1.Effect of Cefradine on pharmacokinetics of baicalin in rats in vivo

Objective To study the effect of oral Cefradine on pharmacokinetics of baicalin in rats in vivo. Methods The rats were divided into two groups: one was supplied with baicalin individually and the other was supplied with the combination of Cefradine and baicalin. Plasma concentrations of two groups were detected by HPLC with electrochemical detection (ECD). The pharmacokinetic parameters were calculated by statistical analysis and compared. Results The pharmacokinetic parameters of combination administration group were C_ max (782.63?469.37) ng/mL, AUC_ 0-24 h (8 407.86?3 476.14) ng/mL?h; while the individual administration group of baicalin was C_ max (2 645.62?601.42) ng/mL, AUC_ 0-24 h (28 952.90 ?5 731.42) ng/mL?h. Objective To observe the pain behavioral performance of rats that different sensory nerve fibers were transected,and examine the expression of brain-derived neurotrophic factor(BDNF) in these models.Methods Twenty-four rats were divided into three groups according to random number table method:SUR group,GS group and SHAM group, which received sural nerve transection, gastrocnemius-soleus nerve transection or sham operation respectively.There were 8 rats in every group.The expression of BDNF in the lumbar 5 DRG and spinal dorsal horn were detected,and the types of damaged cells were also observed.Results In GS group, 50% paw-withdrawal thresholds were significantly decreased on the ipsilateral hind paw compared with baseline and with those in SHAM group,and the paw-withdrawal durations in response to the thermal stimulus increased significantly (P<0.01 =.In contrast, no change was found in SUR group(P>0.05 ).The expression of BDNF in the lumbar 5 DRG ( (37.87 ± 4.23 ) % ) and spinal dorsal horn ( (21.9 ± 3.1 ) % ) was significantly higher in GS group than in SHAM group( ( 17.31 ± 2.12 ) %, ( 12.6 ± 1.3 ) % ), and no significant difference was found between SUR and SHAM groups(P>0.05 ).FG opposite cells which also expressed BDNF in GS group were more than those in SUR group ( (47.7 ± 1.8) % and (26.7 ± 2.3 ) % ) (P < 0.01 =.The percentage in N200 and FG double positive cells to N200 positive cells in GS group was significantly increased in GS group than those in SUR group ( (47.7 ±1.8 ) %, (26.7 ± 2.3 ) % ) (P < 0.01 =.Conclusion The data suggest that injury of the sensory nerve innervating skin does not produce hyperalgesia, but injury of the sensory nerve innervating muscle does.Different kinds of neuron were damaged and the differences of BDNF expression is essential for this difference.
3.Clinical significance of MRI and transrectal ultrasound combined with carcinoembryonic antigen examination on accurate preoperative staging of rectal carcinoma

Objective To study the consistency in the diagnosis of preoperative TNM rectal cancer staging using high resolution magnetic resonance imaging (MRI) and transrectal ultrasound (TRUS) combined with carcinoembryonic antigen (CEA) and postoperative pathological TNM.Methods 156 cases pathologically proven were retrospectively analyzed and divided into 4 groups including preoperative MRI group (39 cases),TRUS group (39 cases),MRI and TRUS group (39 cases),MRI and TRUS combined with CEA group (39 cases).The differences between preoperative T,N staging and postoperative pathologic T,N staging were analyzed.Results There were statistically significant differences in the diagnosis of preoperative T and postoperative pathological T in 4 groups (T: Kappa =0.685,P =0.000; N: Kappa =0.544,P =0.000),but there were no significant differences in preoperative N and postoperative pathological N staging in preoperative MRI group,TRUS group,MRI and TRUS group (Kappa =0.142,P =0.329; Kappa =0.154,P =0.645; Kappa =0.154,P=0.229),and significant difference was observed in MRI and TRUS combined with CEA group (Kappa =0.544,P =0.000).There were no significant differences in the accuracy of T staging among the 4 groups (x2 =0.326,P =0.574; x2 =0.562,P =0.719; x2 =0.287,P =0.986),but significant difference in the accuracy of N staging were showed among the 4 groups (x2 =4.643,P =0.026; x2 =6.643,P =0.026; x2 =5.243,P =0.019).Conclusion Preoperative evaluation by the MRI add TRUS combined with CEA can improve the accuracy of preoperative staging,which can provide more reliable basis for decision-making and improve the coincidence rate of operative procedures in line with the estimate.It also provides the basis fur the accurate preoperative diagnosis and individualized treatment.

Cerebral apoplexy is one of the main causes of death in the middle-aged and elderly population,which has higher mortality and disability rate.The incidence of the disease is increasing year by year and it is a serious threat to human life and health.Therefore,it is of great significance to find an effective target for the diagnosis and treatment of stroke.Thioredoxin (Trx) is the major thiol reducing agent in the cells,it is involved in many signal transduction pathways in the cells by regulating the redox state of the cell.It has disulphide reductase activity,which can reduce the oxidative stress injury in the rats after the stroke.Thioredoxin interacting protein (TXNIP) is an endogenous inhibitor of Trx,it can destroy the redox balance and promote the oxidative stress by binding/inhibiting the activity of Trx,while the inhibition or knockdown of TXNIP has obvious neuroprotective effects.Recent studies suggest that Trx/TXNIP may be involved in the pathophysiology of cerebral apoplexy by a variety of pathways.This article analyses the research status of Trx/TXNIP and studies the localization of Trx system in the central nervous system and the progress of Trx system in ischemic cerebral apoplexy.It reviews the mechanism of Trx/TXNIP in cerebral apoplexy and prospectes the signaling pathways involved in the pathophysiological process of Trx/TXNIP to provide new ideas for the treatment of cerebral apoplexy.
5.Study on cell-penetrating peptides carrying foreign proteins into eyeball

Objective To investigate whether poly arginine as the carrier can carry foreign proteins to penetrate the cell membrane and even penetrate the eyeball barrier.Methods Poly-Args (R9) was used as a CPP in this study.R9-green fluorescent protein (GFP) and GFP were constructed.In vitro,human lens epithelial cells were treated with these two proteins.Then,MTT assay were used to detect whether the protein could affect the proliferation of the cells.Flow cytometry and laser confocal microscopy were used to detect the penetrability of CPPs on the cells.In vivo,eyes of mice were treated with protein in eye drops way for 7 days.Then total protein were extracted,ELISA were used to detect the penetrability of CPPs.Results The results of MTT,flow cytometry and laser confocal microscopy showed that CPPs could carry protein into cells in a dose dependent manner without affecting cell proliferation.In vivo,slit lamp showed that the mice eyeballs had no any abnormal after treated by GFP,R9-GFP,and ELISA results also showed that R9 could effectively get foreign protein into the eyeball.Conelusion R9 can carry foreign protein into the cell membrane and eyeball barrier.This study provides the basis for the eye medication and dosing mode improvement.
6.Correlations of prognosis, outcomes with myocardial dysfunction of aneurysmal subarachnoid hemorrhage

Objective To investigate the correlation between aneurysmal subarachnoid hDepartment of Neurosurgery,Xi Dian Group Hospital,Xi'an 710077,Chinaemorrhage (aSAH) and myocardial dysfunction (MD).Methods Totally 96 cases of aSAH patients meeting the criterion were chosen,and the correlations of electrocardiograph (ECG),and ultrasonic cardiogram (UCG)with delayed cerebral ischemia (DCI),death and poor outcome and death were analyzed.Results Of the 93 cases of aSAH patients meeting the criterion,DCI rate was 59.2%,death rate was 19.4%,and poor outcome rate was 21.5%.With multivariable Poisson regression analysis,the results showed that wall motion score index (WMSI) > 1.1 was correlated with death and poor outcome,mid-ventricular wall motion abnormality (WMAs) was correlated with death,and apical WMAs was correlated with DCI.Conclusions Myocardial dysfunction is significantly related to outcome of aSAH.Thus,this study may provide a reference for prognosis assessment and treatment of aSAH.
7.Effects of Embolization and Clipping Therapy on Cognitive Function in Patients with Intracranial Aneurysms

Objective:To evaluate the effects of endovascular embolization and microsurgical clipping on cognitive function in patients with intracranial aneurysm.Methods:A total of 80 patients with intracranial aneurysms,who were treated in Chongqing Hospital of Traditional Chinese Medicine from March 2014 to September 2015,were selected and randomly divided into control group(n=40) and experimental group (n=40).The control group was treated with microsurgical clipping,and the experimental group,with endovascular embolization.The postoperative cognitive function was assessed by the mini mental state examination scale (MMSE),and length of stay,in-hospital mortality rate,mortality rate in 1 year and the incidence of postoperative adverse reactions were compared between the two groups.Results:The total score (26.78± 0.85) of MMSE in the experimental group was higher than that (22.25± 0.63) of the control group (P<0.05).The cognitive dysfunction rate (37.50%) of the experimental group was lower than that (55.00%) of the control group (P<0.05).The length of stay [(6.7± 3.9) days] of the experimental group was lower than that [(9.6± 4.5) days] of the control group(P<0.05).There were no significant differences in in-hospital mortality rate and mortality rate in 1 year,and the incidence of adverse reactions between the two groups (P>0.05).Conclusion:Endovascular interventional therapy can improve the cognitive function of the patients with intracranial aneurysms and shorten the length of hospital stay.
8.The correlations between the different pathological types gastritis and the expressions of AQP3 and AQP4

Objective To explore the correlations of expressions of gastric mucosa water channel aquaporin AQP3 and AQP4 and different gastritis types. Methods The gastric mucosa was mounted under gastroscope. The types of gastric mucosa pathology and activity were tested by the common pathohistology. The expressions of AQP3 and AQP4 were determined by immunohistology. Results The expressions of AQP3 and AQP4 of chronic superficial gastritis were significantly higher than those in the non-gastritis group and chronic atrophic gastritis group (P < 0.01), especially in the activity period of chronic superficial gastritis. The expressions of AQP3 and AQP4 of chronic atrophic gastritis group were reduced when compared to those in the non-gastritis group, in spite of no statistical differences between them. While compared to the non-gastritis group , the expression of AQP3 of chronic atrophic gastritis group during the active stage was remarkably decreased (P < 0.05). Conclusion The expressions of AQP3 and AQP4 of gastric mucosa in chronic gastritis in various pathological types are different. AQP3 and AQP4 may be the targeted point , which could be used for the differential diagnosis and treatment of chronic gastritis of different pathological types.
9.Association between cerebrospinal fluid viral load and hearing loss in neonates with congenital symptomatic cytomegalovirus infection

Objective To determine the relationship between the viral load of cerebrospinal fluid (CSF) and sensorineural hearing loss (SNHL) in newborns with symptomatic congenital cytomegalovirus (CMV) infection.Methods The study cohort comprised 36 newborns with symptomatic congenital CMV infection.CSF from all of the patients were analyzed for the presence of CMV DNA by PCR.Audiological function were performed on them by brain stem auditory evoked potential at birth,at 6 and 12 months of age.Results (1)Of the 36 newborns,15 cases (41.7%) had positive CSF PCR result,17 cases (47.2%) had SNHL.(2) The prevalence of SNHL in the group of newborns with positive CSF PCR result was 60.0% (9/15),and it was 38.1% (8/21) in the group of newborns with negative CSF PCR result,there was no significant difference of the prevalence of SNHL between the two groups (P =0.194).(3) In newborns with positive CSF PCR result,the amounts of CSF CMV DNA was not different between the newborns with SNHL and normal hearing (3.35 ± 0.68 vs.3.17 ± 0.56,P =0.36).Conclusion A positive CSF PCR result and the CMV viral load of CSF did not correlate with SNHL.
10.Regulation of saw palmetto extract on immune function in GL261 glioma

Objective:To investigate the effect of saw palmetto extract ( SR ) on GL261 glioma in rats and immune system.Methods:The 40 rats were divided into 4 groups randomly,one was the control group without tumor( the blank group) ( n=10) ,the other 30 rats were given subcutaneous inoculation of tumor cells and then divided into 3 groups:the tumor-bearing group( n=10) ,low dose SR group( n=10) ,and high dose SR group( n=10).After 1 week fed,the rats of SR groups were given the saw palmetto extract,low dose group 50 mg/kg once a day every other day and 300 mg/kg of high dose group every other day.The tumor-bearing groups received the same dose of distilled water.After 4 weeks fed, we measured the tumor weight and the inhibition rate was calculated.The tumor cell apoptosis was detected by TUNEL staining.The activity of splenic macrophages was detected by macrophage phagocytosis of chicken erythrocytes and CD4+lymphocytes by flow cytometry.Results:①The SR groups compared with tumor group, the tumor weight was significantly reduced(F=62.678,P=0.000).The tumor inhibition rate was significantly higher in high dose group.②The apoptosis of tumor cells in tumor-bearing group was significantly less than SR groups and the apoptosis was significantly increased after treatment with SR ,especially in high dose SR group(F=1.287,P=0.000).③Compared with the blank group,the macrophage activity decreased remarkablely.SR enhanced the macrophage activity obviously, positively related to doses of SR ( F=141.205,P=0.000;F=126.903,P=0.000).④In the tumor-bearing group,the CD4+lymphocyte count decreased significantly.After SR treatment,the CD4+lymphocyte increased and were positively related to the dose of SR.Conclusion: Saw palmetto extract can strengthen the immune system.
