2.Phosphoglycerate mutase 5 and necroptosis
Chinese Journal of Pathophysiology 2016;32(2):377-380
Necroptosis, or programmed cell death, is a type of cell death with a controllable death signaling pathway and the morphological features similar to necrosis. It is mainly mediated by death receptors or pathogen pattern re-cognition receptors.Among them, tumor necrosis factor receptor 1 (TNFR1)-mediated necroptosis is the most well-studied one.Receptor-interacting protein kinase 1 (RIPK1) and receptor-interacting protein kinase 3 (RIPK3) are the 2 key kina-ses involved in the formation of complex I & II and necrosome in the process of necroptosis.Phosphoglycerate mutase 5 ( PGAM5) , a member of phosphoglycerate mutase gene family, lacks PGAM activity and possesses the phosphatase activi-ty.PGAM5 is anchored in the mitochondrial membrane and is also called mitochondrial phosphoglycerate mutase 5.It has been shown that PGAM5 involves in the formation of necrosome during necroptosis and it is able to accelerate the fission of mitochondria by dephosphorylation of dynamin-related protein 1 (DRP1), thus promoting cell necroptosis.

Basic & Clinical Medicine 2017;37(5):643-647
Objective To investigate the effect of netropsin on migration and invasion ability of gastric cancer cells and its mechanisms. Methods To determine if netropsin inhibits migration and invasion of gastric cancer cells, Transwell migration and invasion assay was performed. Then Western blot was performed to detect expression of E-cadherin and vimentin in gastric cancer cells with or without presence in medium netropsin. Finally, immunofluorescence was performed to detect changes in the cellular localization of β-catenin to validate whether Wnt/β-catenin pathway was suppressed or not. Results Netropsin with the concentration of 25 μmol/L had minimal inhibition effect on cell proliferation and was able to suppress ability of migration and invasion by inhibiting EMT in gastric cancer cells(P<0.05).Meanwhile netropsin was able to down-regulate the expression of epithelial markers E-cadherin and up-regulate the expression of mesenchymal marker vimentin. Finally, immunofluorescence showed that netropsin was able to block translocation of β-catenin from cytoplasm to nuclear. Conclusions Netropsin can inhibit EMT thereby suppressing migration and invasion of gastric cancer cells. The mechanism is that netropsin can compete with HMGA2 for transcription factor binding site thereby suppressing the Wnt/β-catenin pathway.

Journal of Integrative Medicine 2004;2(6):450-2
OBJECTIVE: To study the pharmacological activities and toxicity of the crude alkaloids of Toddalia asiatica and to provide pharmacological data for the further development of this herbal medicine. METHODS: We observed the anti-inflammatory effects of the crude alkaloids of Toddalia asiatica, using xylol and agra to induce the turgidness and sodium carboxymethyl cellulose (CMC-Na) to induce leucocyte strolling in the rats. The analgesic effects were observed by body-distortion methods. The effects of alkaloids of Toddalia asiatica on hepatic function were observed by testing the contents of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) in serum and calculating the liver index. The LD 50 and 95% creditability were calculated with developed Karber Method. RESULTS: The administration of alkaloids of Toddalia asiatica had the function of inhibiting the auricle swelling caused by xylol and joint swelling caused by agar and leucocyte migration caused by CMC-Na, decreasing the body-distortion of the rats. After two weeks administration, the contents of ALT and AST showed that there was no obvious difference between administered group and control group. The LD50 of the crude alkaloids of Toddalia asiatica was 1.622 g/kg and the 95% creditability was 1.29-2.03 g/kg. CONCLUSION: Toddalia asiatica has anti-inflammatory and analgesic effects, and there is no injury to the liver after long-term administration in rats.
