1.Research progress of anti-VEGF for the therapy in wet age - related macular degeneration

Wet age-related macular degeneration is the leading cause of the severe, irreversible vision loss in individuals over the age of 65 years. Anti-VEGF therapy has been shown to play a key role in the pathogenesis of wAMD, which is the front-line therapy admittedly. lt has no clear steady curative effect for some patients even if they accepted repeating Anti-VEGF therapy. For the purpose of visual acuity improved more, a steady flow of new therapy has emerged, such as function towards the same or different targets of antiangiogenesis, consolidating the effect by combination therapy, improving or simplifying the mode of administration, etc. This paper gives a brief review of the progress of anti-VEGF for the therapy in wet age-related macular degeneration.
2.Effects of resveratrol on high glucose memory-induced injury to endothelial cell

Cultured human umbilical vein endothelial cells (HUVECs) were divided and assigned to 6 groups:normal control group,mannitol control group,high glucose group,low dose resveratrol group,medium dose resveratrol group,and high dose resveratrol group.HUVECs were pretreated with or without 30 mmol/L glucose plus various concentrations of resveratrol (0.1,1,10 μmol/L) for 16 hours and then incubated with 5.5 mmol/L glucose for 6 days.The levels of vascular cell adhesion molecule 1 (VCAM-1),monocyte chemotactic protein 1 (MCP-1),and plasminogen activator inhibitor 1 (PAI-1) in the culture supernatant were measured by ELISA.NF-κB expression was detected by Western blot.Compared with normal glucose group,high glucose up-regulated the expression of NF-κB,along with the increased levels of VCAM-1,MCP-1,and PAI-1 (all P<0.05),which were still maintained at high levels even after withdrawal of high glucose.Resveratrol treatment down-regulated the expression of NF-κB and lowered the levels of VCAM-1,MCP-1,and PAI-1 (all P<0.05).These results suggest that resveratrol may decrease the secretion of VCAM-1,MCP-1,and PAI-1 and prevent high glucose memory-induced injury to endothelial cell via NF-κB pathway.

4.Effects of profound hemodilution with 6% HES on coagulation and platelet function curve

Objective To study the effects of profound hemodilution with 6% HES (200/0.5) on coagulation.Methods Ten male new Zealand long-ear rabbits weighing (2.43 ?0.19) kg were anesthetized with intravenous thiopentone 8 mg-1 and tracheotomized and mechanically ventilated (VT = 15 ml-1 , RR = 24 bpm) . Anesthesia was maintained with intravenous infusion of thiopentone and succinylcholine. Femoral artery and vein were cannulated for BP and CVP monitoring. Hemodilution was performed in 6 steps at 30 min intervals. Blood was withdrawn from artery and simultaneously replaced by intravenous infusion of equal volume of 6% HES until Hct was 5 % -8 % . Blood samples were taken before hemodilution and 30 min after each step of hemodilution for determination of coagulation function curve using Sonoclot coagulation and platelet function analyzer. Results When Hct was 25%-30% there was no significant difference in coagulation function before and after hemodilution. 5.Construction of recombinant adeno-associated virus vector with human preproenkephalin gene

Objective To construct the recombinant adeno-associated virus vector with human preproenkephalin gene (rAAV-hPPE). Methods The human preproenkephalin (hPPE) gene was cloned into the adeno-associated virus (AAV) vector plasmid pSNAV which contained neo expression box. The recombinant pSNAV-hPPE was then transfected into BHK cells using lipofectamine?2000. The G418-resistant cells, BHK / SH1, were obtained. The BHK / SH1 cells were infected with HSV1-rc /△UL2 which has the function of packaging the recombinant AAV(rAAV) . After purification, the construction of rAAV-hPPE was achieved.Results The construction of pSNAV-hPPE was confirmed by digestion with restriction enzyme. Southeon bolting was used to detect the virus liters (2.5 ? 1212 v.g /ml) .Conclusion This rAAV-hPPE virus vector with high liter and strong infectivity can be used in transgenic analgesic research.

Objective To study the expression of p73 gene in lung cancer and tumorigenesis, progression of lung cancer in the elderly. Methods The expression of p73 protein in 65 cases of lung cancer, adjacent tissues of cancer and normal tissues was determined by immunohistochemical SABC method. The results were analysed by combining the clinicopathological data and the prognosis. 7.Retinal pigment epithelial and choroidal atrophy in wet age-related macular degeneration treated with multiple transpupillary themotherapy:A case report

AIM: To report a case of wet age-related macular degeneration (AMD) treat with multiple transpupillary themotherapy (TTT) and choroidal neovascularization (CNV) disappeared but retinal pigment epithelial(PRE) and choroidal atrophy occurred with a low vision at the end point.METHODS: Clinical data including fundus hotographs, fundus fluorescein angiography (FFA), indocyanine angiography (ICGA) and optical coherence tomography(OCT) was reviewed.RESULTS: A 72-year-old man complained about blurred vision of his left eye and FFA revealed polypoidal choroidal vasculopathy (PCV) in the macula. His left eye had stable vision of 0.1 for 6 years without any treatment of CNV. About 2 years later, his right eye presented a piece of CNV. During the period of 3 years, the lesion remained more (3×5 PD) and less (1×2PD) in size with remarkable exudates and bleeding, and 7 sessions of TTT were applied with 80-280mW, 2mm of spot, and 60 seconds exposure and with the interval of 3 months or more. The CNV lesion finally disappeared, but there left a white area in the macula and vision decreased from 0.3 to 0.04.CONCLUSION: Although CNV lesion can be eliminated by TTT, obvious atrophy of RPE cells and the choroids can happen and this may not be of help for patient vision. It suggests that the parameters of TTT will be lower than 120 mW/mm and limited less two sessions if applicable, especially for Asia people.
8.The application of mind map in theory course teaching of ophthalmology

Ophthalmology is a predominantly morphology of clinical discipline. The eye has the characteristics of small scale, close structure, complex and diverse diseases, different clinical manifestations and so on. As an effective tool, the mind map can promote thinking visualization, help learners establish the logic relation of things and cultivate their innovative ability. To achieve better teaching effect, we introduce the mind map into theory course teaching of ophthalmology. On the one hand, teachers can optimize the teaching design, clarify the teaching ideas, and on the other hand, students can draw the mind map on their own or with others which will improve their subjective initiative of learning and study efficiency, and at the same time cultivate their ability of mutual collaboration.

Workbench 11.0 finite element analysis software. The models were loaded with six different methods to analyze the stress distribution.
RESULTS AND CONCLUSION:The stress distribution of bending root was mainly concentrated in the tooth
neck, fol owed by the apex. During the overal tooth movement, the stress was smal est on the root, periodontal ligament and alveolar bone. Correction of bending roots needs more accurate positioning and traction.
10.Clinical study on antibiotic-induced endotoxin release in Gram-negative bacterial infection

Objective To observe the various influence of Imipenem/Cilastatin(IPM) and Cefoperazone(CPZ) on Gram-negative bacterial endotoxin release in severely burned patients.Methods Patients were divided into IPM group and CPZ group randomly. The bacterial cultures and microbial drug-sensitivities of burn wound swabs and subeschar tissues were performed on the 1st and 3rd day, At the same time, the levels of LPS in plasma were detected in seven burned patients administrated with IPM and six with CPZ at the time 0?2?4?12?24?48?72h of antibiotics administration.Results Patients were infected by Gram-negative bacterial. The counting of bacteria in subeschar tissues decreased 10~100 times after the administration of antibiotics 3 days; Plasma LPS levels elevated to the peak value at 2h after infusion of IPM or CPZ , and then showed significant decrease. The magnitude of increased LPS in CPZ group was higher than that in IPM group; The LPS levels of 72h were lower significantly than that of 0h.Conclusion IPM and CPZ could effectively control Gram-negative bacterial infection and induce the releases of LPS from bacteria, of which the former induced less LPS release than the latter; The levels of LPS came down with the decreasing of bacterial counting.
