1.Effect of salidroside on autophagy of pancreatic cells in rats with severe acute pancreatitis

Objective:To study the effects of salidroside (Sal) on autophagy of pancreatic cells in rats with severe acute pancreatitis (SAP).Methods:Fifty healthy male Wistar rats were randomly (random number) divided into the sham group, SAP group, Sal 15 group, Sal 30 group and Sal 60 group, 10 rats in each group. The sham group was injected retrogradely normal saline into pancreatic duct, while the 5% sodium taurocholate was injected retrogradely into pancreatic duct to establish severe pancreatitis model in the SAP group and all the Sal groups. Salidroside at different doses were intraperitoneally injected in all the Sal groups at 2 h after surgery. The serum amylase (AMY) activity and plasma tumour necrosis factor-α (TNF-α) level were detected by enzyme-linked immunosorbent assay (ELISA), and Western blot was used to detect the expression of pancreatic tissue protein. The histopathological changes of pancreas were observed under the light microscope. The data were analyzed by SPSS 24.0, and One-way ANOVA was used for comparison between groups. A P value less than 0.05 was considered to be statistically significant. Results:After salidroside intervention, the levels of TNF-α (28.1±0.9) pg/mL, the relative expression of p65 (0.52±0.01), AMY activity (3 801±292) and relative expression of p-mTOR were the lowest in the Sal 60 group (all P<0.05); while the relative expression of LC3 was the highest in the Sal 60 group (all P <0.05). The degree of pathological damage of pancreatic tissues was decreased with the increase of salidroside dose. Conclusions:Salidroside at different doses have protective effects on rats with severe pancreatitis, which seems to be most obvious in the Sal 60 group.
2.Curative effect analysis on different arrangement of hollow screws for femoral neck fracture

Abstact:Objective To explore the effect of different arrangement of hollow screws in treatment of femoral neck fracture.Methods The clinical data of 160 cases with femoral neck fracture from March 2010 to May 2014 in our hospital were collected.They were randomly di-vided into triangle group and inverted triangle group according to arrangement of hollow screws.The operation time,intraoperative blood loss, fluoroscopy frequency in operation,length of stay,time of fracture healing,postoperative complications,Harris score and functional recovery of two groups were recorded and compared.Results The operation time and intraoperative blood loss in inverted triangle group were lower than those in triangle group,the difference was significant(P <0.05).The shortening rate of inverted triangle group and triangle group were 10%and 26.3% respectively.The shortening rate of inverted triangle group was lower than those of triangle group,the difference was significant (P <0.05).For the fluoroscopy frequency in operation,average hospitalization time,fracture healing time,rate of nonunion and femoral head necrosis,Harris score and functional recovery,there was no statistical significance between two groups(P >0.05).Conclusion Inverted tri-angle hollow screws treating femoral neck fracture makes surgery operation easier,operative time shorter and blood loss smaller.

4.Cytotoxic effects of gastric cancer cell-DC fusion vaccines

Objective To study on the cytotoxic effects in vivo and in vitro of the T lymphocytes which were activated by the DC-gastric cancer cell fusion vaccines. Methods The mononuclear cells were separated from the peripheral blood of gastric cancer patients and co-cultured with granulocyte-macrophage colony stimulating factors, interleukin-4 and tumor necrosis factor-? to generate mature dendritic cells. The human gastric cancer SGC7901 cells and the dendritic cells were fusioned by using polyethylene glycol, and the pure fusion cells were screened out by culturing with HAT and HT selective culture systems. The mixture of SGC7901 and DC without fusion and the T lymphocytes served as controls. The ability of fusion cells activated by T lymphocytes to kill SGC7901 gastric cancer cells was investigated by MTT in vitro. The anti-tumor effects were evalutated by detecting the growth of the new planted tumors in the nude mice, the apoptosis and proliferation of the planted cancer cells, the pathological changes of the tumor tissues and the prohibitive rate for planted tumors before and after the application of the fusion cell vaccine. Results The mature dendritic cells were obtained from peripheral blood mononuclear cells of gastric cancer patients successfully. Dendritic cells and SGC7901 cells were fusioned and the pure fusion cells were got. Objective To obtain SGC7901 gastric cancer cell-dendritic cell fusion vaccines by cell fusion technology. The phenotype changes of the fusion cells were detected to provide experimental data for the following research work. Methods The SGC7901 gastric cancer cells and dendritic cells were induced to be fused by PEG and the pure fusion cells were obtained by selective culture with the HAT/HT culture system. The phenotype changes of the fusion cells were detected by flow cytometer. Results The fusion cells had special morphologic characteristic and the phenotypes of the fusion cells were remarkably higher than the parental dendritic cells. Conclusion The SGC7901 gastric cancer cell-dendritic cell fusion cells kept special morphologic characteristic and their phenotypes were significantly higher than the parental dendritic cells, which may be the cellular base for their stronger biological effects.
6.Study on the induction culture and phenotypes of dendritic cells originating from peripheral blood of gastric cancer patients

Objective To analyse the morphological and phenotypic characteristics of dendritic cells obtained by induction of peripheral blood monocytes of gastric cancer patients and provide an experimental basis of (dendritic) cells for the biological treatment of gastric cancer. Methods Peripheral blood monocytes of gastric cancer patients were co-cultured with GM-CSF, IL-4 and TNF-? to obtain the dendritic cells. The (morphological) characteristics of the dendritic cells were observed by light and electron microscopes and the (phenotypes) were studied by flow cytometry. Results Mature dendritic cells were oblained seven days after (induction) culture. These cells showed numerous typical dendritic protuberances on their surface.The (phenotypes) of the cells increased gradually with their maturation and reached stable levels seven days later. Conclusions Mature dendritic cells could be obtained from peripheral blood monocytes of gastric cancer (patients) after induction culture with GM-CSF,IL-4 and TNF-?. The obtained cells had typical morphological and phenotypic characteristics. These cells could be used for further research in the biological treatment of (gastric) cancer.
7.A study on the biological characters of SGC7901 gastric cancer cell-dendritic cell fusion vaccine

Objective To provide experimental basis for the study of SGC7901 gastric cancer cell-dendritic cell fusion vaccine. Methods The fusion of SGC7901 gastric cancer cells and dendritic cells was induced by PEG. We obtained pure fusion cells by selective culture with HAT/HT culture systems. The growth curve characters, clone abilities, abilities to irritate T lymphocytes and the ability to grow were detected. Results The fusion cells had weaker proliferative abilities and clone abilities with increased abilities to irritate T lymphocytes. The CPM value of T lymphocytes were 15083?231、 9608?83、 4214?135、 3020?28 respectively when they were co-cultured with the fusion cells on different proportions. Conclusion SGC7901 gastric cancer cell-dendritic cell fusion cells keep strong abilities to irritate T lymphocytes while inable to grow into new planted tumors in immunodeficiency animals.

9.Detection of STAT3 mRNA and CEA mRNA in peritoneal washings for predicting peritoneal micrometastasis in patients with gastric carcinoma

Objective To compare detection of STAT3 and CEA by nest RT-PCR and peritoneal lavage cytology examination(PLC) in predicting peritoneal micrometastasis in gastric carcinoma patients. Methods Peritoneal washings of 66 patients with gastric carcinoma and 12 patients with benign gastric diseases were collected to detect the mRNA level of STAT3 and CEA by nest RT-PCR.PLC were carried out by H-E staining.Results The positive rates of STAT3 mRNA and CEA mRNA in peritoneal washings were 43.9% and 48.5%,respectively.The positive rates of STAT3 mRNA and CEA mRNA were significantly higher than those of PLC(31.8%).The positive rates of STAT3 mRNA and CEA mRNA were concurrently increased with the depth of tumor invasion,TNM staging and pathogenesis.Conclusion Detecting STAT3 mRNA in peritoneal washings by nest RT-PCR can improve the sensitivity of free tumor cell detection in peritoneal washings,and may be used to predict peritoneal micrometastasis of gastric carcinoma.
10.Detection of Authenticity of Grape Seed Oil with Ion Mobility Spectrometry

A rapid method for authenticity detection of grapeseed oil was proposed based on Ion Mobility Spectrometry ( IMS) . After optimization, the inlet temperature was set at 170 ℃, drift tube temperature was set at 60 ℃. In this method, the oil sample was diluted in hexane (50-fold, V/V) and then directly analyzed in IMS. The detection time was 20 s. To establish an adulteration detection model, recursive Support Vector Machine ( R-SVM) was applied to classifying pure and adulterated grape seed oils. The result of 10-fold cross validation showed that the accuracy of discrimination was up to 91 . 2%. The results in study indicate that IMS method is a new, fast and convenient technique for the adulteration detection of edible oil.
