1.Therapeutic effects of phlorizin on the early stage of diabetic retinopathy in diabetic db/db mice

Objective To investigate therapeutic effects of phlorizin on the early stage of diabetic retinopathy(DR)in diabetic db/db mice.Methods The 24 db/db male mice were divided into 3 groups of phlorizin-treated diabetic group(20mg/kg of phlorizin by intragastric gavage)(DMT,n =8),diabetic group(normal saline solution by intragastric gavage)(DM,n=8),with C57BLKS/J db/ m mice as the control group(CC,n=8)for ten weeks.After ten weeks,all mice were sacrificed and the retina was isolated.The specimen was embedded in paraffin wax and sliced.The sections were dyed with Sudan red and observed for retinal morphological change.Meanwhile,the expressions of GSK-3β and phospho-GSK-3β in retinas were determined by western blot.Results During the study,the CC group showed good condition.The DM group featured with polydipsia,polyphagia and a rapid increase of body weight.Phlorizin-treated diabetic mice displayed much better than the DM group.Compared with CC group,DM group showed cells proliferation in capillary endothelia and obvious thickening in basement membrane.After administration of phlorizin,the vascular lesion of central regions was remarkably improved.The expression level of phospho-GSK-3β protein was lower in DM group than in control group(76.2±15.8 vs.255.4 ± 10.7,t =16.30,P<0.01)and was higher in Phlorizin-treated DM group(188.5±18.4)than in DM group(76.2± 15.8,t=8.05,P<0.01).Conclusions Phlorizin decreases phospho-GSK-3β3expression in retina in db/db diabetic mice and suppresses the GSK-3βactivity,which leads to the protection of DR.
2.Time-dependent changes in CT of radiation-induced liver injury: A preliminary study in gastric cancer patients.

In this study, the time-dependent changes on dynamic computed tomograph (CT) of radiation-induced liver injury in gastric cancer patients was examined. The CT images of 52 gastric cancer patients who had received chemoradiotherapies were reviewed on the PACS system. Dynamic CT scan was performed in all the subjects. Our results showed that 18 patients were found to have radiation-induced liver injury. The CT findings of radiation-induced liver injury in gastric cancer patients tend to show up one month after radiation treatment. The damaged area was of low density on all three phases, and then it was enhanced on portal vein phase or delay phase. The focal radiation reaction of liver without basic disease vanished 9-11 months later after treatment. We are led to conclude that dynamic CT is of help in the diagnosis of CRT-induced liver injury, and it may be the method of choice for following up the whole course of the CRT-induced liver injury, i.e., form hepatic damage to healing. The classification of CT findings we recommend can avoid the influence of technological factors, and thereby serve as a better guide for treatment of CRT-induced liver injury.
3.Cost-effectiveness analysis of sequential therapy of levofloxacin in the treatment of type Ⅲa prostatitis

Objective To evaluate the cost-effectiveness of sequential therapy of levofloxacin in the treatment of type Ⅲa prostatitis.Methods A total of 280 in-patients diagnosed typeⅢa prostatitis were enrolled in this study and randomized to administer with either levofloxacin 500 mg( n =140 ) for intravenous injection once daily for 28 days ( control group) or administered of levofloxacin injection 500 mg ( n=140 ) once daily for 7 days followed by oral levofloxacin 500 mg once daily for 21 days( sequential group).Clinical results and safety were observed and cost-effectiveness analysis was made after 4 -week therapeutic dura-tion.Results The clinical effective rates showed no significant difference between 2 groups ( P<0.05 ).But cost -effectiveness ratio and sensitivity analysis in sequential group was significantly lower than that in control group(P<0.05).The adverse drug reaction rates in con-trol group ( 15.71%) and sequential group ( 5.71%) showed significant difference between 2 groups ( P<0.05 ).Conclusion The sequential therapy with the antibiotic therapeutic of levofloxacin 500 mg once daily achieved similar clinical efficacy to mono-therapy and offered an eco-nomic superiority and less adverse reactions in the treatment of type Ⅲa prostatitis.
4.Quality analysis of 2009 pandemic influenza A(H1N1) vaccines

Objective To analyze the laboratory testing data of 2009 pandemic influenza A (H1N1) vaccines during lot release procedure, thus to know the overall quality status of this vaccines.Methods National Institutes for Food and Drug Control(NIFDC) carried out the laboratory test according to the specifications of each manufacture, and the results was analyzed and compared between manufacturer and NIFDC. Results 99.8% of vaccines batches were released by NIFDC, haemagglutinin contents were between 90% to 103 % of labeled values, and testing results slightly differ between manufactures and NIFDC,other items related to safety were all meet specifications. Conclusion The quality of H1N1 vaccines in China were satisfying, the lot release and independent test by NIFDC play important roles to ensure the vaccines' quality.

6.Therapeutic effect of adipose derived mesenchymal stem cell conditioned medium thermosensitive hydrogel on skin scald

BACKGROUND: Studies have shown that adipose derived mesenchymal stem cells are involved in the skin repair after scald, but the hydrogel made of the excreta by adipose derived mesenchymal stem cells is rarely reported in the treatment of skin scald. OBJECTIVE: To analyze the therapeutic effect of human adipose derived mesenchymal stem cell conditioned medium hydrogel in a mouse model of skin scald. METHODS: Adipose derived mesenchymal stem cells were obtained from adipose tissues by enzyme digestion combined with adherent culture method. Morphological and flow cytometry were used to identify phenotype and induce differentiation. Secondly, the stable proliferative phase cells were harvested to obtain the conditioned medium, and chitosan, mannitol, beta-glycerol phosphate sodium and hyaluronic acid were added to prepare the thermosensitive hydrogel. Then the 95 ℃ aluminum block was used to rapidly establish a model of degree III skin scald on the left (experimental group) and right (control group) sides of the back of 24 C57BL/6 mice. In the experimental group, adipose derived stem cell conditioned medium hydrogel was applied twice a day on the right side of the mouse back, and in the control group, fresh medium hydrogel was applied twice a day on the left side of the mouse back. The treatment period lasted for 7 days. Healing time and healing process were observed to calculate the healing rate. Histopathological changes were observed by hematoxylin-eosin staining at paraffin sections at 4, 14, 28 days after skin scald. RESULTS AND CONCLUSION: (1) The human adipose derived mesenchymal stem cells had fibroblast-like morphology and proliferated vigorously, and the average doubling time was 55 hours. These cells could be induced to differentiate into osteoblasts, chondrocytes and adipocytes. High expression of CD29, CD44, CD90 and CD105 were observed on these cells with low expression of CD31 and CD34, which met the standard of mesenchymal stem cells. (2) Thethermosensitive hydrogel prepared by the conditioned medium was cool and transparent viscous liquid at 4-20 ℃, and was changed into semi-solid gel at( 37 ℃ after 15 minutes. (3) The normal structure) subcutaneous fat and muscle tissue (of 95 ℃ aluminum block scalded mice wer) standards of degree III burns. The wound area was roughly 3 cm2. (4) In the repair process, shorter wound healing time, less scar and better dermis structure were observed in the experimental group compared with the control group. (5) Inflammatory infiltration, thickness of granulation tissue, epidermal thickness, fibroblasts and vascular density were significantly improved in the experimental group as compared with the control group (P < 0.05). To conclude, human adipose derived mesenchymal stem cells conditioned medium hydrogel can promote the wound healing and promote the quality of regenerated skin after skin scald.
7.Effects of silencing inhibitor of DNA binding-1 gene on the growth and invasiveness of adenoid cystic carcinoma cells.

OBJECTIVETo investigate the role of inhibitor of DNA binding-1 (Id-1) gene in adenoid cystic carcinoma cell growth and invasion behavior.
METHODSWith salivary adenoid cystic carcinoma cell lines ACC-M and ACC-2, dedected Id-1 gene expression was screened with immunofluorescence assay. After Id-1 mRNA knocking-down using small interfering RNA, RT-PCR and Western blot were used to detect the different expressions before and after interference, and the growth of cells before and after interference was deceted using the MTT assay, and the cell invasion ability was checked with the use of Transwell chamber assay.
RESULTSId-1 were both expressed in the ACC-M and ACC-2, and the expression in ACC-M was higher than that in ACC-2. After Id-1 RNA interference, the growth and invasiveness of ACC-M and ACC-2 were inhibited with the restrained degree in ACC-M much stronger than that in the ACC-2.
CONCLUSIONIn view of the important role of Id-1 in the behavior of growth and invasion in ACC cell, interfering the expression of Id-1 gene is expected to be a novel and effective means for the treatment of adenoid cystic carcinoma.

8.Iodine nutrition level of children aged 8 - 10 in low-coverage area of iodized salt of Yushu Qinghai province in 2009: an analysis of surveillance results

Objective An analysis was conducted to investigate the iodine nutrition level of children aged 8 - 10 in low-coverage area of iodized salt of Yushu Qinghai province for providing a scientific basis for the development of effective preventive measures. Methods Yushu, Chengduo, Nangqian and Zaduo counties with higher non-iodized salt coverage rate in Yushu Qinghai province were chosen as survey counties in 2009. Three townships were selected in each county, and 2 primary schools were selected in each township and 40 urine samples of children aged 8-10 were collected randomly in each primary school. The content of urinary iodine was analyzed by As-Ce catalytic spectrophotometery. Results Median urinary iodine of children aged 8 - 10 in Nangqian and Zaduo was < 100 μg/L. The percentage of median urinary iodine < 50 μg/L in Yushu was over 20%. Median urinary iodine of children aged 10 in Zaduo was 81.5 μg/L, the percentage of median urinary iodine < 50 μg/L of children aged 9 and 10 was over 20%. The percentage of median urinary iodine < 50 μg/L in children aged 9 and 10 of Yushu was over 20%. Median urinary iodine of girls in Zaduo was 87.1 μg/L, the percentage of median urinary iodine < 50 μg/L of boys in Zaduo was over 20%. The percentage of median urinary iodine < 50 μg/L of girls in Yushu was over 20%. Conclusions The iodine nutrition level of children aged 8 - 10 in Nangqian, Zaduo and Yushu counties were very low due to the impact of non-iodized salt. We propose salt market in the region to strengthen management and improve the coverage and consumption rates of iodized salt to improve the level of iodine nutrition for effective prevention of iodine deficiency disorders.
9.Factors affecting clinical application of sex therapy for erectile dysfunction.

OBJECTIVETo discuss the factors that affect the clinical application of sex therapy for erectile dysfunction (ED).
METHODSUrological researchers recommended free sex therapy to ED patients and their partners in the outpatient department and studied the factors affecting the patients of Medicaid their partnersology, acceptance of sex therapy by interview and observation.
RESULTSMany patients refused sex therapy. Only 11 pairs of volunteers underwent it, of whom 5 pairs gave up halfway, 1 pair were stopped by the therapist due to the breakup of their marriage, and merely 5 pairs accomplished the whole course. Among the 5 pairs of quitters, 2 pairs thought of their problem as organic and turned to drug therapy, 1 pair withdrew because the patient's girlfriend broke up with him, 1 pair gave up because they lived in another city too far away from the clinic, and the other pair discontinued because they could not spare the time from their work.
CONCLUSIONSMany factors affect the clinical application of sex therapy for ED in China. Apart from the shortage of qualified professional sex therapists, some factors from the patients and their partners are at work, such as stress of work, lack of time, home location, education background, relationship between the patient and his partner, attitude to sex therapy, and so on.

10.Sex therapy is effective for erectile dysfunction.

OBJECTIVETo evaluate the sex therapy for erectile dysfunction (ED).
METHODSWe recommended the sex therapy to ED patients and their partners (11 couples in all) in the outpatient department. Of the total number of volunteers, 2 males were accompanied by ejaculation disorder, another 2 with hyposexuality, and 1 female had vaginismus. The effect of the therapy was assessed and the problems with it analyzed by interviews with the subjects and the results of International Index of Erectile Function (IIEF) before and after the treatment.
RESULTSOf the 11 pairs of volunteers, 5 quitted halfway and 1 discontinued at the therapists' decision. The 5 couples who accomplished the whole therapy felt more satisfied with their sexual life and general relationship, with improved scores on all the items of IIEF, particularly on erectile function (EF), with the only exception of sexual desire (SD).
CONCLUSIONThe sex therapy is effective not only for ED but also for other accompanying sexual dysfunctions, the sexual dysfunctions of the patients' partners and the improvement of the general relationship of the couples.
