1.Cost-Effectiveness Analysis of Rabeprazole versus Lansoprazole for Peptic Ulcer

OBJECTIVE: To evaluate the cost-effectiveness of Rabeprazole versus Lansoprazole for peptic ulcer. METHODS: A total of 101 patients with peptic ulcer were randomly assigned to receive either Rabeprazole+Clarithromycin sustained release capsules+tinidazole(treatment group) or Lansoprazole+Clarithromycin sustained release capsules+tinidazole(control group) for 14 days. RESULTS: In the treatment group vs. the control group, the effective rate was 96.0% vs. 84.3%(P0.05); the cost was 285.46 vs. 373.87 yuan; the cost-effectiveness ratio was 297.35 vs. 443.50. The incremental cost-effectiveness ratio for the control group was -755.64 as against the treatment group. CONCLUSION: Rabeprazole group is optimal for peptic ulcer in that it is more economical, more effective and safer than Lansoprazole group.
2.Effect of hypoxia on expression of angiopoietin-2 in human retinal pigment epithelium

Background Choroidal neovascularization(CNV)has been descrihed as a main reason of visual loss in a lot of ocular diseases.Researches showed that local hypoxia and retinal pigment epithelial(RPE) cells play an important role in the formation of CNV.A closely relationship of angiopoietin-2 (Ang-2) and angiogenesis has been proved.However,whether the expression of Aog-2 in hypoxic cultured human RPE cells is associated with the pathogenesis of CNV is still below understood.Objective This study was to investigate the effects of hypoxia on expression of Ang-2 in cultured human RPE cells in vitro,and discuss the possible effects of Ang-2 in the formation of CNV.Methods Human RPE cells were cultured and passaged,and 4-7 generation of cells were used in the experiment.The cells were incubated in cultural plate at the density of 5×107 cells/L.The culture medium containing 200 μmol/L CoCl2 was used to establish the hypoxia model of human RPE cells culturecd in vitro for 0.5,1,2,4,6,12and 24 hours,and the RPE cells cultured under normoxia were as controls.Reverse transcription polymerase chain reaction(RT-PCR) was used to detect the expression of Ang-2 mRNA in cultured human RPE cells,and enzymelinked immunosorbnent assay(ELISA) was used to assay the content of Ang-2 protein in supernatant of cultured human RPE cells.Results The survival rate of human RPE cells was 90% after resoscitation.The fourth generation of cells showed the fusiform with the less pigment in them.The Ang-2 mRNA/β-actin mRNA value in human RPE cells was significantly different among various groups(F=1086.30,P=0.00),The Ang-2 mRN A/β-actin mRNA value in hypoxia cultured for 0.5 hours group began to increase and peaked in hypoxia culture for 4-6 hours group,with the significant differences in comparison with normoxia control group(P<0.05).The Ang-2 mRNA/β-actin mRNA value decreased to the baseline level at hypoxia for 24 hours.The ELISA analysis showed that the concentration of Ang-2 protein in supernatant of RPE cells showed significant difference among groups(F=1034.00,P=0.00).The concentration of Ang-2 protein increased at hypoxia culture for 0.5 hours and peaked at 6 hours,showing significant differences in comparison with the control group (P<0.05).Conclusions Hypoxia could significantly up-regulate the expression of Ang-2 in human RPE cells cultured in vitro.Ang-2 expresses highly in the early stage of hypoxia,implying that Ang-2 participates in the formation of CNV.
3.Effect of Sophoramine on Immune Function of Mice

The effects of sophoramine (SA) on immune system in normal mice were studied. SA 15. 20mg/kg reduced the weight of thymus,decreased the rate of E rosette,and also suppressed T lymphocyte transformation. The weight and the hemolytic plaque of spleen were not influenced by SA at 15. 20mg/kg. At the dose of 25mg/kg,SA reduced the weight of thymus and spleen,decreased the rate of E rosette and suppressed the T lymphocyte transformation. All of the doses of SA didn' t affect the phagocytotic function of peritoneal macrophage.

6.Feasibility of chitosan-beta-tricalcium phosphate as an injectable bone tissue engineering scaffold material

0.05).Under SEM,the BMSCs showed good adhesion to beta-TCP with obvious proliferation.Conclusion BMSCs can grow and proliferate well on the compound BMSCs/beta-TCP and beta-TCP has good biocompatibility with BMSCs in vitro,which may be used as a good scaffold material for injectable tissue engineering bone.
7.Changes of CGRP and NPY expression in tissue-engineered bone with fascia flap

Objective To evaluate the expressive levels of calcitonin gene related peptide (CGRP) and neuropeptide Y (NPY) in tissue-engineered bone with fascia flap in vivo. Method A segmental bone defect 1.5cm in length was made at the both radius of 12 healthy New Zealand rabbits. The defects were repaired by implantation in two ways: for left radius, engineered bone with fascia flap was implanted (served as experiment group), and for right radius only engineered bone was implanted (served as control group). 3, 6 and 12 months after implantation, the expressions of CGRP and NPY in the new bone were determined with immunohistochemistry and semi-quantified using image analysis software. The human bone morphogenetic protein 7 (hBMP 7) gene was reconstructed in retroviral vector and transferred into incasing cells PT67 by liposome mediated method.The clones of the cells transferred with BMP 7 were selected by G418, and targeted rabbit bone marrow stem cells were infected with the virus granules which secreted from PT67 cells and also selected by G418. The mRNA and protein of BMP 7 gene in transferred cells were analyzed with hybridization in situ and immunohistochemistry. BMP 7 retrovirus vetor was successfully reconstructed. Cells transferred by PLNCX 2 hBMP 7 expressed abundant human BMP 7 mRNA and protein in the cytoplasm. However positive findings were not found in those cells that were not transferred. It may be used to increase the osteogenic capability of BMSc in the study of bone tissue engineering.
10.Effect of body mass index on outcomes of in vitrofertilization for infertile females at different ages

BACKGROUND:The effect of body mass index (BMI) on the outcomes ofin vitrofertilization (IVF) is controversial. Some scholars suggested that the higher BMI, the lower rate of pregnancy, and the higher rate of abortion. However, some others got the different results. To data, the effects of BMI on the pregnancy outcomes in females at different ages are rarely reported.
OBJECTIVE:To explore the effect of BMI on the pregnancy outcomes of IVF for infertile women atdifferent ages.
METHODS:A total of 902 cycles undergoing IVF were divided into two groups according to the age:≤ or> 35 years old groups. The patients in each group were then divided into four subgroups according to BMI: low weight, normal weight, excess weight, and obesity subgroups, respectively.
RESULTS AND CONCLUSION: With increasing BMI, the average dosage of gonadotropins was increased gradualy in two both of age groups (P < 0.05). In the≤ 35 years old, the levels of estrogens on the day of injecting human chorionic gonadotropin was significantly lower in obesity group than the other subgroups (P < 0.05); the rates of empty folicle in excess weight and obesity subgroups were significantly higher than that in normal weight subgroup (P < 0.008 23). The fertilization rate and clinical pregnancy rate of in obesity subgroup were significantly lower than those in the other subgroups (P < 0.05). In the > 35 years old group, the empty folicle rate in obesity subgroup was significantly higher than that in normal weight subgroup (P < 0.008 23). Fertilization rate in excess weight and obesity subgroups was significantly lower than that in the normal weight subgroup (P< 0.008 23). There were no significant differences in cleavage rate, cycle cancelation rate, implantation rate, biochemical pregnancy rate and abortion rate between four subgroups (P> 0.05). These findings suggest that in the females at the younger age, BMI not only affects the quality of folicles and fertilization rate; but with increasing BMI, clinical pregnancy rate is decreased obviously. However in the females at the older age, BMI only affects the folicular quality, embryo quality and fertilization rates; there are no significant differences in the outcomes of pregnancy in the four subgroups. BMI have effects on pregnancy outcomes of IVF, and the effects are much greater in the younger female group.
