1.Abnormality of water molecular diffusion of rats after focal cerebral ischemia

BACKGROUND: Some researches report that diffusion-weighted imaging (DWI) can appear abnormal signal in minutes after cerebral ischemia.OBJECTIVE: To investigate the manifestation and regularity of diffusion coefficient in middle cerebral artery occlusion (MCAO) of rats after cerebral ischemia.DESIGN: Randomized controlled study.SETTING: Neurological Department of Clinical College of ChongqingMedical University.MATERIALS: The experiment was conducted at the Radiological Department of the First Affiliated Hospital of Chongqing Medical University from April to July 2004. Totally 18 adult Wistar rats with clean grade were randomly divided into three groups: control group, sham operation group and cerebral ischemia group with 6 rats in each group.METHODS: The origin of right middle cerebral artery was blocked with filament into right internal carotid. In the sham-operated group, the filament was just inserted into the common carotid artery not into the origin of middle cerebral artery, rats in normal control group were not treated with anything after anesthesia. MRI experiments were performed on a 1.5 T GE signa Highspeed NMR spectrometer to obtain DWI and ADC values at the time-point of 7, 15, 30, 60 minutes, 3,6, 12, 24 hours, 3, 7, 15 days, 1 and 2 months after operation.MAIN OUTCOME MEASURES: Changes of ADCs in infarct hemisphere and contralateral hemisphere.RESULTS: There was no significant change in MRI T1-and T2-weighted imaging in rats at 7 minutes post-infarction, but abnormal high intensity was emerged during the period of 1 hour to 3 hours, and enlarged over time. Finally, the infarction area located the right basal ganglia and the frontal-parietal lobes at 12 hours and increased less significantly later.Focal abnormal high signal was noted in DWI 7 minutes after cerebral ischemic insult, and the ADC values was decreased at the same time,reduced gradually after significant recover at 30 minutes, dropped to the lowest level at 12 hours and again increased at 24 hours, since then all the ADC values were remained high throughout follow-up, which increased by 5 times at 2 months than that at 12 hours. Correspondingly,rADC values showed the contrary pattern of evolution.CONCLUSION: It is reflected that ADC is highly sensitive not only to onset of cerebral ischemia but also to chronic cerebral ischemia.
2.Comparative study on three resin cements to restore anterior flared roots

BACKGROUND: Fiber post is widely used in front teeth aesthetic repair, but some defect teeth cannot form ferrule or thick root canal, which resulted in fiber post prosthetic replacement defluxion and a failure of repair. Presently, new-style resin cement had self sticking component, showing good effects in sticking of fiber post. Whether combination of new-style resin cement can repair affected teeth using fiber post deserves further investigation.OBJECTIVE: Through fatigue test, residual flexural strength and scanning electron microscope (SEM) observation to compare the strength of severely weakened roots restored with three resin cements. METHODS: Eighteen intact maxillary central incisors were collected and formed to severely weakened canals at the same size. Three kinds of resin cements were used to restore. Rely X Unicem, Panavia F and Super-Bond C&B resin cement and identical glass fiber post were used. All the specimens were restored using Ni-Cr ceramic crown, and placed on TCML chewing machine that loaded 1 200 000 cycle forces, and the cycle times when failure occurred were recorded. The samples were intact following loading received residual flexural strength testl SEM was employed to observe breakage surface of the samples. RESULTS AND CONCLUSION: Mean cyclic loading was significantly greater in the Super-Bond C&B Group (1 200 000 times) than in the Relyx Unicem Group (640 000 times) and Panavia F Group (550 000 times) (P< 0.05). No significant difference was detected between the Rely X Unicem and Panavia F Groups. Residual flexural strength was 747.99 N in the Super-Bond C&B Group. SEM demonstrated that Super-Bond C&B formed longer resin processes, which were more than other two groups. Results confirmed that Super-Bond C&B demonstrated better results when restoring flared roots with prefabricated glass fiber posts.

It has been a problem in clinical practice that recurrence and distant metastasis of patients with colorectal cancer undergoing surgery.Recently,pericolonic tumor deposits has attracted extensive attention of scholars,and they think pericolonic tumor deposits may be closely related with colorectal cancer postoperative recurrence and metastasis.This review aims to introduce concept of pericolonic tumor deposits,pathological characteristics and its important role in clinical staging and prognosis,so as to provide clinical doctors medical advice to set more reasonable treatment and achieve the purpose of reducing tumor recurrence or metastasis for patients with colorectal cancer.
4.AFFERENT CONNECTIONS OF THE PREOPTIC REGION——STUDIED WITH HRP METHOD IN THE RAT

The afferent connections of the magnocellular preoptic (poma), lateral preoptic (pol), and medial preoptic (pom)nuclei have been investigated with horseradish peroxidase (HRP) method in the rat. Our findings indicated that1. poma mainly receives projection from the rhinencephalic structures including the anterior olfactory nucleus, olfactory tubercle, piriform cortex, entorhinal cortex, the nucleus of lateral olfactory tract, the anterior amygdaloid area, and the cortical amygdaloid nucleus;2. pom mainly receives afferent fibers from some of the medial hypothalamic nuclei;3. pol has the most extensive fiber input, mainly from some of the brain stem nuclei; and4. poma is the only one of the preoptic nuclei which has reciprocal connections with the contralateral forebrain structrues. It is interesting that the poma and pol receive the afferent fibers from different prefrontal cortical regions and the dorsal portion of pom may have input from the lateral reticular nucleus and nucleus tractus solitarii that the ventral portion may not have.In addition, a large number of HRP-labeled cells could be seen in the supraoptic nucleus after injecting any one of the preoptic nuclei and the possible signification was discussed.

6.The analysis of the curative effect for the lipitor in curing essential hypercholesterolemia

Objective To evaluate the curative effect of the atorvastatin calcium tablet(trade name:lipitor) in curing essential hypercholesterolemia.Methods 76 patients with essential hypercholesterolemia were divided into two groups randomly:43 patients in the research group(lipitor tablet 10mg,qn);33 patients in the comparison group (simvastatin tablet 20mg,qn).The period of treatment for both two groups was 6 weeks.The changes of blood fat were detected before and after taking the medicine.The curative effect were compared at the same time.Resnlts The total cholesterol(TC)of the research groups decreased by 37.06%,the TC of the comparison group decreased by 22.05%.The triacylglyceride(TG)of the research group decreased by 24.81%,however,the TG of the compar- ison group decreased by just 14.49%.The low-density lipoprotein cholesterol(LDL-C)of the research group de- creased by 42.30% while the comparison group just decreased by 28.30%(P<0,01).Also,the high-density lipoprotein cholesterol(HDL-C)of the research group increased by 7.80%,and the comparison group increased by 7.13%(P>0.05),The effective rate of the research group was 95.35%.For the comparison group,the effective rate was 90.91%.Conclusion The lipitor has a good curative effect in curing essential hypereholesterolemia.Also, the lipitor has fewer side effect,This medicine is safe and reliable for patients.
7.The impact of serum levels of immunoglobulin and complement during nucleoside antiviral treatment in chronic hepatitis B patients

Objective To evaluate the effects of nucleoside/nucleotide analogue treatment on immunoglobulin and complement in patients with chronic hepatitis B (CHB).MethodsA total of 157 CHB patients were recruited and divided into CHB group,liver cirrhosis (LC) group and severe hepatitis B (SHB) group.There were 50 patients who received oral antiviral treatment (lamivudine 100 mg/d,or entecavir 0.5 mg/d,or telbivudine 600 mg/d).Serum levels of complement 3 and 4 (C3,C4),C-reaction protein (CRP),hemolytic complement (CH50),immunoglobulin G,M,A (IgG,IgM,IgA),hepatitis B surface antigen (HBsAg) and hepatitis B e antigen (HBeAg) were detected by enzyme-linked immunosorbent assay (ELISA) or immunoturbidimetry.Hepatitis B virus (HBV) DNA was quantified by real-time polymerase chain reaction (RT-PCR) before and 1,2,3 and 4 weeks after nucleoside antiviral therapy.Comparison of means was done by t test and Mann-Whitney test.The correlation was analyzed by Pearson correlation coefficient test.ResultsSerum IgA and IgM levels of SHB and LC patients were significantly higher than those of CHB patients (P<0.01).Levels of C3,C4,CH50 and CRP were significantly different among three groups.Levels of C3,IgM,IgG and HBV DNA in HBeAg positive patients were significantly different from those in HBeAg negative patients.There was a statistically significant difference of IgA,IgM,C3 and CH50 levels between patients with high HBV DNA level and low HBV DNA level in HBeAg-positive patients.While in the HBeAg-negative patients,only the IgA level was significantly different with HBV DNA levels.After anti-viral treatment,immunoglobulin and HBV DNA levels were all decreased in three groups,while the serum complement level was increased compared to baseline,and the differences became significant at week 4 of treatment. HBV DNA level was negatively correlated with C3 (r=-0.78,P=0.021) and HBeAg titer was positively correlated with C3 (r=0.87,P=0.015).ConclusionsThe immunoglobulin,CRP,C3,C4,and C H50 could reflect the inflammatory activity in liver.The changes of C3 level can predict the efficacy of antiviral therapy.
8.EFFECTS OF INSULIN-LIKE GROWTH FACTOR 1 ON β-AMYLOID INDUCED NEURON APOPTOSIS AND TAU PHOSPHORYLATION

The aim of this study is to elucidate the protective and anti-apoptotic effects of insulin-like growth factor 1 ( IGF-1 ) against β-amyloid (Aβ) and investigate the effect of IGF-1 on Aβ-induced tau phosphorylation. Cell viability was measured using the MTT (3-(4,5-dimethylthiazolyl-2 )-2,5-diphenyltetrazolium bromide) assay, early apoptosis and late apoptosis/necrosis were analyzed by flow cytometry using Annexin V-FITC and propidium iodide (PI) double staining, and morphology was examined by Hoechst 33342 staining. Tau phosphorylation was detected using AT8 immunostaining. Preincubation of cultured rat hippocampal neurons with IGF-1 for 24 h prevented cytotoxicity induced by Aβ25-35 for 48 h. The MTT value significantly increased from 54.51% to 61.8% of the control group, and the percentage of Hoechst 33342-positive cells decreased from 30.77% to 22.81%. Incubation with Aβ25-35 for 48 h caused a marked increase in the percentages of Annexin V-FITC single-labeled cells (Annexin V +/PI-) and Annexin V/PI double-stained cells (Annexin V +/PI + ) (3.41% and 19.47% , respectively), which were significantly decreased by pretreatment with 100 ng/ml of IGF-1 for 24 h (to 2.98% and 15.16% , respectively). Aβ25-35 treatment increased tau phosphorylation and AT8 positive cells were 41.84%. This effect could be inhibited by different concentrations of IGF-1. Our findings showed that IGF-1 protected against Aβ-induced cytotoxicity, decreased the percentage of early and late apoptosis/necrosis cells, and inhibited tau phosphorylation, which may be the cellular mechanisms for its neuroprotective action.

10.Changes of sex hormone levels and sexual function in male patients after liver transplantation

ObjectiveTo investigate changes of sex hormone levels and sexual function in male patients after liver transplantation.MethodsBlood samples were taken from 69 male patients receiving liver transplantation pretransplantation and postoperative 3rd,7th,14th day,1st and 3rd month respectively.Serum estradiol and testosterone were measured by radioimmunoassay.Serum sex hormone binding globulin was measured by ELISA.Sixty-nine discharged adult male liver transplant recipients ( age ranging 24 -45 years) with a normally functioning allograft for at least 6 months were questioned on their pre-and post-operativesexualfunction compared with 23adulthealthy controls.ResultsPretransplantation serum estradiol and sex hormone binding globulin were significantly higher (87.56±31.21 vs.26.00 ±9.12,u=9.30,P<0.0001,134.50±30.68 vs.51.04 ±12.05,u=12.69,P < 0.0001,respectively),and testosterone was lower than normal controls (2.02 ± 1.28 vs.4.82 ± 1.48,u =-8.73,P <0.0001 ) and levels were correlated with MELD scores (r =0.80,P <0.0001,r =-0.77,P <0.0001,r =0.72,P <0.0001,respectively).Sex hormone levels were back to normal on postoperative 2nd week (serum estradiol) or 1st month (serum testosterone and sex hormone binding globulin).Compared with pretransplant status,sexual function improved significantly when evaluated on postoperative 6th month.ConclusionsLevels of serum sex hormone were correlated with MELD scores before liver transplant.After liver transplant,estradiol was back to normal level on postoperative 2nd week,testosterone and sex hormone binding globulin back to normal level on postoperative 1 st month.Sexual function significantly improves after liver transplantation.
