One hundred and seventeen patients with newly diagnosed autoimmune thyroid diseases(AITDs)and 30 normal controls(NCs)were selected. All the patients were divided into 5 groups:hyperthyroidism group(n=33),subclinical hyperthyroidism group(n=21),hypothyroidism group(n=25),subclinical hypothyroidism group(n=23)and euthyroidism group(n=15).Plasma malondialdehyde(MDA),superoxide(SOD)and inflammatory factors levels were measured. Our results showed that plasma MDA level in dysthyroidism groups was significantly higher than that in NCs group(P<0.05 or P<0.01).SOD/MDA ratio in AITD groups Was significantly lower than that in NCs group(P<0.05 or P<0.01).In a multiple step-wise regression analysis, serum hishly sensitive C-reactive protein(hs-CRP)and tumor necrosis factor-α(TNF-α)were independent factors of MDA.This study might suggest that plasma MDA level in AITD patients receiving no treatment may increase.MDA may be correlated with hs-CRP or TNF-α levels.

Objective To investigate the relationship between oxidative stress and lipid metabolism characteristics in elderly patients under different status of thyroid function. Methods Eighty-six newly-diagnosed elderly patients with thyroid diseases,83 non-elderly patients with thyroid diseases and 20 normal subjects as control were selected.and 47 hyperthyroidism and 39hypothyroidism in elderly,43 hyperthyroidism and 40 hypothyroidism in non-elderly were diagnosed according to diagnostic criteria of the thyroid disease. The levels of fasting plasma malondialdehyde(MDA),superoxide(SOD)and oxidized low density lipoprotein(OX-LDL)were measured by thiobarhiturates, colorimetric method and enzyme linked immunosorbent assay(ELISA).Meanwhile, lipid profile and thyroid function were also measured,SOD/MDA ratio was calculated. Results Lipid profiles in the elderly hyperthyroidism group were significantly higher than those in the non-elderly group(P<O.05 or P<0.01)but were significantly lower than those of normal control group(P<0.05 or P<0.01).The plasma MDA[(10.23±6.29)μmol/L vs.(3.66±2.53)μmol/L vs.(7.37±4.58)μmol/L],free fatty acids(FFA)[(0.86±0.58)mmol/L us.(0.45±0.12)mmol/L vs.(0.61±O.46)mmol/L]and SOD levels were significantly higher in elderly patients with hyperthyroidism than in normal control and non-elderly hyperthyroidism groups(P<0.05 or P<O.01).In the patients with hypothyroidism, the plasma MDA[(9.03±5.98)μmol/L vs.(6.59±3.18)μmol/L vs.(3.66±2.53)μmol/L],OX-LDL[(387.36±71.04)μg/L vs.(355.22±45.01)μg/L vs.(324.53±56.19)μg/L],TC,LDL-C,non-HDL-C,LP(a) and Apo-B100 levels were significantly higher in the elderly group than in the non-elderly and normal control groups(P<0.05 orP<0.01).The SOD/MDA ratio was significantly lower in the elderly patient groups with hyperthyroidism or hypothyroidism than in normal control and non-elderly groups(all P<0.01).In a multiple step-wise regression analysis.FT4 and FFA were independent factors related tO MDA in hyperthyroidism group, and non-HDL-C and LDL-C were independent factors related to MDA inhypothyroidism group. Conclusions Serious lipid metabolic disorder exists in newly-diagnosed and untreated elderly patients with hyperthyroidism and hypothyroidism. It is suggested that oxidative stress is obviously strengthened in elderly patients with thyroid function disorder and that the degree of oxidative stress is correlated with lipid metabolic disorder.

Objective To study influences of intervention in pregnant women with positive thyroid autoantibodies on the thyroid function of babies. Methods A total of 55 pregnant women were enrolled with positive thyroid peroxidase antibody (TPOAb) and/or thyroglobulin antibody (TgAb) during prenatal checkup. They were randomly divided into two groups: intervening group( n= 36, newborn group A) was treated with levothyroxine ( L-T4 ), and non-intervening group ( n= 19, newborn group B) was not treated. 30 cases of pregnant women with negative thyroid autoantibodies served as a normal population control group (newborn group N). Serum TSH, TPOAb, TgAb, TT3, TT4, FT3 and FT4 were measured by high-sensitive immunochemiluminescent assay ,and urinary iodine was also examined in the pregnant women. Fetal plasma TSH, TT3, TT4, FT3, and FT4 levels were measured after cutting the umbilical cord from placenta, and repeated measurements were made by 3-4 weeks and 8-10 weeks postpartum. Results At baseline, serum TSH levels of the pregnant women in intervening and nonintervening groups were significantly higher than that in control group ( P<0.05 ). Non-intervening group had higher TSH and lower TT3, TT4, FT4 compared with the other two groups (P<0. 05 or P<0.01 ). The cord blood TSH levels of the neonates in both group B [(7.06 ± 1.31 ) mIU/L] and group A [(6.23 ± 1.26 ) mIU/L] were significantly higher than that of group N [(5.48±1. 17) mIU/L, P<0.01 and 0. 05]. By 3-4 weeks postpartum, the serum TSH level [(3.21±0.70)mIU/L] in group B was significantly higher than those in group N [(2.72±0.51)mIU/L] and group A [(2.78±0.42) mIU/L, all P<0.05]. The serum TSH level in group B [(2.99±0.57) mIU/L] was still higher than those in group N [(2.48±0.68) mIU/L] by 8 to 10 weeks postpartum (P<0.05 ). Multiple stepwise regression analysis revealed that TSH, TPOAb, and urine iodine levels of mothers were independently related to TSH of their infants. Conclusion When differences in thyroid function exist in pregnant women, these differences also reside in their offspring. The thyroid function in neonates correlates with both the thyroid autoantibodies and thyroid function of their mothers.

Objective To investigate the growth inhibition effect of evodiamine (Evo) on renal carcinoma 786-0 cells and to explore its molecular mechanism. Methods After treated with Evo, methyl thiazolyl tetrazolium ( MTT) assay was used to detect the vitality of 786-0 cells, flow cytometry was employed to examine the cell cycle distribution in 786-0 cells, and immunoblotting was utilized to determine the expression levels of target proteins related to cell cycle progression. Results Evo remarkably inhibited 786-0 cells vitality in dose-dependent manner. Cell cycle analysis indicated that 786-0 cells were arrested in G2/M phase followed by Evo treatment. Furthermore, the results of immunoblotting showed that Evo up-regulated the protein expression levels of P53, P21 and its downstream target gene CyclinB1 in 786-0 cells. Conclusion Evo treatment can induce 786-0 cell cycle G2/M arrest, and its underlying mechanism might be dependent on the P53/P21 signal pathway.

Objective:To reconstruct the three-dimensional (3D) dose distribution in radiotherapy based on the convolutional neural networks (CNN) through multi-perspective scintillation light processing. Methods:First, fluorescence images were captured from three orthogonal perspectives using a complementary metal-oxide-semiconductor (CMOS) imaging sensor. Then, the images were converted into 3D images, which were input to the trained CNN for dose reconstruction. Finally, the reconstructed doses in different fields were evaluated in terms of gamma pass rate, mean-square error (MSE), percentage depth dose (PDD), and cross beam profile (CBP). Additionally, as the CNN model, 3D-Unet was pre-trained on a virtual dataset. Results:With the 50% maximum dose of as the threshold and 3%/3 mm as the standard, the central-plane and stereo-mean gamma pass rates of all field reconstruction distributions were over 90%, with MSEs remained below 1%. Besides, the PDD and CBP curves showed MSEs below 1‰ and below 1%, respectively. Conclusions:The deep learning-based method for 3D dose reconstruction using scintillation light contributes to enhanced verification of instantaneous 3D relative dose based on plastic scintillation detectors.

BACKGROUNDBased on the excellent medical care and management system for Chinese veterans, as well as the detailed medical documentation available, we aim to construct a Chinese Veteran Clinical Research (CVCR) platform on non-communicable diseases (NCDs) and carry out studies of the primary disabling NCDs.
METHODSThe Geriatric Neurology Department of Chinese People's Liberation Army General Hospital and veterans' hospitals serve as the leading and participating units in the platform construction. The fundamental constituents of the platform are veteran communities. Stratified typical cluster sampling is adopted to recruit veteran communities. A cross-sectional study of mental, neurological, and substance use (MNS) disorders are performed in two stages using screening scale such as the Mini-Mental State Examination and Montreal cognitive assessment, followed by systematic neuropsychological assessments to make clinical diagnoses, evaluated disease awareness and care situation.
RESULTSA total of 9 676 among 277 veteran communities from 18 cities are recruited into this platform, yielding a response rate of 83.86%. 8 812 subjects complete the MNS subproject screening and total response rate is 91.70%. The average participant age is (82.01±4.61) years, 69.47% of veterans are 80 years or older. Most participants are male (94.01%), 83.36% of subjects have at least a junior high school degree. The overall health status of veterans is good and stable. The most common NCD are cardiovascular disorders (86.44%), urinary and genital diseases (73.14%), eye and ear problems (66.25%), endocrine (56.56%) and neuro-psychiatric disturbances (50.78%).
CONCLUSIONWe first construct a veterans' comprehensive clinical research platform for the study of NCDs that is primarily composed of highly educated Chinese males of advanced age and utilize this platform to complete a cross-sectional national investigation of MNS disorders among veterans. The good and stable health condition of the veterans could facilitate the long-term follow-up studies of NCDs and provide prospective data to the prevention and management of NCDs.
