Journal of Applied Clinical Pediatrics 2004;0(07):-
Objective To investigate the protective effects and mechanism of shenmai injection(SMI) on cardiomyocytes injured by hydrogen peroxide(H 2O 2) or angiotension Ⅱ(AngⅡ). Methods According to the different concentrations of H 2O 2, AngⅡ,SMI,Vitamin C(VitC) added into the cardiomyocytes culture media(CCM), the cultures were divided into 15 groups. Lactate dehydrogenase(LDH ),malondialdehyde(MDA),superoxide dismutase (SOD) of CCM and cardiomyocytes viability(CMV), Na +-K +-ATPase,Ca 2+-ATPase?cardiomyocytes apoptosis rate(CMAR) ,nitrogen oxide(NO), nitrogen oxide synthesis enzyme(NOS),eNOS expression of cardiomyocytes were detected respectively .Results H 2O 2 or AngⅡ could decrease CMV, SOD, Na +-K +-ATPase,Ca 2+-ATPase,NOS,NO,eNOS expression and increase CMAR,LDH and MDA.SMI could lessen the changes of the items mentioned above ,which were caused by H 2O 2 or AngⅡ.The effects of SMI 10 ml/L were stronger than those of SMI 5 ml/L or VitC 50 mg/L.Conclusion SMI has a significant protective effect on cardiomyocyte injured by H 2O 2 or AngⅡ.

Objective To investigate the therapeutic effect and mechanism of ribavirin to mice viral myocarditis. Methods Onehundred and twenty mice were randomly divided into 3 groups. Group 1 was normal control group 1 .group 2 CVB3 control group, group 3 ribavirintherapy group. In the latter two groups, 0.2 mL CVB3(10-7.5Tcid5o/mL)was infected into abdominal cavity. Ten mice of each group were killed at the 7 th, 14 th,21 st,28 th day. Mice motality, light and electron microscape, myocardial viral tiler, myocardial pathological score, serum cTnI,malonytdialdehyde(MDA) and cardiac myocyte apoptosis rate(CMAR) were examined.Re-sults The motality, myocardial viral titer, myocardial pathological score,cTnI were lower in group 3(ribavirin therapy group) than in group 2(CVB3 control), and MDA and CMAR were no significant differences between ribavirin group and simple CVB3 group. Conclusion Ribavirin has good treatment effect to mice viral myocardilis, and its mechanism is due to decrease viral titer.

Objective To approach a semi-quantitative diagnostic criteria of neonatal hypoxic-ischemic encephalopathy(HIE) by comprehensive scoring, including history, clinical manifestation and laboratory data, which can be used for early diagnosis, severity grading and the institution of therapy. Methods We analyzed history, clinical manifestation and some parameters of laboratory data, and established criteria for scoring and grading, and compared this grading with the grading determined by clinical data and follow-up,which brought forward a semi-quantitative diagnostic criteria, and applied this criteria to other 41 patients with HIE to test its reliability. Results The grading criteria were established as the following:if total score ≥20.0,the patient was graded as severe one;total score between 10.5-19.5, moderate one; between 5.5-10.0,mild one;≤5.0,non-HIE one. The scoring system was used in other 41 patients with HIE,the specificity and sensitivity were 100% and 96 .9%,97.2% and 100%,100% and 100%, respectively. Conclusions For severe HIE cases who are not permitted to receive compated tomography(CT) examination or no CT can be used, this scoring system can be used to diagnoze HIE and grade the severity. This method is simple and easy to perform. Both specificity and sensitivity are high for diagnosis and severity judgment of HIE.

Objective To study the normal values of blood pressure in healthy full-term infants in the first 7 days of life, and to determine the critical hypertension and hypotension in newborn infants. Methods Omni-Trak~(TM)NVS monitor with DINAMAP was used to measure the blood pressure.Systolic, diastolic and mean blood pressure were measured every day by DLNAMAP in a cohort of 50 healthy full-term infants in the first 7 days of life. Results 1.Systolic,diastolic and mean blood pressure of healthy full-term infants were significantly influenced with the age of birth.2.Stepwise linear multiple regressions was used to examine the multiple correlations among days of life, birth weight and gestational age.3.Optimal regression equations were built respectively.SBP=32.4+0.7X_1+(3.7X_2+)(0.6)X_3(mm Hg).DBP=13.1+ 0.6X_1+2.9X_2+0.6X_3(mm Hg).MAP=17.3+0.4X_1+2.4X_2+0.8X_3(mm Hg).(3.MAP=)(DBP+)(0.45)(SBP-DBP)(mm Hg).4.The critical hypertension and hypotension in newborn infants were obtained. Conclusions (Du)ring the first 7 days of life, there is a progressive rise in blood pressure of healthy full-term infants. There are linear reliance correlations between systolic, diastolic, mean blood pressure and day of life, birth weight gestational age.The critical hypertension and hypotension in newborn infants is important for clinic diagnosis.

Objective To realize the myosin heavy chain(MHC) expression and related factors in severe pneumonia. Methods The staphylococcus aureus peumonia models of 30 Wistar rats were established and other 20 rats as control group.MHC expression and myocardial histopathologic score and cardiac function were examined. Results The expression of ?-MHC mRNA were lower in the rats with peumonia than that in the control group, while the expression of ?-MHC mRNA in the cases with peumonia increased significantly. The relative contents of ?-MHC mRNA had negative correlation with myocardial histopathologic score and had positive correlation with EF,FS.The relative contents of ?-MHC mRNA had positive correlation with myocardial histopathologic score and had negative correlation with EF,FS.Conclusions The cases with severe peumonia have abnormal expression of MHC,with the transformation of ?-MHC to(?-MHC).These proved that severe pneumonia can complicated with heart failure from gene level.
