Objective To investigate the effect on cell cycle of hepatitis B virus x protein and ap- proach the molecular mechanism of x protein's carcinogenesis. Methods Gene transfection mediated by the lipofectamine was used to introduce the eukaryotic expression vector of HBV x gene into human hepato- cellular carcinoma cell line HepG2(HepG2X cell).The selective medium containing G418 was used to select the cell clones which the X protein was expressed constantly.Flow eytometry was used to detect the cycle of the cells;raf-1 protein was detected by Western blot.Results X protein could be expressed on 21?10~3 loca- tion in the transfected cells,while there were no protein expressed in the control cells.The proliferation of X cells increased significantly according to MTS method(P

Colorectal cancer, C RC, is a kind of cancer, which has a high rate of morbidity and mortality.Liver metastasis is the most frequent metastasis way in CRC, and related to the prognosis of the patients.There are many studies on the treatment of colorectal liver metastasis. The resection of the metastasis is one of the major treatments. Other treatments, including chemotherapy, alleviative and immunogenic treatments,are beneficial supplements to the resection. We summariz the indications, contraindications, managements and effects of these treatments.
3.The ultrasonographic characteristics and common misdiagnosis reason of superficial neurilemmoma

Objective To investigate the ultrasonic features and differential diagnosis of superficial neurilemmoma ,and improve diagnostic accuracy of superficial neurilemmoma by ultrasound .Methods Retrospectively analyse the ultrasonic findings of 51 ca-ses ,which confirmed for superficial neurilemmoma by pathology ,and analyse the misdiagnosis reason from the aspects of size , shape ,boundary ,internal echo ,blood flow signals .Results Superficial neurilemmoma had some characteristics in the shape ,bound-ary ,internal echo ,blood flow signals and so on .However ,the key point to differentiate it from other diseases was to identify the re-lationship with peripheral nerve .In this study ,the coincidence rate of ultrasonoic diagnosis with pathologic diagnosis was 78 .4% . Conclusion Superficial neurilemmoma have some similarity with neurofibroma or sebaceous cyst on ultrasonographic images .When making diagnosis ,especial attention should be paid to identify the relationship of neurilemmoma with peripheral nerve .
4.Study of the Mechanism of Cassia Obtusifolia L in Decreasing Blood-lipid

OBJECTIVE:To study the mechanism of Cassia obtusifolia L in decreasing blood-lipid.METHODS:Primary cultures of rat hepatocytes in vitro were used to determine the concentrations of proteins in hepatocytes by the method of Folin-phenol reagent and to assay the cholesterol synthesis from 14 C-acetate with liquid scintillation count.RESULTS:Ex?tract of Cassia obtusifolia L inhibited cholesterol synthesis from 14 C-acetate,but cassiaside B didn't affect cholesterol synthe?sis.CONCLUSION:The mechanism of extract of Cassia obtusifolia L in decreasing blood-lipid may result from inhibition of cholesterol synthesis,but inhibiting cholesterol synthesis may be not the main approach of cassiaside B in decreasing blood-lipid.
6.Liver cancer stem cell isolation and biological characteristics

BACKGROUND:Normal stem cells and tumor stem cells have differences in cellsignaling pathway of gene expression and dependence. How to find a therapeutic method of selectively kil ing tumor stem cells is a topic that should be studied greatly. OBJECTIVE:To isolate human liver cancer stem cells MHCC97 and to investigate the biological characteristics of them. METHODS:Tumor stem cells were selected from highly metastatic human hepatoma cellline MHCC97 by flow cytometry. CD133-CD34-MHCC97 from normal human liver stem cells and CD133+CD34+MHCC97 from tumor stem cells were isolated. Their phenotypes, growth curve, cellcycle and multi-lineage differentiation were measured. RESULTS AND CONCLUSION:Phenotypes of CD133+CD34+MHCC97 cancer stem cells were CD133+and CD34+and they had the same growth curve and cellcycle with the liver stem cells CD133-CD34-MHCC97, besides, they had the differential ability to epithelial cells and endothelial cells that proved to be stem cells. These results indicated that human cancer stem cells CD133+CD34+MHCC97 had the specific characteristics of tumor stem cells, could differentiate into epithelial cells and endothelial cells, had the biological property of tumor stem cells, was an origin of tumor relapse and metastasis, and a target of clinical therapy.

8.Stress changes on endplates of fractured vertebra under different bone cement volumes

Objective To investigate the effect of different bone cement volume injected after vertebroplasty on stress distribution of the upper and lower endplates of the injured vertebra in the flexion and extension positions by finite element analysis.Methods Two patients with osteoporotic compression fractures of L2 vertebral body were chosen.Bone cement volumes of 25% and 50% were respectively injected into the fractured vertebral body bilaterally.Pre-and post-operative finite element models of fractured and adjacent vertebral bodies were developed after extracting the CT data.Stress changes at L2 superior-and inferior-vertebral endplates were observed in the vertical,flexion,and extension loading.Results Stress at the upper and lower endplates of the fractured vertebra increased after bone cement injection and more increase was observed in 50% bone cement group than in 25% bone cement group.In the neutral position,the stress mainly concentrated at posterior vertebral body,with stress of the upper endplate improving from 0.91 × 106 N · mm to 0.93 × 106 N · mm and stress of the lower endplate from 0.98 × 106 N ·mm to 1.03 × 106 N · mm.In the anterior flexion,the stress mainly concentrated at anterior vertebral body,with stress of the upper endplate improving from 4.44 × 106 N · mm to 5.55 × 106 N · mm.In the posterior extension,the stress mainly concentrated at medial vertebral body,with stress of the upper endplate improving from 21.95 × 106 N · mm to 22.10 × 106 N · mm and stress of the lower endplate from 30.60 ×106 N · mm to 30.88 × 106 N · mm.The finite element model revealed stress at the upper and lower endplates was higher in the anterior flexion and posterior extension than in the neutral position,while in the anterior flexion the stress was comparatively higher.Conclusions A small amount of implanted bone cement act no evident effect on stress distribution of the upper and lower vertebral endplates.However,the stress changes are significant as cement volume reached up to 50%,especially in the flexion position,as may suggest an enhanced risk of adjacent intervertebral disc degeneration and adjacent vertebral body fractures.

10.The clinical characters of tear film instability in diabetes patients

Background The current epidemiology study had shown the prevalence of age and sex adjusted dry eye was higher in patients with diabetes than population without diabetes.Further researches demonstrated that the tear film disturbance is common after the phacoemulsification or photocoagulation in the eyes of diabetic patients.Objective The aim of this study was to characterize the clinical features of tear film instability in diabetes patients.Methods One hundred and sixty-two patients with tear-film abnormality referred to Tongren Eye Center from January 1,2010 to September 1,2010 underwent questionnaire about diabetes and other diseases,BUT,Schirmer test.Tear film instability was diagnosed as abnormality of either Schirmer test or BUT and showed as M ( Q25,Q75 ).The right eyes of 162 dry eye patients meeting with the including criteria were enrolled.The patients were assigned to two groups according to with ( 80 patients) or without ( 82 patients) diabetes mellitum.DEQ questionnaire were scored.The percentage of cases with meibomain gland abnormal score > 1 was calculated.Mann-Whitney U analysis and Chisquare analysis were used to compare the difference between the two groups.Results The Schirmer test in diabetic group was 8 ( qualities:5,9 )mm and was longer than 6 ( qualities:5,7 ) mm in non-diabetic patients ( U =2452,P =0.00).The result of BUT test was 3 ( qualities:2,4 ) seconds in diabetic patients and was shorter than 4 (qualities:3,5) seconds in non-diabetic patients( U=2104,P<0.01 ).The DEQ score of diabetic patients was 15 ( qualities:1 0,19,which was less than21 ( qualities:19,23.25 ) in non-diabetic patients.51.2 % ( 41/80 ) diabetic participants and 32.9% (27/82) nondiabetic participants appeared meibography ( grade larger than 1 ) (x2 =16.07,P=0.00).The percentages of dry eyes were 51.2% (41/80) and 93.9% (77/82) respectively in diabetes and nondiabetes groups(x2 =37.24,P<0.01 ).No significant correlation was found between the diabetes course and DEQ score or meibography( r =0.16,P =0.16 ; r =0.10,P =0.36 ).Conclusions Diabetes patients with tear film instability have longer Schirmer test results,shorter BUT,more severe meibomain glands damage and lower DEQ scores.The dry eye symptom is lack in the diabetic patients though appearing the tear film and meibomain glands damage.Therefore,more attention should be given to ocular surface health in diabetes patients.
