1.An Analytical Method for Oxygenated Nonhydrocarbonsin Sediments and Crudes

An analytical method for seperating nonhydrocarbons in sediments and crudes into an acidic fraction, a neutral fraction (Ⅰ), and a more polar neutral neutral fraction (Ⅱ) is reported. With this method, components only appear in individual fractions. Fatty acids and polyols and polyones are seperated into the acidic fraction; mono-ketones, esters and straight-chain alkanols fall into the neutral fraction Ⅰ; Streols are found in the neutral fraction Ⅱ. No evident alteration of ratios of the components through the separation procedures is observed. Erucic acid, 5α(H)-androstan-17-one, and 5α(H)-androstan-3β-ol are selected first time as internal standards for sterols in acidic fraction, neutral fraction Ⅰ and Ⅱ, respectively. They are good standards for quantitation of the components with gas chromatography, especially 5α(H)-androstan-3β-ol with in the first time an interal standard for quantitation of sterols. The analytical method has been proved to be effective by seperation of more than hundred sedimental samples
2.Clinicopathological analysis of solid-pseudopapillary tumor of the pancreas

Purpose:To study clinical and pathological features of solid-pseudopapillary tumor(SPT) of the pancreas. Methods:The clinical data were reviewed in four patients with SPT. Immunohistochemistry for Vim, CK8, CK18, Chromogranin, Sy, AAT, NSE, S-100 was performed in all four cases using EnVision. Results:All four patients were female. The mean age was 30 years. The tumors were composed of papillary and solid structures. The uniform cells were strongly diffuse positive for Vim, and strongly positive for focal AAT in four cases. Three cases were positive for NSE. All four cases were negative for Sy , Chromogranin and S-100. One case was poorly positive for CK8 and CK18. Conclusions:SPT of the pancreas, preferentially in young women, is a tumor with low malignant potential. Immunohistochemistry is very important in the diagnosis and differential diagnosis of SPT of the pancreas..
3.The Structural Basis of Ca~(2+) Transport by The Ca~(2+-)ATPase From Skeletal Sarcoplasmic Reticulum

The Ca2+-ATPase of sarcoplasmic reticulum is a Ca2+ pump that plays a key role in regulating cytosol calcium concentration in muscle cells. It undergoes a sequential conformational transition during the transport process. According to the classical E1/E2 theory, in the E1 state the binding sites have high affinity and open to the cytoplasm, whereas in the E2 state the binding sites have low affinity and face the luminal side. Crystal structures of several states during the reaction cycle of Ca2+-ATPase have been solved recently, including a Ca2+-bound form (E1-2Ca2+), a Ca2+-unbound form stabilized by a potent inhibitor thapsigargin (TG) (E2-TG), an ATP-bound form (E1-ATP), an E1-P-ADP state, and an E2-Pi state. The details of these crystal structures and the relationship between structure and function of Ca2+-ATPase during reaction cycle were summarized, and the issues to be addressed in future research were raised.
4.Construction of recombinant lentivirus vector expressing small hairpin RNA against human Bax inhibitor-1 gene expression

BACKGROUND: Lentivirus-mediated RNA interference technology has been widely used in the study of gene function because of its specificity, high inhibition efficiency and persistent effects. Virus packaging, transfection, as well as small hairpin RNA (shRNA)-coding sequence could affect the inhibition efficiency. Therefore, impact factors of RNA interference for human Bax inhibitor 1 (BI-1) was studied in this experiment.OBJECTIVE: To find the valid small interference RNA (siRNA) targeting human BI-1 gene by Lentivirus.DESIGN, TIME AND SETTING: Single sample observation was completed in Department of Medical Genetics, Basic Medical College, Peking University Health Science Center from September 2007 to December 2008.MATERIALS: 293T cell and SH-SY5Y cells were preserved in this laboratory; 4 shRNAs were designed through a use of online design tool by Ambion (www.ambion.com). METHODS: Several recombinant plasmids which expressed shRNAs targeting different regions of the BI-1 gene and labeled enhanced green fluorescent protein as a fusion gene were constructed. Four types of shRNA-expressing virions were obtained by cotransfection of shRNA-expressing plasmid and package protein-expressing plasmids into 293T cells. Green fluorescent protein expression was determined using flow cytometry to search the optimal package conditions. Real-time PCR and beta actin mRNA was used as an internal control, four types of recombinant viral supernatant and control viral supernatant were added on the SH-SY5Y cells, knock-down efficacies of the endogenous BI-1 gene were determined, and the RNA interference effective sequences were optimized. MAIN OUTCOME MEASURES: Intracellular green fluorescent protein expression rate of the cells infected with different packaging virus. RESULTS: The optimal infective packaging virus could be obtained when the ratio of plasmids pLentiLox3.7, REV, VSVG and RRE was 2:1:1:1 and the supernatants were collected 48 hours after transfection. Replacement of culture media 24 hours after transfection could increase the infection efficacy of the virions. shRNA targeting-2-17nt (start coding region) of BI-1 gene was most valid interference efficiency and could decrease the gene expression to 40%. CONCLUSION: Influencing factors of Lentivirus-mediated RNA interference technology virus packages were investigated in this study, which could be a foundation for constructing stable BI-1 knocked down neuronal cell model and for studying the abnormal expression of BI-1 related to the neuron apoptosis disease.
5.Self-efficacy and influencing factors of clinical nurses with needlestick injuries

Objective To investigate the level and influencing factors of self-efficacy among nurses with needlestick injuries and explore effective strategies to improve self-efficacy.Methods 713 clinical nurses with needlestick injuries had been enrolled by convenience and stratified sampling method to comphte the investigation using the self-designed demographic questionnaire and General Self-Efficacy Scale.Results The average score of the self-efficacy was (2.70 ±0.52) points.69.1% of the needlestick injury nurses had the sclf-efficacy score at the middle level; only 19.1% of them had a higher level score on self-efficacy.Gender,title,section,initiative report,work environment,and occupational safety training were identified as the influencing factors of the self-efficiency.Conclusions Even though more than half of the nurses with needlestick injuries had a middle level self-efficacy,they need further improvement.Other influencing factors should be explored in future in order to develop the corresponding management intervention which can strengthen the self-efficacy of nurses with needlestick injuries.
6.The promoting effects of insulin-like growth factor 1 on the biological behaviour of human retinal vascular endothelial cells

Background The suppression of retinal angiogenesis is one of primary treatment targets for retinal vascular diseases,so seeking the intervention targets of retinal neovascularization is a hot research.Studies showed that insulin-like growth factor 1 (IGF-1) can promote the growth and restrain the apoptosis of vascular endothelial cells.However,whether IGF-1 is an intervention target for the treatment of retinal vascular diseases is unelucidated.Objective This study was to address the effects of IGF-1 on the migration,apoptosis and capillary tube formation of human retinal vascular endothelial cells (HRECs) and mechanism.Methods HRECs were cultured in vitro,and the cells in the exponential phase were prepared for subsequent experiments.The expression of IGF-1R mRNA in the cells was examined using reverse transcriptase PCR assay.Different concentrations of IGF-1 were added in the medium based on the difference of tests.The relative free-cell area difference (△S) after test was measured by Photoshop CS4 software and compared among 0,10 and 200 ng/ml IGF-1 groups 12 and 24 hours after cell scratching,respectively.The cell apoptotic rate was assayed by flow cytometry and compared between 0 ng/ml IGF-1 group and 1 000 ng/ml IGF-1 group,and the number of capillary tubes was examined by Matrigel test and assessed among 0,10,100 and 200 ng/ml IGF-1 groups 24 hours after addition of IGF-1.The expressions of platelet derived growth factor (PDGF)-BB mRNA and caspase-3 mRNA in the cells of the 0,500 and 1 000 ng/ml IGF-1 groups were detected by real-time fluorescence quantitative PCR after adding IGF-1 for 6 hours.Results Cultured cells grew well and attached 90% confluence 2-3 days after incubation,and IGF-1R mRNA was positively expressed in the cells.In 12 and 24 hours after scratching,the relative migrating area of the cells was gradually reduced with the increase of IGF-1 contents.The △S was (4.83 ± 0.61) × 105 μm2 in the 200 ng/ml IGF-1 group,which was significantly larger than (3.28±0.64) ×105 μm2 in the 0 ng/ml IGF-1 group 24 hours after stretching (t=-3.707,P=0.021).The apoptotic rate in the 0 ng/ml IGF-1 group and 1 000 ng/ml IGF-1 group was (18.77±2.37) % and (12.05 ±0.88) %,with a significant difference between them (t =2.869,P =0.046).The number of intact tubes was significantly increased in the 200 ng/ml IGF-1 group compared with the 0 ng/ml IGF-1 group ([20.33±2.83]/well vs.[17.94± 1.96]/well;t =-2.940,P =0.042).Compared with 0 ng/ml IGF-1 group,the relative expression level of PDGF-BB mRNA was elevated and that caspase-3 mRNA was evidently reduced in the 1 000 ng/ml IGF-1 group (t=-3.489,P =0.025;t =7.287,P =0.002).Conclusions IGF-1 can promote the migration and angiogenesis of HRECs and inhibit the apoptosis of HRECs.These effects of IGF-1 probably are associated with the up-regulation of PDGF-BB and down-regulation of caspase-3 in the cells.
7.The efficacy of meticulous process management in control of infections in operating room

Objective To discuss the nursing effect and application value of refinement process of nursing management mode in hospital infection control in the operating room.Methods 620 patients who will carry out surgi-cal treatment were selected as the research subjects.310 cases without detailed process of nursing management were included in the control group,and 310 patients with refinement process of nursing management were selected as obser-vation group.The clinical nursing effects of two groups were recorded.Results Observation group surface 88.94%, medical staff hand 92.54%,disinfectant 95.65%,100.00% sterile items.The control surface 81.38%,80.00% of medical personnel hand,disinfectant 75.93%,90.00% sterile items,the monitoring qualified rate between two groups had statistically significant differences (χ2 =5.021,4.596,7.581,6.725,all P <0.05).The sores of nursing safety, operating room management,form filling,ward nursing in the observation group were (95.37 ±4.66 )points, (94.27 ±5.21 )points,(96.13 ±3.29)points,(95.03 ±4.87)points.Those in control group were (90.12 ± 2.03)points,(89.13 ±2.26)points,(90.03 ±1.88)points,(88.17 ±2.25)points,the differences between two groups were statistically significant (t =18.185,15.935,28.343,22.514,all P <0.05).The scores of nurses and patients communication,nursing procedures,ward environment,care attitude in the observation group were (24.19 ± 0.87)points,(24.03 ±0.64)points,(24.51 ±0.45)points,(24.28 ±0.36)points.Those in the control group were (21.54 ±1.65 )points,(20.88 ±1.72)points,(20.95 ±1.66)points,(21.09 ±0.89)points,the differences between two groups were statistically significant (t =25.013,30.220,36.443,58.502,all P <0.05).Conclusion Refinement process of nursing management mode application helps to control nosocomial infection in operating room of hospital infection control,improve the quality of clinical nursing services,improve the clinical nursing satisfaction, which is worthy of popularization and application in clinic.
8.A REPORT OF THE CLAM'S DISEASE CAUSED BY VIBRIO FURNISSII

A gram-negative bacillus was isolated from the bodies of dead clams among clambanks along the south beach of Jiangsu province. It was confirmed to the Vibrio furnissii by morphological and biochemical characteristic examinations. This isolate grows well in clam's body fluid media and may multiply rapidly in sea-water at temperature about 25—37℃. It is of high toxicity. The healthy clams were all diseased with the same syndrome and died as natural illness by inoculate them with the isolate. It is considered that the large enormous death of clams along the south beach of Jiansu province is concerned with the wide spreading of the clam's infectious disease caused by Vibrio furnissii which hasn't been reported both internal and abroad.

The fine ultrastructure and localization of acid phosphatase in cell ultrastructuralevel of a HGPRT-human promyelocytic leukemia cell mutant(HL-60-AR)arestudied by scanning electron microscopy,transmission electron microscopy,freezeetching,and electron microscopic cytochemistry techniques.The results of theobservation show that the ultrastructural characteristics of HL-60-AR cells aresimilar to that of HL-60 cells.There are microvilli and ridges over cell surface.Thecells have large nucleus with prominent nucleoli,and numerous nuclear pores.Thereare less developed Golgi complex,expanded rough endoplasmic reticulum andabundant polyribosomes.After treatment with retinoic acid(RA)at 10~(-6) mol/L for 5days,HL-60-AR cells differentiate along myeloid pathway and have a decreasednucleocytoplasmic ratio accompanied with nuclear condensation and segmention.A (?)ignificant increase of specific granules is demonstrated.Microvilli of the cellsdisappear,surface features of the treated cells become more irregular and largeprotrusion and blunt pseudopodia appear.Increase of acid phosphatase content localizedon azurophilic granules(lysosomes)and Golgi complex is showed.The application offour kinds of electron microscopic techniques might provide the best way foridentifying cell ultrastructure.
10.Study on parents′ knowledge and attitude toward postoperative pain management of children with limb fracture

Objective:To investigate parents′ knowledge and attitude toward post-operative pain management in pediatric orthopedics and analyze influencing factors.Methods:A total of 170 parents of 6 to 16 years old children who underwent selective surgeries and were hospitalized because of limb fracture were conveniently selected in Children′s Hospital of Soochow University from April to December in 2019, They were investigated using the general data questionnaire, Pain Management Knowledge and Attitudes Questionnaire.Results:There were 167 parents completed questionnaires in our study. The mean score of parents′ knowledge, general attitude and negative attitude to pain medication were 15.0(3.00), 24.0(4.00) and 17.0(3.00) respectively. There were statistical differences between different educational levels of parents′ knowledge ( χ2 value was11.599, P=0.021), as well as different age of parent′s general attitude ( χ2 value was 7.053, P=0.029). Spearman-related results showed that parents′ cognition was positively correlated with general attitudes ( r value was 0.394, P<0.01). Conclusions:The majority of parents are at moderate to high level of knowledge, general attitude and negative attitude to pain medication. Pediatric nurses should pay more attention to parents′ knowledge deficit of and negative attitude toward pain assessment tools and pain-drugs to develop targeted educational intervention programs for promoting parental involvement.
