Several M-CAT elements are present in the chicken nicotinic acetylcholine receptor γ-subunit promoter and required for the totipotency of this gene.To understand the binding properties of M-CAT element, the interaction of M-CAT motif,at the position -67/-61 relative to the initiation site of γ-promoter with nuclear proteins, were analyzed.Mutageneses and binding experiments demonstrated that the M-CAT core motif bound nuclear protein (s) from tested chicken tissues to generate a high-mobility complex;while the flanking sequence bound different factors to generate lower-mobility complexes with the exception of liver.Southwestern blotting showed that only a 30 kD protein was expressed in tested tissues of chicken embryo at day 13.These results suggest that a 30 kD factor can directly bind to M-CAT element in the manner of monomer, even homodimer and is ubiquitously expressed in tissues, while the binding of the factor(s) to flanking sequence should probably rely on the presence of a 30 kD ubiquitous factor.

BACKGROUND:There is no clear understanding on the effects of subcutaneous fat and stem cels on wound healing. OBJECTIVE:To explore the therapeutic effects of skin composite prepared with adipose tissue-derived stem
cels on skin defects. METHODS: Epidermal cels, fibroblasts, adipose tissue-derived stem cels as seed cels and bovine colagen gel as a scaffold were used to build a complex with a variety of cels. A 6-mm diameter circular skin defect was made on the both sides of the rat back. The right side as experimental side was implanted with an 8-mm diameter multilayer skin composite, and the left side (control side) was only treated with a simple dressing. RESULTS AND CONCLUSION: For the constructed multi-layer skin composite, the epidermal layer was continuously merged into the multi-layer, the fibroblasts evenly distributed in the corium layer, and lipid droplets existed in the fat layer in which the cels distributed uniformly. Cel aggregation was obviously observed at the junction of different layers. In the experimental side, the rate of wound healing, granulation tissue thickness, the thickness of dermis and the capilary density were significantly higher than those in the control side. Taken together, we can construct multilayer skin composites with a variety of cels as seed cels, such as epidermal cels, fibroblasts and adipose tissue-derived stem cels, and bovine colagen gel as a scaffold, which promote wound healing and increase the thickness of dermis.

Chinese Journal of Tissue Engineering Research 2016;20(11):1538-1543
BACKGROUND:Epidermal stem cels are the source of cels for repairing the skin and its appendages. To study differentiation and migration of epidermal stem cels is crucial for perfect healing.
Objective: To investigate the effects of substance P in skin wound repair.
METHODS:Thirty Wistar rats were randomized into groups of substance P, capsaicin and control (n=10 per group). After the establishment of skin defect models, exogenous neuropeptide substance P, capsaicin, and normal saline were daily injected at the wound for continuous 5 days. Skin thickness, skin wound area, BrdU-positive cels were detected in the three groups at different time after injection.
Results and conclusions:No difference n the skin thickness was found in the three groups at 14 days after injection (P > 0.05), but with time, the epidermal thickness was reduced. At 42 days, the epidermal thickness was reduced remarkably in the substance P group and decreased certainly in the capsaicin group, but unchanged in the control group. The wound area of the substance P group was significantly lower than that in the control and capsaicin groups at 3, 5, 7 days after injection (P < 0.05), but there was no difference between the control and capsaicin group (P > 0.05). The BrdU positive rates in the three groups were significantly different at 3, 5, 7, 12 days after injection: the substance P group > the control group > the capsaicin group (P < 0.05). These results show that substance P can promote epidermal cel proliferation and accelerate wound healing.

Chinese Journal of Parasitology and Parasitic Diseases 1987;0(03):-
This paper deals with the ultrastructure of the body wall of adult Pagumogonimus skrjabini by transmission electron microscopy. Infected crabs were collected from Siyen, Hubei Provine, and adult worms were obtained from the lungs of experimentally infected dogs 90 days post-infection.The normal structure of body wall of the P. skrjabini is composed of tegument, tegument cell, musele, muscle cell and protoplasmic tubules, all of which form together syncytium. The tegument contains external plasma membrane, tegument matrix and basal plasma membrane. The cell coat in fine granules is distributed over the whole external plasma membrane surface. The tegument matrix contains various secretory bodies, vesicles and mitochondria. The tegument cell is irregular in shape. Golgi complex, ribosome, autoly-sosome are seen in the cytoplasm. There are two layers of muscle, the external circular muscle and the inner longitudinal muscle layers. The nucleus of immature muscle cell has many heterochromatins, while the nucleus of mature muscle cell is large and round in shaps. Mitochondria and glyco-gen granules are transmitted from muscle cell proper to the muscle by protoplasmic tubules (Figs. 1-7).

Objective: To observe the relationship between homocysteine(Hcy)and spatial cognitive ability and to evaluate whether vitamin B12 supplementation could protect spatial cognitive ability in rats. Methods: The rats were randomized into three groups. The experimental group was given methionine subcutaneously. The intervention group was given methionine and vitamin B12 subcutaneously. The control group received isometric normal saline. After being raised for 8 weeks, all rats were examined for plasma Hcy and serum vitamin B12 and scores of Y-maze test. The brain tissues of hippocampus were checked immunohistochemically for acetylcholinesterase (AChE). Results: The experimental group developed hyperhomocysteinemia and descreased spatial cognitive ability significantly (P0.05). Conclusion: Homocysteine can impair the spatial cognitive ability in rats and vitamin B12 is effective in lowering Hcy level and protect the spatial cognitive ability of rats.

Objective:To analyze the clinical characteristics and treatment effect between bilateral(bi-)and unilateral(uni-) sudden sensorineural hearing loss(SSNHL).Method:Four hundred and eighty cases of SSNHL were retrospective study, which were divided into two groups of bi-SSNHL(n=40) and uni-SSNHL(n=440).Clinical characteristics and treatment effects were compared of the two groups.Result:The incidence rate of bi-SSNHL was 8.3 percent and uni-SSNHL was 91.7 percent of all patients with SSNHL. Bi-SSNHL occurs more commonly in patients of old age, diabetes mellitus, and lipid panes abnormalities compared to uni-SSNHL. Twenty-eight ears in the bi-SSNHL group showed hearing recovery (35%),compared with 56.4 percent of patients with uni-SSNHL.Conclusion:Bi-SSNHL and uni-SSNHL may have a completely different clinical characteristics and treatment effect, that implies a different pathophysiology and prognosis. Recognition their different clinical characteristics and treatment effect between bilateral and unilateral SSNHL can help in counseling and managing the patients and correctly evaluate the prognosis.

ATP-binding cassette transporter A1 (ABCA1) and scavenger receptor class B type I (SR-BI/CLA-1) are the key proteins in reverse cholesterol transport (RCT). The high expression of ABCA1 and SR-BI/CLA-1 can decrease the danger of atherosclerosis. The purpose of the study is to find ABCA1 and CLA-1up-regulators for treating atherosclerosis by using cell-based high throughput screening models. Among 20 000 compounds screened, E0869 [1-(3, 4-dimethylphenyl)-1-oxopropan-2-yll4-((methylsulfonyl)methyl)benzoate] was found as the positive hit. The up-regulated activities of E0869 in ABCAl1-LUC and bCA-l1-LUC HepG2 cell were 160% and 175%, respectively. The EC50 values of E0869 in ABCAl1-LUC and CLA-l1-LUC HepG2 cell were 3.79 and 1.42 pμol- x ,(-1) respectively. E0869 could upregulate the mRNA and protein levels of ABCA1, SR-BI/CLA-1 and ABCGJ1genes in HepG2 and RAW264.7 cells by Real-Time Quantitative PCR and Western blotting analysis, but could not influence the expression of FAS, SREBP-l1 and CD36. Foam cell assay showed that E0869 could inhibit lipids accumulation in mouse peritoneal macrophages RAW264.7. Cholesterol efflux assay showed that E0869 could induce HDL-mediated cholesterol efflux in mouse peritoneal macrophages RAW264.7. In conclusion, E0869 could up-regulate ABCA1 and CLA-1 activity, and had good anti-atherosclerotic activity in vitro.
