Objective:To study the effects of 1?,25-dihydroxyvitamin D 3 and UVB on cell proliferation and melanin synthesis of normal human melanocytes. Methods:Melanocytes of foreskins of healthy men were cultured and treated with various concentration of 1?,25-dihydroxyvitamin D 3 or UVB (55 mJ/cm 2),or both.Cell proliferation was measured with MTT assay .The synthesis of melanin was determined by chromatography. Results: 1?,25-dihydroxyvitamin D 3 and UVB could promote the proliferation of cultured melanocyte,and 1?,25-dihydroxyvitamin D 3 could promote the melanin synthesis.The significant concentration of 1?,25-dihydroxyvitamin D 3 ranging from 10 -7 to 10 -10 mol/L.Conclusion:1?,25-dihydroxyvitamin D 3 and UVB irradiation could promote the proliferation of melanocyte,which indicates that they might be effective in the treatment of vitiligo.

Objective To study the effects of endothelin-1 (ET-1) and adrenocorticotropic hormone (ACTH) on the proliferation of cultured normal human melanocytes. Methods Normal human melanocytes were obtained from the foreskins of healthy men, and cultured in minimum essential medium (MEM) with 10% bovine serum. The cultured melanocytes of the 3rd passage were treated with various concentrations of ET-1 and ACTH. Cell proliferation was assessed by the MTT method. Objective:To investigate the effect of the integrin-linked kinase(ILK) small interfering RNA(siRNA) on prostate cancer in nude mice by orthotopic injection of human cell line DU145.Methods:The cultured human cell line DU145 was knocked down for ILK using a siRNA.Cellular ILK expression was quantified by RT-PCR and Western blot analysis.Moreover,cell attachment,invasiveness and microfilament dynamics assays were performed.Furthermore,the impact of the ILK siRNA on the prostate cancer was tested using a nude mice model in which prostate cancer was induced by orthotopic injection of human prostate cancer cell line DU145.Gross tumor volume of prostate in nude mice,cell differentiation,the state of apoptosis and proliferation were tested after 5 weeks of injection.Results:The expression of ILK was suppressed significantly by siRNA,cellular mRNA and protein of ILK decreased 87% and 81% separately.The knockdown of ILK also induced the attachment and invasiveness of DU145 cell growing down.The tumor volume,cell differentiation,apoptosis index and proliferation index of prostate in nude mice of ILK siRNA orthotopic injection model were significantly smaller,better,increased and decreased separately than those in control group.Conclusion:Targeting inhibition of ILK not only decreases attachment and invasiveness of human DU145 cells,but also suppresses the growth and development of prostate cancer of orthotopic injection human DU145 cell line model in nude mice.
4.Clinical application of 320-detector CT in interventional treatment of bronchial artery hemoptysis

Objective To evaluate the clinical value of 320-detector CT in interventional treatment of bronchial artery hemoptysis. Methods CTA and DSA images of 30 patients with bronchial artery hemoptysis were retrospectively analyzed.Spatial anatomical characters of the bronchial arteries,such as the type of branches,origin and opening positions of the bronchial arteries were observed and recorded.Results In 30 patients,6 bronchial arteries distribution patterns were found,and the most common type was R1 L1 (43.3%).83 bronchial arteries were identified using CTA,including 38 on the right and 45 on the left.The right bronchial arteries mainly originated from the intercostal artery (52.6%),while the left bronchial arteries mainly from the descending aorta and aortic arch (82.2%).The opening positions of right and left bronchial arteries were mainly located at the right wall of the descending aorta (78.9%),and anterior wall of the descending aorta (62.2%),respectively.When the cacarina of trachea was used as the reference position,the left and right bronchial arteries were mainly located in the range of above 2 cm to below 1 cm from tracheal bifurcation, accounting for 80% and 89.5%,respectively.Compared with DSA,the sensitivity and specificity of CTA were 97.5% and 100%, respectively.Conclusion 320-detector CT can be used to clearly display the distribution patterns,origin and opening positions of bronchial arteries,and especially to find bronchial arteries with ectopic origin.It is possible to apply 320-detector CT in preoperative routine examination and postoperative evaluation of massive hemoptysis.
5.BODY-COMPOSITION ASSESSMENT BY DUAL-ENERGY X-RAY ABSORPTIOMETRY IN PRESCHOOL CHILDREN

Objective: Our study described the body composition of 50 preschool children aged from 4 to 6 years old and evaluated their nutritional and developing status. Methods: Body composition was measured with dual energy X ray absorptiometry(DXA). Results: BMC, BMD, FTM, %BF, LTM of all children were (463.3?80.9)g、(0.69?0.04)g/cm 2、(3917.1?1281.6)g、(21.8?4.9)、(13391.4? 1823.8 )g respectively. The relationships between BMC and age and between LTM and age were positively correlated. BMD and LTM of the boys were significantly higher than those of the girls. On the contrary, FTM, %BF, F t/l and F t/a of the girls were obviously greater than those of the boys. Conclusion: DXA method may be used to evaluate the nutritional and developing status in preschool childran.
6.Efficacy and safety of two kinds of homemade sirolimus-eluting stents for treatment of acute ST segment elevation myocardial infarction

Objective To compare the efficacy and safety of two kinds of homemade sirolimus-eluting stents (Firebird and Excel) for treatment of acute ST segment elevation myocardial infarction (STEMI) in patients who underwent percutaneous coronary intervention (PCI). Methods The 249 consecutive patients with STEMI who underwent PCI were randomly divided into two groups: Excel group (n=136) and Firebird group (n=113). They were followed up for 6-24 months, and coronary angiography was reviewed average 12 months later. The primary endpoints were major adverse cardiac events, including death, reinfarction and target vessel revascularization. The second endpoints included late luminal loss and restenosis 12 months after treatment. Results There were no significant differences in baseline data, coronary arterial lesion before operation, and immediateness condition after PCI between the two groups (all P>0.05). Within follow-up, there were 2 (1.47%) death cases and 1 (0.88%) death case, 1 (0.74%) and 1 (0.88%) nonfatal myocardial infarction case, 2 (1.47%) and 2 (1.77%) target vessel revascularization cases in the two groups respectively (all P>0.05). There were no significant differences in late luminal loss of in-stent and in-segment, the rates of in-stent restenosis, in-segment restenosis and stent thrombosis, the in-stent minimal lumen diameter and in-segment minimal lumen diameter between the two groups (all P>0.05). Conclusions The two kinds of homemade sirolimus-eluting stents may have similar efficacy and safety in patients with STEMI treated with primary PCI.
7.Expressions of desmoglein 1 and desmoglein 3 in primary keratinocytes and two keratinocyte cell lines HaCaT and A431

Objective To detect the mRNA and protein expressions of desmoglein 1 (DSG1) and DSG3 in different types of keratinocytes (KCs).Methods Two keratinocyte cell lines HaCaT and A431,as well as primary keratinocytes from human abdomenal skin served as the object of this study.Direct immunofluorescence assay was performed to observe and quantify the expressions of DSG1 and DSG3,and quantitative PCR (qPCR) to determine the mRNA expressions of DSG1 and DSG3,in these cells.Results Both DSG1 and DSG3 were expressed in all the three types of keratinocytes,and the fluorescence intensity of DSG1 and DSG3 in HaCaT cells was higher than that in primary keratinocytes but lower than that in A431 cells.Similarly,all the keratinocytes expressed DSG1 and DSG3 mRNA,with the relative expression levels of DSG1 and DSG3 mRNA in primary keratinocytes being 291.7% and 237.4% of those in HaCaT cells respectively (both P < 0.01),and those in A431 cells being 0.1% and 18.8% of those in HaCaT cells respectively (both P < 0.05).Conclusions HaCaT cells,A431 cells and primary keratinocytes all can be used for the study of DSG1 and DSG3,of which,A431 cells show the strongest expressions of DSG1 and DSG3,and primary keratinocytes display the highest expressions of DSG1 and DSG3 mRNAs.
8.Targeting of integrin-linked kinase with a small interfering RNA inhibits human penile smooth muscle cell contraction and stretch in vitro

Objective To investigate the changes of human penile smooth muscle cell contraction and stretch resulting from the integrin-linked kinase (ILK) gene knock-down.Methods Cultured human penile smooth muscle cells were knocked down for ILK using a small interfering RNA (SiRNA).Cellular ILK expression was quantified by Western blot analysis.Cell attachment,spreading and migration were also performed.Furthermore,microfilament dynamics was tested by means of Alexa Fluor 488 phalloidin stain.Results First,blocking the expression of ILK by siRNA significantly inhibited human penile smooth muscle cell attachment,speading and migration; moreover,ILK down-regulation affected actin cytoskeleton reorganization and changed cell morphology in human penile smooth muscle cell.Conclusions Targeting of ILK with a small interfering RNA not only inhibits human penile smooth muscle cell attachment,spreading and migration,but also effectively suppresses microfilament dynamics.This may be of potential therapeutic usefulness in treating erectile dysfunction (ED).
9.Expression of ET_(A-R) Gene in Cultured Normal Human Melanocytes

Objective To explore the expression of high selective A receptor of endothelin-1 (ETA-R) in melanocytes from normal human beings. Methods Normal human melanocytes were cultured and mR-NA expression of ETA-R in melanocytes was measured by reverse transcription-polymerase chain reaction. Results The mRNA expression of ETA-R was seen in melanocytes. Conclusion This study combined with other experiments suggests that endothelin-1 may affect melanocytes by high selective ETA-R which may con-tribute to the pathogenesis of vitiligo.
10.Study on Activities of Phase Ⅰ and Ⅱ Drug-metabolic Enzymes in Spleen of Rats with Spleen-Kidney Yang Deficiency Syndrome

Objective To study the changes of activities of phase Ⅰ and Ⅱ drug-metabolic enzymes in the spleen of rats with spleen-kidney yang deficiency syndrome.Methods The rat model of spleen-kidney yang deficiency syndrome was established by gastric gavage of Radix et Rhizoma Rhei decoction combined with injection of hydrocortisone for 17 continuous days.And then we detected the activities of 6 kinds of phase Ⅰ drug-metabolic enzymes of CYP2C19,CYP2D6,CYP2C9,CYP1A2,CYP2C8,CYP3A4,and 4 kinds of phase Ⅱ drugmetabolic enzymes of phenol sulfotransferase (PST),uridine diphosphate glucuronosyl transferase 1 (UGT1),glutathione transferase (GST),estrogen sulfotransferase (SULT1E1) in the spleen.Results Compared with the normal control group,the activities of PST,UGT1,GST and SULT1E1 in the model group were significantly decreased (P < 0.05 or P < 0.01),and the activity of CYP1A2 was significantly increased (P < 0.01),while CYP2C19,CYP2D6,CYP2C8,CYP3A4,CYP2C9 enzymes showed no obvious changes(P > 0.05).Conclusion The activities of splenic drug-metabolic enzymes,in particular the phase Ⅱ enzymes,are significantly varied under the state of spleen-kidney yang deficiency.
