vitro and stimulated by different concentrations of stromal cel derived factor-1 (100, 200, 300 μg/L). MTT was used to detect the influence of stromal cel derived factor-1 on the proliferation of bone marrow stem cels so as to screen the best concentration of stromal cel derived factor-1. Then, stromal cel derived factor-1 at the best concentrations was used to intervene the bone marrow stem cels, and MTT was used again to detect the proliferation of bone marrow stem cels.
RESULTS AND CONCLUSION:Stromal cel derived factor-1 at concentrations of 100, 200, 300 μg/L could promote the proliferation of bone marrow stem cels, and the effect was more notable at 200 and 300 μg/Lbut withno significant difference. Therefore, 200 μg/L was considered to be the best concentration of stromal cel derived factor-1 for intervention of bone marrow stem cels. Compared with the blank control group, 200 μg/L stromal cel derived factor-1 could significantly promote the proliferation of bone marrow stem cels. Taken together, stromal cel derived factor-1 can promote the proliferation of bone marrow stem cels, and its best concentration is 200 μg/L.
