Objective To observe the changes of chief chemical components(hypaconitine,ephedrine,pseudoephedrine,peoniflorin,glycyrrhetic acid) in the combinations of different-dose herbs of Wutou Decoction(WD). Methods Uniform design method was adopted to design the experiments,and HPLC was applied to determine the contents of chief chemical components of hypaconitine,ephedrine,pseudoephedrine,peoniflorin,glycyrrhetic acid. DPS V7.55 was used to carry out the nonlinear regression analysis of experimental data such as decocting rates of the chief chemical components. The mathematical model was established to analyze the experimental results. Finally,with the decocting rates of the chief chemical components as the optimizing indexes,the combination of optimal-dose herbs of WD was seeked. Results There existed an interaction between each herb of WD,and each herbs as well as the interaction between the herbs had a certain effects on the decocting rate of the chemical components. In particular,the interaction of prepared Radix Aconiti and prepared Radix Glycyrrhizae had obvious effects on the decocting rates of hypaconitine and glycyrrhetic acid. On the basis of the different optimization index,different combinations of optimal-dose herbs of WD can be obtained. Conclusion Through the method of nonlinear regression analysis,rich connotation of compatibility in Chinese herbal formula can be obtained.The optimization of optimal-dose herbs combination of the formula should be based on the pharmacological actions.

Objective To investigate the diagnostic and therapeutic methods of small intestinal hemorrhage.Method Clinical data of forty-nine cases of small intestinal hemorrhage were analyzed retrospectively.Results Small intestinal tumors were found in 21 cases (42.9%),among wtich,most were benign;infectious diseases in 12(24.5%);diverticula's in 8(16.3%);and vascular malformations in 6(12.2%).Radionuclide scanning was positive in 10 out of 13 cases (76.9%).Angiography was performed in 12,7 of them were abnormal (58.3%).Small bowel series made diagnosis in 25% of 36 cases.Postoperative bleeding was found in 5 cases and short bowel syndrome in 1 case.Conclusions Tumors are the most common cause of small bowel bleeding,and acute hemorrhagic nercotizing enteritis and typhoid often cause small bowel massive bleeding.Radionuclide scanning is one of the most useful diagnostic methods for diverticula's.Angiography is a valuable procedure in diagnosis of vascular lesions and malignants.Small bowel series is also helpful in diagnosis of solid lesions and diverticula's. Exploratory surgery coupled with intraoperative endoscopy can be helpful in diagnosis of small bowel bleeding.

Objective To determine the contents of strychnine and brucine in Xiaobining Spray.Methods A HPLC method was adopted and the chromatogram conditions were as follows:column of Kromasil C18(4.6 mm? 250 mm,5 ? m),mobile phase consisting of acetonitrile-0.01 mol/L sodium heptanesulfonate and 0.02 mol/L potassium dihydrogen phosphate equivalent volume mixture(10 % phosphoric acid adjusting pH to 2.8)(21:79),column temperature of 30 ℃,flow rate of 1.0 mL/min,detection wavelength of 260 nm.Results A good linearity was obtained form 0.109 2 ? g to 0.546 0 ? g for brucine(r=0.999 6)and 0.173 6 ? g to 0.868 0 ? g for strychnine(r=0.999 7).The average recovery of brucine was 100.59 % with RSD being 1.81 %,and of strychnine was 100.83 % with RSD being 1.69 % .Conclusion This method is simple,reproducible,and can be used to determine the strychnine and brucine in Xiaobining Spray.

Objective To determine the contents of hypaconitine and mesaconitine in Shubining Granules. Methods The HPLC method was established for the determin ation of hypaconitine and mesaconitine in Shubining Granules. The HPLC was perfo rmed on Akasil C18(4.6 mm?250 mm,5 ?m) column,mobile phase consisted of meth anol-0.1 %triethylamine (70 ∶30),column temperature 30 ℃,and the flow rare was 1.0 mL/min.The detection wavelength was 230 nm.Results A good linearity w as obtained in the range of 0.126～2.520 ?g of Hypaconitine and 0.152～3.04 ?g of Mesaconitine.The average recovery of Hypaconitine was 101.12 %.The average recovery of Mesaconitine was 99.68 %. Conclusion This method is simple and qui ck,and can be used as quality control of Shubining Granules.

【Objective】To observe the effect of Ma Xing Shi Gan Decoction(MXSGD)on metabolism of oxygen free radicals in mice with bronchial asthma.【Methods】Forty BALB/c mice were randomized into 4 groups:normal group,model group,low-dose MXSGD group(20?g?kg~(-1)?d~(-1))and high-dose MXSGD group (80?g?kg~(-1)?d~(-1)).Except the normal group,the mice in other 3 groups received sensitization by intraperitoneal injection of ovalbumin(OVA)and challenge by inhalation of OVA to induce bronchial asthma.Meanwhile,MXSGD groups were given the corresponding doses according to the experimental design by gastric gavage,and normal group and model group received the same volume of normal saline for 27 days.Twelve-four hours after last challenge,blood sample was taken and homogenate of lung tissue was prepared for the detection of superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(GSH-Px)contents in the plasma and the lung tissue.【Results】High-and low-dose MXSGD increased SOD and GSH-Px contents and decrease MDA content in the lung tissue and plasma(P

Efforts on directed evolution of sortase A to optimize its catalytic properties have been undertaken and shown the promise. To facilitate screening of sortase A mutants with expected catalytic properties, a novel ligation efficiency monitoring system, including reporter substrates GFP-LPETG and GGGYK-Biotin, was developed. GFP-LPETG, wild type sortase A, and a recently reported high activity sortase A mutant were prepared recombinantly from Escherichia coli BL21 (DE3). Taking advantage of the newly designed reporter system, the ligation efficiency catalyzed by wild type and mutant form of sortase A could be sensitively monitored via SDS-PAGE directly. Consistent with previous report, the mutant sortase A displayed much higher catalytic activity compared to wild type enzyme, indicating the new reporter system is easily and fast handled and sensitive. The application of this reporter system into systemic screening will facilitate future directed optimization of sortase A.

Objective To investigate the effect of Heat shock protein 70 induced by hypoxic precondition on apoptosis of the remnant livers after the hepatectomy of transplanted hepatic cancer models in SD rants and it's possible mechanisms.Methods Sixty SD rats of transplanted hepatic cancer models were divided into group A (sham operation group),group B (IR + hepatectomy group),and group C (hypoxia precondition + IR + hepatectomy group),with 20 rats in each group.C group was given 10％ oxygen atmosphere for 90 minutes before operation,In 15 minutes of hepatic vascular occlusion with pringle method,Left lateral of liver which tumor was located was resected.At 1,8,12,24 hours after operation,Five rats of each group were killed respectively,then the remnant liver tissues were sampled to measured the hepatocytes apoptosis rate with flow cytometry,expression of heat shock protein 70 (HSP70) and Bax were detected by Western blotting.The serum AST and ALT activities were measured by an automatic analyzer.The histological changes of the hepatocytes were also observed by optical microscopy.All the data were statistically analyzed.Results (1) Compared with A and B group,the HSP70 expression increased obviously in C group at each time point (all P ＜ 0.05).The protein showed a weak expression in group A and B group,while the index had no significant differences between two groups (P ＞ 0.05).(2) In B group compared with A group,the expression of Bax and the hepatocytes apoptosis rate were significantly increased (all P ＜ 0.01 or P ＜0.05).However,amplitudes of the above changes in C group were significantly lower than those of B group (all P ＜ 0.05).(3)The liver function was improved in group HP.The pathological examination showed that the liver tissues in A group were general normal and in C group were showed mild changes,but in B group presented evident pathological changes.Conclusions HP can protect against ischemia-reperfusion injury after the hepatectomy of transplanted hepatic cancer models in SD rants,one of possible mechanisms is that HP can induce HSP70 expression at great intensity,which may decrease expression of Bax protein and the apoptosis of hepatocytes.

Objective To investigate the level of soluble interleukin 7 receptor (sIL-7R) in serum of lupus nephritis(LN)patients and evaluate its clinical significance. Methods sIL-7R level in serum of LN patients and healthy controls were measured by ELISA , while total 24 hours urinary protein and complement C3 of LN patients were measured by BN ProSpec. The level of sIL-7R correlation with SLEDAI, total 24 hours urinary protein and complement C3 were analyzed respectively. Results The levelof sIL-7R was higher in serum of LN patients than healthy controls (P < 0.01). Moreover, its expression in serum was increased in LN patients in active stage than in LN patients in stable stage (P < 0.05). The level of sIL-7R was positively assosicated with SLEDAI, total 24 hours urinary protein(P < 0.01, P < 0.05) and negatively with complement C3 (P < 0.05). Conclusion The level of sIL-7R is upregulated in serum in LN patients and correlated with disease activity and progression, so it may be expected to become a potential marker of disease in prediction.
