1.Investigation and Analysis of Physical and Mental Health of Nurses

Objective to investigate the situation of occupational health of nurses,so as to provide a reference for protecting the health of nurses. Methods Cluster sampling method was employed in this study. totally 1 204 nurses of two three-level general hospital and a cancer hospital in Bei-jing were randomly surveyed with homemade health of nurses′cross-sectional questionnaire,and a nurse medical examination report was access at the same time. Results there was statistically significant difference in the incidence of nurses work pressure and work fatigue in different hospitals and different departments(P < 0.05). 95.18% of the nurses undergo regular medical examinations,of which 56.97% were found abnormal. the dif-ferences in the incidence rate of abnormal medical examination results in nurses were statistically significant among different hospital and different de-partments(P < 0.05). Conclusion Great job stress and work fatigue are observed in nurses,and they have health problems. It is necessary for the hospital to take appropriate interventions to improve physical and mental health nurses.
2.Laparoscopic radical prostatectomy for high-risk prostate cancer (49 cases)

Objective To evaluate the safety and effect of extraperitoneal laparoscopic radical prostatectomy for high-risk prostate cancer. Methods From January 2012 to August 2015, 49 patients diagnosed high-risk prostate cancer underwent extraperitoneal laparoscopic radical prostatectomy with 'sandwich' urethra reconstruction. Results None converted to open surgery and the mean operative time was (2.15 ±0.29) h, mean intraoperative blood loss was (60.25 ± 20.29) ml. No rectal injury was observed. The patients were ambulant 1 to 2 days postoperatively. Pelvic lymph nodes metastasis was found in 5 cases. Positive margin was found in 13 cases. Mild urinary incontinence oc-curred in 3 cases. Urethral stenosis occurred in 2 cases. Biochemical relapses occurred on 5 cases during the follow-ing period of 1~43 months for 46 cases. Conclusions Extraperitoneal laparoscopic radical prostatectomy is safe and effective in treatment of high-risk prostate cancer. The very key is to master the anatomy of prostate and laparoscop-ic techniques.
3.Chemical constituents in stem of Acanthus ilicifolius

Objective To study the chemical constituents in the stem of Acanthus ilicifolius.Methods Some compounds were repeatedly isolated and purified on silica gel column chromatography and vacuum liquid chromatography,their chemical structures were elucidated by their physicochemical properties and spectral data,such as IR,(()~1HNMR),(()~(13)C-NMR),and ESI-MS.Results Eight compounds were obtained from the stem of A.ilicifolius and identified as hexacosanoic acid(Ⅰ),stigmasterol(Ⅱ),tetratriacontanol(Ⅲ), 2-benzoxazolinone(Ⅳ),stigmasterol-3-O-?-D-glucopyranoiside(Ⅴ),vanillic acid(Ⅵ),4-hydroxy-2-benzoxazolone(Ⅶ),quercetin(Ⅷ).Conclusion Compound Ⅶ is first reported as a natural product.Compounds Ⅰ,Ⅲ,and Ⅵ-Ⅶ are reported from this plant for the first time.
4.Effects of Adiponectin on Cell Activity and Expression of T-cadherin in Human RPE Cells Cultured with High D-glucose

Objective To investigate the effects of adiponectin on cell activity and expression of T-cadherin in human retinal pigment epithelial(hRPE) cells cultured with high glucose.Methods ①The hRPE cells were divided into three groups:normal glucose group(5.5mmol/L glucose),high glucose group (33mmol/L glucose),high glucose plus adiponectin group (33mmol/L glucose+2.5ug/ml adiponectin).48h later,the cell activity was observed by MTT assay.②After the above groups were cultured respectively for 24h,48h,72h again,the expression of T-cadherin mRNA was observed by RT-PCR. Results ①Cell activity in high glucose group significantly decreased. Cell activity adiponectin group was higher than that in the high glucose group.②With the extension of time ,the expression of T-cadherin decreased in high glucose group,but increased in adiponectin group. Conclusion Adiponectin may be protect the hRPE cells by T-cadherin.

7.EFFECTS OF INSULIN-LIKE GROWTH FACTOR 1 ON β-AMYLOID INDUCED NEURON APOPTOSIS AND TAU PHOSPHORYLATION

The aim of this study is to elucidate the protective and anti-apoptotic effects of insulin-like growth factor 1 ( IGF-1 ) against β-amyloid (Aβ) and investigate the effect of IGF-1 on Aβ-induced tau phosphorylation. Cell viability was measured using the MTT (3-(4,5-dimethylthiazolyl-2 )-2,5-diphenyltetrazolium bromide) assay, early apoptosis and late apoptosis/necrosis were analyzed by flow cytometry using Annexin V-FITC and propidium iodide (PI) double staining, and morphology was examined by Hoechst 33342 staining. Tau phosphorylation was detected using AT8 immunostaining. Preincubation of cultured rat hippocampal neurons with IGF-1 for 24 h prevented cytotoxicity induced by Aβ25-35 for 48 h. The MTT value significantly increased from 54.51% to 61.8% of the control group, and the percentage of Hoechst 33342-positive cells decreased from 30.77% to 22.81%. Incubation with Aβ25-35 for 48 h caused a marked increase in the percentages of Annexin V-FITC single-labeled cells (Annexin V +/PI-) and Annexin V/PI double-stained cells (Annexin V +/PI + ) (3.41% and 19.47% , respectively), which were significantly decreased by pretreatment with 100 ng/ml of IGF-1 for 24 h (to 2.98% and 15.16% , respectively). Aβ25-35 treatment increased tau phosphorylation and AT8 positive cells were 41.84%. This effect could be inhibited by different concentrations of IGF-1. Our findings showed that IGF-1 protected against Aβ-induced cytotoxicity, decreased the percentage of early and late apoptosis/necrosis cells, and inhibited tau phosphorylation, which may be the cellular mechanisms for its neuroprotective action.
8.Pre-mRNA splicing and retinitis pigmentosa

Retinitis pigmentosa (RP) is a large group of common hereditary eye diseases with highlyheterogeneous genetic background. Over forty genes with diverse functionalities are associated with RP and they include a set of ubiquitously expressed genes. These include five genes involved in the precursor messenger RNA( premRNA) splicing. Recent progress in disease gene identification for RP has established the involvement of pre-mRNA splicing as one important mechanism in the disease etiology and has shed light on the splicing process itself, a fundamental biological process. To this date, studies in this field have been focused on two major issues. First, how do the mutations of the adRP associated splicing factors (adRP-SF) affect the splicing function? Second, how do the mutations in these ubiquitously expressed genes lead to specific retinopathy? The two topics fit with the two continuous important steps of the disease pathogenesis. Recently, researchers have made a dramatic progress in the first topic. The identification of the SNRNP200 gene,the fifth adRP-SF and its relevant functional study has shown significance to the progress in the study of RP. Numerous investigations are also being carried out in addressing the second issue.Generation of a variety of models led to a better description of the pathological process of the disease. However, in respect to the key pathogenic mechanism,researchers are still puzzled with a number of confusing questions. In this commentary,the results from the latest investigations were summarized, and in particular,the difficulties in studying the molecular mechanism by which the pre-mRNA splicing deficiency causes RP were detailed.
9.Effect of 5-lipoxygenase and 12-lipoxygenase on proliferation and apoptosis of pancreatic cancer cell

Objective To examine the expression of lipoxygenases (LOXs)on human pancreatic carcinoma and their effects on proliferation and apoptosis of human pancreatic carcinoma in vitro.Methods Expression of 5- LOX and 12-LOX in the tissue of human pancreatic carcinoma and pancreatic cancer cell line (SW1990) was detected by immunohistochemical staining and RT-PCR.To examine the effects of polyunsaturated fatty acids on pancreatic cancer cell line,the proliferation rate of SW1990 cells treated with arachidouic acid (AA),docosahexaenoic acid (DHA) and eicosapentenoic acid (EPA) was analyzed by MTT and the incorporation of bromodeoxyuridine (BrdU) methods.An annexinV/propidium iodide (PI) assay with flow cytometry and in situ terminal deoxynucleotidyl transferase-mediated dUTP-digoxigenin end labeling (TUNEL) assay were used to identify potential induction of apoptosis by the unsaturated fatty acids.The effects of 5-LOX inhibitor (MK-886) and 12-LOX inhibitor (baicalein) on the proliferation of SW1990 cells were determined by above methods.Results Both 5-LOX and 12-LOX were over expressed in the tissue of human pancreatic carcinoma and the pancreatic cancer cell line.AA had a stimulatory effect on the growth of SW1990 cells while EPA and DHA had an inhibitory effect in a dose-dependent manner.The apeptosis of SW1990 cells co-cultured with DHA or EPA lasting for 24 hours was increased markedly.Furthermore, both MK-886 and baicalein in a dose-dependent-manner inhibited the growth of SW1990 cells ard induced a significant increase of apoptotic cells rate.Conclusions Expression of lipoxygenase is up-regulated in human pancreatic cancer. Polyunsaturated fatty acids have regulatory effects on the growth of pancreatic carcinoma in vitro.AA stimulates proliferation of pancreatic cancer cell line,while DHA and EPA suppress proliferation,simultaneously inducing apoptosis of pancreatic cancer cell line.Blocking the functions of lipoxygenase with its inhibitor exerts a negative regulatory effect on the growth of pancreatic carcinoma in vitro.Lipoxygenase might be a novel therapeutic target for the pancreatic cancer.
10.Effect of Q Switch Nd∶YAG solid laser in treatment of naevus fusco-caerulens zygomaticus

Objective To observe the effect of laser of Q Switch Nd:YAG 1 064nm wave length laser in the treatment of naevus fusco caerulens zygomaticus. Methods Since October 2000, forty nine cases of naevus fusco caerulens zygomaticus had been treated once every three months. Results Thirty two cases had been treated three times, and seventeen cases, four times. The total effective rate was 100 %. The pigmentation of all the cases were darker after the first treatment, later as the treatment progressed, the pigment became lighter. No pigment loss was seen. Conclusion Q switch Nd:YAG 1 064 nm wavelength laser has a good effect in the treatment of naevus fusco caerulens zygomaticus. It is safe and reliable.
