1.Expression of hTERT mRNA in salivary gland tumors

Objective:To investigate the expression of human telomerase reverse transcriptase(hTERT)gene in salivary gland tumors and to study its clinicopathological significance.Methods:In situ- mRNA hybridization method was used to detect the expression of hTERT mRNA in 10 cases of normal salivary gland tissues, 30 of benign salivary gland tumors and 43 of malignant salivary gland tumors.The relationship between hTERT mRNA expression and sex,age, tumor size, location, histologic differentiation of the malignant tumors was analyzed by using ?2 test.Results:hTERT expression was negative in all normal tissues. Positive rate of hTERT mRNA expression was 3.3%(1/30) and 83.7%(36/43) in benign and malignant salivary gland tumors respectively(P0.05).Conclusion:hTERT mRNA expression may be used to distinguish malignant salivary gland tumors from benign.
2.Inhibitory effect of antisense oligonucleotide of integrin-linked kinase on cell proliferation of human epithelial ovarian cancer

Objective To explore the inhibitory effect of integrin-linked kinase antisense oligonucleotide(ILK-ASODN)on cell proliferation in human ovarian cancer cell line(HO8910).Methods We transfected ILK-ASODN into HO8910 to block ILK gene expression.measured the expression levels of integirn-linked kinase(ILK)mRNA by RT-PCR and ILK protein by western-blotting;the inhibiting effects of the transfection on HO8910 proliferation,the cell cycles,and cell apoptosis were assessed by water soluble tetrazolium-1(WST-1)and flow cytometry(FCM).Results After transfection of ILK-ASODN,the expression levels of ILK mRNA decreased significantly in groups D,E,F being 0.307±0.011,0.198±0.008,0,respectively,when compared with those of the two control groups of A and B(P<0.05).The expression levels of ILK protein of the groups D,E and F decreased significantly also,being 26.3 ± 0.8,20.6±0.4 and O,respectively.HO8910 cell proliferation was inhibited significantly,and the rates of apoptosis of the groups D,E and F increased significantly,being 7.31%,8.84%and 11.27%respectively.The cell population increased in G0/G1 phase of the groups D,E and F,being 49.25%,56.28%and 67.61%respectively.significantly different in comparison with those of groups of A and B (P<0.01).Conclusions Transfection of ILK-ASODN into human ovarian cancer line inhibited cancer cell proliferation significantly.
3.Expression of glucose-regulated protein 78 in ovarian cancer tissues and its clinical significance

Objective To explore the expression of glucose-regulated protein (Grp) 78 in ovarian cancer tissues and its clinical significance.Methods The expression of Grp78 in 60 cases of ovarian cancer tissue,15 cases of ovarian borderline tumor tissue,10 cases of normal ovarian tissue,10 cases of ovarian cyst tissue,was detected by immunohistochemistry,and analyzed the relationship between its expression and clinicopathological features of ovarian cancer.Results The expression of Grp78 in ovarian borderline tumor and ovarian cancer tissue was significantly higher than that in normal ovarian and ovarian cyst tissue[7/15 and 68.3% (41/60) vs.1/10 and 1/10] (P <0.05).The expression of Grp78 was positively correlated with clinicopathological staging and lymphatic metastasis of ovarian cancer (P < 0.05),negatively correlated with histological differentiation (P < 0.05).No correlation with age and ascites (P > 0.05).Conclusions The levels of Grp78 are elevated in ovarian cancer specimens; high expression of Grp78 maybe participate in the occurrence,development,and prognosis of ovarian cancer.Increased expression of Grp78 might be a useful marker for predicting the carcinogenesis and progression of ovarian cancer.
4.Small interfering RNA inhibits glucose regulated protein 94 expression in transplantable models of human ovarian carcinoma in nude mice

BACKGROUND:After glucose regulated protein 94 (GRP94) was knockout in model mice of transplanted tumor, cel ular adhesion is terminated, thus stimulating the proliferation of liver-derived cel s and promoting the development of liver cancer. We speculate that GRP94 plays a protective role against liver cancer.
OBJECTIVE:To investigate the expression of endoplasmic reticulum molecular chaperone GRP94 mRNA and protein with smal interfering RNA technique in nude mice model of transplantable human ovarian carcinoma, and to explore the effect of GRP94 mRNA and protein expression on the growth of transplanted tumor.
METHODS:The gene sequences of human GRP94 were obtained from Gene Bank. psiSTRIKETM/GRP94 was constructed, which is eukaryotic expression vector control ed by the U6 promoter of human RNA polymerase Ⅲ. The transplantable model of human ovarian carcinoma in nude mice was established using human ovarian cancer HO-8910 cel line. The eukaryotic expression vector was transfected into the transplanted tumors in nude mice, and the growth of the tumor was observed. The nude mice models were divided into three groups, specific smal interfering RNA group, non-specific smal interfering RNA group and saline control group. The volumes of the subcutaneous tumor were determined. RT-PCR and immunohistochemistry were used to detect the mRNA and protein expression of GRP94 respectively.
RESULTS AND CONCLUSION:The recombinant plasmid of RNA interference specific for GRP94 was successful y constructed. The subcutaneous tumors appeared in al the nude mice 5 days after transplantation. The diameter of subcutaneous tumors was 7-10 mm 14 days after transplantation. The growth of subcutaneous tumors in nude mice with interference specific for GRP94 treatment was significantly decreased as compared with non-specific smal interfering RNA group and control group (P<0.05). The proliferation activity was inhibited by 65.1%. The expression of GRP94 mRNA and protein was significantly down-regulated after treatment of psiSTRIKETM/GRP94 (P<0.01). The transfection of psiSTRIKETM/GRP94 could significantly induce inhibitory effects on the growth of ovarian carcinoma in nude mice, and the underlying mechanism is associated with the down-regulated expression of GRP94 mRNA and protein.
5.Alveolar bone defect repair using autologous bone marrow mesenchymal stem cells combined with platelet-rich fibrin

Abstract BACKGROUND: Alveolar bone deficiency wil not meet aesthetic and functional requirements for dental implants. OBJECTIVE:To observe the repair effect of passage 3 autologous bone marrow mesenchymal stem cels (BMSCs) and platelet-rich fibrin (PRF) on alveolar bone defects in rabbits. METHODS:Twenty-seven New Zealand rabbits were randomly divided into BMSCs/PRF group, PRF group and model group (n=9 per group). The left mandible incisors were extracted in al the rabbits under general anesthesia. BMSCs/PRF group was immediately implanted BMSCs/PRF composite into the alveolar socket, PRF group only implanted PRF, and model group implanted nothing. RESULTS AND CONCLUSION: In the model group, the alveolar crest and alveolar mucosa become sunken notably and narrowed. In the BMSCs/PRF and PRF groups, the thickness of alveolar bone wal, alveolar bone width, alveolar bone height difference, and bone mineral density were al increased, especialy in the former group. In addition, the trabecular arrangement was better in the BMSCs/PRF groups than the model and PRF group. Our findings indicate that alveolar socket filing with composite of BMSCs and PRF can achieve preservation of alveolar bone width and height after tooth extraction in rabbits.
6.Nursing of patients with heat illness combined with respiratory failure

ObjectiveTo summarize the experience of nursing patients with heat illness combined with respiratory failure. Method The nursing records of 13 patients with heat illness combined with respiratory failure were reviewed for summarizing the nursing measures.Result After treatment and nursing care,12 of them were discharged and 1 died from multiple organ dysfunction. Conclusions Rapid cooling is key to the success of rescue.The nursing measures include keeping respiratory tracts unobstructed and preventing complications from artificial airway and mild hypothermia therapy.

Objective: To study the relationship between the individual psychological characteristic of medical students and the way parents bring up them. Methods: Data on SCL-90, 16PF,EMBU of 1550 medical students were analysed. Results: It is showed that girl students have experienced more of the parents' emotion, understanding and favouritism, while the boy students have experienced more of their parents' refusal, deny, punishment, severity and excessive interference. There are more only-one-children who experience parents' emotion, understanding, interference and excessive protection. Conclusions: Parents' bad breeding attitude and behavior influence children's psychological health and forming of the individual characteristic directly or indirectly
8.The value of combined MRS and DWI in diagnosis of cervical cord injury

Objective To explore the value of MRS and DWI in diagnosis of cervical spinal cord injury.Methods The MRI plain scan images of 95 cases who suffering from cervical spinal cord injury were analyzed.They were divided into two groups according to the T2 signal of cervical cord,63 cases in Group A(incomplete injury group)with no abnormal on T2 ,32 cases in Group B(complete injury group)with high signal on T2 .The other 50 cases with no injury of cervical cord were recruited as control group (Group C). All the cases were performed with MRS and DWI.The N-acetyl aspartic acid(NAA),choline compounds(Cho)and creatine(Cr)as well as the peak area of lactic acid (Lac)on MRS,and apparent diffusion coefficient(ADC)on DTI were measured separately.Mean-while,NAA/Cho,NAA/Cr,Cho/Cr,Lac/Cho and ADC were analyzed.Results On MRS sequence,the values of NAA/Cho and NAA/Cr were lower in Group B (P <0.05)when compared with Group A.The values of NAA/Cho and NAA/Cr were also lower in Group B,when compared Group C.However,the value of Lac/Cho in Group B was higher than it in Group C(P <0.05).When com-pared Group A with Group C,the value of Lac/Cho only increased in the former.However,on DWI,the value of ADC in Group A was(0.79±0.1 7)×10 -3 mm2/s,lower than that of Group C(0.93±0.1 5)×10 -3 mm2/s (P =0.026);the value of ADC in Group B was(1.21±0.20)×10 -3 mm2/s,significantly higher than that of Group C(P =0.01 7).When compared with Group B,the value of ADC in Group A was a lightly lower (P =0.143)with no statistical significance.Conclusion MRS can noninvasively and quantita-tively reflect the changes of metabolism of medium in injured cervical cord and help characterize the severity and degree of cervical cord injury from metabolic and molecular level.DWI has high sensitivity on early diagnosis of cervical cord injury.The combination of MRS and DWI has important value for formulating reasonable treatment plan and prognostic evaluation.
9.The discrimination of ~(18)F-FDG PET/CT detection of incidental ileocecal fluorodeoxyglucose uptake

Background and purpose: As the most common tracer used for PET/CT to detect malignant tumors. ~(18)F-fluorodeoxyglucose (~(18)F-FDG) can reflect the metabolism of glucose. However, there exists physiologic uptake in the intestinal tract of healthy people. Because ileocecal foci is predilection site of malignant tumors and inflammatory disease, the purpose of this study was to investigate the discrimination of PET/CT detection of incidental ileocecal high FDG uptake. Methods: We retrospectively analyzed 28 unexpected ileocecal focal uptake of ~(18)F-FDG localized by PET/CT, and then semi-quantitatively analyzed the extent of radioactive uptake in ileocecal foci. After PET/CT, the etiology of the findings was confirmed histologically by surgery, endoscopic or by long-term follow-up. All the data were assessed for statistical significance using one-way ANOVA. Results: In all the ileocecal foci, there were 14(50%) physiologic FDG uptake, 8 (28.6%) benign diseases and 6 (21.4%) malignant tumor. Maximal standardized uptake value was 5.2±1.6 in physiologic uptake, 6.8±4.1 in benign lesions and 12.8±5.5 in malignant lesions. There was statistically significant difference between malignant tumors and the other 2 groups. According to ROC analysis, when SUN_(max) was 6.75, the sensitivity and specificity of ileocecal malignant tumor were 100% and 86.4%, respectively. Conclusion: The significant value of discrimination between benign and malignant lesions of ileocecal focal FDG uptake by PET/CT was affirmative. It is very important to recognize physiologic uptake of ileocecal conjunction in the interpretation of image. Semi-quantitative analyze was not helpful to identify benign lesions from physiologic uptake.
10.Effect of platelet-rich fibrin released transforming growth factor beta 1 on the proliferation of bone marrow mesenchymal stem cells in vitro

BACKGROUND:Bone marrow mesenchymal stem cel s are the ideal cel s for tissue repair. Whether the ability of in vitro proliferation can be enhanced is a key factor to promote tissue repair.
OBJECTIVE:To observe the effect of transforming growth factorβ1 on the proliferation of bone marrow mesenchymal stem cel s in vitro.
METHODS:Blood samples were taken from the central artery of rabbits to prepare platelet-rich fibrin by centrifugation method which was then placed into fresh DMEM at 37℃for 7, 14, 21, 28 days to col ect exudates. The mass concentrations of transforming growth factor-β1 in the exudates of platelet-rich fibrin were detected. Rabbit bone marrow mesenchymal stem cel s were col ected and cultured in the conditioned medium made by the exudates of platelet-rich fibrin, and the proliferation of cel s was observed.
RESULTS AND CONCLUSION:Concentration of transforming growth factorβ1 was increased with time increasing, increased fastest at 21-28 days, and peaked at 28 days. Under the same stimulus concentration, the proliferation of bone marrow mesenchymal stem cel s was reduced at 0-1 day, increased obviously at 1-2 days, and entered into a steady phase at 2-3 days. Under 150 ng/L transforming growth factorβ1, bone marrow mesenchymal stem cel s proliferated fastest. Experimental findings indicate that with the increase of time, the concentration of transforming growth factorβ1 in the exudates of platelet-rich fibrin increase gradual y, and the conditioned media containing different concentrations of transforming growth factorβ1 play different roles in
promoting the proliferation of bone marrow mesenchymal stem cel s. Bone marrow mesenchymal stem cel s cultured in the conditioned medium containing 150 ng/L transforming growth factorβ1 for 2-3 days can proliferate fastest.
