1.Effect of Shiwei Longdanhua particles on serum inflammatory factors and immune function in elderly patients with acute bronchitis

Objective To observe the effect on serum inflammatory factors and immune function of Shiwei Longdanhua particles in elderly patients with acute bronchitis.Methods 120 cases of elderly patients with acute bronchitis from November 2014 to October 2016 were selected and randomly divided into the observation group and the control group with 60 cases in each group, according the order of single and double number.The control group was given conventional treatment, and the observation group was given Shiwei Longdanhua particles based on the control group.Treatment for seven days, to observe the differences of serum inflammatory factors (hs-CRP, TNF-αand IL-6, IL-1) and immunologic function indexes (CD3 +, CD4 +, CD8 +, CD4 +/CD8 +and IgM, IgG, IgA) between the two groups.Results After treatment, hs-CRP, TNF-αand IL-6, IL-1 of the two groups decreased with before treatment, the difference was statistically significant (P<0.05), and all indexes of the observation group were lower than those of the control group, the difference was statistically significant ( P<0.05 ) .After treatment, CD3 +, CD4 +, CD4 +/CD8 + of the two groups increased with before treatment except CD8 +decreased, the difference was statistically significant (P<0.05), there was no significant difference between the control group and before treatment, there was significant difference between the two groups after treatment (P<0.05).After treatment, IgM, IgG and IgA of the two groups decreased with before treatment, the difference was statistically significant (P<0.05), and all indexes of the observation group were lower than those of the control group, with significant differences, the difference was statistically significant (P<0.05).Conclusion Shiwei Longdanhua particles has the effects of inhibiting inflammatory reaction and improving the immune status in the treatment of elderly patients with acute bronchitis .
2.Culture and differentiation of cynomolgus monkey neural stem cells

BACKGROUND:Studies regarding neural stem cells (NSCs) mainly focus on rodents and human,few of which addressing non-human primate animals.OBJECTIVE:To explore a culture system of cynomolgus monkey NSCs (cmNSCs),additionally,to observe its differential potential.DESIGN,TIME AND SETTING:An in vitro cytology observation was performed at the Canter for Stem Cell Biology and Tissue Engineering,Sun Yat-sen University between July 2008 and April 2009.MATERIALS:Spontaneously aborted cynomolgus monkey fetus with 3-5 months old (gestational age),was provided by Landao Biotechnology Co.,Ltd.METHODS:The subventricular zone and the hippocampus were dissected aseptically from the brain of spontaneously aborted cynomolgus monkey fetus,then triturated with fire-polished glass Pasteur pipettes to single cells,and cultured in 25 cm~2 culture flask with concentration of (2.0-5.0)×10~5.The neurospheres were transferred into polyornithine and fibronectin precoated cell culture dish after 2 weeks when they formed.Each 20 μg/L of epithelium growth factor (EGF) and basic fibroblast growth factor (bFGF) was daily added.The adhesive cells were passagad when they reached about 90% confluency.MAIN OUTCOME MEASURES:The NSCs and its differential potential were identified by surface marker nestin and immunocytochemistry.RESULTS:The neurospheres formed after 1 week culture,and grew with time prolonged.The culture system was changed into adherent culture at 4th week.At 24 hours after plated on the poly-omithine/fibronectin pre-coated dish,neurospheras adhered to the dish and many cells migrated out of neurnshperes and proliferated as monolayer.The immunocytochemistry showed that NSCs which culture medium contain EGF and bFGF preserve neatin positive,and the cmNSCs that without EGF and bFGF differentiated into astrocytes (GFAP positive),neurons (Tuj-1 positive) and oligodendrocytes (O4 positive).CONCLUSION:The culture system which combines suspension culture and adherent culture is fit for the culture of cmNSCs.The cultured cmNSCs has potential to differentiate into astrocytes,neuronal cells and oligodendrocytes.

5. Protective effect of Pterocarya hupehensis Skan extract on the immune liver injury induced by concanavalin A

Objective: To investigate the protective effect and mechanism of Pterocarya hupehensis Skan extract on liver injury induced by concanavalin A (conA). Methods:Forty C57BL/6 male mice were randomly divided into 5 groups: normal group, model group, and groups of low-, medium- and high-dose extract, with 8 mice in each group. Except for the normal group that received intravenous injection of normal saline, the other mice were injected with conA (12 mg/kg) for replication of the immunological liver injury model. The treatment groups were intragastrically administered with corresponding concentrations of extract. After 12 hours, the mice were subjected to eyeball extraction and dissection of liver tissue to prepare HE stained sections. The liver index was collected and calculated. Serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) were also detected. The content of STAT-3 and the level of phosphorylation in liver tissue were also tested. Results: HE staining showed that the liver tissue of the model group was significantly damaged compared with the normal group, and that of the treatment groups were ameliorated. Compared with the model group, each treatment group had reduced liver index and serum ALT, AST, IL-6 and TNF-α levels (P<0.05 or P<0.01). Significant phosphorylation of STAT-3 was observed in the model group, but it was inhibited in the treatment groups. Conclusion: The extract of Pterocarya hupehensis Skan can effectively treat immunological liver injury induced by conA, which may be related to the inhibition of phosphorylation of STAT-3.

Objective To investigate the surgical procedure of restoring stenosed hilar bile duct by gallbladder lamella with cystic artery.Methods Based on extensive incising and hilar cholangioplasty,stenosed hilar bile duct was restored by gallbladder lamella with cystic artery.Results Among all the 12 pa tients,including 10 intrahepatic bile duct calculi patients,1 right hepatic duct injury and 1 left intrahepatic chaolangiocarcinoma,no cystic fistula,reflexed cholangitis and recurrent stone were noticed by post operative follow-up.Conclusion This procedure is a ideal method by remaining the normal function of Oddis sphincter while removing stenosis.

9.Application of video-based teaching in the training of nursing operation skills for newly-contracted nurses

Objective To investigate the effect of video-based teaching in the training of nursing operation for newly-contracted nurses. Methods One hundred and twenty one nurses newly recruited in September 2011 to September 2012 were set as the control group, another 128 in October 2012 to October 2013 were set as the experiment group. The former were trained and assessed with traditional training method and the latter were trained for 1 year in video-based teaching methodology. After training, both groups were examined about their operation skills and meanwhile a survey on video-based teaching was conducted. Result After training, the results in operating skills evaluation in the experiment group were significantly better those of the control group (Z=2.82, P<0.05). Conclusion Compared with traditional training method, the video-based teaching can raise the level of nursing operation skills and significantly improve the quality of nursing, thus worthy of popularization and application.
10.Role of multi-modality magnetic resonance imaging for early diagnosis and response evaluation of neoadjuvant chemotherapy in inflammatory breast cancer

Objective:To investigate the characteristics of magnetic resonance imaging and clinical application of multi-modality magnetic resonance imaging (MRI) for evaluating the response of neoadjuvant chemotherapy (NACT) in inflammatory breast cancer (IBC).Methods:A total of 36 IBC patients were enrolled in the study.The morphological, hemodynamic and diffusion-weighted imaging features of MRI were analyzed. Eleven patients underwent MRI examination before and after NAT. The imaging changes were analyzed and the efficacy of NACT was evaluated.Results:There were 38 identified breast carcinoma in these 36 cases, among which abnormal skin thickening and enhancement, extensive edema was found in 37 breast lesions. Enhancement of breast lesions in 25 cases was non-mass-like enhancement. Diffusion limitation was found in all lesions. The number of vessels in affected side was more than that in healthy side in MIP images. Thirty three cases had axillary lymph node enlargement.The sensitivity and specificity of MRI in evaluating residual breast tumors and vascular thrombus were high, but the evaluation of axillary lymph nodes was relatively low.Conclusions:Multi-modal MRI can be used for early and accurate diagnosis of IBC. It can also be used to predict and evaluate the effect of neoadjuvant chemotherapy.
