1.Effects of GLP-2 on the mRNA expression of GLP-2R and PG in postburn rats

Objective To investigate the effects of glucagon like peptide 2(GLP 2) on the mRNA expressions of GLP 2 receptor (GLP 2R) and proglucagon gene(PG) in postburn rats. Methods A total of 55 Wistar rats were randomly divided into three groups including burn group, GLP 2 treatment group(treated with GLP 2, 200 ?g/kg, b.i.d ) and normal control group. Rats were sacrificed at 6, 12 h and 1, 3 and 5 d after burn injury. The proliferating cell nuclear antigen(PCNA) expression and the histological changes of the intestinal mucosa were determined by immunohistochemical staining. mRNA expressions of GLP 2R and PG were detected by RT PCR at 6, 12 h and 1, 3 and 5 d after burn injury. Results The expression of PCNA in rat intestinal mucosa decreased at 1 d after burn injury, but it was stronger in GLP 2 treatment group than that in burn group. Histological observation revealed that intestinal villi in GLP 2 treatment group were regularly arranged without obvious epithelial shedding. PG mRNA expression peaked at 12 h, 1 and 3 d after burn injury. No change of PG mRNA expression was found after treatment with GLP 2. Decreased GLP 2R mRNA expression was found in postburn rats, but after treatment with GLP 2, increased GLP 2R mRNA expression was found. Conclusion GLP 2 supplementation can keep the structure of the postburn rat intestinal mucosa without affecting PG gene expression. The mechanism may probably be related to the promotion of the mRNA expression of GLP 2R of the intestinal mucosa in rats.
2.Prevalence and subtype distribution of HPV infection among women in Beijing urban area and their correlation with age.

OBJECTIVETo investigate the prevalence and subtype distribution of human papillomavirus (HPV) infection and its correlation with age among women in Beijing urban area, and provide some epidemiological evidence for the clinical application of HPV vaccines.
METHODSWe collected cervical specimens from 1999 women in the Outpatient Department of our hospital, performed genetyping of HPV-DNA, and analyzed the incidence of HPV infection in different age groups.
RESULTSHPV infection was detected in 502 (25.2%) of the 1999 women patients, with 391 (19.6%) cases of high-risk HPV, which included 326 (83.4%, 326/391) cases of single infection. HPV-16 was the most common type (21.2%, 69/326), followed by HPV-52 (19.3%, 63/326) and HPV-58 (16.0%, 52/326). The prevalence of HPV infection was the highest among the women aged 41 -50 years and the lowest among those over 60 years.
CONCLUSIONThe subtype- and age-specific distribution of HPV infection among women in Beijing urban area shows an obvious heterogeneity, which deserves due consideration in the clinical application of HPV vaccines.

3.Monitoring Indexes of Plasma CsA Concentration in Patients Receiving Hematopoietic Stem Cell Transplantation

OBJECTIVE:To study the optimal monitoring indexes of plasma concentration of cyclosporine A(CsA) in patients receiving hematopoietic stem cell transplantation.METHODS:The lowest CsA concentration(C0) and the highest CsA concentration(C2) in 23 hematopoietic stem cell transplant recipients were detected by FPIA.All the data were analyzed statistically.RESULTS:Within 6 months after hematopoietic stem cell transplanttion,C0,C2,C0+C2 and C2/C0 in the recipients were(228.84?142.48) ?g?L-1,(741.50?294.42) ?g?L-1,(970.34?391.18) ?g?L-1 and(3.88?1.94) ?g?L-1,respectively.CONCLUSION:As reasonable monitoring indexes for the plasma concentration of CsA,C0+C2 and C2/C0 can comprehensively reflect the exposure of drug in body and monitor the toxicity of CsA in liver and kidney.
4.Digital Subtraction Angiography of Meningioma and Preoperative Embolotherapy

Objective To investigate the features of digital subtraction angiography(DSA) of meningioma and the application of therapy in preoperative embolization.Methods DSA in 36 cases with meningioma proved pathologically was performed.Of them,22 cases which the blood supply of tumor was mostly by external carotid artery were undergone external carotid artery embolization and surgical incision 3~10 days after embolization.Results Among 36 cases,the blood supply of tumor was only from external carotid artery in 9 cases,both of external and internal carotid artery supply in 25,and only internal carotid artery in 2.Among 22 cases undergone embolization of supplying artery the tumor stain absolutely disappeared in 16,evidently decreased in 6.22 cases undergone surgical incision after embolization,the tumors were wholly resected,average bleeding 150~500 ml during surgery.Conclusion The blood supply of meningioma can be only by external or internal carotid artery,or both.Preoperative cerebral angiographies and embolization are beneficial for the total resection of meningioma and reduction of bleeding during surery.
5.Observation of relationship between morphology of ciliary body and glaucoma with ultrasound biomicroscopy

Objective To observe the relationship between morphology of ciliary body and glaucoma with ultrasound biomicroscopy (UBM). Methods Eighty shallow-anterior chamber patients and 80 normal-anterior chamber patients were enrolled as shallow-anterior chamber group and control group, respectively. All patients were greater than 40 years old. The shallow-anterior chamber group included 28 patients of acute angle-closure glaucoma (AACG subgroup), 26 patients of chronic angle-closure glaucoma (CACG subgroup), and 26 patients of primary angle-closure suspects (PACS subgroup). Both eyes of all patients were checked with UBM, and position of each eye on sagittal and coronal section of superior, nasal, inferior and temporal were recorded. Then ciliary process height (CPH), ciliary process thickness (CPT), ciliary process interval (CPI), ciliary process number (CPN), A angle and α angle were measured. Furthermore, the difference between shallow-anterior chamber group and control group, the difference between AACG, CACG and PACS subgroup were compared, as well as the difference between PACS subgroup and control group, and the correlation between A angle and α angle was analyzed. Results Compared with control group, CPH and CPT of shallow-anterior chamber group increased, while CPI, CPN, A angle and α angle decreased (P<0.05). Except CPH and CPN between CACG and PACS subgroup, there were significant differences of parameters between subgroup of AACG, CACG and PACS (P<0.05). CPH of PACS subgroup increased than those of control group, while CPI, CPN, A angle and α angle decreased (P<0.05). There was positive correlation between A angle and α angle. Conclusion Morphologic parameters of ciliary process may be sensitive indexes which can early screen primary angle-closure glaucoma.
6.Establishment of network laboratory for blood cell analysis and its quality assurance

Objective To establish a network laboratory for blood cell analysis and better calibrate haematology analyzers in local lab.Methods According to GB/T 15481《General requirements for the competence testing and calibration laboratories》(idt ISO/IEC 17025),we established a network laboratory providing traceability for blood cell analysis.Complete blood count was traced to Calibration Laboratory in NCCL;The secondary standard haematology analyzer with the same model and calibrator with same lot number were used for verification for a long period.Fresh blood from healthy people was used to calibrate haematology analyzers.Results Gradually we have improved our laboratory quality management system, precision as well as accuracy,which was satisfactory.The unified blood sample was adopted to calibrate different equipments in our hospital and showed consistence when compared with calibration analyzer.The correlation coefficient of all tests is more than 0.99.The relative deviation of WBC,RBC,HCT,HGB and PLT are within?7%,?3.5%,?4%,?3% and?15%,respectively.Conclusions Secondary standard systems provides good comparable results with calibration laboratory.Its tracing mode and quality control scheme could ensure the traceability and accuracy of completed blood count.Furthermore,using elective fresh blood from healthy people,the comparable results from different analyzers were achievable.
7.Effects of L-glutamine Granules on Protein Metabolism and Immunofunction in Severely Burned Patients

OBJECTIVE:To study the effect of L-glutamine granules on protein metabolism and immunofunction in severely burned patients.METHODS:39 severe burn patients(total burn surface areas 30%~60%,full thickness burn areas 20%~50%) were randomly divided into two groups:control group(C group,19 patients) and glutamine treatment group(GLN group,20 patients).GLN group patients were given glutamine granules 0.5g/kg daily for 7 days,and C group were given same weight placebo for 7 days.The concentrations of plasma glutamine,prealbumin,transferrin,immunological globulin and IL-2 were determined.Moreover,the wound healing rate of burn area was observed and then hospital stay days were recorded.RESULTS:After 7 days of taking glutamine,the concentrations of plasma glutamine,prealbumin,transferrin IgG,IgM and IL-2 in GLN group were significant higher than those in before medication and C group(P0.05).The wound healing rate was faster and hospital stay days were shorter in GLN group than those in C group(P
8.Novel Evidence Suggests Hepatitis B Virus Surface Proteins Participate in Regulation of HBV Genome Replication

Naturally occurring mutations in surface proteins of Hepatitis B virus(HBV)usually result in altered hepatitis B surface antigen(HBsAg)secretion efficiency.In the present study,we reported two conserved residues,M75 and M103 with respect to HBsAg,mutations of which not only attenuated HBsAg secretion(M75 only),but also suppressed HBV genome replication without compromising the overlapping p-gene product.We also found M75 and M103 can initiate truncated surface protein(TSPs)synthesis upon over-expression of full-length surface proteins,which may possibly contribute to HBV genome replication.However,attempts to rescue replicationdefective HBV mutant by co-expression of TSPs initiated from M75 or M103 were unsuccessful,which indicated surface proteins rather than the putative TSPs were involved in regulation of HBV genome replication.
9.The Protamine-like DNA-binding Protein P6.9 Epigenetically Up-regulates Autographa californica Multiple Nucleopolyhedrovirus Gene Transcription in the Late Infection Phase

Protamines are a group of highly basic proteins first discovered in spermatozoon that allow for denser packaging of DNA than histones and will result in down-regulation of gene transcription[1].It is well recognized that the Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) encodes P6.9,a protamine-like protein that forms the viral subnucleosome through binding to the viral genome[29].Previous research demonstrates that P6.9 is essential for viral nucleocapsid assembly,while it has no influence on viral genome replication[31].In the present study,the role of P6.9 in viral gene transcription regulation is characterized.In contrast to protamines or other protamine-like proteins that usually down-regulate gene transcription,P6.9 appears to up-regulate viral gene transcription at 12-24 hours post infection (hpi),whereas it is non-essential for the basal level of viral gene transcription.Fluorescence microscopy reveals the P6.9's co-localization with DNA is temporally and spatially synchronized with P6.9's impact on viral gene transcription,indicating the P6.9-DNA association contributes to transcription regulation.Chromatin fractionation assay further reveals an unexpected co-existence of P6.9 and host RNA polymerase Ⅱ in the same transcriptionally active chromatin fraction at 24 hpi,which may probably contribute to viral gene transcription up-regulation in the late infection phase.
