Objective To observe the expression of vascular endothelia l growth factor (VEGF)and transforming growth factor a 1 (TGF-a 1 )in callus of non-union,bone defect for investigation of the molecular mechanisms of bone defect resulted from non-union,and further explore the co rrelative influences of VEGF and TGF-a 1 in fracture healing.Methods The bone defect resulted from non-union model and the fracture healing model were established in the radius of rabbit.The sp ecimens of callus and soft tissue were collected postoperatively at1,2,4, 6,8and12weeks.The expressions of VEGF and TGF-a 1 were in-vestigated by means of immunohistochemistry(SP method).Results There was no sign of subperiosteal healing except a large segment filled with hemorrhage and granulation tissue in bone defect area.There was only a little amount of granulation tissues transforming into chondrocytes.Some chondrocyt es stopped prolif-erating after reaching hyper-trophic chondrocyte stage but changed into fibrous connective tissues.Most of the granulation tissue finally changed to fibrous connective tissues in the fracture gap.In addition,there w ere a large number blood vessels invading the normal fracture healing site than that in non-union.In im muno-histochemistry observation,there were significa nt decreasing expressions of VEGF and TGF-a 1 in callus of different period of non-union comparing to union(P
2.Clinical effect of pseudomonas aeruginosa injection on malignant pleural effusion

Objective:To observe the effect of thoracic infusion of pseudomonas aeruginosa injection in malignant pleural effusion. Methods:A total of 90 patients with malignant pleural effusion were randomly divided into treatment group (31 cases),control group A (29 cases) and control group B(30 cases). Treatment group was treated with pseudomonas aeruginosa through intrathoracic infusion. Control group A and B were respectively treated with cisplatin and interleukin-2 through intrathoracic infusion. The clinical efficacy and adverse reaction were compared among the three groups. Results:The total effective rate of treatment group was 80.6%,the total effective rates of the control group A and B were 51.7%and 56.7%respectively.Compared with that of contral groups, the total effective rate of treatment group was higher, and the differences were statistically significant (P<0.05). The incidence of serious side effects and toxicity was lower in treatment group than in control groups. Conclusion:The effect of thoracic infusion of pseudomonas aeruginosa injection for malignant pleural effusion is significant, and the adverse reaction is mild. Thus it is worth to be promoted clinically.
3.Effect of high load application of atorvastatin on the clinical outcome of patients with acute myocardial infarction after percutaneous coronary intervention

Objective To evaluate the effect of high load application of atorvastatin on the clinical outcome of patients with acute myocardial infarction (AMI) after percutaneous coronary intervention (PCI).Methods Eighty patients diagnosed with AMI were continuously enrolled in this study and randomly divided into two groups:high dose group(n=40) and control group(n=40).Application of high load atorvastatin before operation in loading group.The levels of serum lipid and cardiac function were measured and analyzed.Results The levels of BNP((204±60.3) pg/mL vs.(328.3±67.5) pg/mL;t=1.938,P=0.0315) on 7 days after PCI and levels of LVEF((50.3±6.0)% vs.(56.9±7.3)%;t=2.169,P=0.019) on 1 month after PCI in high dose group were significantly better than those in control group.Correlation analysis showed that the administration of statin was negatively associated with levels of BNP(r=-0.157,P=0.021) on 7 days after PCI and positively associated with LVEF(r=-0.328,P=0.026) on 1 month after PCI.Conclusion The treatment of high dose statin before PCI may reduce the ischemia-reperfusion injury and prevent the no-reflow development,which therefore improve the cardiac function of AMI patients.

5.HPLC fingerprinting of radix paeoniae alba.

To establish a sensitive and specific HPLC method for quality control of Radix Paeoniae Alba, HPLC method was applied for quality assessment of Radix Paeoniae Alba. HPLC analysis was performed on a Symmetry C18 column (250 mm x 4. 6 mm ID, 5 microm, Waters, USA). The mobile phase consisted of acetonitrile (solvent A) and water containing 0.1% (v/v) phosphoric acid (solvent B) at a constant flow rate of 0.8 mL x min(-1). An increasing linear gradient (v/v) of solvent A was used (t/min, % A): (0,10), (5,10), (25,15), (45, 22), (46, 65), (50, 80) and (60, 80). The column temperature was set at 25 degrees C. The chromatograms were monitored at 230 nm and the on-line UV spectra were recorded in the range of 190 - 400 nm. The HPLC chromatographic fingerprinting of Radix Paeoniae Alba, showing 11 characteristic peaks, was established from 28 lots of Radix Paeoniae Alba. The areas of main chromatographic peaks were found to complied with the following rule: paeoniflorin > 1, 2, 3, 4, 6-penta-O-galloyl-glucos > albiflorin > methyl gallate > other compounds. The chromatographic fingerprinting of Radix Paeoniae Alba with high specificity can be used to control its quality and assure lot-to-lot consistency.

6.Correlation of adiponectin, monocyte chemoattractant protein-1, and endothelial function to vascular remodeling in coronary in-stent restenosis.

OBJECTIVETo investigate the correlation between vascular remodeling index (RI) and serum adiponectin, plasma monocyte chemoattractant protein-1 (MCP-1), endothelial function and evaluate the mechanism of coronary in-stent restenosis.
METHODSRI 6 months after percutaneous coronary intervention (PCI), serum adiponectin, plasma MCP-1 and flow-mediated dilation (FMD) before and 3 days,6 months after PCI were measured in 30 patients with and 30 without coronary in-stent restenosis.
RESULTSCompared with patients without restenosis and those with restenosis before PCI, the patients with coronary in-stent restenosis showed significantly increased plasma MCP-1 3 days and 6 months after PCI (P<0.05) and reduced RI 6 months after PCI, serum adiponectin and FMD 3 days and 6 months after PCI (P<0.05). RI was positively correlated to serum adiponectin and FMD and inversely to MCP-1.
CONCLUSIONThe occurrence of coronary in-stent restenosis is the result of the interrelations between multiple factors.

7.Expression of recombinant human zona pellucida-3 protein (rhZP3) in Pichia pastoris.

Human Zona Pellucida(ZP), which is a complex matrix surrounding oocytes,is comprised of three immunologically distinct glycoproteins(hZP1, hZP2 and hZP3). Because hZP3 possesses the sperm receptor activity and the acrosome-inducing activity, it has long been used as a candidate antigen to develop an immunocontraceptive vaccine. However, a large amount of native hZP3 protein is unavailable. It is an effective way to express hZP3 protein directly in vitro. Nevertheless, it had been reported that the rhZP3 protein produced in Pichia pastoris was not secreted but accumulated in the cells and could only be purified after being solubilized by strong denaturants. More unfortunately, after purification the final product required 6mol/L urea to maintain solubility. An improved project was advanced with the aim to express secreted and soluble rhZP3 protein in yeast. In this study, the fragment of hZP3 cDNA coding for aa 23 - 408, which the N-terminal leader was removed and most of the C-terminal transmembrane-like domain was reserved, was amplified by two PCR primers including EcoR I and Not I sites respectively and a His6 codon cassette was added to 5'-terminal. The hZP3 insert was incorporated into expression vector pPIC9K. The resulting recombinant yeast expression vector was designated pPIC9K-rhZP3. Linearized pPIC9K-rhZP3 was transformed into Pichia pastoris. After G418 selection, the recombinant Pichia pastoris strains were identified by PCR and the rhZP3 was expressed following the manufacturer' s protocol. Following induction with methanol, the rhZP3 protein was secreted and dissolved into the culture supernatant. SDS-PAGE and Western blot analyses showed that the apparent molecular weight of the expressed rhPZ3 proteins in yeast was smaller and a little size heterogeneity than native ones; after purified with Ni-chelating affinity chromatography, the final product's apparent molecular weight was about 32 - 34KD and their yield more than 20mg/L. We supposed that the C-terminal transmembrane-like domain be useful for secretion of rhZP3 into the culture supernatant and the expressed rhZP3 protein be incompletely digested by proteinases of Pichia into shorter fragments which all were glycosylated inhomogeneously. Fortunately, the fragments of rhZP3 protein can be recognized in Western blot by the polyclonal antibodies to porcine ZP3 which has showed a cross-reactivity with human ZP in vitro. It will be expected that the rhZP3 protein expressed in Pichia pastoris not only has immunogencity, say, it can rise antibodies in vivo to prevent spermatozoa-ovum binding, but also does not contain ovarian factors that might be the cause of undesired side effects, e.g. ovaritis and can be used as a safe immunogen in human antifertility vaccine research.

9.Metallothionein and its isoform genes expression in the human pancreatic cancer cell strains and their function.

OBJECTIVETo compare the expression of metallothionein (MT) genes and proteins in six human pancreatic cancer cell strains and two human pancreatic cancer drug-resistant cell strains and to explore the relationship between the expression of the MT and pancreatic cancer cell chemo-resistance.
METHODSReverse transcriptase-polymerase chain reaction (RT-PCR) was used to determine the MT isoform-specific mRNA, and cadmium/hemoglobin saturation-electrochemistry to determine MT protein levels.
RESULTSMT protein expression in the pancreatic cancer cell strains was encoded by MT-1A, MT-1B, MT-1E, MT-1F, MT-1G, MT-1X, and MT-2A genes. The expression of MT proteins was upregulated and MT-1B, MT-1E, MT-1X, MT-2A genes overexpressed in human pancreatic cancer drug-resistant cell lines (P < 0.05).
CONCLUSIONExpressions of MT proteins and genes correlate with the proliferation and chemoresistance of human pancreatic cancer cell strains.

Adjustable spine prone frame is used in posterior lumbar surgery, during operation, X-ray can shoot fast and internal fixation implants can fix accurately, the vacant abdominal ensure normal respiratory and circulatory function, and the lumbar curvature can adjust conveniently. It meets the surgical position and care requirements, can prevent the complications effectively.
