Two pairs of primers specific to small subunit ribosomal DNA of Plasmodium falciparum were designed and the expected SSUrDNA fragment was amplified for detecting P.falciparum infection with double-ternperature-nested polymerase chain reaction using DNA prepared by boiling method. The results showed that the nested PCR could amplify a constant size of desired SSUrDNA fragment of P. falciparum which was further confirmed by digestion of restrlction endonuclease and could detect parasitemia level of 0. 8 ? 10-6. It has great potentials for identifying Plasmodium species in ring form of erythrocytic stage and detecting mixed Plasmodium infections. Therefore, it is suggested that this method is sensitive, accurate, simple and rapid in detecting Plasmodium falciparum in blood samples for malaria diagnosis.

ObjectiveTo evaluate the therapeutic effect of Salvia miltiorrhiza and ligustrazine combined with vinpocetine in patients with cerebral infarction.MethodsAtotal of 206 patients with cerebral infarction were randomly divided into a controlgroup and a combined treatment group according to the random number table, 103 in each group. The control group was treated with aspirin and vinpocetine, the combined treatment group received Salvia miltiorrhiza and ligustrazine injection on the basis of the control group. The serum contents of malondialdehyde (MDA) and nitric oxide (NO), as well as the serum levels of superoxide dismutase (SOD) and nitric oxide synthase (NOS) were detected before and after the treatment. The electroencephalography (EEG) amplitudes and the regional cerebral blood flow (rCBF) were recorded before and after the treatment .ResultsIn the combined treatment group, the serum content of MDA was significantly decreased (151.36 ± 11.36 mmol/Lvs.62.23 ± 4.11 mmol/L;t=74.878,P<0.01), while the serum content of NO (110.31 ± 9.51 mmol/Lvs.154.23 ± 12.21 mmol/L;t=28.801,P<0.05) and the serum levels of SOD (55.52 ± 4.47 U/mlvs.85.39 ± 7.21 U/ml;t=35.735,P<0.05) and NOS (115.21 ± 8.39 mmol/Lvs.190.12 ± 11.29 mmol/L;t=54.049,P<0.01) were significantly increased than before the treatment. The rCBF (39.39 ± 2.45 ml/100 g?minvs. 69.95 ± 4.25 ml/100 g?min;t=32.385,P<0.05) and EEG (62.41% ± 4.58%vs.97.02% ± 9.67%;t=31.012,P<0.05) in the combined treatment group were significantly increased than before the treatment. The total efficiency rate in the combined treatment group was significant higher than that in the control group (88.3% vs. 66.9%;χ2=12.343, P=0.004).ConclusionsSalvia miltiorrhiza and ligustrazine combined with vinpocetine has a beneficial therapeutic effect in patients with cerebral infarction, and it is probably associated withincreasing cerebral blood flow and enhancing antioxidant activity.

Objective To improve the specific nursing standards of neostomy and chronic wounds, make patients receive professional guidance and standard treatment. Methods To establish professional team in the neostomy and conduct systematic and specialized training for team members, to work out nursing standards in the neostomy and execute nursing consultation and discussion in team on special types of chronic wounds. Results The professional knowledge of the team members and clinical nurses was enhanced, 18lessons about 50 class-hours were given, the attendance rote reached above 96% since the establishment of professional team in the neostomy, the quality of clinical nursing was improved and the complications were reduced. Conclusions The establishment of professional team provides clinical nurses with a platform of communication and learning, reflect their own value, and promote development of nursing science.

Objective:To explore the value of flexible endoscopic evaluation of swallowing (FEES) in guiding the decannulation of neurological disease patients with dysphagia after a tracheotomy. Methods:The FEES results of 188 neurological disease patients with dysphagia who had undergone a tracheotomy were analyzed retrospectively. The utility of FEES evaluation indexes (including glottis activity, the classification of pharyngeal secretions and residues as well as penetration-aspiration grade) for predicting the success of decannulation was explored. Results:One hundred and nine of the patients (the success group) were decannulated successfully and 79 (the failure group) were not, a success rate of 57%. The abnormal glottis activity rate among the failure group was 55%, significantly higher than among the success group (21%). The pharyngeal secretion classifications and penetration-aspiration grades among the success group were also significantly lower than among the failure group, on average. The average course of recovery from the tracheotomy was 184 days in the success group, significantly shorter than that of the failure group (292 days). No significant differences in residues were observed. Conclusion:The glottis activity, secretions and intake aspiration evaluated using FEES are of great value for guiding the decannulation of neurological disease patients with dysphagia after a tracheotomy.

Objective:To evaluate the effect of transcutaneous auricular vagus nerve stimulation (taVNS) on tourniquet-induced hypertension (TIH) in the patients undergoing anterior cruciate ligament reconstruction.Methods:Seventy-four patients of either sex, aged 18-60 yr, of American Society of Anesthesiologists Physical Status classification I or II, with body mass index of 18-30 kg/m 2, undergoing elective anterior cruciate ligament reconstruction under general anesthesia combined with preoperative femoral nerve block, were divided into 2 groups ( n=37 each) using a random number table method: sham stimulation group (group SS) and group taVNS. Group SS received stimulation on the ear lobe and the tail of the helix of the left ear. Group taVNS received stimulation on the cymba concha and the earlobe of the left ear. Both groups received stimulation from 1 h before induction of anesthesia until the end of the procedure (frequency of 30 Hz, pulse width of 300 μs, and amplitude of the strongest current that could be tolerated by the patient in the absence of pain). The tourniquet inflation pressure was 280 mmHg, with an inflation time of 60-90 min. Systolic blood pressure, diastolic blood pressure and heart rate were recorded before tourniquet inflation to assess the development of intraoperative TIH. The consumption of intraoperative propofol, remifentanil, nitroglycerin, esmolol, norepinephrine and atropine was recorded, and the occurrence of postoperative nausea and vomiting, skin itching and headache and dizziness was also recorded. Results:Compared with group SS, the incidence of TIH and the number of patients used nitroglycerin were significantly reduced ( P<0.05), and no significant changes were found in the other parameters in group taVNS ( P>0.05). Conclusions:taVNS can decrease the occurrence of TIH in the patients undergoing anterior cruciate ligament reconstruction.

DNA assembly is the core technology of synthetic biology. With the development of synthetic biology, researchers have developed different DNA assembly technologies that rely on DNA polymerase or DNA ligase, and also have developed some non-enzyme-dependent DNA assembly techniques to facilitate the automation of DNA assembly. The assembly of large fragments of DNA from a few hundred kb to Mb is mostly dependent on microbial recombination. In this paper, the three types of DNA assembly technologies, including enzyme-dependent, non-enzymatic and in vivo homologous recombination, are reviewed.
