1.Effect of salidroside on autophagy of pancreatic cells in rats with severe acute pancreatitis

Objective:To study the effects of salidroside (Sal) on autophagy of pancreatic cells in rats with severe acute pancreatitis (SAP).Methods:Fifty healthy male Wistar rats were randomly (random number) divided into the sham group, SAP group, Sal 15 group, Sal 30 group and Sal 60 group, 10 rats in each group. The sham group was injected retrogradely normal saline into pancreatic duct, while the 5% sodium taurocholate was injected retrogradely into pancreatic duct to establish severe pancreatitis model in the SAP group and all the Sal groups. Salidroside at different doses were intraperitoneally injected in all the Sal groups at 2 h after surgery. The serum amylase (AMY) activity and plasma tumour necrosis factor-α (TNF-α) level were detected by enzyme-linked immunosorbent assay (ELISA), and Western blot was used to detect the expression of pancreatic tissue protein. The histopathological changes of pancreas were observed under the light microscope. The data were analyzed by SPSS 24.0, and One-way ANOVA was used for comparison between groups. A P value less than 0.05 was considered to be statistically significant. Results:After salidroside intervention, the levels of TNF-α (28.1±0.9) pg/mL, the relative expression of p65 (0.52±0.01), AMY activity (3 801±292) and relative expression of p-mTOR were the lowest in the Sal 60 group (all P<0.05); while the relative expression of LC3 was the highest in the Sal 60 group (all P <0.05). The degree of pathological damage of pancreatic tissues was decreased with the increase of salidroside dose. Conclusions:Salidroside at different doses have protective effects on rats with severe pancreatitis, which seems to be most obvious in the Sal 60 group.

3.Clinical survey of the effect of acupuncture,moxibustion combined with bladder function training on the neu-rogenic bladder

Objective To observe the effect of acupuncture, moxibustion combined with bladder function training on the neurogenic bladder.Methods 64 patients were randomly divided into the treatment group and control group,treatment group was given acupuncture,moxibustion combined with bladder function training,while the control group was treated with simple bladder function training, then comparing the bladder function changes between two groups after a month.Results In the treatment group,the bladder capacity,residual urine volume,urinate rate were respectively ( 349.4 ±13.5 ) mL, ( 98.7 ±15.3 ) mL, 92.26% and the control group respectively ( 323.4 ± 17.8)mL,(127.8 ±17.8) mL,51.85%,the difference between the two groups were statistically significant ( t =5.48,5.60,χ2 =3.97,all P<0.05).Conclusion Acupuncture,moxibustion combined with bladder function train-ing can be effective in the treatment of neurogenic bladder,while improve the quality of patient's life.
4.Short-term efficacy of da Vinci robotic and laparoscopic radical gastrectomy for gastric cancer: a Meta analysis

Objective To compare the short-term efficacy between da Vinci robotic and laparoscopic radical gastrectomy for gastric cancer.Methods Database including PubMed,EMBASE,the Cochrane Library,Medline,the China National Knowledge Infrastructure (CNKI),Wanfang Database,VIP and China Biomedicine were searched with the key words of gastric cancer,gastrectomy,da Vinci surgical system,laparoscopic,laparoscopy,胃癌,胃切除术,达芬奇机器人手术系统and腹腔镜.Literatures published between May 2002 and March 2014 were retrieved.Related controlled trials containing the clinical efficacy of da Vinci robotic and laparoscopic radical gastrectomy for gastric cancer were enrolled in the study,and then the literatures were screened and the data were extracted by 2 independent reviewers.The quality of the literatures was assessed,and the data were analyzed using the RevMan 5.2 software.Patients receiving da Vinci robotic surgery and laparoscopic surgery were allocated into the robotic group and laparoscopy group,respectively.The count data were analyzed using the relative risk (RR) and 95% confidence interval (95% CI),and the measurement data were analyzed using weighted mean difference (WMD) and 95% CI.The heterogeneity of the data was analyzed using the I2 test.Results Eleven literatures including 3 698 cases were enrolled in the study.There were 953 patients in the robotic group and 2 745 patients in the laparoscopic group.There were significant differences in the operation time,volume of intraoperative blood loss,time of postoperative flatus,time of first oral intake and duration of postoperative hospital stay between the 2 groups (WMD =51.23,-36.96,-0.24,-0.26,-1.11,95% CI:28.01-74.46,-61.28--12.65,-0.42--0.06,-0.39--0.14,-1.92--0.29,P < 0.05).There was no significant difference in the number of lymph nodes resected,the distance to proximal and distal resection margin and overall incidence of postoperative complications (WMD =1.63,0.21,0.06,RR =1.11,95% CI:-0.49-3.76,-0.65-1.06,-0.13-0.25,0.90-1.37,P > 0.05).Conclusion Da Vinci robotic radical gastrectomy for gastric cancer is safe and feasible,with the advantages of minimal invasion and rapid postoperative recovery.
5.Clinical efficacy of single-port Da Vinci robotic surgical system in the radical resection of rectal cancer

Objective To explore the clinical efficacy of single-port Da Vinci robotic surgical system in the radical resection of rectal cancer.Methods The retrospective descriptive study was conducted.The clinical data of one patient with rectal cancer who was admitted to the Southwest Hospital of the Third Military Medical University in June 2017 was collected.The patient underwent the first case of radical resection of rectal cancer with the single-port la Vinci robotic surgical system,and intraoperatively received laparoscopy-assisted digestive tract reconstruction.Observation indicators:(1) intra-and post-operative situations;(2) follow-up and survival.Follow-up using outpatient examination and telephone interview was performed to detect patients' survival up to July 2017.Results (1) Intra-and post-operative situations:the patient underwent successful single-port Da Vinci robotic radical resection of rectal cancer.The operation time and volume of intraoperative blood loss were 145minutes and 20 mL,without blood transfusion.The using time of nonsteroidal analgesic drug,time for out-of-bed activity,time to anal exsufflation,time of fluid diet intake and time of pelvic drainage tube removal were 1 day,24hours,48 hours,72 hours and 4 days after operation,respectively.No postoperative complication was detected.Results of postoperative pathological examination:moderate-differentiated rectal adenocarcinoma invaded serosal layer,with negative circumferential margin,negative distal margin of rectum and mesenteric lymph node metastasis (1 of 16 harvested lymph nodes was positive).Pathologic staging was T4aN1M0 (stage Ⅲb).The patient was discharged from hospital at postoperative day 6.(2) Follow-up and survival:the patient was followed up for 1month,with a good survival.Conclusion Single-port Da Vinci robotic surgical system in the radical resection of rectal cancer is safe and feasible,with good short-term outcomes.
6.Meta-analysis of Acupuncture as Adjuvant Therapy in Improving Gastrointestinal Function of Acute Pancreatitis Patients

Objective To evaluate the clinical effect of acupuncture as an adjuvant therapy for acute pancreatitis (AP).Methods We retrieved the databases ofCNKI,VIP,Wanfang,PubMed,EMBase,and Cochrane Library,selected the relevant randomized clinical trials (RCTs) of AP treated with routine western medicine as control group and treated with routine western medicine combined with acupuncture therapy as trial group.And then,the quality of the qualified RCTs was evaluated,and the end-point outcomes of the RCTs were analyzed by Meta-analysis.Results A total of 7 RCTs involving 371 AP patients were included into the analysis.The time for abdominal pain relief(WMD =-1.44;95%CI:-2.38,-0.50),time for abdominal distension relief (WMD =-2.50;5%CI:-4.07,-0.73),time for the restart of anal exhaust(WMD =-1.79;95%CI:-3.73,-0.14),time for defecation (WMD =-1.95;95%CI:-3.51,-0.39),time for the restart of bowel sound(WMD =-1.39;95%CI:-2.44,-0.34),time for blood amylase becoming normal(WMD =-2.09;95%CI:-3.22,-0.96),and hospitalization time (WMD =-3.70;95%CI:-6.04,-1.36) in the trial group were less than those in the control group,and the differences of the above indexes except for the time for the restart of anal exhaust were significant (P < 0.05).Conclusion Acupuncture as an adjuvant therapy is effective for improving the gastrointestinal function of AP patients,and shortening time for blood amylase becoming normal and hospitalization time.However,due to the low methodological quality,more well-designed randomized controlled trials are needed to confirm the therapeutic effect of acupuncture therapy for AP.
7.Effect of interleukin-17 in the migration and invasion of gastric cancer cell via regulating epithelial-mesenchymal transition

Objective To investigate the effect of interleukin-17 (IL-17) in the gastric cancer cell migration and invasion via regulating epithelial-mesenchymal transition (EMT) and its potential function.Methods (1) Human gastric cancer cell MGC-803 lines in the logarithmic growth phase were stimulated by 0, 1 ng/mL, 10 ng/mL,100 ng/mL and 1μg/mL of IL-17 for 48 hours, and the phenotypic changes were observed.The concentration of IL-17 was selected for follow-on experiments based on the most obvious phenotypic changes.Gastric cancer cell MGC-803 which were stimulated by 100 ng/mL of IL-17 and PBS for 48 hours were allocated into the experimental group and control group, respectively.(2) The expressions of E-cadherin and Vimentin mRNA in gastric cancer cells were assayed through real-time PCR (RT-PCR).(3) The relative expressions of E-cadherin and Vimentin proteins in gastric cancer cells were assayed by the Western blot.(4) The scratch test and Transwell detection were also utilized to study the migration and invasion of gastric cancer cell MGC-803 in vitro.Measurement data with normal distribution were presented as-x ± s and comparison between groups was analyzed using the t test.Results (1) There were significant phenotypic changes in the gastric cancer cell after the different concentration of IL-17 stimulated gastric cancer cell MGC-803 for 48 hours.Cells were changed from polygonal and tight junction to spindle and loosely junction with a deterioration of cell adhesion.Cell phenotypes were gradually changed as the concentration of IL-17 was changed from 0 to 100 ng/mL.Phenotypic changes were the most obvious when 100 ng/mL of IL-17 was used, but these were non-significant as the concentration of IL-17 increased to 1 μg/mL with the death and floating of some cells.(2) The relative expressions of E-cadherin mRNA and Vimentin mRNA in RT-PCR were 0.45 ±0.13 and 1.06 ±0.23 in the experimental group and 2.39 ±0.55 and 1.23±0.41 in the control group, respectively, with significant differences (t =3.811, 2.923, P <0.05).(3) The results of Western blot showed the relative expressions of E-cadherin and Vimentin proteins were 0.86 ± 0.17 and 1.56 ± 0.29 in the experimental group and 1.01 ± 0.12 and 0.56 ± 0.17 in the control group, respectively, with significant differences (t =3.551, 3.601, P < 0.05).(4) Cell migration in the 2 groups were detected by the scratch test at 36 hours after scratch test, and the width of scratch in the experimental and control groups were (0.76 ± 0.13) mm and (0.40 ± 0.15) mm, showing a significant difference (t =3.095, P < 0.05).Transwell detection showed number of transmembrane cell in the experimental and control groups were 159 ±28 and 94 ± 18, respectively, with a significant difference (t =3.307, P < 0.05).Conclusion IL-17 can promote the migration and invasion of gastric cancer cells via stimulating alteration of EMT.
8.Clinical efficacy of single-port Da Vinci robotic surgical system in the radical gastrectomy of gastric cancer

Objective To explore the clinical efficacy of single-port Da Vinci robotic surgical system in the radical gastrectomy of gastric cancer.Methods The retrospective descriptive study was conducted.The clinical data of the first patient in China who underwent single-port radical gastrectomy of gastric cancer using Da Vinci robotic surgical system in the Southwest Hospital of the Third Military Medical University in June 2017 were collected.Patient underwent radical gastrectomy of gastric cancer using single-port Da Vinci robotic surgical system +D2 lymph node dissection + Billroth Ⅱ anastomosis.Observation indicators:(1) intra-and post-operative situations;(2) follow-up and patients' survival.Follow-up using outpatient examination and telephone interview was performed to detect the patients' postoperative survival up to July 2017.Results (1) Intra-and postoperative situations:patient underwent radical distal subtotal gastrectomy of gastric cancer using single-port Da Vinci robotic surgical system.Operation time and volume of intraoperative blood loss were respectively 303 minutes and 100 mL.There was no intraoperative complication.The distances from tumor to proximal margin and distal margin were 5 cm and 6 cm.Number of lymph node dissected and length of abdominal incision were 51 and 3 cm.Time of gastric tube removal,time for out-of-bed activity,time of gastrointestinal function recovery,time of drainage tube removal and postoperative pain score were 17 hours,24 hours,36 hours,36 hours and 3,respectively.Patient took a little fluid diet after gastric tube removal.There was no occurrence of postoperative complication.Results of pathological examination showed that tumor invaded deep muscular layer,with 2 positive lymph nodes in No.3 and negative proximal and distal margins.Pathological staging was pT2N1M0 (Ⅱa staging).Duration of hospital stay was 6 days.(2) Follow-up and patients' survival:patient was followed up for 1 month,with a good survival.Conclusion The single-port Da Vinci robotic surgical system is safe and feasible in the radical gastrectomy of gastric cancer,with good short-term outcomes.

Silica dioxide bound graphene oxide ( GO-SiO2 ) was applied as an effective adsorbent for determination and quantiflcation of aflatoxin B1 , B2 in edible oil by HPLC. The optimized conditions were GO-SiO2 0. 15 g, extraction time 20 min, elution reagent acetonitrile, elution cycles two times. Results showed under the optimum conditions, the detection limits of aflatoxin B1 , B2 were 0. 17 and 0. 05 μg/L, respectively. The method was successfully applied to the detection of the actual edible oil, the spiked recoveries of aflatoxin B1 and B2 were 81. 4%-105. 3% and the relative standard deviations were 1. 3%-8. 6%.

AIM: To observe the relationship between the immune deviation of Th1 and Th2 cell clones and class Ⅱ major histocompatibility complex (MHC) antigen expression in different stages of acute rejection in transplanted hearts. METHODS: Heart transplantation were performed in rats.Isografts and non-transplanted animals were used as control group. Donor class II MHC antigen expression were detected with monoclonal antibodies and immunostaining technique and the amount of type Ⅰ and Ⅱ cytokines mRNA expression were detected by semiquantitative RT-PCR in cardiac allografts. RESULTS: Myocardial IL-2 mRNA and donor class Ⅱ MHC antigen expression were significantly in creased, accompanied with development of acute rejection( P
