3.A PRELIMINARY RESEARCH IN CULTIVATION AND DIFFERENTIATION OF CORTICAL AND MESENCEPHALIC NEURAL STEM CELLS IN VITRO

Objective To explore the differences of cultivation and differentiation between cortical neural stem cells(C-NSCs) and mesencephalic neural stem cells(M-NSCs). Methods We dissociated embryonic cortex and ventral mesencephalon respectively,and single cell suspensions were achieved by trypsin digestion and mechanical dissociation in sterile condition.These cells were planted into DMEM/F12 culture medium with B27,bFCF,and then were passaged by mechanical methods and induced to differentiation in DMEM/F12 medium with 10%FBS(without growth factors).For examining the features of C-NSCs and M-NSCs,reverse phasecontrast microscopy,immunocytochemical techniques were applied to detect nestin,NSE,GFAP antigens and the specific antigen(TH) of neurons differentiated from the two kinds of NSCs.We observed and compared the difference of the two kinds of NSCs. Results The NSCs existed both in cortex and ventral mesencephalon of embryonic mice,which expressed nestin and possessed the ability of self-renewing and multipotent, but the density of NSCs in cortex was higher than that in ventral mesencephalon.There were some TH positive cells differentiated from M-NSCs but none came from C-NSCs.Conclusion The proliferative activity of C-NSCs is higher than that of M-NSCs,but more number M-NSCs differentiated into dopaminergic neurons in serum medium.
4.Therapeutic study of laparoscopic hernioplasty for inguinal hernia in aged males

Objective To evaluate the therapeutic effect of laparoscopic hernioplasty for inguinal hernia in aged males.Method Forty cases of inguinal hernia in aged males were treated with laparoscopic hernioplasty(group L),while other 46 cases were treated with traditional inguinal hernioplasty(group S).Results To compare with group S,the time be in hospital and the time leave off the bed were significantly shorter in group L(P<0.01 or<0.05),but the mean operation time were longer and the operation expenses Was higher in group L(P<0.05).Followed up for 2-47 months,there were no recurrence in two groups.Conclusions Laparoscopic hernioplasty for inguinal hernia in aged males can shorten the hospitalization time,quicken postoperative recovery.The comphcation is little and the wound ache is light.Along with the further improvement of laparoseopic technique and the amelioration of laparoscopic instrument.the opera-tion time will further shorten,the expenses also will descend.It will be a ideal way for the therapy of inguinal hernia in aged males.
5.Nationwide external quality assessment for drug susceptibility testing of Neisseria gonorrhoeae from 2007 to 2009

Objective To make a nationwide external quality assessment for drug sensitivity testing of Neisseria gonorrhoeae, analyze the problems in and factors associated with the drug sensitivity testing, and to enhance the quality of drug sensitivity testing of N. gonorrhoeae at different monitoring sites. Methods Samples were uniformly delivered to monitoring sites by express mail service. Test results were analyzed in the National Center for STD Control, and the evaluation results were fed back to these monitoring sites. Results A total of 105 quality control samples were delivered from 2007 to 2009, with a response rate of 88.57% (93/105). Thirteen monitoring sites were enrolled in the external quality assessment, including 9 laboratories in 2007, 9 in 2008 and 13 in 2009. The total percentage amounted to 77.42% (24/31) for qualified laboratories during the 3 years, including 6 laboratories in 2007, 7 in 2008 and 11 in 2009. The coincidence rate increased for the detection of penicillinase-producing N. gonorrhoeae (PPNG), N. gonorrhoeae with chromosome-mediated ciprofloxacin resistance, and N. gonorrhoeae with chromosome-mediated spectinomycin resistance, and declined for the detection of N. gonorrhoeae with plasmid-mediated high level tetracycline-resistance (TRNG) and N. gonorrhoeae with chromosome-mediated ceftriaxone resistance. Conclusions The 3-year external quality assessment reveals an improvement in the overall quality of drug sensitivity testing of N. gonorrhoeae at national monitoring sites; the accuracy is improved markedly for the detection of PPNG, N. gonorrhoeae with resistance to spectinomycin and ciprofloxacin, but is needed to increase for the detection of ceftriaxone-resis- tant N. gonorrhoeae and TRNG.
6.Application and preventive effect of hammock firmness sheets for pressure ulcer

Objective To know the preventive effects of using hammock firmness sheets for pa-tients with pressure ulcer.Methods Divided 98 patients into the application group (31 cases), the treatment group(30 cases) and the control group(37 cases) according to themselves condition.Hammock firmness sheets was used in the application group, while the routine preventive method of pressure ulcer was used in the control group.To know the preventive effects of pressure ulcer betweent the two groups.The patients in the treatment group was patients had pressure ulcer, hammock firmness sheets combined with local massage were used for them, observed the treatment effects for them.Results The condi-tion of preventive effect of pressure ulcer in the application group was better than those of in the control group.The cure rate of pres sure ulcer in the treatment group was 90%.Conclusions The hammock firmness sheets can prevent pressure ulcer effectively.
7.Study on regulatory effects of interleukin-6 on placental growth factor expression in trophoblast cells

Objective To investigate the expression pattern of Placental growth factor (PlGF) and regulatory effects of interleukin-6 (IL-6)on PlGF expression in the primary cultured trophoblast cells. Methods Cytotrophoblast cells werecollected by trypsin-collagenase digestion and Percoll gradient centrifugation for primary culture. Reverse transcriptase polymerase chain reaction(RT-PCR) were resorted to demonstrate the expression pattern of PlGF mRNA in trophoblast cells cultured in vitro. The effects of IL-6 with different concentration(100,10,1 and 0.1 ?g/L) and duration (6,12,24 and 48 h) on PlGF expression were observed. Results The 185 bp and 248 bp bands of PlGF were showed by RT-PCR. PlGF expression correlated with IL-6. PlGF began to increase at 6 h, and reached the climax at 12 h when recultured with 100 ?g/L IL-6. Conclusions PlGF expression has time and dose dependance on IL-6. It may play an important role in early pregnancy.

Objective To study the growth inhibitory effect of chemotherapeutic drugs on the pancreatic cancer cell line Aspc-1 and Bxpc-3, and the relations to the varied concentration and exposere time of chemotherapeutic drugs administrated in single or in combination. Methods Four chemotherapeutic drugs included 5-fluorouracil (5-FU), epirubicin (E-ADM), mitomycin(MMC) or cisplatin (DDP) administrating in single or in combination and using varied concentrations(d1、d2、d3 and d4) and exposure times(24、28 and 72 hours); and then the growth inhibitory effect on the two cell lines. resulted from these drugs were assayed by MTT colorimetry and analysed by “ t ” test. 9.A Case-control Study on Risk Factors of Lung Cancer in Rural Area of Heilongjiang Province

Objective To explore the risk factors of lung cancer in rural area. Methods A 1∶1 matched case_control study was conducted in 106 individuals in Heilongjiang province. Results The amount of passive smoking (OR:2.48,95%CI:1.51~4.08),history of mental scar (OR:4.63,95%CI:1.51~14.15), smoking from Kang (OR:1.69,95%CI:1.10~2.59), smoking indexes (OR:1.75,95%CI:1.10~2.79) and chronic bronchitis (OR:4.67,95%CI:1.12~19.49) had a closely correlation with lung cancer in rural area.Conclusions Lung cancer might be caused by multiple factors synergetically.The main countermeasures for controlling lung cancer were to give up smoking and to improve the conditions of heatable brick bed and heating method.
