There are some problems in the teaching of medical physics course,such as low learning initiative,deficient grasp of concept,and obstacles to realize the internalization and practical application of knowledge.The PAD (presentation-assimilation-discussion) class is applied in the course,in order to improve the students' learning state.PAD class is a teaching mode proposed from a psychological point,which advocates that teaching is the first before students start to learn and has an equal treatment on students' discussions and teachers' instruction,supporting independent learning.The teaching measures include:selecting course content to reach the balance between simplification and detailed interpretation;assigning open homework based on the course;dividing the class discussion into four steps;assessing students based on the performances of homework and final examinations.The research has shown that the PAD class facilitates improving the students' initiative of study and promoting the absorption and application of knowledge.

The high molecular weight genomic DNA of yeast was extracted using three methods.Products were separated on agarose gel electrophoresis,quantified by spectrophotometer ND-1000 and restricted by EcoRⅠand MesⅠ.The result was shown that the genomic DNA extracted by modified benzyl chloride method was the best.The products of wild isolates supported it,too.This method was suitable for restriction of genomic DNA from yeast.

A quantitative real-time PCR assay was developed to measure the proviral load of human T-lymphotropic virus type I (HTLV-I) in peripheral blood. The technology utilizes special primers and Taqman MGB fluorescence probe to measure amplification products from the gag-pro-pol polyprotein gene of HTLV-I. HTLV-I copy number was normalized to the amount of cellular DNA by quantitation of the beta-actin gene, The amplification system was sensitive to detect 5 copy/microL. The standard curve had a good linearity when the quantity for the gene was between 10(3) and 10(7) copy/microL (R2 = 0.999). Good reproducibility was observed in each intra- and inter-assay. We also measured proviral load in peripheral blood in 12 HTLV-I seropositive former blood donors. Proviral load for HTLV-I infected donors ranged from 0.015 to 12.819 copy/cell in WBC with the mean of 3.116 copy/cell.

The research for efficient and low-cost electrocatalysts for water splitting are essential for the exploitation and application of hydrogen energy. Transitional metal phosphides are considered as one of the most promising bifunctional water splitting electrocatalysts. Herein, we reported a facile two-step method (firstly hydrothermal preparation, then phosphorization under low temperature) to synthesize the bead-chain like nanoarrays of CoP supported on three dimensional Nickel foam (CoP/NF). The as-prepared CoP/NF could act as an efficient bifunctional catalyst for overall water splitting. The catalyst exhibited remarkable activity for both the oxygen evolution reaction (OER) and hydrogen evolution reaction (HER) in alkaline media,delivering a current density of 10 mA/cm2at an overpotential of 281 mV for OER and 95 mV for HER, respectively. Efficient water splitting in alkaline system was realized by applying a two-electrode system with the CoP/NF acting as both the anode and cathode. The applied potential was 1.63 V to obtain the current density of 10 mA/cm2,and good stability was also testified.

Filamin A and 14-3-3-σ are closely associated with the development of breast cancer.However,the exact relationship between them is still unknown.The present study aimed to examine the interaction of filamin A with 14-3-3-σ in the invasion and migration of breast cancer.RNA interference technology was employed to silence filamin A in MDA-MB-231 cells.Real-time PCR and Western blotting were used to detect the expression of filamin A and 14-3-3-σ at mRNA and protein levels,respectively.Double immunofluorescence was applied to show their colocalization morphologically.Wound healing assay and Trans-well assay were used to testify the migration and invasion of MDA-MB-231 cells in filamin A-silenced cells.The results showed that silencing filamin A significantly increased the mRNA and protein levels of 14-3-3σ.In addition,double immunofluorescence displayed that filamin A and 14-3-3σ were predominantly colocalized in the cytoplasm of MDA-MB-231 cells.Silencing filamin A led to the enhanced fluorescence of 14-3-3σ.Furthermore,cell functional experiments showed that silencing filamin A inhibited the migration and invasion of MDA-MB-231 cells in vitro.In conclusion,silencing filamin A may inhibit the invasion and migration of breast cancer cells by upregulating 14-3-3σ.

Objective To construct double gene deletions at the htrA and clpP loci on the chromosome of Streptococcus mutans(Sm) and to remove the antibiotic resistance markers with the Cre-loxP*site-specific recombination system. Methods The htrA gene was cloned into the pGEM-T-Easy TA cloning vector and then inactivated via the insertion of a kanamycin resistance cassette(lox71-Km-lox66), yielding pGEM-T-△htrA/Km for deleting the htrA gene. Using the same method, the pGEM-T-△clpP/Sp was constructed for deleting the clpP gene. Following the transformation of pGEM-T-△htrA/Km in Sm, the homologous recombination event was selected. One such mutant was transformed with a cre expression plasmid (pCrePA). The kanamycin resistance gene was then excised. The pCrePA was then easily eliminated at nonpermissive temperatures, resulting in a mutant strain (MS △ htrA) carrying a deletion at the htrA loci without a selectable marker. This mutant was verified by PCR and DNA sequencing. Then, the clpP and spectinomycin resistance gene were deleted from MS △htrA, yielding markerless mutant strain lacking clpP and htrA. Results The deletion of htrA, clpP and antibiotic resistance markers were confirmed by PCR analysis and DNA sequencing. Conclusions A mutant of Sm was constructed successfully which contained a deletion of the htrA and clpP gene without selectable marker. The Cre-loxP*system can be applied to Sm, which provides experimental evidence for generating markerless multiple gene deletion mutants.

Objective To explore the effect of family-centered nursing on family function and family burden of schizophrenic patients.Methods A total of 108 schizophrenic patients with the whole course less than 5 years were randomly divided into study group(n =53)and control group(n =55).The two groups received the routine antipsychotic drugs maintenance therapy and the general psychological education.In addition,the study group received the family-centered nursing intervention,and the total following-up course was 1 year.To evaluate the family situation and psychotic symptoms of patients,all the participants were assessed by using Family Assessment Device (FAD),Family Burden Scale of Disease (FBS) and Brief Psychiatric Rating Scale (BPRS) at entry and 1 year after discharged.Results There was no statistically significant difference between the two groups in scores of BPRS,FAD and FBS at baseline (P ＞ 0.05).After 1 year,the scores of BPRS (24.60 ± 7.87),total function of FAD (26.8 ± 3.7) and FBS (20.60 ± 9.91) in the study group were lower than those in the control group:BPRS (28.91 ± 8.72),FAD (30.1 ± 3.4) and FBS (30.76 ± 12.27),and the difference was statistically significant (t =2.61,4.67,4.57,respectively; P ＜0.05).The scores of each factor of FAD and four domains of FBS (such as family economic burden,family daily activities,familyent ertainment and mental health of family member) of the study group were lower than those of the control group (P ＜ 0.05).Conclusions The family-centered nursing model is helpful to improve family function,the prognosis of the disease and to decrease family burden of schizophrenia.

Lattice strain ruthenium nanoparticles uniformly and stably supported on nitrogen-modified carbon nanosheets(RuNPs/NC)were prepared via simple wet-chemical and subsequent pyrolysis method.The nitrogen doped NC could effectively improve their uniform dispersion and lattice compression of RuNPs.Through changing the pyrolysis temperature,the nitrogen content,types and degree of lattice strain of RuNPs could be effectively tuned,which could be used to adjust and control their peroxidase-like activity.The as-prepared RuNPs/NC-900 exhibited highest peroxidase-like activity,and could catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine(TMB)to produce a blue product with the maximum absorption at 652 nm in the presence of H2O2.The steady-state kinetic analysis indicated that the reaction catalyzed by RuNPs/NC followed the Michaelis-Menten kinetic model.Tannic acid(TA),gallic acid(GA)and ascorbic acid(AA)could effectively inhibit the RuNPs/NC-H2O2-triggered chromogenic reaction of TMB,resulting in color fading and decrease in absorbance.Based on this,a sensitive and accurate system was constructed for detection of TA,GA and AA.The detection limits(3σ/S)for TA,GA and AA were 0.014,0.014 and 0.29 μmol/L,respectively.This study not only developed a colorimetric sensing method based on RuNPs/NC nanozyme but also offered a new approach for the sensitive detection of antioxidants in food.

OBJECTIVETo validate the value of the Oxford classification of IgA nephropathy in predicting the renal outcome in Chinese population.

METHODSRetrospective study was done in patients with IgA nephropathy. All slides were re-assessed according to the Oxford classification of IgA nephropathy. The primary end point is doubling serum creatinine, or a 50% reduction in estimated glomerular filtration rate (eGFR), or end-stage renal disease. Pathologic predictors for the progression to the end point were determined by univariate and multivariate Cox regression.

RESULTSTotally 533 patients were enrolled in the study. During the follow-up (median: 39 months; range: 12-263 months), 5.07% of the patients reached the end point. While tubular atrophy and interstitial fibrosis and arterial/ arteriolar lesion were associated with the endpoint in univariate analysis, only the T score was predictive of the renal outcome in multivariate Cox regression. Combination of the patho- logic lesions had no impact on renal outcome.

CONCLUSIONAccording to the Oxford classification of IgA nephropathy, the degree of tubulointerstitial fibrosis is the only feature independently predictive of renal outcome.

A water-soluble fluorescent probe (7-diethyl amino-3-formaldehyde coumarin) for Fe3+ detection was designed and synthesiZed, and its structure was confirmed by 1 HNMR, 13 CNMR and MS spectra.This probe showed high emission intensity at 471 nm.With the continuous addition of Fe3+, the emission intensities at 471 nm decreased gradually, and showed an excellent linearity with Fe3+in the range of 0.02-60 μmol/L, and the regression equation was I=322.56-4.86CFe3+.This probe was able to detect Fe3+ qualitatively and quantitatively with the detection limit as low as 22 nmol/L.Besides, this probe showed high sensitivity to Fe3+over other metal ions.The detection process was reversible, which could be recycled for the Fe3+detection.In terms of good optical properties and the strong fluorescence in physiological pH, the probe was successfully applied in imaging Fe3+ of living Ramos cells.
