1.Experimental study on fixation of clavicular fractures using self-made intramedullary nails

[Objective]To design an intramedullary nail for mid-shaft clavicular fractures based on the anatomic study of human clavicles.[Method]Sixty adult clavicles and 5 fresh cadaveric clavicles were used to survey data about full length,angle between lateral and middle segments,angle between middle and medial segments and the narrowest outer and inner diameter of clavicle.The designed interlocked intramedullary nail was used to fix mid-shaft clavicular fractures.[Result]The full length of clavicle was about(14.82?1.15) cm.The included angle between lateral and middle segment and included angle between middle and medial segment were all about 30?,the narrowest outer diameter of clavicle was about(1.00~0.16) cm,the narrowest inner diameter of clavicle was about(0.50~0.18) cm.After conversely placing guide pin from fracture end,expanding medullary cavity and adjusting the nail,the designed nail was inserted into the medullary cavity from the distal or proximal ends of clavicle,and the proximal and distal ends of the nail were locked by interlocked screws at last.[Conclusion]Treatment of mid-shaft clavicular fractures with the self-made nail has such advantages as less surgical invasion,visible fracture reduction and stable fracture fixation.The biomechanical properties of the nail will be further investigated.
2.Detection of Bacterial Endotoxins in the Levofloxacin Hydrochloride Injection by Limulus Test

OBJECTIVE:To establish the method for assaying bacterial endotoxins in levofloxacin hydrochloride injec?tion.METHODS:The interference test of3batches of levofloxacin hydrochloride injection with2kinds of limulus test agent was studied.RESULTS:The interference between the samples and limulus test agents was eliminable(below1.0mg/ml).The detection results were up to standard.CONCLUSION:The bacterial endotoxin in the sample can be examined by limulus test instead of pyrogen test in rabbits.
3.A biomechanical investigation of reconstruction of anterior cruciate ligament

Objective To investigate two fixation methods for reconstruction of anterior cruciate ligament(ACL)with autogenous patellar tendon graft and their instant stability.Methods 15 specimens were divided into three groups.The first group was treated with 1/3 bone-patellar ligament-bone replacement and in terference screw fixation,the second group was treated with 1/3 bone-patellar ligament and Krackow locked bilateral suture,and in the third group the intact ACL was not treated.The mono-axial tensile tests were performed on a versatile hydraulic material testing machine(WE-5A)to compare the linear load,maximum load,linear straining,maximum straining,linear displacement,maximum displacement,rupture displacement,destructive energy,elastic modulus,and ultimate strength of ACL in the three groups.Results The ACL stability and rigidity in the group of 1/3 bone-patellar-bone replacement and interference screw fixation were higher than those in the other two groups.The differences between test results were statistically significant.Conclusion The ACL reconstruction with 1/3 bone-patellar ligament-bone replacement and interference screw fixation is a good method to ensure higher strength of graft and better joint stability.
4.ANALYSIS AND CHARACTERIZATION OF CAROTENOIDS IN GINGKO SEED

Objective: To analyze and identify the carotenoid in gingko seed.Methods: The chemical composition of the pigment from gingko seed was identified as carotenoid by UV absorption spectrum. Its content was determined by spectrophotometric method and its composition was analyzed and identified by thin-layer chromatography and HPLC method.Results: Gingko contained carotenoids, roughly about 489 ?g/100 g in which lutein, ?-carotene and ?-carotene amounted to 69.20%,15.80% and 7.45% respectively. Conclusion: Gingko contained large quantity of carotenoids, mainly lutein, and next in order ?-carotene and ?-carotene.
5.Effect of isoflurane on memory of rats and expression of acetylcholine receptor in hippocampus

Objective To observe the change in memory of aged rats and the expression of alpha 4 acetylcholine receptor in hippocampus 5 h after inhalation of 1.2% isoflurane.Methods Fifty healthy adult and 50 aged SD rats were randomly divided into aged normal control group(A0),aged water labyrinth normal control group(WA0),1 day isoflurane aged water labyrinth group(WA1),3-day isoflurane aged water labyrinth group(WA3),7-day isoflurane aged water labyrinth group(WA7),adult normal control group(B0), adult water labyrinth normal control group(WB0),adult 1 day isoflurane water labyrinth group(WB1),3-day isoflurane water labyrinth group(WB3),and 7-day isoflurane water labyrinth group(WB7),10 rats in each group.Memory of rats in WA1,WA3,WA7 groups was detected 5 h after inhalation of 1.2% isoflurane while memory in rats of WB1,WB3 and WB7 groups was detected during the Morris water maze testing.The rats in A0,WA0,WA7,B0,WB0 and WB7 groups were killed on day 7 to detect the expression of ?4 acetylcholine receptor by Westernblotting for observation of changes in distribution of positive immune reaction cells in hippocampus.Results Water maze test showed that there was no significant difference in expression of alpha 4 acetylcholine receptor between WA0 and WB0 groups.However,the expression level of alpha 4 acetylcholine receptor was significantly lower in latent period of WA1 than in latent period of WA0,WB0 and WB7 groups(P
6.Biomechanical study on clavicle intramedullary nail

ObjectiveTo discuss the biomechanical properties of clavicle intramedullarynail in fixing middle fracture of clavicle and compare with the traditional plate and screw and single Kirschner wire,in order to provide scientific evidence for clinical application.MethodsFifteen adult cadaver were collected to make middle fracture of clavicle models.According to the clinical operation methods,the fracture models were divided into three groups with 5 specimens each:clavicle intramedullary nail group,plate and screwfixation group,and Kirschner wire internal fixation group.Biomechanical properties of three groups were tested and compared by experimental stress analysis method.ResultsIn the load 1000 N,the bending strength and bending stiffness of clavicle intramedullary nail group[ (57.41±4.72),( 2.28±0.17) N ] had no significant difference compared with those of plate and screw fixation group[(58.50 ±t4.14),(2.26 ±0.20) N ] (P > 0.05),while had statistical significance compared with Kirschner wire internal fixation group [(51.14±4.32),(1.54±0.16) N] (P < 0.05).The bending strength and bending stiffness had statistical significance between plate and screw fixation group and Kirschner wire internal fixation group (P< 0.05).In the torsional mechanical test,torque of clavicle intramedullary nail group had no significant difference compared with plate and screw fixation group (P > 0.05 ),but had statistical significance compared with Kirschner wire internal fixation group (F =14.88,P <0.05).In compression test of the clavicle,the compression limit load of clavicle intramedullary nail grotup [ ( 1246±87) N ] had no statistical significance compared with plate and screw fixation group [ ( 1348±121 ) N ] ( P > 0.05 ),and had significant difference compared with Kirschner wire internal fixation group [ ( 870±72 ) N ] (F =14.74,P < 0.05 ).Conclusions Clavicle intramedullary nailing for treating middle fracture of clavicle can provide better biomechanical strength,stiffness and stability.Meanwhile as a minimally invasive operation,this operation method is feasible.Compared to other internal fixation methods,it has certain advantages.
7.Effects of isoflurance on learning and memory function and receptor for advanced glycation end products expression of hippocampus in rats

Objective To explore the effect of isofluranee on receptor for advanced glycation end products (RAGE) expression of hippocampus and learning and memory function in rats. Methods 24-month Sprague-Dawley male rats (n=45) and 4-month Sprague-Dawley male rats (n=45) were randomly divided into six groups with 15 rats each group. Group C1 (aged control group),group C2(adult control group) breath 30% oxygen and air mixed gas; Group S1(single inhalation of isoflurane aged group),Group S2(single inhalation of isoflurane adult group)were anesthetized with 1.5% isoflurane,breath 30% oxygen and air mixed gas for 2h;Group R1(Repeated inhalation of isoflurane aged group), group R2(Repeated inhalation of isoflurane adult group) were anesthetized with 1.5% isoflurane,breath 30% oxygen and air mixed gas 2h a day for three days. Eight rats randomly selected from each group were killed and their hippocampus were immediately isolated for detection of RAGE expression by immunohistochemistry and RT-PCR after accomplished treatment 24h. The remained rats' learning and memory function were assessed using Morris water-maze test. Results The results of Morris water-maze test showed that the times of acerossing the original platform and the time consumption of staying the original platform quadrant was shorter in group S1 and group R1,but the escape latency was longer than group C1(escape latency C1 (9.42± 2.63)s,S1(13.20±3.85)s,R1(17.20±3.44)s, F=12.773, P<0.05). In the group R2,the escape latency was longer but the times of accrossing the original platform and the time consumption of staying the original plat-form quadrant was shorter than group C2 (times of accrossing the original platform C1(7.30±2.40), S1(3.90± 2.42),R1(3.44±2.40), F=7.448, P<0.01).To contrast with the group C2,there were no significant differ-enees in spatial probe test in the group S2(P>0.05). The levels of mRNA and protein of RAGE in hippocampus was significantly higher in group S1 and group R1 than group C1(RAGE mRNA expression C1(0.11±0.02),S1 (0.56±0.09), R1(0.73±0.14), F=179.447, P<0.01). To contrast with the group C2, there were no differ-ences found in the levels of mRNA and protein of RAGE in group S1(P>0.05), but it was higher in the group R2 (RAGE mRNA express C2(0.22±0.04), R2 (0.41±0.08), F=40. 209, P < 0. 01). Conclusion Isoflurane can reduce learning and memory function in both aged and adult rats, but aged rats are particularly significant im-pacted. This effect may be induced by the increase of RAGE expression in hippocampas.
8.Effects of isoflurane on expression of receptor for advanced glycation endproducts in hippocampus in rats

Objective To investigate the effects of isoflurane on receptor for advanced glycation endproducts (R(A)GE) expression in the hippocampus in rats. Methods Forty-five male 4-month-old and 45 male 24-month-old rats were used in this study. The animals were divided into 2 age groups ( n = 45 each): the aged group (group O) and the adult group (group A). Each group was further divided into 3 subgroups ( n = 15 each):Ⅰ control subgroup (group OC,AC) inhaled 30% O2 in air; 1 single isoflurane inhalation subgroup (group OS,AS) inhaled 1.5 % isoflurane for 2 h and Ⅲ repeated isoflurane inhalation subgroup (group OR, AR) inhaled 1.5 % isoflurane 2 h per day for 3 days. One day after isoflurane inhalation, learning and memory function was assessed using Morris water maze test in 8 animals in each subgroup. The rest of each subgroup were killed and their hippocampi were immediately isolataed for detection of RAGE mRNA and protein expression by RT-PCR and immuno-histochemistry. Results The cognitive function was impaired after signle or repeataed isoflurane anesthesia as compared with control animals in both aged and adult groups. The expression of RACE mRNA and protein in hippocampus was significantly increased after either single or repeated isoflurane anesthesia in aged group but only after repeated isoflurane anesthesia in adult gpoup. There was no significant difference in RAGE mRNA and protein expression in the hippocampus between control and single isoflurane inhalation animals in adult group. Conclusion Isoflurane can reduce learning and memory function in both aged and adult rats by increasing RAGE expression in hippocampus especially in aged rats.
9.Effects of inhalation of different concentrations of isoflurane on hippocampal cytochrome c expression in aged rats

Objective To investigate the effects of inhalation of different concentrations of isoflurane on the expression of cytochrome c ( Cyt c) in hippocampus in aged rats.Methods Sixty-three aged male SD rats (20 months) weighing 500-600 g were randomly divided into 3 groups(n=21each):control group inhaling 30%O2 for 2h (group C) and 2 isoflurane groups anesthetized with 0.75 % and 1.5 % isoflurane in 30 % O2 for 2 h respectively (group Ⅰ1 and Ⅰ2 ).Arterial blood samples were obtained from 5 rats at 30 min, 1 and 2 h of anesthesia for blood gas analysis. Eight animals were killed at 24 h after anesthesia in each group.Their hippocampi were immediately removed for determination of Gyt c expression by immuno-histochemistry and Western blot analysis.Cognitive function was assessed by Morris water maze test the day before experiment and once a day for 6 consecutive days starting from the 1st postoperative day.Results The Cyt c expression in hippocampus was significantly increased in Ⅰ1 and Ⅰ2 groups in a concentration-dependent manner as compared with group C.The escape latency was significantly prolonged and the frequency of crossing the original platform and the duration of staying at the original platform quadrant were decreased in group Ⅰ1 and Ⅰ2 compared with group C.Conclusion Inhalation of isoflurane anesthesia can decrease cognitive function through up-regulating the Gyt c expression in hippocampus in aged rats.
10.Expressions of miRNA27a and miRNA451 in ovarian cancer and breast cancer cells and its correlation with drug resistance by targeting MDR1/P-glycoprotein

Background and purpose: Resistance of cancer cells to chemotherapy is a major clinical obstacle to successful treatment and leads to poor prognosis for the patients. MicroRNA (miRNA) plays a vital role in tumor cells response to chemotherapeutic agents. This study plans to investigate the expressions of miRNA27a and miRNA451 in ovarian cancer and breast cancer cells and its correlation with drug resistance. Methods:A2780/Taxol cells were established using stepwise selection;Stem-loop quantitative real-time PCR (stem-loop RT-PCR) was used to detect expression of miRNA27a and miRNA451 in ovarian cancer and breast cancer cells. The A2780 and A2780/Taxol cells were transfected with the mimics or inhibitors of miRNA27a or negative control (NC) RNA and the mimics of miRNA451 or NC were transfected into MCF-7/ADM cells by LipofectamineTM 2000. The expression levels of MDR1 mRNA and P-glycoprotein (P-gp) were examined using RT-PCR and Western blot respectively. MTT method was used to analyze drug sensitivity. Results:The expression of miRNA27a was an average of (2.2±0.30) times higher in A2780/Taxol cells than in A2780 cells, with a significant difference between the two groups (P<0.05). The expression of miRNA451 was lower by 84%in MCF-7/ADM cells than in MCF-7 cells, with a signiifcant difference between the two groups (P<0.05). A2780/Taxol cells transfection with inhibitors of miRNA27a showed that the levels of MDR1 mRNA was decreased by (39±0.14)%, P-gp level [(26±5.3)%] decreased compared with the NC group [(43±6.7)%], the IC50 (0.53μmol/L) was less than the NC group (6.8μmol/L), and there was a signiifcant difference between two groups (P<0.05). Transfection of A2780 cells with mimics of miRNA27a led to increase MDR1 mRNA expression by (121±0.11)%and decrease the sensitivity to paclitaxel (IC50:0.2μmol/L vs 0.06μmol/L). There was a signiifcant difference between two groups (P<0.05). Transfection of MCF-7/ADM cells with mimics of miRNA451 showed that expression of MDR1 mRNA was decreased by (65±12)%, P-gp [(31±19)%] was less than the NC group [(83±12)%], the sensitivity of cells to adriamycin enhanced and the IC50 of adriamycin (4.61μmol/L) was less than the NC group (26μmol/L), and there was a signiifcant difference between two groups (P<0.05). Conclusion:The expressions of miRNA27a and miRNA451 are deregulated in A2780/Taxol and MCF-7/ADM cells respectively, which may play vital roles in drug resistance by regulating MDR1/P-gp expression directly or indirectly.
