1.Investigation of Distributing Characteristics of Blood Stasis Syndrome on 300 Cases of Advanced Stage Lung Cancer Patiens

Objective To investigate the distributing characteristics of blood stasis syndrome on 300 cases of advanced stage lung cancer patiens. Method Through filling in the table-syndrome of distributing characteristics of primary pulmonary carcinoma for stage Ⅲ and Ⅳ patients,the distributing characteristics of blood stasis syndrome in advanced lung cancer patiens were observed. Result The constituent ratio of blood stasis syndrome increased significantly following the worse patho-typing degree,the cell differentiation degree,the occurrence of hepatic metastasis,opposite pulmonary metastasis,osseous metastasis and troisier sign. Conclusion The syndrome of blood stasis of lung cancer has the distinct feature. Accordingly,the diagnostic standard which can be qualified of syndrome of blood stasis should be established. Only in that way can we guide the clinical diagnosis,the evaluation of therapeutic effect and the clinical trial of new drug of TCM preferably.
2.Dual-dissociation method to record whole-cell current of wistar rat neurons

BACKGROUND: The main difficulties of whole-cell patch clamP are cell culture and whole-call current recording. OBJECTIVE: To reduce the difficulty of patch-clamp experiment by combining acute cell isolation with current separation technique. METHODS: A total of 40 wistar rats aged 4-7 days, irrespective of genders, were selected. The pallium of wistar rats were cut into slices with 400-600 μm, rested for 1 hour in artificial cerebrospinal fluid with gas mixture, simultaneously put in oxygen. The brain tissues were placed into artificial cerebrospinal fluid containing 16 u/mL(type X) and 2 u/mL(type XIV) proteinase, incubated for 60 minutes, and cleared the digestive enzyme. Under the whole-cell voltage clamp mode, the potential was hold at -80mV, depolarizing pulse stimulation from -60 mV to 60 mV, 10 mV step and 160 ms width. The total Tran membranes current was recorded, and then 70 mmol/LCsCI, 70mmol/L CsF was 11 μmol/L Na~+ channel blockers tetrodotoxin, 30 mmol/L tetraethylammonium, 1 mmol/L 4-AP was successively added into extracellular fluid. The inward sodium current, transient outward potassium current and delayed rectifier potassium current was recorded, and then the result was analyzed using clampfit. MAIN OUTCOME MEASURES:①Cell morphological observation; ②Whole-ceil current recording;③Inward sodium currentrecording; ④Outward potassium current recording.RESULTS AND CONCLUSION: The cell had clearly three-dimensional structure, smooth surface and whole neurodendrite or neuraxon. The cell viability could maintained for 8-10 hours at 25 ℃ temperature. The added 11 tetrodotoxin in extracellular fluid could block sodium Current. The outward potassium current could be blocked by tetraethylammonium with 30 mmol/L and 4-AP with 1 mmol/L. The calls harvested by modified rapid dissociation have good functions. Using current separation method, only specific blockers are needed, without extracellular fluid or electrode solution replacing, which can record the inward sodium current, transient outward potassium current, as well as delayed rectifier potassium current. This method can obviously improve the work efficiency than traditional one.
3.Study on contemporary TCM academic researching team of lung cance based on the literature metrology and social network analysis

Objective To analyze the contemporary TCM academic team of lung cancer, summarizes its main academic thoughts. Methods To represent characters, important documents, and the main academic ideas as the basis of a team. From the perspective of the literature, literature of Chinese medicine against lung cancer, adopt the method of literature metrology, and social network analysis the candidate teams were found;the method of literature investigation evaluation candidate team heritage was adopted, the main academic thoughts from each team representative was summarized. Results A total of 8 441 researchers published 5 030 papers about treatment of lung cancer with TCM. 91 researchers with large amount papers and seven academic team were found. Conclusion Chinese medicine treatment of lung cancer made a great progress in the contemporary, team enriched the TCM system of principle, method, square, medicine.
4.Research progress of application of E-cadherin, N-cadherin and vimentin in tumor metastasis

The invasion and metastasis of tumor as the primary cause of death in cancer increases not only the patient's suffering, but also the difficulty of clinical treatment. It is an important guiding to find the molecular biological tumor markers and gene targets of antirecurrence and antimetastasis for clinical treatment. The invasion and metastasis of tumor is a complex process including the biological characteristics of the cancer, interaction among host cells, extracellular matrix and cancer cells, etc. Recently, epithelial mesenchymal transition (EMT) is becoming a hot spot in oncology research by virtue of its effect on genesis and development of tumor. Currently, the known markers of EMT include epithelial marker proteins mainly containing E-cadherin and interstitial marker proteins mainly containing N-cadherin and vimentin. It is necessary to make a summary among the studies on three proteins.
5.Effects of edaravone on JAK2/STAT3 signaling pathway in transplanted donor liver of rats with different cold ischemia times

Objective To investigate the effect of edaravone on the JAK2/STAT3 signaling pathway after ischemia-reperfusion injury in donor rat liver under different cold ischemia times. Methods A total of 102 SD rats were randomly divided into sham operation group,control group and experimental group. Six rats were in sham operation group with free liver operation and no transplantation. Forty-eight rats were in control group and experimental group respectively, and divided into subgroups according to the different cold ischemia times (30 min, 6 h, 12 h and 18 h). There were 6 donors and 6 recipients in each group. The rat model of orthotopic liver transplantation was established by modifiedtwo cuff method. All the donors were perfused by abdominal aorta and the warm ischemia time was 3-5 min. After different cold ischemia times, the experimental group was treated with edaravone (3 mg/kg) at 5 min before the opening of the new hepatic artery, and control group was injected with 3 mg/kg saline. Recipients of each group were sacrificed after 6 h. Finally, real-time fluorescence quantitative PCR was used to analyze the relative expression of JAK2/STAT3 mRNA of donor liver. Results The GAPDH gene and JAK2/STAT3 were well amplified. Under the same cold ischemia time, compared with the control group, the relative expression of JAK2/STAT3 was significantly decreased in the experimental group (P<0.05). With the prolongation of cold ischemia time, the relative expressions of JAK2 and STAT3 mRNA showed a decreasing trend in control group and experimental group, while the relative expression of JAK2 mRNA increased first and then decreased in the experimental group (P<0.05). Conclusion Edaravone has a protective effect on transplanted donor liver during different cold ischemia times, and extends the cold ischemia time for 18 h, which may be related to the inhibition of JAK2/STAT3 signal transduction pathway.
6.Clinical Observation of Intranasal Dexmedetomidine and Sufentanil for Pediatric Sedation Undergoing Stomatology Operation of Outpatient Department

[Objective] To research the sedative effect of intranasal dexmedetomidine and sufentanil for pediatric sedation for stomatological operation of outpatient department.[Methods] 60 children undergoing stomatological operation of outpatient department,age 3 ~ 7 years,weighing 10 ~ 32 kg,of ASA physical status Ⅰ ~ Ⅱ,were divided into the three groups (n =20) randomly using a random number table:group dexmedetomidine and sufentanil (group DS),group dexmetomidine (group D),group sufentanil (group S).Recorded the children's behavior using the Ohio State University behavior rating score (OSUBRS),the University of Michigan Sedation Score (UMSS),SBP,HR and side-effects when entry,during and leave operation and in post-anesthesia care unit,side-effects,the satisfaction of stomatological doctors and parents.[Results] The OSUBRS of group DS when entry,during operation were lower than group D (P < 0.01).The UMSS of group DS were higher than group D and group S when entry and during operation (P < 0.05).The success rate of group DS was higher than group D and group S (P < 0.01).There was no different of mean percentage change in systolic blood pressure and heart rate from baseline between group DS and group D (P > 0.05).There were no instances of respiratory depression,hypotension and bradycadia.[Conclusion] Intranasal dexmedetomidine and sufentanil provides satisfactory pediatric sedation for stomatological operation of outpatient department without side effects such as respiratory depression,bradycadia and hypotension.
7.A prospective cohort study on hormone-dependant metastatic breast cancer treated with Shuganyishen formula

Objective Observing the effect of Shuganyishen formula on hormone-dependant metastatic breast cancer and the mechanisms of anti-resistance and endocrine therapy. Methods 226 ITT cases were divided into two cohorts by treatment. 126 cases were treated with the combination of Shuganyishen formula and standard western medicine including endocrine therapy, and 100 cases were treated with western medicine alone. The end points were time to progress (TTP) and overall survival and quality of life (QOF). The effect of Shuganyishen formula on growth of TAM-resistance cell line in vitro was observed. Results There were statistical significances of median TTP between combination and western medicine in Luminal A (ER+ /PR+, HER2-) as well as Luminal B (ER+/PR+, HER2+), but no significances of OS; The extent and incidence of hot flush, sweat and syndromes related to bone were decreased significantly in combination treated group. Shuganyishen formula could reverse resistance to TAM. Conclusion Patients with LuminalA and LuminalB metastatic breast cancer benefit from Shuganyishen formula, which maybe related with its reverse resistance to endocrine therapy.
8.Effects of Dredging Collaterals and Activating Blood Worm Chinese Materia Medica on Angiogenesis Related Factors of Lung Cancer in Hypoxic Environment

Objective To observe the effects of dredging collaterals and activating blood worm Chinese materia medica on angiogenesis related factors of lung cancer in hypoxic environment. Methods The lung cancer A549 cells were cultured in vitro to simulate tumor hypoxia microenvironment by the hypoxia workstation, and different concentrations of Scorpio, Scolopendra and Gecko medicated serum were added. MTT method was used to detect cell proliferation and screen the best medicine concentration and duration of action. Lung cancer A549 cells were administrated by the three kinds of medicated serum, and cells were collected and supernatant was cultured. Contents of VEGF, TGF-β1, and bFGF were detected by ELISA. Results Three kinds of medicated serum had the inhibitory effect on both added normoxia and hypoxia in cultured A549 lung cancer cells. 7.5% concentration of medicated serum was selected, and 24 h later were used in later experiments. Scorpio, Scolopendra and Gecko medicated serum can more reduce the contents of VEGF, TGF-β1 and bFGF in the supernatant of A549 cell compared with the control group (P<0.05, P<0.01). Conclusion Dredging collaterals and activating blood worm Chinese materia medica had inhibitory effect on cancer cells and the regulation of angiogenesis related cytokines in the condition of normoxia and hypoxia.
9.A discussion on the tactics for cultivating medical students' autonomous learning ability

This article analyzes the limitations of traditional medical theory teaching, and proposes the strategies for cultivating medical students' autonomous learning ability, i.e., informatization-based flipped classroom, problem-oriented teaching, mind mapping training, semi-open book examination, exploitation of the clinical and scientific thinking, and practice activities of medical humanities. The strategies of "problem oriented teaching" and "mind mapping training" were integrated into the practice teaching of hematology. Compared with the traditional medical teaching mode, students' feedback after class showed that the teaching mode incorporating new cultivation strategies was more conducive to the improvement of students' self-learning ability ( P = 0.008), and their satisfaction with teaching mode, learning interest, and self-learning ability were all improved. Thus, the appropriate application of the above strategies can help improve students' autonomous learning ability and optimize the effect of medical theory teaching.
10.Effects of Xianglian Huazhuo Decoction on Wnt1, GSK-3β and β-catenin in chronic atrophic gastritis rats

Objective:To investigate the mechanism of Xianglian Huazhuo Decoction in treating chronic atrophic gastritis (CAG) rats by detecting the protein and gene expressions of Wnt1, GSK-3β, β-catenin in Wnt signaling pathway.Methods:Totally 60 Wistar rats were randomly divided into blank group and model group. The CAG model was established by MNNG free drink combined with sodium salicylate gavage combined with abnormal hunger and satiety. After model establishment, the rats were randomly divided into model group, Xianglian Huazhuo Group and folic acid group. Folic acid group received the folic acid water solution 1.3 mg/kg for gavage; the Xianglian Huazhuo Group received Xianglian Huazhuo Decoction 15.5 g/kg for gavage; the blank group and the model group received the same volume of purified water for gavage. After 8 weeks of intervention, the general state and the atrophic degree of gastric mucosa were observed. The expressions of Wnt1, GSK-3β and β-catenin were detected by immunohistochemical method, and the mRNA expression of β-catenin expression was detected by real-time quantitative polymerase chain reaction PCR.Results:The general condition of rats in Xianglian Huazhuo group was improved, the atrophy of gastric mucosa was improved, and the effect was better than that in folic acid group. Compared with the model group, the positive area ratio of Wnt1 decreased in Xianglian Huazhuo group ( P<0.05), and the positive area ratio of β-catenin decreased in folic acid group and Xianglian Huazhuo group ( P<0.05 or P<0.01), the positive area ratio of GSK-3β increased ( P<0.05), and the β-catenin mRNA level of Xianglian Huazhuo group decreased ( P<0.05). Conciusion Xianglian Huazhuo Decoction may increase GSK-3β by downregulating abnormal expression of Wnt1, promoting β-catenin to degrade normally, thereby blocking the abnormal activation of the Wnt signaling pathway, delaying or reversing CAG carcinogenesis to a certain extent, and exerting therapeutic effects.
